Validamycin A Induced Resistance in Rice against Rhizoctonia solani via the Salicylic Acid-Depended Signaling Pathway.

Validamycin A (VMA), the major active component of validamycin, provides effective control of rice sheath blight; however, the mechanisms underlying VMA-induced resistance to Rhizoctonia solani remain poorly understood. Here, we integrated antioxidant enzyme assays, phytohormone profiling, transcriptomics, and metabolomics to investigate VMA-induced immunity in rice. VMA improved oxidative stress tolerance by enhancing antioxidant enzyme activities and activating glutathione metabolism. Moreover, VMA established a primed multihormonal state characterized by elevated salicylic acid (SA), jasmon

Citral-based acylthiourea derivative enhances the efficacy of thiophanate-methyl against Rhizoctonia solani through membrane destabilization.

Rhizoctonia solani is a highly destructive fungal pathogen of rice, and effective management currently depends heavily on chemical fungicides such as thiophanate-methyl (TM). To establish a sustainable approach for enhancing fungicide efficacy, we screened 22 citral-based acylthiourea derivatives and identified compound e3 as a highly effective synergist of thiophanate-methyl (TM), achieving a synergistic ratio of 3.88. When the molar ratio of e3 is 20 %, the mixture of e3 and TM (SETM) exhibits a pronounced antifungal activity against R. solani, both in vivo and in vitro. Integrated transcrip

Hydrogen peroxide contributes to SYP-14288-induced multidrug resistance in Rhizoctonia solani by modulating detoxification and efflux pathways.

SYP-14288 is an effective antifungal compound against Rhizoctonia solani, the causal agent of rice sheath blight. The chemical acts through its unique uncoupling mechanism. Previous studies have demonstrated that SYP-14288-resistant mutants of R. solani develop multidrug resistance (MDR), but the underlying mechanisms remain unclear. In this study, in SYP-14288-resistant mutants, hydrogen peroxide (H₂O₂) accumulated at higher levels, which may activated oxidative stress signaling and upregulated detoxification pathways. Compared with the parental strain, these mutants also showed significantly

Cinchona-based liquid formulation exhibits antifungal activity through Tryptophan starvation and disruption of mitochondrial respiration in Rhizoctonia Solani.

This study investigates the antifungal properties of a liquid Cinchona-Based Formulation (CBF) against Rhizoctonia solani. CBF, a stable acidic extract of Cinchona bark (0.1 N HCl), effectively suppressed mycelial growth (58.75 ± 0.48%) and sclerotia biomass (22.1 ± 0.65 mg), with an EC50 of 217.14 µg/mL. GC-MS and HPLC revealed quinine as the primary active alkaloid. Molecular docking revealed a strong binding affinity of quinine to the fungal tryptophan (Trp) transporters (- 6.5 to - 7.8 Kcal/mol). Supplementation of 400 µg/mL Trp in the media reversed the fungal growth with increased fungal

Identification of Genes Involved in Antifungal Activity of Burkholderia seminalis Against Rhizoctonia solani Using Tn5 Transposon Mutation Method.

Rhizoctonia solani is the causative agent of rice sheath blight disease. In a previous study, we found that the growth of R. solani was inhibited by Burkholderia seminalis strain R456. Therefore, the present study was conducted to identify the genes involved in the antifungal activity of B. seminalis strain R456 by using a Tn5 transposon mutation method. Firstly, we constructed a random insertion transposon library of 997 mutants, out of which 11 mutants showed the defective antifungal activity against R. solani. Furthermore, the 10 antagonism-related genes were successfully identified based o

Metabolites contributing to Rhizoctonia solani AG-1-IA maturation and sclerotial differentiation revealed by UPLC-QTOF-MS metabolomics.

Rhizoctonia solani is a causative agent of sheath blight, which results in huge economic losses every year. During its life cycle, the formation of sclerotia helps Rhizoctonia solani withstand a variety of unfavorable factors. Oxidative stress is a key factor that induces sclerotium formation. The differentiated and undifferentiated phenotypes of R. solani AG-1-IA were obtained by controlling aerial conditions. Metabolomics based on the mass spectrometry technique combined with multivariate and univariate analyses was used to investigate the metabolic variation in vegetative, differentiated an

Synergistic antifungal activity of citral combined with its acylthiourea derivative against Rhizoctonia solani and its underlying mechanisms.

Rhizoctonia solani is a significant soil-borne plant pathogenic fungus. Its sclerotial structure confers strong environmental adaptability and persistent infectivity, posing a serious threat to the production of numerous crops. Citral, a plant-derived monoterpene aldehyde, exhibits antifungal activity; however, its antifungal potency is limited. In this study, citral was combined with its acylthiourea derivatives, leading to the screening and construction of an efficient synergistic antifungal system, CE4. CE4 significantly inhibits mycelial growth of R. solani and reduces its capacity for scl

The OsAGO2-miR11337-5p-OsLRR Module Confers Rice Resistance to Rhizoctonia solani by Modulating Vitamin B6 Biosynthesis.

Argonaute (AGO) proteins play a pivotal role in small RNA (sRNA)-mediated RNA silencing and plant immunity. However, their roles in rice defense against sheath blight remain largely unknown. In this study, we found that the osago2 mutant exhibited enhanced resistance to Rhizoctonia solani (R. solani). Small RNA sequencing revealed that miR11337-5p, an OsAGO2-associated sRNA, was significantly upregulated upon R. solani infection. OsLRR, which encodes a leucine-rich repeat protein, was identified as the direct target of miR11337-5p. Functional analyses demonstrated that miR11337-5p negatively r

Rhizoctonia solani Secretes RsCAP3 to Target Nb14-3-3b, Interfering with Hormone-Mediated Resistance in Nicotiana benthamiana.

The necrotrophic phytopathogen Rhizoctonia solani causes leaf spot and root rot in tobacco, which significantly reduce the yield and quality of the crop. Recently, much attention has been paid to R. solani effectors, but only a few of them have been characterized. Here, we identified a CAP domain-containing protein, RsCAP3, with a functional signal peptide, which triggers slight cell death on tobacco leaves. Meanwhile, RsCAP3 dramatically induces salicylic acid (SA) signaling but inhibits jasmonic acid (JA) signaling, increasing the plant's susceptibility to R. solani. Moreover, we demonstrate

Synthesis and Preliminary Antifungal Evaluation of Acetylated Glucopyranoside Amidrazones against Pythium aphanidermatum, Rhizoctonia solani and Fusarium solani in Cucumber.

Soilborne damping-off caused by Pythium aphanidermatum, Rhizoctonia solani, and Fusarium solani remains a major constraint to cucumber production and shows the need for new chemistries with tractable safety profiles. We synthesized and characterized eight acetylated glucopyranoside amidrazones (AM 1-AM 8) and conducted a preliminary antifungal screen. In dual-culture assays on PDA, halogenated analogues (AM 2:4Br; AM 6:3,5Cl2; AM 7:3Cl) produced the largest inhibition zones against R. solani and F. solani, whereas P. aphanidermatum was less responsive. Scanning electron microscopy revealed wri

Automatic Scoring of Rhizoctonia Crown and Root Rot Affected Sugar Beet Fields from Orthorectified UAV Images Using Machine Learning.

Rhizoctonia crown and root rot (RCRR), caused by Rhizoctonia solani, can cause severe yield and quality losses in sugar beet. The most common strategy to control the disease is the development of resistant varieties. In the breeding process, field experiments with artificial inoculation are carried out to evaluate the performance of genotypes and varieties. The phenotyping process in breeding trials requires constant monitoring and scoring by skilled experts. This work is time demanding and shows bias and heterogeneity according to the experience and capacity of each individual person. Optical
