Exploring the Individual Bacterial Microbiota of Questing Ixodes ricinus Nymphs.

Ixodes ricinus is the most common hard tick species in Europe and an important vector of pathogens of human and animal health concerns. The rise of high-throughput sequencing has facilitated the identification of many tick-borne pathogens and, more globally, of various microbiota members depending on the scale of concern. In this study, we aimed to assess the bacterial diversity of individual I. ricinus questing nymphs collected in France using high-throughput 16S gene metabarcoding. From 180 dragging-collected nymphs, we identified more than 700 bacterial genera, of which about 20 are abundan

Borrelia miyamotoi in host-seeking Ixodes ricinus ticks in England.

This paper reports the first detection of Borrelia miyamotoi in UK Ixodes ricinus ticks. It also reports on the presence and infection rates of I. ricinus for a number of other tick-borne pathogens of public health importance. Ticks from seven regions in southern England were screened for B. miyamotoi, Borrelia burgdorferi sensu lato (s.l.), Anaplasma phagocytophilum and Neoehrlichia mikurensis using qPCR. A total of 954 I. ricinus ticks were tested, 40 were positive for B. burgdorferi s.l., 22 positive for A. phagocytophilum and three positive for B. miyamotoi, with no N. mikurensis detected.

Absence of Lyme disease spirochetes in larval Ixodes ricinus ticks.

To determine which kind of spirochete infects larval Ixodes ricinus, we examined questing larvae and larvae derived from engorged females for the presence of particular spirochetal DNA that permitted species differentiation. Borrelia miyamotoi was the sole spirochete detected in larval ticks sampled while questing on vegetation. Questing nymphal and adult ticks were infected mainly by Borrelia afzelii, whereas larval ticks resulting from engorged females of the same population were solely infected by B. miyamotoi. Since larvae acquire Lyme disease spirochetes within a few hours of attachment t

Natural transmission of Zoonotic Babesia spp. by Ixodes ricinus ticks.

To determine characteristics of natural transmission of Babesia sp. EU1 and B. divergens by adult Ixodes ricinus ticks, we examined tick salivary gland contents. We found that I. ricinus is a competent vector for EU1 and that their sporozoites directly invade erythrocytes. We conclude that EU1 is naturally transmitted by I. ricinus.

[Pandora's Box: pathogens in Ixodes ricinus ticks in Central Europe].

Among the various species of hard ticks, Ixodes ricinus is the most frequently found tick throughout Europe. As with other ixodid ticks, the developmental cycle runs through three stages. In each stage a blood meal is required in order to develop to the next stage. Ixodes ricinus has been found to feed on more than 300 different vertebrate species. Usually, larval ticks feed on small mammals such as mice and become infected with various microorganisms and viruses, of which some are substantial pathogens to humans. The pathogens remain in the tick during molting and are thus transstadially tran

Surveillance for Ixodes ricinus ticks (Acari, Ixodidae) on the Faroe Islands.

Ixodes ricinus ticks are expanding their geographic range in Europe, both latitudinally in Scandinavia, and altitudinally in the European Alps. This paper details the findings of both passive and active surveillance on the Faroe Islands. Active field surveillance, using tick dragging, was conducted at 38 sites across the main seven inhabited islands of the Faroes during June-August 2015. Field sampling was conducted at all wooded sites on the islands of Vágar, Streymoy, Eysturoy, Borðoy, Kunoy and Suðuroy as well as in urban parks in the capital Tórshavn, among seabird colonies and at a bird o

[Threat of transmission of infectious pathogens by Ixodes ricinus ticks in Germany].

Tick-transmitted diseases are of great importance for the general health of the German population. Several viruses, such as tick-borne encephalitis virus (TBEV), Uukuniemi virus, Tribec virus, Eyach virus or bacteria, such as Borrelia, Rickettsiae, Francisella tularensis, Anaplasma phagocytophilum, Candidatus Neoehrlichia mikurensis (CNM) and Coxiella burnetii were detected in the most prominent tick in Germany, the hard tick Ixodes ricinus. While infections, such as TBE and Lyme disease are well known, other infections are hardly known even among experts. Although there have been a few descri

Coxiella burnetii and coinfections in Ixodes ricinus ticks in Central Germany.

A total of 1000 Ixodes ricinus ticks were collected in 2006 and 2007 in a forest region of Central Germany and investigated for Coxiella burnetii. The transposase element IS1111 and isocitrate dehydrogenase gene were targets of the real-time polymerase chain reaction. The pathogen was detected in 19 ticks (1.9%), and interestingly, in 10 of these samples, coinfections with Borrelia spp., spotted fever group rickettsiae, or Babesia spp. were present. Our study reports on C. burnetii infections in I. ricinus ticks in an area where cases of Q fever occur regularly and Dermacentor marginatus is no

Coexistence of emerging bacterial pathogens in Ixodes ricinus ticks in Serbia.

The list of tick-borne pathogens is long, varied and includes viruses, bacteria, protozoa and nematodes. As all of these agents can exist in ticks, their co-infections have been previously reported. We studied co-infections of emerging bacterial pathogens (Borrelia burgdorferi sensu lato, Anaplasma phagocytophilum and Francisella tularensis) in Ixodes ricinus ticks in Serbia. Using PCR technique, we detected species-specific sequences, rrf-rrl rDNA intergenic spacer for B. burgdorferi s.l., p44/msp2 paralogs for A. phagocytophilum, and the 17 kDa lipoprotein gene, TUL4, for F. tularensis, resp

[Biology of Ixodes ricinus (L.) and its pathogens in Wrocław area].

Within 2001-2003, a total of 4743 ticks of Ixodes ricinus were trapped from the forest area in Masyw Sliy (Lower Silesia, Poland) near Wrocław. Common distribution of ixodid ticks in all examined habitats, located near touristic routes, are potential loci of Lyme borreliosis in these recreational areas. Out of 1108 tick samples examined by the observation in DFM for the presence of spirochetes, 6.7% were positive. The highest percent of infected ticks (13.1%) was found in 2003.

Tick-borne encephalitis virus in Ixodes ricinus (Acari: Ixodidae) ticks, Tunisia.

Tick-borne encephalitis virus (TBEV) is the etiologic agent of tick-borne encephalitis (TBE) and transmitted by Ixodes ricinus (Linnaeus, 1758) in Europe. The geographical distribution of I. ricinus in the Palearctic region covers also northern Africa, including northwestern Tunisia. While the eco-epidemiology of TBE in Europe is well documented, no data concerning TBEV from northern Africa are available. We investigated whether TBEV is circulating in Tunisia. A total of 877 adult I. ricinus collected from northwestern Tunisia were examined in pools for the presence of TBEV by nRT-PCR. Viral R

Pathogens carried by Ixodes ricinus and Dermacentor reticulatus ticks including coinfections.

Ticks and tick-borne pathogens are becoming an emerging threat to the health of both humans and animals. The number of cases of tick-borne diseases, especially Lyme disease, is constantly growing. Over the last several years, coinfections, which could be explained as presence of two of more pathogens in one organism are being observed with growing interest. Their occurrence may lead to severe or unusual symptoms, prolonged disease duration, diagnostic and therapeutic difficulties. Research on this subject concerns mainly ticks of the species Ixodes ricinus, however, literature data suggest tha

Survival dynamics of tick-borne encephalitis virus in Ixodes ricinus ticks.

Biotic factors contributing to the survival of tick-borne viruses in nature are poorly understood. Using tick-borne encephalitis virus (TBEV) and its principal European vector, Ixodes ricinus, we examined the relative roles of salivary gland infection, co-feeding transmission, and moulting in virus survival. Virus titres in the salivary glands increased after blood-feeding in a time- and dose-dependent manner. This was observed in ticks infected by inoculation but not in ticks infected by the natural route of co-feeding. Amplification of infection prevalence occurred via co-feeding. However, w

Babesia sp. EU1 from roe deer and transmission within Ixodes ricinus.

We report in vitro culture of zoonotic Babesia sp. EU1 from blood samples of roe deer in France. This study provides evidence of transovarial and transstadial transmission of the parasite within Ixodes ricinus, which suggests that this tick could be a vector and reservoir of EU1.

First isolation of Rickettsia helvetica from Ixodes ricinus ticks in France.

Two rickettsial isolates recovered from Ixodes ricinus ticks in Puy-de-Dôme (Central France) were characterized by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, Western blot immunoassay, restriction fragment length polymorphism analysis, and sequencing of a portion of the citrate-synthase gene. By these methods, the isolates appeared to be identical to a member of the spotted fever group rickettsiae, Rickettsia helvetica. This first isolation of Rickettsia helvetica in France has epidemiologic importance; a serosurvey on Mediterranean spotted fever conducted previously in Puy-de-D
