Riemerella anatipestifer (RA) infection causes high mortality and poor feed conversion, leading to great economic losses to the duck industry. This study investigated the effects of RA on the intestinal morphology and immune response of ducks. Histological examination showed that RA infection caused intestinal injury, including significantly reduced mucosal thickness on days 2, 3 and 5, significantly reduced villus height on days 1, 2, 3 and 5 (P < 0.05) and significantly reduced villus height to crypt depth ratios on days 2, 3, 5 and 9 of RA infection (P < 0.05). The expression of intestinal

A novel acquired aminoglycoside resistance gene, aph(3')-IVb, was identified via whole-genome sequencing of a multidrug-resistant Riemerella anatipestifer isolate from a duck. The gene encodes a 262-amino-acid phosphotransferase, APH(3')-IVb, sharing only 39.9% amino acid identity with its closest known homolog, APH(3')-IVa. Heterologous expression of aph(3')-IVb in Escherichia coli and a susceptible R. anatipestifer strain conferred resistance to neomycin, paromomycin, and ribostamycin, a phenotype validated by gene deletion and complementation experiments. Kinetic analysis of the purified AP

Riemeralla anatipestifer, a predominant bacterium with multidrug resistance, has caused tremendous economic losses in the poultry farming industry. However, there are few studies on its identification, pathogenic mechanisms, and virulence factors and effective and systematic prevention and control strategies. The emergence and spread of antibacterial resistance has prompted increased focus on R. anatipestifer. However, studies on the mechanisms underlying gene aggregation and dissemination are lacking. This review summarizes recent studies on R. anatipestifer and explores its epidemiology, pat

BACKGROUND: Tigecycline is regarded as one of the last-resort antimicrobials clinically. Emergence of plasmid-mediated tet(X) undermines such an important drug. However, the origins of tet(X) remain largely unexplored.
METHODS: Riemerella anatipestifer strains were characterized by PCR, antimicrobial susceptibility testing, WGS and bioinformatics analysis. Functional analysis of tet(X) was verified by cloning experiments. Genomic structures of chromosome- and plasmid-mediated tet(X) were analysed.
RESULTS: Thirty-eight R. anatipestifer strains were collected and found to be positive for tet(

Riemerella anatipestifer is an important pathogenic bacterium that infects ducks. It exhibits resistance to multiple classes of antibiotics. Multidrug efflux pumps play a major role as a mechanism of antimicrobial resistance in Gram-negative pathogens and they are poorly understood in R. anatipestifer. In this study, a gene encoding the B739_0873 protein in R. anatipestifer CH-1, which belongs to the resistance-nodulation-cell division (RND) efflux pump family, was identified. With respect to the substrate specificity of B739_0873, the antibiotic susceptibility testing showed that the B739_087

Riemerella anatipestifer (RA) is an economically important avian pathogen that has historically caused significant disease in waterfowls but is increasingly recognized in various poultry production systems and wild bird species, highlighting its expanding host range and adaptive potential. Despite the availability of vaccines and antimicrobial interventions, RA remains a persistent challenge due to serotype diversity, limited cross-protective immunity, evolving antimicrobial resistance patterns, and an incomplete understanding of the host-pathogen interactions that govern disease outcome. OBJECTIVES: To elucidate the role of a novel carbapenem-hydrolysing class D β-lactamase (RAD-1) from Riemerella anatipestifer.
METHODS: We applied WGS and bioinformatic analysis to screen putative β-lactamase genes in R. anatipestifer SCVM0004. A putative class D β-lactamase gene was cloned into pET24a and transferred into Escherichia coli BL21 (DE3) for antibiotic susceptibility determination and protein purification. Meanwhile, the purified native protein was used to determine the enzymatic activities.
RESULTS: A class D β-lactamase, RAD-1, was identified in the genome of R. anatipestifer

A novel chromosomally-located β-lactamase gene, blaRASA-1, was identified in Riemerella anatipestifer RA-CH-1. The RASA-1, encoded by blaRASA-1, was a class A extended-spectrum β-lactamase (ESBL), which shared 42.7% and 40.5% identities with the RAA-1 and CGA-1 β-lactamase, respectively. Overexpression of RASA-1 in Escherichia coli confers broad resistance to β-lactams and the purified native RASA-1 revealed ESBL-like hydrolysis activity. Blasting in GenBank showed that blaRASA-1 was exclusively detected in Riemerella anatipestifer. Moreover, sequence analysis revealed that this gene was locat

Riemerella anatipestifer, which causes New Duck Syndrome, poses a significant threat to poultry production, causesing respiratory distress, neurological signs, and septicemia primarily in ducks and occasionally chickens. R. anatipestifer outbreak control requires knowledge about their genetic diversity, serotypes, and antibiotic profiles, which are currently unavailable. In this study, R. anatipestifer was isolated from ducks and chickens in Thailand between 2021 and 2023 to characterize their genetic relatedness, antimicrobial profiles, and resistance genes. Seven different serotypes were ide

Riemerella anatipestifer (RA) infections cause major economic losses in the duck industry. In this study, we developed an RA vaccine to control virulent serotype 1 and 2 RA, which predominate in worldwide prevalence. We established a strategy for vaccine candidate screening, and selected strains D15-RDA-92 (serotype 1) and D14-RDA-8 (serotype 2). These strains were characterized by ≤50% embryo mortality and <3.0 serum resistance assay values in in vitro screening. We evaluated the protective efficacy of live bivalent RA vaccines against virulent homologous serotype RA. Ducklings received two o

Chloramphenicol resistance in Riemerella anatipestifer is primarily mediated by chloramphenicol acetyltransferases (CATs). Inconsistent nomenclature of the CatB superfamily in public databases currently complicates the genomic surveillance of these resistance determinants. In this study, we tested the subtype scheme recorded in the NCBI Reference Gene Catalog against a complete pairwise amino acid identity matrix, applying a ≥ 80% identity criterion. The analysis supports the existing type B-1 to type B-5 subtypes, assigns two previously unassigned sequences to Type B-2, and recommends retirem

Riemerella anatipestifer (R. anatipestifer) is a Gram-negative bacterial pathogen for ducks and other birds, the clinical isolates of this bacterium are commonly multidrug-resistant. In the previous study, it was showed that intracellular excess iron increased the sensitivity of R. anatipestifer to aztreonam and this process depended on the respiratory complex subunits nuoB and sdhC. However, the underlying mechanisms of respiratory complex involvement in antibiotic resistance was not to be fully understood. In this study, it was shown that under iron-limited conditions, R. anatipestifer exhib

Riemerella anatipestifer (R. anatipestifer) affects the duck farming industry worldwide, causing substantial economic losses. The current disease prevention and treatment strategies primarily include vaccines and antibiotics. However, the large number of serotypes and increasing resistance to R. anatipestifer make it challenging to prevent and treat the infection. This study carried out the serotyping and molecular typing of 51 R. anatipestifer strains and predicted vaccine proteins based on pan-genome analysis and cross-immune protection potential. For serotype identification, the rabbits wer

Riemerella anatipestifer infection is one of the main infectious diseases that threaten the poultry industry at present. Serotypes 1 and 2 are the main serotypes causing global outbreaks of R. anatipestifer. In this study, we designed the primers specific for R. anatipestifer serotypes 1 and serotype 2 strains, and for R. anatipestifer 16S rRNA (for the species identification) to establish a multiplex PCR assay for rapid detection of the serotype 1 and serotype 2 strains. The specificity test showed that a 505 bp fragment was amplified from serotype 1 strains, and a 1125 bp fragment was amplif

Riemerella anatipestifer (RA) is one of the most important pathogens of 1- to 8-wk-old ducklings that severely affects the development of the duck industry in China. Every year, antibiotic medicines including tetracycline and doxycycline are used in the duck industry. Few reports compare the expression of multidrug-resistant genes in RA before and after addition of chemical drugs. With this in mind, the direct effects of gradient concentration of tetracyclines on the expression of tetracycline resistance genes (TETr) in RA at the cDNA level were studied by using a quantitative real-time PCR me

Several outbreaks of Riemerella anatipestifer in commercial geese occurred within a short time period. A serious disease was recognized in the affected birds, mainly characterized by depression and severe neurologic disturbances. The morbidity ranged from 20 to 30% and the mortality from 5 to 20%. Generally, the clinical signs started at the age of 8 to 10 days. Post-mortem examination revealed fibrinous pericarditis, perihepatitis and airsacculitis in all birds. Some of the birds also had synovitis of the tibio-tarsal joints and oedematous swelling of the subcutaneous tissues around these joi

Riemerella anatipestifer (RA) is a common disease causing economic losses to duck farms worldwide. Novel supplements are crucially needed to control this bacterium, enhance poultry performance, and produce synergistic effects with vaccines in stimulating the immune system. This study investigated the effect of nano-selenium (Nano-Se) on the vaccinated (VAC) and challenged (Ch) Pekin ducklings (Anas platyrhynchos) with RA. Five experimental groups (G1-G5) were included in this study: G1 was the control group, G2 was the RA-challenged group, G3 was the Nano-Se+Ch group, G4 was the VAC+Ch group,

Mucosal immunity serves as the first line of defense against pathogenic infections in animals. Respiratory mucosal vaccines elicit multi-layered immune responses that confer distinct advantages for disease prevention and control. Riemerella anatipestifer (RA) infection causes duck infectious serositis, leading to sustained economic losses in the duck industry. Commercially available inactivated RA vaccines are hampered by drawbacks such as stress induction and adjuvant residue. Targeting the RA-YM strain of serotype 1, the dominant epidemic RA serotype in China, this study employed markerless
