Tissue expression of S100 proteins in gallbladder mucosa of the patients with calculous cholecystitis.

Proteins of S100 group, produced by phagocytes represent endogenous activators of innate immune responses. Role of these proteins in the etiopathogenesis of cholelithiasis remains unknown. The studies aimed at the morphometric evaluation of S100A8 and S100A9 protein expression in gallbladder mucosa in patients with acute and chronic calculous cholecystitis (n = 71). The presence of proteins was detected by immunohistochemistry while quantitative analysis employed the spatial visualization technique. We found the immunopositive expression of the two studied S100 proteins in neutrophils and mono…

Phagocyte-derived S100 proteins in autoinflammation: putative role in pathogenesis and usefulness as biomarkers.

The cytoplasmic S100 proteins derived from cells of myeloid origin are promising new markers of (auto-)inflammation. S100A8/A9 and S100A12 are released from monocytes and granulocytes during activation of the innate immune system. Tissue and serum concentrations correlate to disease activity, both during local and systemic inflammation. In autoinflammatory diseases such as Familial Mediterranean Fever (FMF) and Systemic onset Juvenile Idiopathic Arthritis (SJIA), a dysregulation of alternative secretory pathways may be involved in pathogenesis and lead to hypersecretion of S100 proteins. Since…

Elevation of S100 calcium-binding protein A7 in recurrent pterygium.

Recurrent pterygium, a common ophthalmic disease, is difficult to treat as its pathogenesis is unclear. To investigate the key genes responsible for the recurrence of pterygium, tissue samples were collected from six patients with primary pterygium (primary group), six patients with recurrent pterygium (recurrent group) and six patients with ocular trauma (control group) who underwent surgery between December 2014 and June 2017. The differentially expressed genes amongst these tissues were detected using expression profiling microarrays and verified by reverse transcription-quantitative PCR (R

Multicentric reticulohistiocytosis with S100 protein positive staining: a case report.

Multicentric reticulohistiocytosis is a very rare systemic disease that affects skin, mucosa and joints. We reported a case of a woman with multicentric reticulohistiocytosis who presented typical skin syndromes and arthralgia. Immunohistochemical analysis showed positive staining for S100 protein, which was reported negative in the majority of previously presented cases. Other immunohistochemical markers (CD68(+), CD1a(-), lagerin (-) and complete histologic and clinical picture were specific enough to make the definitive diagnosis of multicentric reticulohistiocytosis. The patient was admini

Immunohistochemical localization of S100 protein in benign and malignant conditions of the breast.

A peroxidase--antiperoxidase technique for S100 protein has been applied to 122 breast lesions from 122 patients. These included 35 cases of fibrocystic disease, 16 cases of sclerosing adenosis, 24 cases of papilloma and papillomatosis, 43 intraduct carcinomas, and four intralobular carcinomas. In fibrocystic disease, S100 protein was demonstrable in large amounts in cells between the duct lining cells and the basement membrane of the ducts, being most pronounced in those exhibiting adenosis. Areas of epitheliosis showed scattered positive cells within the ducts with more strongly positive cel

Altered expression of S100 fused-type proteins in human skin due to ultraviolet B irradiation.

Ultraviolet B (UVB) radiation profoundly affects epidermal homeostasis by inducing DNA damage, reactive oxygen species generation, and stress signalling. This review summarizes recent findings on the dynamic regulation of S100 fused-type proteins, including filaggrin (FLG), filaggrin 2 (FLG2), cornulin (CRNN), repetin (RPTN), hornerin (HRNR), trichohyalin (TCHH), and trichohyalin like protein 1 (TCHHL1) in human skin after UVB irradiation. In experiments using human skin xenografts, UVB (500 mJ/cm²) irradiation transiently upregulates proliferation-related proteins (TCHHL1, RPTN, and HRNR) and

Changes in S100 Proteins Identified in Healthy Skin following Electrical Stimulation: Relevance for Wound Healing.

OBJECTIVE: Targeted electrical energy applied to wounds has been shown to improve wound-healing rates. However, the mechanisms are poorly understood. The aim of this study was to identify genes that are responsive to electrical stimulation (ES) in healthy subjects with undamaged skin. METHODS: To achieve this objective, study authors used a small, noninvasive ES medical device to deliver a continuous, specific, set sequence of electrical energy impulses over a 48-hour period to the skin of healthy volunteers and compared resultant gene expression by microarray analysis. MAIN RESULTS: Applica

The human epidermal differentiation complex: cornified envelope precursors, S100 proteins and the 'fused genes' family.

The skin is essential for survival and protects our body against biological attacks, physical stress, chemical injury, water loss, ultraviolet radiation and immunological impairment. The epidermal barrier constitutes the primordial frontline of this defense established during terminal differentiation. During this complex process proliferating basal keratinocytes become suprabasally mitotically inactive and move through four epidermal layers (basal, spinous, granular and layer, stratum corneum) constantly adapting to the needs of the respective cell layer. As a result, squamous keratinocytes co

Mechanistic insights and clinical implications of the S100 protein family in breast cancer.

The S100 family, the largest subgroup of calcium-binding proteins, plays a pivotal role in maintaining cellular homeostasis through regulation of calcium-dependent signaling pathways. However, dysregulation of S100 proteins is increasingly implicated in breast cancer, the leading cause of female malignancy-related morbidity and mortality worldwide. Notably, majority of breast cancer-related deaths are attributed to metastasis and recurrence Aberrant S100 expression has emerged as a key contributor in such processes. Beyond promoting tumor progression by modulating proliferation, angiogenesis, …

Protein S100 immunoreactivity in glial cells and neurons of the Japanese quail brain.

In mammals, sparse data illustrated the neuronal expression of S100 protein in central and peripheral nervous system. Similar studies have not been performed in other vertebrate species, in particular in birds. We provide here a detailed description of the distribution of the calcium-binding protein S100 in neuronal and glial elements in the central nervous system of an avian species, the Japanese quail (Coturnix japonica) largely used for neuroanatomical and functional studies. The distribution of S100-like immunoreactivity was analyzed by three different antisera: a polyclonal, against S100

Retinoic Acid Regulates Immune Responses by Promoting IL-22 and Modulating S100 Proteins in Viral Hepatitis.

Although large amounts of vitamin A and its metabolite all-trans retinoic acid (RA) are stored in the liver, how RA regulates liver immune responses during viral infection remains unclear. In this study, we demonstrated that IL-22, mainly produced by hepatic γδ T cells, attenuated liver injury in adenovirus-infected mice. RA can promote γδ T cells to produce mTORC1-dependent IL-22 in the liver, but inhibits IFN-γ and IL-17. RA also affected the aptitude of T cell responses by modulating dendritic cell (DC) migration and costimulatory molecule expression. These results suggested that RA plays a

Pro-inflammatory S100 proteins are associated with glomerulonephritis and anti-dsDNA antibodies in systemic lupus erythematosus.

Objectives Systemic lupus erythematosus (SLE) is associated with elevated levels of S100A8/A9, pro-inflammatory proteins mainly secreted by activated polymorphonuclear neutrophils (PMNs). The underlying mechanisms for increased S100A8/A9 levels and their relation to the clinical phenotype have not been carefully investigated. We assessed S100A8/A9 and S100A12 levels in SLE patient sera in relation to disease activity, clinical phenotype, presence of anti-dsDNA antibodies and ability to promote phagocytosis of necrotic cells (NCs) by PMNs. Methods Serum levels of S100A8/A9 and S100A12 were meas

Effects of Ca2+ and Zn2+ on trifluoperazine-S100 proteins interactions: induced circular dichroism and fluorescence spectra.

Interactions of trifluoperazine (TFP) with S100 proteins, EF-hand type Ca2+-binding proteins, in the presence of Ca2+ and Zn2+ were studied with induced circular dichroism (CD) and fluorescence spectra. The positive CD bands of TFP were induced at around 265 nm by adding either S100a or S100a0 protein in the presence of Ca2+. No CD band of TFP was, however, induced by adding S100b protein in the presence of Ca2+. Addition of Zn2+ to the TFP/S100 protein solutions did not induce any CD band at all. The fluorescence intensity of 2-p-toluidinylnaphthalene 6-sulfonate (TNS) bound to S100a or S100a…

Trichohyalin-like 1 protein, a member of fused S100 proteins, is expressed in normal and pathologic human skin.

Trichohyalin-like 1 (TCHHL1) protein is a novel member of the fused-type S100 protein gene family. The deduced amino acid sequence of TCHHL1 contains an EF-hand domain in the N-terminus, one trans-membrane domain and a nuclear localization signal. We generated specific antibodies against the C-terminus of the TCHHL1 protein and examined the expression of TCHHL1 proteins in normal and pathological human skin. An immunohistochemical study showed that TCHHL1 proteins were expressed in the basal layer of the normal epidermis. In addition, signals of TCHHL1 proteins were observed around the nuclei
