In this study, six candidate DNA aptamers specific to trichlorfon were successfully obtained through 16 rounds of capture-SELEX. Their binding affinities were rigorously evaluated and comparatively analyzed using three orthogonal techniques: quantitative real-time polymerase chain reaction (qPCR), gold nanoparticle-based colorimetric assay (AuNP assay), and SYBR Green I (SGI)-mediated fluorescence assay. Among these identified aptamers, APTri-7 exhibited the highest binding affinity, with a dissociation constant (Kd) of 0.597 ± 0.206 µg mL-1. Subsequently, using APTri-7, a highly sensitive ele

BACKGROUND: Brucellosis is a zoonosis, caused by Brucella spp. which are small aerobic intracellular coccobacilli, localized in the reproductive organs of host animals, causing abortion and sterility. The diagnosis of this zoonosis is based on microbiological, serological or real time-polymerase chain reaction (RT-PCR) laboratory tests. Although the common microbiological and serological based assays have advantages, they are not able to solve the diagnosis problems.
AIMS: To overcome some of the limitations of present techniques, in this study, we developed an aptamer through whole-cell syst

Cerebrospinal fluid (CSF) leakage is a serious medical condition that, if left undiagnosed or untreated, can lead to severe complications, including life-threatening intracranial infections (e.g., meningitis). Current diagnostic methods rely on immunofixation electrophoresis or nephelometry for detecting beta-2 transferrin or beta-trace protein, respectively, but these techniques are time-consuming, require specialized laboratory equipment, and may yield false-negative results due to sample contamination. To address these limitations, we developed a novel electrochemical aptamer-based (E-AB) b

Exosome (EXO) membrane proteins are attractive biomarkers for liquid biopsy, yet their heterogeneity makes it difficult to develop reliable antibody-based recognition reagents. Aptamers provide high-affinity and highly specific alternatives through the systematic evolution of ligands by exponential enrichment (SELEX), but the nanosized EXOs introduce substantial separation challenges that complicate SELEX workflows. Here, we present DeteRministic Evolution of Aptamers via a Microfluidic-integrated robotic platform (DREAMbot), an automated system engineered to execute multiround EXO-targeted ap

Epinephrine (EP) and norepinephrine (NE) are two essential catecholamine neurotransmitters. Accurate monitoring of their levels is crucial for clinical diagnosis. In this study, we employed the library-immobilized SELEX technique to select DNA aptamers for EP and NE. A representative aptamer, named EP-1, showed a strong affinity to EP and a slightly weaker binding to NE, with dissociation constant (Kd) values of 12.2 μM and 36.6 μM, respectively, as determined by the thioflavin T (ThT) assay. The Kd values obtained from isothermal titration calorimetry (ITC) were 9.4 μM EP and 58.1 μM NE, comp

BACKGROUND: Most biological molecular complexes consist of multiple functional domains, yet rationally constructing such multifunctional complexes is challenging. Aptamers, the nucleic acid-based functional molecules, can perform multiple tasks including target recognition, conformational changes, and enzymatic activities, while being chemically synthesizable and tunable, and thus provide a basis for engineering enhanced functionalities through combination of multiple units. However, the conventional approach of simply combining aptamer units in a serial manner is susceptible to undesired cros

Aptamers are nucleic acid-based affinity reagents that have been incorporated into a variety of molecular sensor formats. However, many aptamer sensors exhibit insufficient sensitivity and specificity for real-world applications, and although considerable effort has been dedicated to improving sensitivity, sensor specificity has remained largely neglected and understudied. In this work, we have developed a series of sensors using aptamers for the small-molecule drugs flunixin, fentanyl, and furanyl fentanyl and compare their performance─in particular, focusing on their specificity. BACKGROUND: Aptamers are not so new a concept, however, it is scarcely discussed by medical fraternity. Aptamers are potent, new identification molecules set to rope in a new technique in the diagnostic arena. Aptamers have started almost a revolution in diagnostic assays since their discovery in the 90s. (Radu S. Phalloidin (PHD), a potent bicyclic peptide from Amanita species, poses severe food safety risks when misidentified as edible mushrooms. However, no aptamer-based detection system has been reported to date. Here, we report the isolation of a high-affinity and highly specific DNA aptamer (Apt-PHD) selected through 15 rounds of Capture-SELEX using a structured 97-nt ssDNA library. High-throughput sequencing identified PHD-3 as the optimal candidate, exhibiting dissociation constants of 0.292 ± 0.0034 μM (isothermal titration calorimetry) and 42.7 ± 6.7 nM (SYBR Green I fluorescence assay). BACKGROUND: Interleukin-17A (IL-17A) is an important pro-inflammatory cytokine observed in the development of many disorders, such as psoriasis, rheumatoid arthritis, and multiple sclerosis. The anti-IL-17A biological drugs, including Secukinumab, Ixekizumab, and Brodalumab, are monoclonal antibodies approved for several disease treatments. Due to the disadvantages of biological therapies, including their immunogenicity, difficulties in scale generation, and high production costs and time, it is necessary to find new alternative anti- IL-17A agents for these monoclonal antibodies. Aptamers are single-stranded DNA or RNA molecules that bind specific ligands with high affinity. DNA aptamers are especially promising due to their enhanced stability and cost-effectiveness, yet their practical use often necessitates coupling with an expression platform to transduce ligand-binding events into detectable signals. To overcome this limitation, we developed Expression-SELEX, an efficient selection method that integrates a DNAzyme-based expression platform directly into the screening process. Previously, we employed this approach to isolate a phenylalanine-responsive allosteric DNA

Legionella pneumophila (Lp) is a water borne bacterium causing Legionnaires' Disease (LD) in humans. Rapid detection of Lp in water system is essential to reduce the risk of LD outbreaks. The methods currently available require expert skills and are time intensive, thus delaying intervention. In situ detection of Lp by biosensor would allow rapid implementation of control strategies. To this end, a biorecognition element is required. Aptamers are considered promising biorecognition molecules for biosensing. Aptamers are short oligonucleotide sequence folding into a specific structure and are a

The abuse of ribavirin, an antiviral drug, in poultry breeding can cause quality degradation and drug resistance. So it is of great importance to establish a simple and effective method for detecting ribavirin in foods. In this work, aptamers that could especially bind to ribavirin with high affinity were obtained by the Capture-SELEX method. After 15 rounds of enrichment, ssDNA library pool was enriched and then analyzed by high-throughput sequencing. The seven most enriched sequences were selected as candidate aptamers for affinity and specificity characterization. Among the candidate aptame
