Modulation of carbon tetrachloride-induced oxidative stress by dietary fat in rats(open star).

Oxidative stress is believed to be involved in the pathophysiology of a number of chronic diseases including atherosclerosis, diabetes, and cataracts and to accelerate the aging process. The aim of this study was to elucidate the role of various dietary fats in the in vivo modulation of CCl(4) induced oxidative stress using rat as a model. Rats were raised on diets enriched with saturated (Beef Tallow), n-9 (Sunola oil), n-6 (Safflower oil) or n-3 (Flaxseed oil) fatty acids and exposed to elevated oxidative stress by administration of CCl(4.) Plasma concentration of 8-iso-PGF(2alpha), antioxid

Positional distribution of CLA in TAG of lamb tissues.

The content and positional distribution of CLA in TAG fractions of lamb tissues was examined with either preformed CLA or the linoleic acid precursor of CLA in the diet as experimental treatments. The CLA content of phospholipid (PL) from these tissues was also examined. Thirteen lambs were randomized to the following dietary treatments: (i) control diet (no supplement); (ii) CLA supplementation (0.33 g d(-1) for 21 d prior to weaning) to milk-replacer of pre-ruminating lambs, or (iii) feeding linoleic acid-rich oil (6% safflower oil on a dry matter basis) to weaned ruminating lambs. Effect of vegetable oils on the experimental infection of mice with Trypanosoma congolense.

Vegetable oils are frequently used as solvents for lipophilic materials; accordingly, the effects of their components should be considered in animal experiments. In this study, the effects of various vegetable oils on the course of Trypanosoma congolense infection were examined in mice. C57BL/6J mice were orally administered four kinds of oils (i.e., coconut oil, olive oil, high oleic safflower oil, and high linoleic safflower oil) with different fatty acid compositions and infected with T. congolense IL-3000. Oil-treated mice infected with T. congolense showed significantly higher survival ra

Rapid determination of volatile constituents in safflower from Xinjiang and Henan by ultrasonic-assisted solvent extraction and GC-MS.

The total volatile components were extracted from safflower by ultrasonic-assisted solvent extraction (USE) and their chemical constituents were analyzed by gas chromatography-mass spectrometry (GC-MS) to provide scientific basis for the quality control of safflower. Five different solvents (diethyl ether, ethanol, ethyl acetate, dichloromethane and acetone) were used and compared in terms of number of volatile components extracted and the peak areas of these components in TIC. The results showed that USE could be used as an efficient and rapid method for extracting the volatile components fro

[Biliary calculi in th golden hamster (M. a. auratus). XXXV. Effect of the hydrogenation of Polifat KA-02 on its preventive activity against pigment cholelithiasis].

It has been previously reported that Polifat KA-02 (fatty acids derived from safflower oil) prevents the pigment cholelithiasis produced by vitamin A in the golden hamster. In order to establish whether the saturation or the unsaturation of Polifat is directly related with its preventive action, in the present study the effect of the hydrogenation of the derivative on this preventive action was tested. Two experiments were carried out in which the animals received the different experimental diets ad libitum during 70 days. In the 1st. experiment the effect of the additions to the lithogenic di

Genome-Wide Identification and Functional Characterization of the TIFY Gene Family Associated with Abiotic Stress and Hormone Response in Carthamus tinctorius L.

Jasmonate-ZIM domain (JAZ) proteins act as core negative regulators of jasmonic acid signaling and mediate plant stress adaptation, developmental progression, and secondary metabolism. Safflower (Carthamus tinctorius L.) is an economically important Asteraceae crop with medicinal, edible oil, and ornamental values; however, the evolutionary characteristics and molecular functions of its TIFY gene family remain poorly characterized. In the present study, we performed a genome-wide identification of the safflower CtTIFY family, followed by systematic analyses of phylogenetic relationships, chrom

A historical perspective and review of the safety of lipid emulsion in pregnancy.

Total parenteral nutrition for pregnant patients is essential to maintain or restore optimal nutritional status for both the mother and fetus when adequate protein and caloric intake by the oral or enteral route is not feasible. This report reviews the safety of intravenous lipid emulsions in total parenteral nutrition programs for pregnant patients. Controversy regarding the adverse effects of intravenous lipid emulsion when administered to pregnant patients is based on early reports of the use of cottonseed oil lipid emulsions. The adverse effects reported included infarction of the placenta

Dietary fat and colon cancer: animal model studies.

Since it was first suggested that high dietary fat is a risk factor in colon cancer, there have been several studies to test this hypothesis. Epidemiologic studies suggested a positive association between dietary fat and colon cancer. Laboratory animal model studies demonstrated that not only the amount of fat, but also types of fat differing in fatty acid composition are important determining factors in colon tumor development. Chemically-induced colon tumor incidence was increased in rats fed the semipurified diets containing 23% corn oil, safflower oil, lard or beef tallow (high-fat) as com

Effect of Lifelong Exposure to Dietary Plant and Marine Sources of n-3 Polyunsaturated Fatty Acids on Morphologic and Gene Expression Biomarkers of Intestinal Health in Early Life.

Altered intestinal health is also associated with the incidence and severity of many chronic inflammatory conditions, which could be attenuated via dietary n-3 PUFA interventions. However, little is known about the effect of lifelong exposure to n-3 PUFA from plant and marine sources (beginning in utero via the maternal diet) on early life biomarkers of intestinal health. Harems of C57Bl/6 mice were randomly assigned to one of three isocaloric AIN-93G modified diets differing in their fat sources consisting of the following: (i) 10% safflower oil (SO, enriched in n-6 PUFA), (ii) 3% flaxseed oi

Physiological effects of γ-linolenic acid and sesamin on hepatic fatty acid synthesis and oxidation.

Interrelated effects of γ-linolenic acid (GLA) and sesamin, a sesame lignan, on hepatic fatty acid synthesis and oxidation were examined. Rats were fed experimental diets supplemented with 0 or 2 g/kg sesamin (1:1 mixture of sesamin and episesamin) and containing 100 g/kg of palm oil (saturated fat), safflower oil rich in linoleic acid, or oil of evening primrose origin containing 43% GLA (GLA oil) for 18 days. In rats fed sesamin-free diets, GLA oil, compared with other oils, increased the activity and mRNA levels of various enzymes involved in fatty acid oxidation, except for some instances.

Isolation, Characterization, and Anti-Inflammatory Effects of Carthamus tinctorius L. Leaf-Derived Exosome-like Nanoparticles in ETEC-Challenged IPEC-J2 Cells.

Safflower (Carthamus tinctorius L.) is a cash crop grown worldwide. Its seeds and flowers are primarily processed for edible oil and medicinal raw materials, while stems and leaves are discarded as agricultural waste. Recently, plant-derived exosome-like nanoparticles (PELNs) have gained growing research interest in food nutrition owing to their exceptional biocompatibility and relatively low cost for large-scale production. This study aimed to explore the potential functional value of safflower leaf agricultural waste by isolating safflower-derived PELNs. Firstly, a protocol was established f

CtChi19: a valuable gene for improving resistance to Botrytis cinerea and Alternaria alternata in Carthamus tinctorius.

Safflower (Carthamus tinctorius L.) is an economically important crop with both medicinal and oil-producing values, However, its production is severely threatened by fungal diseases, including those caused by Alternaria alternata and other pathogenic fungi. To date, molecular investigations into the disease resistance mechanisms of safflower remain largely limited. In this study, proteomic analysis was performed on safflower following infection by A. alternata, identifying 490 and 921 differentially expressed proteins (DEPs) at 48 hours post-infection (hpi) and 72 hpi, respectively, Among thes

Dietary high oleic canola oil supplemented with docosahexaenoic acid attenuates plasma proprotein convertase subtilisin kexin type 9 (PCSK9) levels in participants with cardiovascular disease risk: A randomized control trial.

Proprotein convertase subtilisin/kexin type 9 (PCSK9) is a novel circulating protein which plays an important role in regulation of cholesterol metabolism by promoting hepatic LDL receptor degradation. However, the action of dietary fat composition on PCSK9 levels remains to be fully elucidated. The objective was to investigate the action of different dietary oils on circulating PCSK9 levels in the Canola Oil Multicenter Intervention Trial (COMIT). COMIT employed a double-blinded crossover randomized control design, consisting of five 30-d treatment periods. Diets were provided based on a 3000

Omega-3 polyunsaturated fatty acids and the eicosapentaenoic acid metabolite 18-hydroxyeicosapentaenoic acid (18-HEPE) attenuate experimental necrotizing enterocolitis.

Necrotizing enterocolitis (NEC), a devastating gastrointestinal disease in infants born prematurely, is linked to intestinal inflammation. Maternal intake of omega-3 polyunsaturated fatty acids (PUFAs) has been reported as protective against inflammatory reactions in their offspring. We hypothesized that omega-3 fatty acids would prevent intestinal injury in an experimental model of NEC. Elevated omega-3 levels in mouse pups were achieved either by enriching dam-fed diets with omega-3 or by endogenous production in transgenic fat-1 mice able to convert omega-6 to omega-3. Mucosal injury scores

Genome-Wide Identification, Expression Profiling, and Functional Validation of Oleosin Gene Family in Carthamus tinctorius L.

Carthamus tinctorius L., commonly known as safflower, is an important oilseed crop containing oil bodies. Oil bodies are intracellular organelles in plant cells for storing triacylglycerols (TAGs) and sterol esters. Oleosins are the most important surface proteins of the oil bodies. We predicted and retrieved the sequences of eight putative C. tinctorius oleosin (Ctoleosin) genes from the genome database of safflower. The bioinformatics analyses revealed the size of their open reading frames ranging from 414 to 675 bp, encoding 137 to 224 aa polypeptides with predicted molecular weights of 14.

Hypertension induced by omega-3 polyunsaturated fatty acid deficiency is alleviated by alpha-linolenic acid regardless of dietary source.

Omega-3 polyunsaturated fatty acid deficiency, particularly during the prenatal period, can cause hypertension in later life. This study examined the effect of different sources of alpha-linolenic acid (canola oil or flaxseed oil) in the prevention of hypertension and other metabolic symptoms induced by an omega-3 fatty acid-deficient diet. Dams were provided one of three experimental diets from 1 week before mating. Diets were either deficient (10% safflower oil-DEF) or sufficient (7% safflower oil+3% flaxseed oil-SUF-F; or 10% canola oil-SUF-C) in omega-3 fatty acids. The male offspring were

Impact of dietary n-3 FA deficiency on rat bone tissue FA composition.

The effect of dietary n-3 FA deficiency on bone tissue FA composition was evaluated in growing rats. Two mixtures combining hydrogenated coconut oil with safflower oil served as the n-3-deficient dietary treatments and provided two levels of linoleic acid (LA). The n-3 treatments were formulated with added alpha-linolenic acid (LNA) from flaxseed oil (diet LNA) or LNA plus DHA, and both were balanced for LA. This study showed that bone is sensitive to changes in dietary n-3 FA and that DHA is more effective than LNA in maintaining DHA levels in these tissues.
