Sarsasapogenin Suppresses RANKL-Induced Osteoclastogenesis in vitro and Prevents Lipopolysaccharide-Induced Bone Loss in vivo.

INTRODUCTION: Osteoclasts are giant polynuclear cells; their main function is bone resorption. An increased number of osteoclasts and enhanced bone resorption exert significant effects on osteoclast-related bone-lytic diseases, including osteoporosis. Given the limitations of current therapies for osteolytic diseases, it is urgently required to develop safer and more effective alternatives. Sarsasapogenin, a major sapogenin from Anemarrhena asphodeloides Bunge, possesses potent antitumor effects and inhibits NF-κB and MAPK signaling. However, the manner in which it affects osteoclasts is uncle

Discovery of a Neuroprotective Diosgenin Derivative as a Novel Antidepressant Candidate Targeting LPS-TLR4 Signaling.

Depression is a widespread and increasing mental disorder, yet current antidepressants, including tricyclic antidepressants (TCAs) and selective serotonin reuptake inhibitors (SSRIs), often cause notable side effects and limited efficacy. Hence, safer therapeutic options are needed. Diosgenin, a phytosteroid sapogenin from the Dioscoreaceae plants, has demonstrated therapeutic potential for neurological disorders but is hindered by unclear target mechanism, poor solubility, and limited bioavailability. Here, we synthesized diosgenin derivatives and evaluated their biological activities. Among

Diosgenin enhances transplanted human oligodendrocyte precursor cell-mediated myelination in preterm white matter injury by inhibiting matrix metalloproteinase-9-driven TGF-β/Smad2/3 signaling.

Preterm white matter injury (PWMI) is a major cause of long-term motor and cognitive disability, and there are no therapies that directly promote oligodendrocyte-lineage maturation. Here, we identify the natural steroid sapogenin diosgenin as a potent pro-myelinating modulator for transplanted human oligodendrocyte precursor cells (hOPCs) using a hypoxia-ischemia mouse model of PWMI, together with in vitro microglia-OPC co-culture systems. Diosgenin crosses the blood-brain barrier and promotes the differentiation of both transplanted hOPCs and endogenous OPCs into myelinating oligodendrocytes,

Unlocking the antimalarial potential of novel steroid-tetraoxane hybrids through consensus molecular docking and molecular dynamics investigation.

The emergence of Plasmodium falciparum resistance to artemisinin-based combination therapies necessitates the development of novel antimalarial agents. This study presents the first computational investigation of steroid-tetraoxane hybrids targeting cyclophilin, a key protein implicated in artemisinin resistance mechanisms. We designed a library of 127 steroid-1,2,4,5-tetraoxane hybrid compounds combining steroidal sapogenin (∆5,(6)-diosgenin-3-one) and gem-dihydroperoxides, and employed consensus molecular docking across eight platforms to minimize algorithm-specific biases. Diosgenone synthesis, anti-malarial activity and QSAR of analogues of this natural product.

Solanum nudum Dunal steroids have been reported as being antimalarial compounds; however, their concentration in plants is low, meaning that the species could be threatened by over-harvesting for this purpose. Swern oxidation was used for hemisynthesis of diosgenone (one of the most active steroidal sapogenin diosgenin compounds). Eighteen structural analogues were prepared; three of them were found to be more active than diosgenone (IC50 27.9 μM vs. 10.1 μM, 2.9 μM and 11.3 μM). The presence of a 4-en-3-one grouping in the A-ring of the compounds seems to be indispensable for antiplasmodial a

Diosgenin exerts an antihypertensive effect in spontaneously hypertensive rats via gut-brain communication.

Gut microbiota is well-established to regulate host blood pressure. Diosgenin is a natural steroid sapogenin with documented anti-inflammatory, antioxidant and antihypertensive properties. We aimed to investigate whether the antihypertensive effects of diosgenin are mediated by the microbiota-gut-brain axis in spontaneously hypertensive rats (SHR). 15-Week-old male Wistar Kyoto rats (WKY) and age-matched SHR were randomly distributed into three groups: WKY, SHR treated with a vehicle, and SHR treated with diosgenin (100 mg kg-1). Our results showed that diosgenin prevented elevated systolic bl

Report: structures and hepatocytotoxicity of co-occurring substances in oleanolic acid tablets.

Tablets of oleanolic acid (OA) have been approved by SFDA in China as an adjuvant therapy for acute and chronic hepatitis. Co-occurring substances present in the tablets of OA and their hepatocytotoxicity have not yet been reported. In the current investigation, the crude OA drug was separated by repeated column chromatography. The structures of the isolated compounds were characterized by spectral analysis and the cytotoxicity of each compound was evaluated in vitro against the human normal liver cell L02 at concentrations from 0.125 to 1000 μmol/L using the MTT method. As a result, OA and it

Functional characterization of a novel β-glucosidase converting trillin to diosgenin from Enterococcus faecalis.

BACKGROUND: Diosgenin, known as “medicinal gold”, is the main precursor for the synthesis of steroid hormone drugs. Currently, diosgenin is primarily produced industrially via the extraction of plant-derived steroidal saponins followed by acid hydrolysis, which generates substantial acidic wastewater. RESULTS: To address this environmental challenge, we sought to discover efficient glycosidases capable of converting steroidal saponins to diosgenin as a sustainable alternative to conventional acid hydrolysis. Through selective enrichment using dioscin as the sole carbon source, we isolated a st

Evaluation of aridanin, a glycoside, and Aridan, an aqueous extract of Tetrapleura tetraptera fruit, on Schistosoma mansoni and S. bovis.

Experiments were conducted on different stages of Schistosoma mansoni and S. bovis using various concentrations of aridanin and Aridan. Aridanin and Aridan were active against Schistosoma mansoni and S. bovis miracidia. A low concentration of aridanin (0.25 micrograms/ml) reduced the production of cercariae by snails already shedding cercariae. Aridanin and Aridan also produced profound reduction in the worm recovery of mice infected with pretreated cercariae of S. mansoni and S. bovis. Higher concentrations of the molluscicides were biocidal to the cercariae of these schistosomes. These resul

Ginseng-derived cholesterol analogues enhance pulmonary siRNA delivery and alleviate allergic asthma.

BACKGROUND: Asthma patients exhibit elevated airway mucus secretion. Small interfering RNA (siRNA) targeting the mucin MUC5AC delivered by lipid nanoparticles (LNPs) is promising but limited by low transfection efficiency. Protopanaxadiol (PPD), protopanaxatriol (PPT), and ginsenoside Rh2 (GR2) were chosen because their dammarane skeleton preserves membrane-insertion capacity for endosomal escape, whereas bulkier multi-glycosylated ginsenosides adversely affect LNP size and stability. PURPOSE: In this study, we designed novel inhaled LNPs incorporating ginseng-derived cholesterol analogs to e

Engineering Salvia miltiorrhiza hairy roots as a scalable platform for high-yield production of paclitaxel and ginsenoside precursors.

Plant-derived natural products offer a rich source of therapeutic agents. However, sustainable and high-yield production remains a grand challenge. We engineered Salvia miltiorrhiza hairy roots to produce taxadiene, a key precursor for the anticancer drug paclitaxel, and protopanaxadiol, a precursor for ginsenosides. The heterologous expression of two key biosynthetic genes, taxadiene synthase from Taxus wallichiana and protopanaxadiol synthase from Panax notoginseng, enabled the production of taxadiene and protopanaxadiol, respectively. Our strategy combined multiple approaches to enhance ter
