Receptor-mediated release of inositol phosphates in the cochlear and vestibular sensory epithelia of the rat.

Various neurotransmitters, hormones and other modulators involved in intercellular communication exert their biological action at receptors coupled to phospholipase C (PLC). This enzyme catalyzes the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PtdInsP2) to inositol 1,4,5-trisphosphate (InsP3) and 1,2-diacylglycerol (DG) which act as second messengers. In the organ of Corti of the guinea pig, the InsP3 second messenger system is linked to muscarinic cholinergic and P2y purinergic receptors. However, nothing is known about the InsP3 second messenger system in the vestibule. In this stud

Designing proteins to combat disease: Cardiac troponin C as an example.

Throughout history, muscle research has led to numerous scientific breakthroughs that have brought insight to a more general understanding of all biological processes. Potentially one of the most influential discoveries was the role of the second messenger calcium and its myriad of handling and sensing systems that mechanistically control muscle contraction. In this review we will briefly discuss the significance of calcium as a universal second messenger along with some of the most common calcium binding motifs in proteins, focusing on the EF-hand. We will also describe some of our approaches

Involvement of second messengers in the signaling pathway of vitellogenesis-inhibiting hormone and their effects on vitellogenin mRNA expression in the whiteleg shrimp, Litopenaeus vannamei.

We incubated fragments of Litopenaeus vannamei ovary to investigate second messengers involved in the regulation of vitellogenin (vg) mRNA levels. The use of 100nM recombinant vitellogenesis-inhibiting hormone (VIH) (corresponding to recombinant L. vannamei sinus gland peptide-G: rLiv-SGP-G) significantly reduced vg mRNA expression in sub-adults after 8h incubation to less than 20% of the control. The concentration of intracellular cyclic guanosine monophosphate (cGMP) increased 3.2-fold relative to the control after 2h incubation with rLiv-SGP-G. However, it reached levels 18-fold relative to

The surface interface and swimming motility influence surface-sensing responses in Pseudomonas aeruginosa.

Bacterial biofilms have been implicated in several chronic infections. After initial attachment, a critical first step in biofilm formation is a cell inducing a surface-sensing response. In the Gram-negative opportunistic pathogen Pseudomonas aeruginosa, two second messengers, cyclic diguanylate monophosphate (c-di-GMP) and cyclic adenosine monophosphate (cAMP), are produced by different surface-sensing mechanisms. However, given the disparate cellular behaviors regulated by these second messengers, how newly attached cells coordinate these pathways remains unclear. Some of the uncertainty rel

Network-level divergence in cyclic di-GMP signalling drives ecological versatility in Acinetobacter baumannii.

Second messenger signalling pathways are known to play a fundamental role in governing bacterial physiology and mediating rapid adaptive responses to stimuli. Despite the extensive characterisation of second messenger signalling systems such as the cyclic di-GMP (c-di-GMP) signalling system in pathogens, relatively little is known about the role of these pathways in the priority pathogen Acinetobacter baumannii. To address this, we carried out a comprehensive exploration of c-di-GMP signalling across multiple modern A. baumannii clinical isolates. We elucidate this second messenger's regulon a

Inhibition by aspirin of the stimulated PGI2 synthesis after administration of kallikrein to hypertensive patients (new aspects of chemical pathogeny concerning the messenger systems: A and C).

The vascular smooth muscle cells belong to the category of "bidirectionally controlled cells", which can be activated (leading to contraction) by some first-messengers (such as Catecholamine-alpha 1, or Angiotensin II) in connection with a recently discovered intracellular second-messenger system-C (sms-C), namely, Diacylglycerol-dependent Protein Kinase-C + Calcium mobilization. On the other hand, these cells can be inhibited (leading to relaxation) by other first-messengers (such as Catecholamine-beta or PGI2) related to the classical "second-messenger system-A" (sms-A), namely: Ns--Adenylat

Increased G alpha q/11 immunoreactivity in postmortem occipital cortex from patients with bipolar affective disorder.

As disturbances in guanine nucleotide binding (G) protein-coupled phosphoinositide second messenger systems have been implicated in bipolar disorder, we examined whether the abundance of G alpha q/11 and phospholipase C (PLC)-beta 1 two key transducing proteins in this signaling pathway, are altered in this disorder. Compared with the controls, immunoreactive levels of G alpha q/11 were significantly elevated by 62% (p = .047) in occipital cortex of bipolar subjects. A similar increase (52%) in the PLC-beta 1 immunolabeling was also found in the occipital cortex of the bipolar subjects, but on

Prostaglandin E receptors in myometrial cells.

Prostaglandins (PGs) exert their effects via binding to specific cell surface receptors and influencing second messenger systems through G-proteins. PGE2 may interact with at least four receptor subtypes (EP1, EP2, EP3, EP4), each showing different pharmacological profiles. The second messengers calcium, inositol phosphates (InsPs) and cyclic nucleotides play decisive roles in uterine contractility. The question in this investigation was, which EP receptors, G-proteins and second messenger systems transmit PGE2 induced signals in human myometrium. We have measured changes in InsPs and cAMP for

Mechanism of melatonin action.

Melatonin transduces the effect of photoperiod on the neuroendocrine system. Synthesis of melatonin in the pineal gland is well described, but the location of its target(s) and the mechanism of its action are little known. In attempt to localize melatonin target(s), the presence of high affinity binding sites in rat brain was determined. Such sites were detected in discrete brain areas, including the hypothalamus and anterior pituitary. Subcellular analysis indicated these binding sites were on plasma membranes, which suggests that melatonin modulates cell functions through intracellular secon

Inositol 1,4,5-trisphosphate receptors: immunohistochemical localization to discrete areas of rat central nervous system.

The second messenger inositol 1,4,5-trisphosphate triggers the release of intracellular Ca2+ stores upon binding to the inositol 1,4,5-trisphosphate receptor protein, a calcium channel that has been purified and molecularly cloned. To clarify the roles of inositol 1,4,5-trisphosphate receptor in the central nervous system, we have examined in detail the distribution of inositol 1,4,5-trisphosphate receptors in the rat brain and spinal cord using immunohistochemical methods. Inositol 1,4,5-trisphosphate receptors are present in neuronal cells, fibers and terminals in a wide distribution of area

Cholinergic stimulation of phosphoinositide hydrolysis in rabbit kidney.

An injection of acetylcholine (ACh) into renal artery is known to cause diuresis. In brain and other organs, cholinergic agents have been shown to produce their actions through the phosphoinositide (PI) second messenger system. To determine if cholinergic agents also produce activation of the PI messenger system in the kidney, we investigated the effects of carbachol (a stable analog of acetylcholine) on PI hydrolysis in the cortex, outer medulla and inner medulla of the rabbit kidney. PI hydrolysis was determined by measuring the formation of inositol phosphates in response to stimulation by

Developmental changes in signalling transduction factors in maturing sperm during epididymal transit.

Testicular spermatozoa are incapable of fertilising eggs in vivo and the epididymal epithelium is responsible for creating a luminal environment conducive to the acquisition of fertilising capacity by the spermatozoa within it. It does this by modifying sperm membranes so that they can interact with stimuli from the female tract, and by supplying or removing low molecular weight compounds that modify sperm cytoplasmic composition. Both these extracellular and intracellular events associated with signal transduction and second messenger systems in maturing spermatozoa are reviewed here.

Phosphoinositide signalling system in human erythrocyte and its role in cell pathology.

Human erythrocytes contain 217.2 +/- 70.5 pmole of inositol 1,4,5-trisphosphate per ml of packed cells (n = 14). The increased generation of Ins 1,4,5P3, was induced by 1 microM A 23187, 20 microM Pb2+, and by AlF4-. Ins 1,4,5P3, Pb2+ and AlF4- evoke shape changes, disorganisation of spectrin network and vesiculation. In erythrocytes of patients with hereditary spherocytosis the level of Ins 1,4,5P3 was increased to 556.7 +/- 374 pmole per ml of packed cells. We suggest that activation of the phosphoinositide signalling system may represent a common denominator for many divergent stimuli and d

Aging does not alter phosphoinositide hydrolysis in the rat cochlear lateral wall.

We have previously reported that the inositol 1,4,5-trisphosphate (InsP3) second messenger system is coupled to purinergic P2y receptors in the cochlear sensory epithelium and lateral wall. The tissues of the cochlear lateral wall (stria vascularis and spiral ligament) are responsible for maintaining the ionic composition of the cochlear endolymph. Both the endolymphatic potential and signal transduction processes are well known to be affected by aging. Furthermore, intracellular inositol concentrations decrease with age in the cochlear sensory epithelia. The present study compared the puriner

Serotonin transporter messenger RNA in the developing rat brain: early expression in serotonergic neurons and transient expression in non-serotonergic neurons.

Serotonin has been shown to affect the development of the mammalian nervous system. The serotonin transporter is a major factor in regulating extracellular serotonin levels. Using in situ hybridization histochemistry the rat serotonin transporter messenger RNA was localized during embryogenesis, the first four weeks postnatally and adulthood. Three general classes of serotonin transporter messenger RNA expression patterns were observed: (i) early detection with continued expression through adult age, (ii) transient expression colocalized with vesicular monoamine transporter 2 messenger RNA but

Effects of subarachnoid hemorrhage on vascular responses to calcitonin gene-related peptide and its related second messengers.

Calcitonin gene-related peptide (CGRP) is a potent vasodilator and a primary signaling molecule in neurovascular communication. In the present study, the authors examined cerebrovascular responses to CGRP and its related second messenger systems during cerebral vasospasm induced by subarachnoid hemorrhage (SAH). Tension measurements were performed in vitro on ring strips of basilar arteries obtained from rabbits subjected to artificial SAH and from control (non-SAH) animals. In vessels from SAH animals, which were preconstricted with serotonin, the vasorelaxant response to CGRP was attenuated.

Compartmentalized cAMP signalling in regulated exocytic processes in non-neuronal cells.

Cyclic adenosine monophosphate (cAMP) is a central second messenger controlling a plethora of vital functions. Studies of cAMP dynamics in living cells have revealed markedly inhomogeneous concentrations of the second messenger in different compartments. Moreover, cAMP effectors such as cAMP-dependent protein kinase (PKA) and cAMP-activated GTP-exchange factors (Epacs) are tethered to specific cellular sites. Both the tailoring of cAMP concentrations, and the activities of cAMP-dependent signalling systems at specific cellular locations are prerequisites for most, if not all, cAMP-dependent pr

Secretory activity in salt glands of birds and turtles: stimulation via cyclic AMP.

O2 consumption in tissue slices from the nasal salt gland of the duck and the lachrymal salt gland of Malaclemys is stimulated by methacholine, a stimulation that is inhibited by bumetanide and by ouabain. In addition, the calcium ionophore A23187 mimics the action of methacholine in stimulating this secretion-related O2 consumption in both glands, suggesting a second-messenger role for this ion in the cholinergic response. However, the adenylate cyclase activator, forskolin, and the cyclic AMP analogue, 8-cpt-cAMP, also stimulate ouabain-sensitive and bumetanide-sensitive O2 consumption in bo
