Effects of secretagogues on cytosolic Ca2+ levels in rat submandibular granular ducts and acini.

Saliva is thought to be formed by a two-stage process, with the secretion of a "primary fluid" by the acinar cells followed by various ionic modifications in the salivary ducts. Both of these processes are under the control of autonomic stimuli. Although the role of the acini in salivary secretion has been studied in some detail, little is known about properties of ducts, particularly the intralobular ducts that make up the bulk of the ductal tissue. In the present study, microfluorometric methods were employed to examine the responses of intracellular Ca2+ concentration ([Ca2+]i) in individua

Secretagogues increase apical membrane conductance of isolated bullfrog corneal epithelium pretreated with loop diuretics.

Bullfrog (Rana catesbeiana) corneas were mounted in an Ussing type chamber and impaled with an intracellular microelectrode and the short circuit current was inhibited by pretreatment with the loop diuretics furosemide (0.3 to 1 mM) or bumetanide (10 to 100 microM). Subsequent addition of the secretagogues prostaglandin E2, forskolin, or 3-isobutyl-1-methylxanthine (IBMX) caused the fractional voltage drop of the apical barrier to decrease from 0.72 +/- 0.05 to 0.48 +/- 0.04 and the chloride-dependent conductance to increase by 0.15 +/- 0.03 mS/cm2, but caused only a small, transient increase

Papain reduces gastric acid secretion induced by histamine and other secretagogues in anesthetized rats.

We studied the effect of papain on rats' gastric acid secretion and found that: 1. Feeding of latex of unripe papaya fruit significantly reduced gastric acid secretion induced by methacholine; 2. Feeding of crystalline papain in doses of 3.2 mg/kg reduced gastric acid secretion induced by histamine, methacholine and tetragastrin; 3. The reduction of gastric acid secretion was observed as early as 2 hours after papain feeding, lasted up to 48 hours, and waned within 96 hours; 4. Intraperitoneal injection of papain had no effect on acid secretion. These results led us to believe tha the effect o

Permeability of the peptidic GH secretagogues hexarelin and EP 51389, across rat jejunum.

The intestinal permeability of hexarelin and EP 51389, two growth hormone releasing hexa- and tri- peptide analogues, was assessed in vitro with side-by-side diffusion chambers in the apical-to-basolateral (AP-to-BL) and in the basolateral-to-apical (BL-to-AP) direction using excised rat jejunal segments. The effect of EP 51389 on P-glycoprotein (P-gp) was evaluated by rhodamine 123 accumulation on monolayers of CH(R)C5 cells with increasing concentrations of EP 51389. Hexarelin and EP 51389 permeability were found to be < 1%. Permeability coefficients (P(app)) were 18.87 +/- 2.86 (x10(-7) cm/

Restoring the mucin barrier: Advances in secretagogue therapies for ocular surface disorders.

Mucins are essential components of the ocular surface, ensuring epithelial protection, tear film stability, and immune regulation. Mucin deficiency contributes to a spectrum of ocular surface disorders, including dry eye disease, allergic conjunctivitis, and cicatricial conditions such as Stevens-Johnson syndrome and ocular graft-versus-host disease. This review outlines current knowledge of mucin biology, regulatory mechanisms, and emerging therapeutic applications of mucin secretagogues in ocular surface disorders. Mast cells contain large quantities of secretagogue-sensitive N-acetylaspartate.

Mast cells play a central role in both immediate allergic reactions and inflammation. A functional nerve-mast cell interaction has been proposed, given the morphological association between mast cells and neuropeptide-containing peripheral nerves. We now show that purified rat peritoneal mast cells contain large quantities of N-acetylaspartate (NAA; 747.50 nmol/mg of protein). Mast cell levels of NAA were rapidly reduced, by 64.0 and 86.4%, following treatment with compound 48/80 and mastoparan, respectively. These secretagogues strongly decreased mast cell histamine content over the same time

Arginine-vasopressin release mediates the aldosterone secretagogue effect of neurotensin in rats.

Acute and chronic systemic administrations of neurotensin (NT) and arginine-vasopressin (AVP) significantly increases plasma aldosterone concentration (PAC) in rats. Deamino-Pen1, Val4, D-Arg8-vasopressin (AVP-A), a potent AVP antagonist, completely reversed both acute and chronic aldosterone secretagogue actions of NT and AVP. AVP-A acute administration did not affect basal PAC, while chronic AVP-A treatment significantly lowered it. Taken together our findings suggest that both NT and AVP exert a marked aldosterone secretagogue effect in rats, and that the mechanism underlying NT action may

[Secretagogue action of glyceryl guaiacolate in tracheal submucosal glands (author's transl)].

The effects of glyceryl guaiacolate on secretory activities of tracheal secretory cells and on behavior of mucus glycoprotein in these cells were investigated histologically and histochemically using isolated canine trachea. Following glyceryl guaiacolate treatment, the number of total glycoprotein-containing goblet cells (GC) did not change. The numbers of acid glycoprotein (AGP)-, neutral glycoprotein (NGP)-, and sulphated glycoprotein (SGP)-containing GC were also unaltered in a concentration range of 10(-7) to 10(-4)M. On the other hand the acinar inner diameter of the submucosal gland (SG

Secretagogue stimulation of [14C]aminopyrine accumulation by isolated canine parietal cells.

Isolated canine gastric mucosal cells accumulate [14C]aminopyrine (AP) when treated with histamine, gastrin, and carbachol. In fractions of varying parietal cell content, this accumulation of AP correlated with the parietal cell content. Cimetidine caused parallel displacement of the dose-response curve to histamine, but failed to alter the response to carbachol or gastrin. Atropine caused parallel displacement of the dose-response curve to carbachol, but failed to inhibit the response to histamine or gastrin. The dissociation constants (Kb) for cimetidine inhibition of histamine and for atrop

Release of substance P by granuloma eosinophils in response to secretagogues in murine schistosomiasis mansoni.

In murine schistosomiasis, granulomas form around parasite ova which lodge in the liver and intestines. The granulomas contain eosinophils which produce substance P. In order to demonstrate substance P release by individual granuloma eosinophils and mechanisms regulating this release, a reverse hemolytic plaque assay was developed. Release of substance P was demonstrated by plaque formation around granuloma eosinophils only when a specific substance P antiserum was used. Few cells released substance P in the basal state. However, eosinophils produced plaques in the presence of calcium ionophor

Cytotoxicity of ricinoleic acid (castor oil) and other intestinal secretagogues on isolated intestinal epithelial cells.

Epithelial cells were isolated from hamster small intestine by a technique of vibration and used to measure cytotoxicity in vitro of certain substances known to stimulate intestinal fluid secretion. These secretagogues have laxative properties and produce mucosal damage in vivo. Compounds tested were ricinoleic acid (caster oil), dioctyl sodium sulfosuccinate, oleic acid, sodium deoxycholate and sodium cholate. Cytotoxicity was assessed by: 1) exclusion of trypan blue; 2) release of intracellular (prelabeled) 51Cr; and 3) inhibition of cellular uptake of 3-O-methylglucose. Ricinoleate produced

Short-term effects of secretagogues on the mouse parotid and sublingual gland tissular water.

The effects of isoproterenol and pilocarpine on the tissular water of mouse parotid and sublingual glands were studied by thermogravimetry (TG) coupled with Fourier transform infrared spectroscopy (FTIR). Results demonstrated that the short-term effects partly resemble the long-term action of these substances on the above-mentioned organs. Due to the different stages examined, it is difficult to speculate on the comparison between previous and the present findings. The original data resulting from the present research corresponded to the dimorphism expressed in the thermoanalytical profile of

Gastric protective effects of gastric secretagogues on 0.6N HCl-induced gastric lesions in rats.

The effects of gastric secretagogues on 0.6N HCl-induced gastric lesions and gastric mucosal prostaglandin E2 (PGE2) contents were investigated in rats. Secretagogues such as histamine (Hist) and amogastrin (Gast) significantly inhibited the formation of gastric lesions induced by 0.6N HCl. The time course of the gastric protective effect of these secretagogues paralleled the increase of gastric acid secretion. This increase was due to the increase in acidity, not to the volume of the gastric juice. The gastric protective effects of Hist and Gast were inhibited by pretreatment with cimetidine,

Cytomorphological study on human submandibular gland following treatment with secretagogue drugs.

Using specimens of human submandibular glands, we have investigated in vitro the morphological modifications induced by clozapine, a dibenzodiazepine derivative that is used in psychotic patients and that provokes hypersalivation, a side-effect of therapy. The effects of the drug, used alone or in combination with carbachol, have been compared with those observed after treatment with drugs acting on specific receptors. To quantify the response to stimulation, we have calculated (with statistical methods) the number of microvilli and microbuds (corresponding to pits seen in images obtained by t

Secretagogue-induced [14C]aminopyrine uptake in isolated equine parietal cells.

Equine oxyntic mucosal cells were obtained by sequential exposure to pronase and collagenase. Acid production by parietal cells was assessed by uptake of [14C]aminopyrine (AP), a weak base that accumulates in intracellular acidic spaces. Incubation for various times revealed a maximal AP uptake at 10 minutes for histamine and carbachol. Similar secretagogue responses were observed for parietal cells from the mucosal cell preparation or after enrichment by elutriation. Histamine and isobutyl-methylxanthine (IBMX) stimulated AP uptake with a dose-dependent response and maximal effective concentr

Effect of aging on the sensitivity of pancreatic beta-cells to insulin secretagogues in rats.

Glucose-stimulated insulin release from rat pancreas is known to be blunted by aging. In the present study, we examined the effect of aging on insulin release induced by various secretagogues using the isolated perfused pancreas of female rats. Insulin release from the perfused pancreas in response to 16.7 mM glucose in 8-month-old rats (older rats) was much less than that in 2-month-old rats (young rats). The first phase of insulin release after glucose stimulation was attenuated in older rats. The addition of 0.1 mM 3-isobutyl-1-methylxanthine (IBMX) potentiated glucose-induced insulin secre

Growth hormone secretagogues modulate inflammation and fibrosis in mdx mouse model of Duchenne muscular dystrophy.

INTRODUCTION: Growth hormone secretagogues (GHSs) exert multiple actions, being able to activate GHS-receptor 1a, control inflammation and metabolism, to enhance GH/insulin-like growth factor-1 (IGF-1)-mediated myogenesis, and to inhibit angiotensin-converting enzyme. These mechanisms are of interest for potentially targeting multiple steps of pathogenic cascade in Duchenne muscular dystrophy (DMD). METHODS: Here, we aimed to provide preclinical evidence for potential benefits of GHSs in DMD, via a multidisciplinary in vivo and ex vivo comparison in mdx mice, of two ad hoc synthesized compoun
