Activity-guided native-PAGE electroelution coupled with DIA proteomics uncovers the superoxide dismutase and antioxidant network in mature peanut seeds.

Mature peanut (Arachis hypogaea L.) seeds accumulate abundant superoxide dismutase (SOD), but the high abundance of storage proteins masks trace antioxidant enzymes and prevents accurate profiling of endogenous SOD isoforms by conventional proteomics. Here we report an activity-guided workflow that combines native-PAGE NBT zymography, low-temperature dialysis-bag electroelution and DIA-based nano-LC-MS/MS to enrich native SOD complexes from seed crude extracts. The two SOD-active gel bands, B1 and B2, were excised, embedded in agarose and electroeluted to recover intact proteins before proteom

From grain to malt: Tracking changes of ultra-low-gluten barley storage proteins after malting.

Barley (Hordeum vulgare L.) is a major cereal crop produced globally. Hordeins, the major storage proteins in barley, can trigger immune responses leading to celiac disease or symptoms associated with food allergy. Here, proteomics approaches were employed to investigate the proteome level changes of grain and malt from the malting barley cultivar, Sloop, and single-, double- and triple hordein-reduced lines. The triple hordein-reduced line is an ultra-low gluten barley cultivar, Kebari®. Using discovery proteomics, 2,688 and 3,034 proteins in the barley and malt samples were detected respecti

Cross-reactivity in cashew nut allergy.

BACKGROUND: Cashew nut allergy is increasing globally and may cause cross-allergic reactions to seed kernels from botanically related plants, including citrus. CASE PRESENTATION: A woman in her twenties with a known nut allergy developed anaphylaxis after consuming a sports drink containing citrus-derived pectin. Skin prick testing revealed reactions to the sports drink (6 mm), lemon seed (14 mm), citrus pectin (6 mm), cashew (16 mm) and pistachio (11 mm). Serum-specific IgE testing confirmed sensitisation to storage proteins in cashew and other nuts, including Ana o 3, Cor a 9, Cor a 14, Jug

New insights into the role of cyanide in the promotion of seed germination in tomato.

BACKGROUND: Cyanide is a natural metabolite that exists widely in plants, and it is speculated to be involved in the regulation of various growth and development processes of plants in addition to being regarded as toxic waste. Previous studies have shown that exogenous cyanide treatment helps to improve seed germination, but the mechanism is still unclear. In this study, tomato (Solanum lycopersicum cv. Alisa Craig) was used as the material, and the effects of cyanide pretreatment at different concentrations on tomato seed germination were investigated. RESULTS: The results showed that exoge

Advancing allergen characterization in Perilla seed: an oil body purification approach.

Perilla seed (PS), a common food in East Asia, has emerged as a leading cause of seed-induced anaphylaxis in Korean children, yet its allergenic proteins remain unidentified. Oleosins, hydrophobic oil-body (OB)-associated storage proteins, are recognized allergens in peanuts, hazelnuts, and sesame, and preliminary evidence suggests a similar role in PS. However, the intrinsic challenges of extracting lipophilic proteins have hindered systematic investigation. This study aimed to purify PS OBs, isolate oleosin under optimized conditions, and characterize candidate major allergens. Commercial Ko

Characterization of β-amino- and γ-amino butyric acid-induced citrus seeds germination under salinity using nanoLC-MS/MS analysis.

BABA or GABA induces salinity acclimation during citrus seeds germination via alternation of specific proteins (e.g., citrin). The impact of four elicitors, namely hydrogen peroxide (H2O2), β-amino butyric acid (BABA), γ-amino butyric acid (GABA) and hydrogen sulfide (H2S) donor, sodium hydrosulfide (NaHS), in citrus seed germination under salinity (150 mM NaCl) was tested. The germination potential was adversely affected by NaCl-alone treatment. Pretreatment with H2O2 or the NaHS-H2S donor prior to salinity had no significant effect in germination process, however, BABA and GABA substantially

Transcriptional coordination enhances folate biosynthesis in dry common bean grains during storage.

Postharvest storage critically influences seed physiology and nutritional quality. Common beans (Phaseolus vulgaris) are a rich source of folates, vitamins that are typically unstable and degrade during storage. We investigated three varieties (from Mesoamerican and Andean origins) over 180 days under different conditions and found significant accumulation of folates and their more stable polyglutamylated derivatives. Transcriptomic analysis under controlled storage (15 °C, 55% relative humidity) revealed consistent upregulation of genes catalyzing key steps in folate biosynthesis (GTP cyclohy

New protocol for kinetic assay seeding ability recovery "KASAR" from formalin-fixed paraffin-embedded tissues.

The real-time quaking-induced conversion (RT-QuIC) alpha-synuclein (aSyn) protein kinetic seeding assay has been very useful for detecting pathological aggregates in various synucleinopathies including Parkinson's disease (PD). This biomarker assay relies on fresh frozen tissue to effectively seed and amplify aSyn aggregating protein. With vast repositories of formalin-fixed paraffin-embedded (FFPE) tissues, it is paramount to harness the power of kinetic assays to unlock the diagnostic potential of archived FFPE biospecimens. However, the major challenge posed by significantly reduced amplifi

Vacuolar processing enzyme is up-regulated in the lytic vacuoles of vegetative tissues during senescence and under various stressed conditions.

Vacuolar processing enzyme (VPE) has been shown to be responsible for maturation of various seed proteins in protein-storage vacuoles. Arabidopsis has three VPE homologues; betaVPE is specific to seeds and alphaVPE and gammaVPE are specific to vegetative organs. To investigate the activity of the vegetative VPE, we expressed the gammaVPE in a pep4 strain of the yeast Saccharomyces cerevisiae and found that gammaVPE has the ability to cleave the peptide bond at the carbonyl side of asparagine residues. An immunocytochemical analysis revealed the specific localization of the gammaVPE in the lyti

Beyond Mixed Utilization: Fractionation, Functional Divergence, and Precision Applications of Soybean 7S and 11S Globulins.

Soybean β-conglycinin (7S) and glycinin (11S) differ markedly in their structure, physicochemical properties, and bioactivity; however, industrial practice still treats them as a mixed ingredient, obscuring their differentiated functional potential. This review critically evaluates fractionation technologies through the lens of the "purity-yield-sustainability" trilemma. First-generation chemical precipitation methods achieve high purity at the laboratory scale but suffer from heavy reagent use, environmental burden, and poor scalability. Second-generation green and physical techniques, such a

Soy protein amyloid fibrils for the design of innovative soft matter systems.

The self-assembly of soy proteins into amyloid fibrils (SAFs) has emerged as an effective strategy for engineering advanced amyloid-based soft matter systems. Under controlled acidic and thermal conditions, soy proteins undergo structural reorganization into β-sheet-rich fibrillar architectures with high aspect ratios, enhanced mechanical properties, and tunable interfacial properties. The compositional complexity of soy proteins, particularly their β-conglycinin (7S) and glycinin (11S) fractions, can contribute to the formation of structurally distinct fibrils with diverse morphologies and fu

ROS production and protein oxidation as a novel mechanism for seed dormancy alleviation.

At harvest, sunflower (Helianthus annuus L.) seeds are dormant and unable to germinate at temperatures below 15 degrees C. Seed storage in the dry state, known as after-ripening, is associated with an alleviation of embryonic dormancy allowing subsequent germination at suboptimal temperatures. To identify the process by which dormancy is broken during after-ripening, we focused on the role of reactive oxygen species (ROS) in this phenomenon. After-ripening entailed a progressive accumulation of ROS, namely superoxide anions and hydrogen peroxide, in cells of embryonic axes. This accumulation,

lpa1-5525: A New lpa1 Mutant Isolated in a Mutagenized Population by a Novel Non-Disrupting Screening Method.

Phytic acid, or myo-inositol 1,2,3,4,5,6-hexakisphosphate, is the main storage form of phosphorus in plants. It is localized in seeds, deposited as mixed salts of mineral cations in protein storage vacuoles; during germination, it is hydrolyzed by phytases to make available P together with all the other cations needed for seed germination. When seeds are used as food or feed, phytic acid and the bound cations are poorly bioavailable for human and monogastric livestock due to their lack of phytase activity. Therefore, reducing the amount of phytic acid is one strategy in breeding programs aimed

Minerals and chosen heavy metals retention in immature common bean (Phaseolus vulgaris L.) seeds depending on the method of preservation.

BACKGROUND: Legumes are a good source of protein, and are also abundant in carbohydrates, B-group vita-mins, dietary fibre and mineral compounds. MATERIAL AND METHODS: This work evaluates the retention of ash, eleven minerals and two heavy metals in products obtained from two common bean cultivars harvested before reaching full maturity, with a dry matter content of about 40%. Analyses were conducted on raw, blanched and cooked seeds and three products prepared for consumption after 12-month storage: two frozen and one canned (sterilized). The former comprised two types of frozen product: one

Metabolic profiling of the Arabidopsis pkl mutant reveals selective derepression of embryonic traits.

Embryos express several unique differentiation characteristics, including the accumulation of a number of metabolites that are generally considered to be unique to seeds. PICKLE (PKL) codes for a CHD3-chromatin remodeling factor that is necessary for repression of embryonic traits in seedlings of Arabidopsis thaliana (L.) Heynh. In pkl mutants, primary roots are capable of expressing many embryonic traits after germination and are referred to as "pickle roots". In an attempt to examine the breadth of PKL-dependent repression of embryo-specific differentiation pathways, we determined the extent

Time-Dependent Proteomic Signatures Associated with Embryogenic Callus Induction in Carica papaya L.

Sex segregation increases the cost of Carica papaya production through seed-based propagation. Therefore, in vitro techniques are an attractive option for clonal propagation, especially of hermaphroditic plants. Here, we performed a temporal analysis of the proteome of C. papaya calli aiming to identify the key players involved in embryogenic callus formation. Mature zygotic embryos used as explants were treated with 20 μM 2,4-dichlorophenoxyacetic acid to induce embryogenic callus. Total proteins were extracted from explants at 0 (zygotic embryo) and after 7, 14, and 21 days of induction. A t

Relationship between Protein Digestibility and the Proteolysis of Legume Proteins during Seed Germination.

Legume seed protein is an important source of nutrition, but generally it is less digestible than animal protein. Poor protein digestibility in legume seeds and seedlings may partly reflect defenses against herbivores. Protein changes during germination typically increase proteolysis and digestibility, by lowering the levels of anti-nutrient protease inhibitors, activating proteases, and breaking down storage proteins (including allergens). Germinating legume sprouts also show striking increases in free amino acids (especially asparagine), but their roles in host defense or other processes are

Proteomic Profiling of Celiac-Toxic Motifs and Allergens in Cereals Containing Gluten.

Cereal-based foods can cause immune-mediated adverse reactions, including celiac disease and IgE-mediated allergies, but the potency of different cereal species to cause such reactions appears to vary, with oats being less celiac-toxic and allergenic than wheat. In order to define differences in the immunological potential of wheat, barley, rye, and oats, proteomic profiling of proteins carrying celiac-toxic motifs and allergens has been undertaken. Total protein extracts were subjected to chymotryptic digestion and analyzed using data-independent ion mobility mass spectrometry and a pipeline

Optimal Preservation of PFFs in Glycerol Enhances Suitability for Modeling Parkinson's Disease.

Injecting α-synuclein pre-formed fibrils (αSyn PFFs) into various tissues and organs involves converting monomeric αSyn into a fibrillar form, inducing extensive αSyn pathology that effectively models Parkinson's disease (PD). However, the distinct physicochemical properties of αSyn amyloid fibrils can potentially reduce their seeding activity, especially during storage. In this study, it is demonstrated that αSyn PFFs exhibit significant sensitivity to low temperatures, with notable denaturation occurring between -20 and 4 °C, and gradual disassembly persisted even under storage conditions at

Development of low-prolamin rice germplasm via CRISPR/Cas9 editing to improve eating and cooking quality.

Simultaneous editing of two major prolamin-encoding genes in rice using a single-guide RNA suppressed prolamin accumulation and reconfigured seed storage protein composition. This targeted modification markedly improved rice eating and cooking quality while maintaining the levels of total starch, total protein, and key agronomic traits.
