Contextualizing selection bias in Mendelian randomization: how bad is it likely to be?

BACKGROUND: Selection bias affects Mendelian randomization investigations when selection into the study sample depends on a collider between the genetic variant and confounders of the risk factor-outcome association. However, the relative importance of selection bias for Mendelian randomization compared with other potential biases is unclear. METHODS: We performed an extensive simulation study to assess the impact of selection bias on a typical Mendelian randomization investigation. We considered inverse probability weighting as a potential method for reducing selection bias. Finally, we inve

Non-recruitment to and selection bias in studies using echocardiography in haemodialysis patients.

BACKGROUND: It is unknown whether patients recruited to renal cardiac imaging studies are fully representative of the investigated population and whether there are differences in characteristics and survival between participants and non-participants (excluded or declined consent). Subjects and Methods Four hundred thirty-five maintenance haemodialysis patients were screened in an observational, prospective study. Baseline demographics, laboratory results, social deprivation scores and survival data were collected from patient records. All patients were followed-up until death, renal transplant

Contralateral prophylactic mastectomy and its association with reduced mortality: evidence for selection bias.

Contralateral prophylactic mastectomy (CPM) refers to removal of the opposite uninvolved breast in women with unilateral breast cancer, and rates are increasing worldwide. In observational studies, CPM is often associated with reductions in breast cancer-specific and all-cause mortality when compared to unilateral surgical treatment alone, but this may reflect selection of a healthier cohort for CPM (selection bias). To further explore this possibility, we examined the association between CPM and non-cancer mortality, an indicator of selection bias. We identified 449,178 adult women diagnosed

Cold-selective topological bias and the emergence of the first membranes.

The emergence of the first membranes requires more than the synthesis of amphiphiles; it requires a mechanism that stabilizes bilayer-forming geometries within chemically diverse prebiotic mixtures. We present a minimal thermodynamic model coupling amphiphile topology to self-assembly, incorporating packing, curvature, and steric contributions. Changes in temperature alter the balance between ordering and disorder within amphiphile aggregates, thereby modifying the relative stability of lamellar, micellar, and dispersed states. The model identifies a bounded temperature regime in which geometr

IUPHAR themed review: Opioid efficacy, bias, and selectivity.

Drugs acting at the opioid receptor family are clinically used to treat chronic and acute pain, though they represent the second line of treatment behind GABA analogs, antidepressants and SSRI's. Within the opioid family mu and kappa opioid receptor are commonly targeted. However, activation of the mu opioid receptor has side effects of constipation, tolerance, dependence, euphoria, and respiratory depression; activation of the kappa opioid receptor leads to dysphoria and sedation. The side effects of mu opioid receptor activation have led to mu receptor drugs being widely abused with great ov

Percutaneous vs. surgical management in acute cholecystitis: addressing selection bias in meta-analyses.

This commentary addresses the recent research titled "Comparing percutaneous treatment and cholecystectomy outcomes in acute cholecystitis patients: a systematic review and meta-analysis" published in the World Journal of Emergency Surgery. The review confirms that cholecystectomy (CC), particularly laparoscopic, is associated with lower mortality and readmission rates compared to percutaneous cholecystostomy (PC). However, this commentary emphasizes that the interpretation of these findings must account for inherent selection bias: in clinical practice, PC is typically reserved for higher-ris

Antithrombotic Drug Eruptions in Dermatology Practice: Selection Bias, Clinical Phenotypes, and Diagnostic Approaches.

Antithrombotic agents are among the most frequently prescribed medications in elderly patients and are essential for the prevention and treatment of cardiovascular and cerebrovascular diseases. Although cutaneous adverse reactions associated with anticoagulants and antiplatelet agents are generally considered uncommon, dermatologists frequently encounter these medications when evaluating patients presenting with pruritic eruptions. This apparent discrepancy may be explained by differences in patient populations. While cardiologists and neurosurgeons manage all patients receiving antithrombotic

How Medical School Experiences Bias Specialty Selection: Perspectives From Current U.S. Otolaryngology Residents.

OBJECTIVES: This study examined the medical school experiences of current U.S. otolaryngology residents to better understand gender biases that may persist within the specialty and to inform strategies that could improve workforce excellence. METHODS: A voluntary, IRB-approved survey was distributed to U.S. otolaryngology residency program directors and coordinators for distribution to their residents. Gender-stratified analyses were performed using Chi-squared tests, Mann-Whitney tests, and thematic analysis. A P-value < .05 was considered statistically significant. RESULTS: Of 137 complete

Selection bias in microbial mutation accumulation studies and the impact of colony growth.

In microbial mutation-accumulation (MA) studies, it is widely thought that natural selection is silenced by repeated single-cell bottlenecks. In support of this claim, 40 published tests for selection bias failed to detect a deficit of non-synonymous relative to synonymous mutations in wild-type microbes. Here we show that this most likely reflects selective reporting and lack of power. Our meta-analysis of 10,856 mutations from wild-type microbial MA reveals a clear signal of selection: non-synonymous mutations are observed 7.7\% less often than synonymous mutations. However, our inference of

Minimizing Selection Bias in Inclusive Jets in Heavy-Ion Collisions with Energy Correlators.

The first-ever measurement of energy correlators within inclusive jets produced in heavy-ion collisions, revealed by the CMS Collaboration, shows a clear enhancement at large angles relative to the proton-proton (p-p) baseline. However, interpreting this enhancement is complicated due to selection bias from energy loss, which also distorts the energy correlator heavy-ion to p-p ratio in the hadronization region, hindering our understanding of parton/hadron dynamics in a colored medium. In this Letter, we introduce a new ratio of energy correlator observables that removes the leading effects of

Sex and smoking bias in the selection of somatic mutations in human bladder.

Men are at higher risk of several cancer types than women1. For bladder cancer the risk is four times higher for reasons that are not clear2. Smoking is also a principal risk factor for several tumour types, including bladder cancer3. As tumourigenesis is driven by somatic mutations, we wondered whether the landscape of clones in the normal bladder differs by sex and smoking history. Using ultradeep duplex DNA sequencing (approximately 5,000×), we identified thousands of clonal driver mutations in 16 genes across 79 normal bladder samples from 45 people. Men had significantly more truncating d

Assessing and attenuating the impact of selection bias on spatial cluster detection studies.

Spatial cluster analyses are commonly used in epidemiologic studies of case-control data to detect whether certain areas in a study region have an excess of disease risk. Case-control studies are susceptible to potential biases including selection bias, which can result from non-participation of eligible subjects in the study. However, there has been no systematic evaluation of the effects of non-participation on the findings of spatial cluster analyses. In this paper, we perform a simulation study assessing the effect of non-participation on spatial cluster analysis using the local spatial sc

Bias in Radiology Resident Selection: Do We Discriminate Against the Obese and Unattractive?

PURPOSE: To evaluate for appearance-based discrimination in the selection of radiology residents. METHOD: A deception study simulating the resident selection process examined the impact of attractiveness and obesity on resident selection. Seventy-four core faculty from 5 academic radiology departments reviewed mock residency applications in September and October 2017. Each application included demographic information and a photograph, representing a prespecified distribution of facial attractiveness and obesity, combined with randomized academic and supporting variables. Reviewers independent

Preoperative Breast MRI: Surgeons' Patient Selection Patterns and Potential Bias in Outcomes Analyses.

OBJECTIVE: The purpose of this study is to determine which patient- and tumor-related and clinical variables influence dedicated breast surgeons' and general surgeons' referrals for preoperative breast MRI for patients with newly diagnosed breast cancer. MATERIALS AND METHODS: Surgeons who perform breast surgery responded to a survey from June 16, 2014, through August 11, 2014. Participants self-identified as breast or general surgeons and provided professional practice details. They used Likert scores (range, 1-7 with increasing likelihood to order MRI) to weigh numerous patient- and tumor-r

The BioMedical Admissions Test for medical student selection: issues of fairness and bias.

BACKGROUND: The BioMedical Admissions Test (BMAT) forms part of the undergraduate medical admission process at the University of Cambridge. The fairness of admissions tests is an important issue. AIMS: Aims were to investigate the relationships between applicants' background variables and BMAT scores, whether they were offered a place or rejected and, for those admitted, performance on the first year course examinations. METHOD: Multilevel regression models were employed with data from three combined applicant cohorts. Admission rates for different groups were investigated with and without c

Physical activity and premenopausal breast cancer: an examination of recall and selection bias.

BACKGROUND: Compared with cohort studies, case-control investigations have tended to report clearer protective associations for the relationship between physical activity and premenopausal breast cancer risk. METHODS: We conducted a case-control study within the Nurses' Health Study II cohort to examine whether recall or selection bias could explain the stronger protective associations. Self-reported total recreational physical activity during adulthood and over a woman's lifetime (ages 12 years to current) were assessed in 1997 before diagnosis and, again, from one to seven years after breas
