Selenium-sensitive miRNA-181a-5p targeting SBP2 regulates selenoproteins expression in cartilage.

Selenium (Se) deficiency brings about defects in the biosynthesis of several selenoproteins and has been associated with aberrant chondrogenesis. Selenocysteine (Sec) Insertion Sequence (SECIS) and SECIS binding protein 2 (SBP2) interaction is a very critical node for the metabolic balance between Se and selenoproteins. The Gpx1, Gpx4 and SelS have different binding affinities with SBP2 in cells. According to our results, both miR-181a-5p and SBP2 appeared to be selenium-sensitive and regulated the expression of selenoproteins in C28/I2 cells under Se sufficient environment. However, they show…

New insights into the physiological actions of selenoproteins from genetically modified mice.

Selenium (Se) is an essential trace element in mammals. Dietary Se restriction or conditions of Se malabsorption lead to deficiency syndromes or exacerbate established diseases in humans and in many animal models. It is assumed that most, if not all, physiological actions of Se are mediated by selenocysteine (Sec) containing proteins. However, the exact role of particular selenoproteins for certain molecular pathways, for the metabolism of nutrients, hormones or cellular components and for the development and adaptive responses of the organism have often remained elusive. Through the use of tr…

Selenoprotein levels in patients with colorectal adenomas and cancer.

OBJECTIVES: Selenium is a trace mineral that, as a constituent of certain selenoproteins, acts as an antioxidant. Results of studies addressing a cancer protective effect of selenium have been controversial. The present study measured selenoprotein-P, extracellular glutathione peroxidase, and plasma selenium in patients with colon cancer and adenomatous colon polyps to determine whether patients who develop colorectal adenomas or cancer are selenium deficient. METHODS: Patients who presented to an endoscopy center for colonoscopy or who were referred to our institution with a newly diagnosed …

The Regulation of Selenoproteins in Diabetes: A New Way to Treat Diabetes.

Selenium is an essential micronutrient required for the synthesis and function of selenoproteins, most of which are enzymes involved in maintaining oxidative balance in the body. Diabetes is a group of metabolic disorders characterized by high blood glucose levels over a prolonged period of time. There are three main types of diabetes: type 1, type 2, and gestational diabetes. This review summarizes recent advances in the field of diabetes research with an emphasis on the roles of selenoproteins on metabolic disturbance in diabetes. We also discuss the interaction between selenoproteins and gl…

The role of selenium metabolism and selenoproteins in cartilage homeostasis and arthropathies.

As an essential nutrient and trace element, selenium is required for living organisms and its beneficial roles in human health have been well recognized. The role of selenium is mainly played through selenoproteins synthesized by the selenium metabolic system. Selenoproteins have a wide range of cellular functions including regulation of selenium transport, thyroid hormones, immunity, and redox homeostasis. Selenium deficiency contributes to various diseases, such as cardiovascular disease, cancer, liver disease, and arthropathy-Kashin-Beck disease (KBD) and osteoarthritis (OA). A skeletal dev…

Selenium and selenoproteins in age-related metabolic diseases: mechanistic insights and translational perspectives.

Selenium is an essential trace element incorporated into selenoproteins, which regulate redox balance, thyroid hormone metabolism, protein folding, and stress signaling. The relationship between selenium and metabolic health is complex, with growing evidence of a U-shaped association. This review synthesizes current evidence on selenium and selenoproteins in age-related metabolic diseases, with a focus on MASLD, obesity, and cardiovascular disease. Mechanistic data from experimental models are integrated with human observational and intervention studies to evaluate how selenium status, chemica…

Comprehensive Analysis of Selenium Metabolism and Selenoproteins-Associated Gene Signatures in Ulcerative Colitis.

BACKGROUND: Ulcerative colitis (UC) is a chronic inflammatory bowel disease marked by persistent mucosal inflammation. Oxidative stress plays a vital role in UC pathogenesis. Selenium (Se), an essential trace element, functions via selenoproteins and metabolites. Selenocysteine (Sec), the 21st amino acid, is incorporated into selenoproteins with strong antioxidant and anti-inflammatory properties. However, the role of selenium metabolism and selenoproteins (SeMet) in UC remains poorly understood. METHODS: Gene expression and clinical data from UC patients and healthy controls were obtained fr…

A positive effect of selenoprotein on mammary gland: Selenoprotein P stabilizes tight junctions by reducing cell death through inflammation mitigation in mice.

Lactational mastitis threatens maternal-infant health and animal husbandry efficiency, with traditional antibiotics limited by milk residues and bacterial resistance. Selenium (Se) regulates inflammation via selenoproteins, but its core effector in mammary inflammation remains unclear. This study investigates the role and mechanism of Se and selenoprotein P (SeP) in LPS-induced mastitis. Tissue pathology, inflammation, apoptosis, and tight junctions (TJs) are assessed via H&E staining, qPCR, Western blot, etc., in mice with varying Se diets (deficient, basal, enriched) subjected to LPS-induced…

Novel selenoprotein neighborhoods suggest specialized biochemical processes.

UNLABELLED: Prokaryotic genomes encode selenoproteins sparsely, roughly one protein per 5,000. Finding novel selenoprotein families can expose unknown biological processes that are enabled, or at least enhanced, by having a selenium atom replace a sulfur atom in some cysteine residues. Here, we report the discovery of 18 novel selenoprotein families or second selenocysteine sites in previously unrecognized extensions of protein translations. Most of these families had some confounding factors-too small a family, too few selenoproteins in the family, selenocysteine (U) too close to one end, a s…

The Effect of tRNA[Ser]Sec Isopentenylation on Selenoprotein Expression.

Transfer RNA[Ser]Sec carries multiple post-transcriptional modifications. The A37G mutation in tRNA[Ser]Sec abrogates isopentenylation of base 37 and has a profound effect on selenoprotein expression in mice. Patients with a homozygous pathogenic p.R323Q variant in tRNA-isopentenyl-transferase (TRIT1) show a severe neurological disorder, and hence we wondered whether selenoprotein expression was impaired. Patient fibroblasts with the homozygous p.R323Q variant did not show a general decrease in selenoprotein expression. However, recombinant human TRIT1R323Q had significantly diminished activit

Human Genetic Disorders Resulting in Systemic Selenoprotein Deficiency.

Selenium, a trace element fundamental to human health, is incorporated as the amino acid selenocysteine (Sec) into more than 25 proteins, referred to as selenoproteins. Human mutations in SECISBP2, SEPSECS and TRU-TCA1-1, three genes essential in the selenocysteine incorporation pathway, affect the expression of most if not all selenoproteins. Systemic selenoprotein deficiency results in a complex, multifactorial disorder, reflecting loss of selenoprotein function in specific tissues and/or long-term impaired selenoenzyme-mediated defence against oxidative and endoplasmic reticulum stress. SEP…

Selenium and selenoprotein P in nonalcoholic fatty liver disease.

Conflicting data link nonalcoholic fatty liver disease (NAFLD), a disease with no currently approved treatment, with selenium (Se) and selenoprotein P (SELENOP), a glycoprotein synthesized and primarily secreted by the hepatocytes, functioning as a Se transporter from the liver to other tissues. This review aims to summarize the evidence between Se, SELENOP, and NAFLD, which may hopefully clarify whether current data on Se and SELENOP in NAFLD warrant further investigation for their diagnostic and therapeutic potential. Most, albeit not all, experimental data show a favorable effect of Se on h

Consequences of mutations and inborn errors of selenoprotein biosynthesis and functions.

In its 200 years of history, selenium has been defined first as a toxic element and finally as a micronutrient. Selenium is incorporated into selenoproteins as selenocysteine (Sec), the 21st proteinogenic amino acid codified by a stop codon. Specific biosynthetic factors recode UGA stop codon as Sec. The significance of selenoproteins in human health is manifested through the identification of patients with inborn errors in selenoproteins or their biosynthetic factors. Selenoprotein N-related myopathy was the first disease identified due to mutations in a selenoprotein gene. Mutations in GPX4 …

A spatial ecological study of selenoprotein P and Keshan disease.

Few spatial ecological studies on selenoprotein P (SePP) and Keshan disease (KD) have been reported. The main objective of this study is to investigate the relationships of SePP with KD, economic indicators and soil selenium and to visualize the evidence for KD precise prevention and control. An ecological study design was employed. The serum SePP of 2351 subjects living in rural areas, general cities and developed cities in 15 KD endemic provinces and 13 KD non-endemic provinces in China were measured. Spatial description and spatial analysis of SePP were conducted. The subjects were adults a

Identification of Genetic Disorders Causing Disruption of Selenoprotein Biosynthesis.

Disorders of selenoprotein biosynthesis in humans, due to mutations in three genes (SECISBP2, TRU-TCA1-1, and SEPSECS) involved in the selenocysteine insertion pathway, have been described. Patients with SECISBP2 and TRU-TCA1-1 defects manifest a multisystem disorder with a biochemical signature of abnormal thyroid function tests due to the impaired activity of deiodinase selenoenzymes, myopathic features linked to SEPN1 deficiency and phenotypes resulting from increased levels of reactive oxygen species attributable to lack of antioxidant selenoenzymes. In patients harboring SEPSECS mutations

Aminoglycoside-driven biosynthesis of selenium-deficient Selenoprotein P.

Selenoprotein biosynthesis relies on the co-translational insertion of selenocysteine in response to UGA codons. Aminoglycoside antibiotics interfere with ribosomal function and may cause codon misreading. We hypothesized that biosynthesis of the selenium (Se) transporter selenoprotein P (SELENOP) is particularly sensitive to antibiotics due to its ten in frame UGA codons. As liver regulates Se metabolism, we tested the aminoglycosides G418 and gentamicin in hepatoma cell lines (HepG2, Hep3B and Hepa1-6) and in experimental mice. In vitro, SELENOP levels increased strongly in response to G418,

Nonsense-mediated decay: assaying for effects on selenoprotein mRNAs.

If the abundance of a particular selenoprotein mRNA is reduced during selenium deprivation, then the mRNA is likely to be a natural substrate for NMD. One assay for NMD involves changing the TGA Sec codon(s) to either a TGC cysteine codon or a TAA nonsense codon. If selenium deprivation elicits NMD and has no other effect on selenoprotein gene expression, then, regardless of selenium concentration, the level of UGC-containing mRNA should be most abundant, the level of UGA-containing mRNA should be intermediate in abundance, and the level of UAA-containing mRNA should be least abundant. Binding sites for the (Hg-Se) complex on selenoprotein P.

The mechanism underlying the interaction between mercury (Hg), selenium (Se) and selenoprotein P (Sel P) in the bloodstream has been explained by the formation of the [(Hg-Se)n]m-Sel P complex. In the present study, the binding sites for the (Hg-Se)n complex on Sel P were studied by competitive assay of the binding of the (Hg-Se)n complex to Sel P with polymeric and monomeric amino acids with simultaneous detection of the Hg, Se of selenite origin and Se of Sel P origin by the high performance liquid chromatography-inductively coupled argon plasma-mass spectrometry method.
