Cellular Senescence in Diabetes Mellitus: Distinct Senotherapeutic Strategies for Adipose Tissue and Pancreatic β Cells.

Increased insulin resistance and impaired insulin secretion are significant characteristics manifested by patients with type 2 diabetes mellitus (T2DM). The degree and extent of these two features in T2DM vary among races and individuals. Insulin resistance is accelerated by obesity and is accompanied by accumulation of dysfunctional adipose tissues. In addition, dysfunction of pancreatic β-cells impairs insulin secretion. T2DM is significantly affected by aging, as the β-cell mass diminishes with age. Moreover, both obesity and hyperglycemia-related metabolic changes in developing diabetes ar

Exploring the Relevance of Senotherapeutics for the Current SARS-CoV-2 Emergency and Similar Future Global Health Threats.

The higher death rate caused by COVID-19 in older people, especially those with comorbidities, is a challenge for biomedical aging research. Here we explore the idea that an exacerbated inflammatory response, in particular that mediated by IL-6, may drive the deleterious consequences of the infection. Data shows that other RNA viruses, such as influenza virus, can display enhanced replication efficiency in senescent cells, suggesting that the accumulation of senescent cells with aging and age-related diseases may play a role in this phenomenon. However, at present, we are completely unaware of

Multiomics and cellular senescence profiling of aging human skeletal muscle uncovers Maraviroc as a senotherapeutic approach for sarcopenia.

Cellular senescence is a hallmark of organismal aging but how it drives aging in human tissues is not fully understood. Here we leverage single nucleus multiomics to profile senescence in mononucleated cells of human skeletal muscle and provide the first senescence atlas. We demonstrate the intra- and inter-populational transcriptomic and epigenomic heterogeneity and dynamics of cellular senescence. We also identify commonalities and variations in senescence-associated secretory phenotypes (SASPs) among the cells and elucidate SASP mediated cellular interactions and niche deregulation. Further

Homoharringtonine exhibits senotherapeutic activity that mitigates diet- and age-associated obesity and insulin resistance and extends lifespan in mice.

The accumulation of senescent cells in white adipose tissue (WAT) is closely associated with the functional decline of WAT and plays a causal role in the pathogenesis of metabolic diseases. Therefore, the elimination of senescent cells in WAT holds promise for the treatment and prevention of age-related metabolic diseases. Using a drug-repositioning strategy for 2150 clinically applied compounds, we discover that homoharringtonine (HHT), an FDA-approved anti-leukemic drug, manifests senotherapeutic activity in vitro in multiple cell types including human preadipocytes, while inflicting minimal

Beneficial Effects of Fisetin, a Senotherapeutic Compound, in Women's Reproductive Health and Diseases: Evidence from In Vitro to Clinical Studies.

Fisetin is a naturally occurring flavonoid, a type of polyphenol found in fruits and vegetables such as strawberries, apples, persimmons, and onions. It has gained increasing attention for its antioxidant properties (enhancement of SOD1 and CAT activity and reduction of ROS), anti-inflammatory effects (suppression of NF-κB signaling), and senotherapeutic activity (senolytic and senomorphic effects). Although numerous studies have examined fisetin in the context of aging and chronic diseases, its role in women's reproductive health has not been systematically explored. Mechanistically, fisetin

From antioxidant to senotherapeutic: Repurposing n-acetylcysteine to counteract senescence via the HuR pathway in human endothelial cells.

Cellular senescence plays a dual role in physiology and pathology. While it contributes to tumour suppression and tissue repair, its persistence promotes chronic inflammation, tissue dysfunction, and age-related diseases. Oxidative stress is a major driver of this process, and the RNA-binding protein HuR serves as a regulator by stabilizing transcripts encoding antioxidant and stress-response proteins. In this study, we established an in vitro model of replicative senescence using human umbilical vein endothelial cells (HUVECs) and investigated the potential anti-aging effects of N-acetylcyste

hESC-derived extracellular vesicles enriched with MFGE-8 and the GSH redox system act as senotherapeutics for neural stem cells in ischemic stroke.

Human embryonic stem cells (hESCs) and their extracellular vesicles (EVs) hold significant potential for tissue repair and regeneration. Neural stem cells (NSCs) in the adult brain often acquire senescent phenotypes after ischemic injuries, releasing neurodegenerative senescence-associated secretory phenotype factors. In this study, we investigated the senotherapeutic effects of hESC-EVs on NSCs and confirmed their neuroprotective effects in neurons via rejuvenation of NSC secretions. Proteomic profiling of hESC-EVs identified MFGE-8 as a critical bridging molecule to NSCs. We also found that

HSP90-specific inhibitor, PSY-2-12, ameliorates cellular senescence via activating autophagy: A potential senotherapeutic target against acute kidney injury.

An unexpected senotherapeutic function of heat shock protein 90 (HSP90) inhibitors-drugs long developed as cancer therapeutics-has recently been revealed. In this study, we employed an HSP90-specific inhibitor, PSY-2-12 (PSY), to explore its potential therapeutic application as a senomorphic agent. We demonstrated that PSY, at non-toxic concentrations, reduces cellular senescence in both cisplatin (CP)-induced senescent kidney proximal tubular cells and angiotensin II-induced senescent human umbilical vein endothelial cells. The anti-senescent effect of PSY was further confirmed in a mouse mod

Targeting Cellular Senescence to Enhance Human Endometrial Stromal Cell Decidualization and Inhibit Their Migration.

Cellular senescence leads to stable cell cycle arrest and an inflammatory senescence-associated secretory phenotype that varies with stressor and cell type. To mitigate these effects and improve health, senotherapeutics (e.g., senolytics and senomorphics) have been developed. Senescent-like endometrial stromal cells (eSCs) lining the uterus of patients with endometriosis and infertility are proposed to impair decidualization, a differentiation process required for uterine receptivity in humans. Quercetin, a natural flavonoid senolytic, dramatically improves decidualization and reduces endometr

Senolytics and senomorphics: Natural and synthetic therapeutics in the treatment of aging and chronic diseases.

Cellular senescence is a heterogeneous process guided by genetic, epigenetic and environmental factors, characterizing many types of somatic cells. It has been suggested as an aging hallmark that is believed to contribute to aging and chronic diseases. Senescent cells (SC) exhibit a specific senescence-associated secretory phenotype (SASP), mainly characterized by the production of proinflammatory and matrix-degrading molecules. When SC accumulate, a chronic, systemic, low-grade inflammation, known as inflammaging, is induced. In turn, this chronic immune system activation results in reduced S

Targeting Senescent Cells in Fibrosis: Pathology, Paradox, and Practical Considerations.

PURPOSE OF THE REVIEW: Senescent cells have the capacity to both effect and limit fibrosis. Senotherapeutics target senescent cells to improve aging conditions. Here, we review the contexts in which senescent cells mediate wound healing and fibrotic pathology and the potential utility of senotherapeutic drugs for treatment of fibrotic disease. RECENT FINDINGS: Multi-action and temporal considerations influence deleterious versus beneficial actions of senescent cells. Acutely generated senescent cells can limit proliferation, and the senescence-associated secretory phenotype (SASP) contains fa

Cellular senescence in bone.

Cellular senescence refers to a process induced by various types of stress that causes irreversible cell cycle arrest and distinct cellular alterations, including profound changes in gene expression, metabolism, and chromatin organization as well as activation/reinforcement of anti-apoptotic pathways and development of a pro-inflammatory secretome or senescence-associated secretory phenotype (SASP). However, because of challenges and technical limitations in identifying and characterizing senescent cells in living organisms, only recently have some of the diverse in vivo roles of these unique

Cellular Senescence in Health, Disease, and Lens Aging.

Background: Cellular senescence is a state of irreversible cell cycle arrest that serves as a critical regulator of tissue homeostasis, aging, and disease. While transient senescence contributes to development, wound healing, and tumor suppression, chronic senescence drives inflammation, tissue dysfunction, and age-related pathologies, including cataracts. Lens epithelial cells (LECs), essential for maintaining lens transparency, are particularly vulnerable to oxidative stress-induced senescence, which accelerates lens aging and cataract formation. This review examines the dual role of senesce

Addition of Polyphenols to Drugs: The Potential of Controlling "Inflammaging" and Fibrosis in Human Senescent Lung Fibroblasts In Vitro.

The combination of a polyphenol, quercetin, with dasatinib initiated clinical trials to evaluate the safety and efficacy of senolytics in idiopathic pulmonary fibrosis, a lung disease associated with the presence of senescent cells. Another approach to senotherapeutics consists of controlling inflammation related to cellular senescence or "inflammaging", which participates, among other processes, in establishing pulmonary fibrosis. We evaluate whether polyphenols such as caffeic acid, chlorogenic acid, epicatechin, gallic acid, quercetin, or resveratrol combined with different senotherapeutics

Targets of senescence in cardiac muscle: insights for regenerative treatments in the aged heart.

INTRODUCTION: Despite considerable investment of time and funding, advancement in cardiac regenerative therapies has been slow. Results of large animal and human studies have failed to live up to the expectations of the positive pre-clinical animal studies. There are many reasons to explain the discrepancies, however a key factor is that patients in need of regenerative therapies are mostly aged, being 70 years and older. AREAS COVERED: The aging heart undergoes structural and functional changes due to biological mechanisms associated with aging. Of these, increased senescent cell burden, inf

Chlorogenic acid attenuates endometrial senescence via mTOR modulation: A combined network pharmacology, in vitro, and in vivo study.

Advanced maternal age is correlated with increased infertility, often due to impaired decidualization and placentation. This study elucidated the senotherapeutic potential of chlorogenic acid (CGA), a phenylacrylate polyphenol as major compound of coffee, in endometrial stromal cells (ESCs). Using a passage number-induced senescence model, CGA reduced senescence markers (P16, P21), promoted proliferation, and upregulated decidualization markers (PRL, IGFBP1, FOXO1). In aged hTERT-immortalized ESCs, CGA enhanced mitochondrial biogenesis by upregulating mitochondrial complex genes and restoring

MM-129 Counteracts 5-Fluorouracil-Induced Cellular Senescence in Colon Cancer via SIRT1/STAT3 Signaling Pathway.

Cellular senescence plays a critical role in tumorigenesis and is recognized as a hallmark of colorectal cancer (CRC). Emerging evidence suggests that 5-fluorouracil (5-FU)-induced senescence may contribute to chemoresistance and tumor recurrence. Here, we investigated the effect of 5-FU on colon cancer cell senescence and whether MM-129 (pyrazolo[4,3-e]tetrazolo[4,5-b][1,2,4]triazine sulfonamide) can antagonize this activity. Senescence was identified by the expression of senescence-associated β-galactosidase (SA-β-gal) and cyclin-dependent kinase inhibitor 1A (p21) using qPCR, microscopy, fl

The natural flavonoid dihydromyricetin targets senescent cells via PRDX2 and alleviates age-related diseases.

Aging is a primary risk factor for chronic diseases, with cellular senescence as an effective target to delay, prevent or alleviate age-related disorders. Here we report in vitro screening outputs from a natural medicinal agent library, wherein dihydromyricetin, a natural flavonoid, showed senotherapeutic potential. Dihydromyricetin protects senescent fibroblasts against further DNA damage and attenuates the senescence-associated secretory phenotype, acting as a senomorphic agent. Proteomics suggests that dihydromyricetin promotes nuclear translocation of peroxiredoxin 2 (PRDX2) to facilitate

Microenvironmental barriers to healing in osteoradionecrosis and opportunities for regenerative intervention.

Osteoradionecrosis (ORN) is a debilitating sequela of radiotherapy (RT) for head and neck cancer (HNC), characterized by persistent bone exposure or radiographic non-viability and profound tissue repair failure. Despite advances in radiation delivery and clinical management, ORN remains challenging to treat, with therapeutic strategies largely focused on symptom control, infection mitigation, or surgical reconstruction rather than restoration of regenerative capacity. Emerging evidence suggests that radiation-induced cellular senescence represents a central and underrecognized driver of ORN pa

Could Hyaluronic Acid Be Considered as a Senomorphic Agent in Knee Osteoarthritis? A Systematic Review.

BACKGROUND: Knee osteoarthritis (KOA) is one of the most common causes of disability in elderly patients and tends to be a major burden on social and health care spending. Despite its severe socioeconomic impact, KOA remains, to date, an incurable disease. Due to its proper characteristics, KOA represents a favorable disease model for experimenting with senotherapeutics, a group of treatments that counteract the development of age-related disorders and chronic diseases. In recent years, the use of intra-articular hyaluronic acid (IAHA) in the treatment of diseases related to the wear and tear
