A case of Setaria equina in a 13-year-old, UK-resident mare.

Setaria equina is a species of filarial nematode parasite transmitted by mosquitoes, which infects horses and other equids. Adult worms reside in the abdominal cavity with apparently little pathogenic significance, but larvae can migrate to other regions of the body with resulting clinical disease. Cases in the UK are rare, with the last published report in 2001. A recent case of S. equina infection was detected in a 13-year-old, UK-resident mare during postmortem examination in Hertfordshire, UK. Adult worms were collected from the abdominal cavity, their DNA extracted and subjected to PCR an

Molecular and morphological detection of Setaria tundra in roe deer (Capreolus capreolus) from Lithuania.

Setaria spp. are vector-borne parasitic roundworms commonly found in the abdominal cavities of wild and domesticated ungulates globally. The objective of this study was to conduct a morphological and molecular analysis of Setaria tundra Issaitshikoff et Rajewskaya, 1928, collected from roe deer Capreolus capreolus Linnaeus in Lithuania. The morphological characteristics of S. tundra described and illustrated. The amplification of the 12S rRNA and 18S rRNA genes confirmed the presence of S. tundra. This is the first case report of S. tundra in roe deer in Lithuania.

Antifilarial activity of Azadirachta indica on cattle filarial parasite Setaria cervi.

Alcohol and aqueous extracts of flowers of Azadirachta indica were tested in vitro for their potential antifilarial activity against whole worm, nerve muscle (n.m.) preparation and microfilariae of Setaria cervi. The effects of alcohol and aqueous extracts were similar in nature on the spontaneous movements of whole worm and nerve muscle preparation. On the whole worm, the response was characterized by initial increase in tone, rate and amplitude of contractions followed by reversible paralysis. The initial stimulant effect is likely to be due to irritant effect on the cuticle. Nerve muscle pr

Demonstration of vector competence of Culex quinquefasciatus (Diptera: Culicidae) for Setaria digitata.

In Taiwan, Setaria digitata infection causes a lumber paralysis in increasing number of cattle. Culex quinquefasciatus is one of the predominant mosquitoes, and it has been suspected that C. quinquefasciatus acts as a vector to Setaria nematodes prevalence but this was not confirmed. C. quinquefasciatus, Aedes albopictus and A. aegypti of various strains were investigated using an artificial infection system to evaluate their vector competence. After blood feeding at day 14, the number of larvae (stage III) per infected mosquito in A. aegypti (Liverpool strain), A. aegypti (Kaohsiung strain),

Ocular infection of cattle with Setaria digitata.

One 5-month-old female native Korean calf and a 2-year-old female Holstein cow raised in two farms about 4 km apart from each other in Korea, were found to have the left eye opaque, which included motile white worms in the aqueous humor. The parasite removed from the left eye of the calf was identified as Setaria digitata based on both light and electron microscopic features. The ocular infection with S. digitata reported herein may document the first aberrant case in Korean cattle.

Excretory/secretory antigens from a bovine filarial parasite cross react with human antifilarial antibodies.

Certain excretory/secretory proteins released by adult females of the bovine filarial parasite, Setaria digitata, along with the release of microfilariae when chromatographically analysed has three major protein fractions of molecular weights 70 kD (ESF1), 16.5 kD (ESF2) and 11 kD (ESF3). Of these ESF2 and ESF3 cross reacted with antibodies from Wuchereria bancrofti infected humans. ESF2 was more specific and accurate in detecting human filarial infection. Similar proteins secreted by human filarial parasites could be targets for combating the disease by cure or control.

Antigenic characterization of excretory-secretory products of Setaria cervi.

Excretory-secretory products (ES) of Setaria cervi, a bovine filarial parasite, were prepared by in vitro maintenance of the worms in a protein free defined medium at 37 degrees C. RPMI-1640 medium proved to be the best as the worms remained motile in it for the maximum period of time without change of medium, and approximately 12-15 mg protein per 100 adult worms could be obtained. Crossed immunoelectrophoretic analysis of ES products revealed 11-15 antigens and most of them were common to somatic antigenic preparations from both adult worms and microfilariae. The ES products were also found

Immunodiagnosis of Bancroftian filariasis by enzyme linked immunosorbent assay using Litomosoides carinii and Setaria cervi antigens.

An ELISA was developed using cross-reacting antigens of Litomosoides carinii and Setaria cervi to detect antibodies in filarial patients. The diagnostic potential of the test was investigated in 80 symptomatic cases of filariasis, 40 asymptomatic carriers and 50 normal subjects of an area around Lucknow endemic for Bancroftian filariasis. By the criteria laid down for ELISA positivity for filariasis, 86.2% of the symptomatic cases could be detected by this test. Only four of 40 asymptomatic carriers and one of 50 normal subjects gave a positive ELISA response. Purified antigenic fractions of S

Oxidative activities in mitochondria-like particles from Setaria digitata, a filarial parasite.

The oxidative metabolic potential of Setaria digitata, a filarial parasite found in the intraperitoneal cavity of cattle, was investigated. These worms showed active wriggling movements which were not affected by respiratory poisons such as cyanide, rotenone and malonate. They also possessed cyanide-insensitive and glucose-independent oxygen consumption pathways. By differential centrifugation of sucrose homogenates, a fraction containing mitochondria-like particles was obtained in which the activity of the marker enzyme, succinate dehydrogenase, was recovered. This fraction catalysed succinat

Screening of mosquitoes for filarioid helminths in urban areas in south western Poland-common patterns in European Setaria tundra xenomonitoring studies.

In recent years, numerous studies screening mosquitoes for filarioid helminths (xenomonitoring) have been performed in Europe. The entomological monitoring of filarial nematode infections in mosquitoes by molecular xenomonitoring might serve as the measure of the rate at which humans and animals expose mosquitoes to microfilariae and the rate at which animals and humans are exposed to the bites of the infected mosquitoes. We hypothesized that combining the data obtained from molecular xenomonitoring and phenological studies of mosquitoes in the urban environment would provide insights into the

Inhibition of Setaria cervi protein tyrosine phosphatases by Phenylarsine oxide: A proteomic and biochemical study.

Phenylarsine oxide (PAO), a specific protein tyrosine phosphatase (PTP) inhibitor significantly decreased the motility and viability of Setaria cervi ultimately leading to its death. The PTP activity present in the cytosolic and detergent soluble fractions as well as on surface of these parasites was significantly inhibited by PAO. A marked alteration in protein spots abundance after proteomic analysis showed 14 down-regulated and 9 upregulated spots in the treated parasites as compared to the control. The PTP inhibition led to increase in the cytosolic and mitochondrial calpain activity in th

Setaria equina: in vivo effect of diethylcarbamazine citrate on microfilariae in albino rats.

Although diethylcarbamazine citrate (DEC) is successful drug in eliminating human filariasis, yet, its mode of action is still debatable. Herein, the effect of DEC to treat albino rats infected with the animal filarial parasite Setaria equina was tested. Microfilarial (mf) counts and sections from liver, lung, kidney as well as spleen were investigated at different time points after treatment by light microscopy. After 45 and 300min of treatment, a significant decrease in blood mf was observed accompanied by adherence of degenerated mf to both kupffer cells and leukocyte in liver sections. In

Four cases of human filariosis due to Setaria labiatopapillosa found in Bucharest, Romania.

We record the occurrence of four cases of subconjunctival eye infection due to Setaria labiatopapillosa. The patients are all women, between 35 and 68 years old, all inhabitants of the same neighbourhood of the lake Pantelimon, a well known area for its blood feeding vector insect population and all complained about the same eye affliction: photophobia, eye swelling, tearing and foreign body sensation accompanied by some rash and low eosinofilia (6-8%). The treatment was surgical (excision) and DEC 1 mg/kg body weight. The vector insect cannot be specified. We stress that to our knowledge thes
