Electrodeposited silver amalgam particles on pyrolytic graphite in (spectro)electrochemical detection of 4-nitrophenol, DNA and green fluorescent protein.

Catalytic properties and high adsorption affinity of nucleic acids and proteins to silver amalgam electrode surface make this kind of electrified interface perspective for bioanalytical and biomedical applications. For the first time, a basal-plane pyrolytic graphite electrode (bPGE) has been used as a substrate for electrodeposition of silver amalgam particles (AgAPs). Optimization of the resulting composition, surface morphology and electrochemical properties of the AgAPs was done by scanning electron microscopy with energy disperse X-ray spectroscopy, image processing software and voltammet

Comparative decreases in tyrosinase, TRP-1, TRP-2, and Pmel 17/silver antigenic proteins from melanotic to amelanotic stages of syngeneic mouse cutaneous melanomas and metastases.

Malignant cutaneous melanomas and metastases were taken directly from in situ lesions of genetically identical (C57BL/6 strain) Tyr-SV40E transgenic mice, and samples were analyzed by Western immunoblotting with antisera specific for the COOH terminus of each of four melanocytic proteins. These were tyrosinase, TRP-1, TRP-2, and Pmel 17/silver. Of the 13 melanomas examined, there were 5 melanotic primary tumors, 5 amelanotic primary tumors, and 3 amelanotic metastases. The melanotic tumors expressed all of the markers to some extent. In contrast, the amelanotic tumors lacked detectable levels

Immunolocalization of Gla proteins (osteocalcin) in rat tooth germs: comparison between indirect immunofluorescence, peroxidase-antiperoxidase, avidin-biotin-peroxidase complex, and avidin-biotin-gold complex with silver enhancement.

Odontoblasts and osteoblasts synthesize gamma-carboxyglutamatic acid (Gla)-containing proteins which are partially deposited in the mineralizing tissues and partially released into the plasma. Using four immunostaining techniques, we have evaluated the question of whether dentin Gla proteins (DGP) are transported to the mineralization front through the odontoblast processes. Undecalcified sections of rat incisors and molar tooth germs were immunostained with affinity-purified antibodies to DGP using the following methods: indirect immunofluorescence; peroxidase-antiperoxidase (PAP); avidin-bio

Novel Antibacterial, Cytotoxic and Catalytic Activities of Silver Nanoparticles Synthesized from Acidophilic Actinobacterial SL19 with Evidence for Protein as Coating Biomolecule.

Silver nanoparticles (AgNPs) have potential applications in medicine, photocatalysis, agriculture, and cosmetic fields due to their unique physicochemical properties and strong antimicrobial activity. Here, AgNPs were synthesized using actinobacterial SL19 strain, isolated from acidic forest soil in Poland, and confirmed by UV-vis and FTIR spectroscopy, TEM, and zeta potential analysis. The AgNPs were polydispersed, stable, spherical, and small, with an average size of 23 nm. The FTIR study revealed the presence of bonds characteristic of proteins that cover nanoparticles. These proteins were

Modification of myofibrillar protein structural characteristics: Effect of ultrasound-assisted first-stage thermal treatment on unwashed Silver Carp surimi gel.

The hardness properties of unwashed surimi gel are considered as the qualities of gelation defect. This research investigated the effect of ultrasound-assisted first-stage thermal treatment (UATT) on the physicochemical properties of unwashed Silver Carp surimi gel, and the enhancement mechanism. UATT could reduce protein particle size, which significantly reduced from 142.22 μm to 106.70 μm after 30 min of UATT compared with the nature protein. This phenomenon can promote the protein crosslinking, resulting in the hardness of surimi gel increased by 15.08 %. Partially unfolded structure of my

Involvement of Ubiquitin-Editing Protein A20 in Modulating Inflammation in Rat Cochlea Associated with Silver Nanoparticle-Induced CD68 Upregulation and TLR4 Activation.

Silver nanoparticles (AgNPs) were shown to temporarily impair the biological barriers in the skin of the external ear canal, mucosa of the middle ear, and inner ear, causing partially reversible hearing loss after delivery into the middle ear. The current study aimed to elucidate the molecular mechanism, emphasizing the TLR signaling pathways in association with the potential recruitment of macrophages in the cochlea and the modulation of inflammation by ubiquitin-editing protein A20. Molecules potentially involved in these signaling pathways were thoroughly analysed using immunohistochemistry

In-vivo evaluation of silver nanoparticles-enhanced Chlorella protein nanovaccine for immunological defense in grouper (Epinephelus sp) against RNA viral infections.

Viral RNA diseases, notably Viral Nervous Necrosis (VNN), routinely produce very high losses (≈80-100% mortality) in cultured groupers (Epinephelus sp.), driving major economic impacts across Asia. Conventional vaccination has been unreliable, largely because of antigen instability and suboptimal, short-lived immunity. Nanovaccine platforms-here, Chlorella-derived proteins augmented with silver nanoparticles (AgNPs)-offer a route to better antigen delivery and immune modulation in aquatic hosts. This study evaluated the efficacy of an AgNPs- Chlorella vulgaris protein nanovaccine in VNN-challe

Synthesis of Hot-Spot-Rich Silver Nanogap Networks within Flat Nanoframes and a Strategy for Targeted Protein Positioning near Plasmonic Hot Spots.

We propose a surface-enhanced Raman spectroscopy (SERS) detection strategy that assembles Ag nanogap-integrated triangular nanoframes (NITNs) on Ni substrates. The Ag NITNs were synthesized via a one-pot tandem reaction followed by Ag coating. By tuning the void area fraction of the Ag NITNs, we engineered narrow intraparticle nanogaps (∼4 nm), which significantly concentrate the electromagnetic near-field. Then, we employed Ag NITNs for liquid-phase, SERS-based protein screening─a task hindered by the inherently weak Raman signals of proteins and their low affinity for plasmonic nanoparticle

Synergistic silver nanoparticle-graphene quantum dot composites in silk fibroin/lathyrus protein-oxidized alginate hydrogels for accelerated diabetic wound healing.

Diabetic wounds require an advanced wound care solution for faster recovery and reduced complications. In this direction, an injectable, self-healing hydrogel (HG) platform based on dynamic Schiff-base crosslinking between oxidized alginate (oAlg) and natural proteins (silk fibroin or Lathyrus sativus-derived plant protein) is developed for the treatment of infected diabetic wounds. The mild one-pot in situ fabrication enables the reduction of silver ions to silver nanoparticles (hereafter denoted as Ag) and the incorporation of graphene quantum dots (GQDs) into the HG matrix. This creates a s

Expression Analysis of Cyst Specific Protein (CSP21) and Cellulose Synthase II (CSII) Genes in Acanthamoeba castellanii Trophozoites Exposed to Silver Nanoparticles Conjugated with Elaeagnus umbellata.

PURPOSE: Acanthamoeba species are eucaryotic protozoa found predominantly in soil and water. They cause ulceration and vision loss in the cornea (Acanthamoeba keratitis) and central nervous system (CNS) infection involving the lungs (granulomatous amoebic encephalitis). Antiparasitic drugs currently used in the treatment of infections caused by Acanthamoeba species are not effective at the desired level in some anatomical regions such as the eye and CNS. The existence of an agent effective against both cysts and trophozoites has not yet been proven. Drugs used for treatment of Acanthamoeba inf

Oral exposure to silver nanoparticles increases oxidative stress markers in the liver of male rats and deregulates the insulin signalling pathway and p53 and cleaved caspase 3 protein expression.

The present study was aimed at assessing the impact of AgNPs on the liver of male rats orally exposed to 0, 50, 100 and 200 mg/kg/day of polyvinyl pyrrolidone coated AgNPs (PVP-AgNPs) for 90 days. The induction of apoptotic cell death -by measuring the protein levels of the active form of caspase 3- and the levels of the microtubule-associated protein 1A/1B-light chain (LC3) protein were measured as a marker of the induction of autophagy. PVP-AgNPs caused an increase of the activity of superoxide dismutase (SOD) and catalase (CAT) in the liver of male rats. However, the activity decreased afte

Analytical technique for label-free multi-protein detection based on Western blot and surface-enhanced Raman scattering.

We have developed a new analytical procedure for label-free protein detection designated "Western SERS", consisting of protein electrophoresis, Western blot, colloidal silver staining, and surface-enhanced Raman scattering (SERS) detection. A novel method of silver staining for Western blot that uses a silver colloid, an excellent SERS-active substrate, is first proposed in the present study. During the process of silver staining, interactions between proteins and silver nanoparticles result in the emergence of SERS of proteins. In the present study, we use myoglobin (Mb) and bovine serum albu

Silver staining of the nucleolus organizer regions (NOR) requires clusters of sulfhydryl groups.

Silver stainability of nucleolus organizer regions (NORs) appears to be correlated with the presence of grouped sulfhydryl (SH) side chains of proteins. In fact, heavy metals with high affinity for SH groups, such as Hg and Cu, do prevent the silver staining reaction. Ferricyanide, which is known to oxidize SH to disulfides, also prevents any further reaction with silver. On the other hand, alkali and reducing agents (mercaptoethanol, cyanide) do not affect silver stainability of the NORs. These results show that the silver staining reaction is not related to disulfide or persulfide groups and

The Effect of Silver Nanoparticles on Listeria monocytogenes PCM2191 Peptidoglycan Metabolism and Cell Permeability.

Listeria monocytogenes is Gram-positive bacterial pathogen, a causative agent of food poisoning and systemic disease - listeriosis. This species is still susceptible to several conventionally used antibiotics but an increase in its resistance has been reported. For this reason the search for new, alternative therapies is an urgent task. Silver nanoparticles seem to be the promising antibacterial agent. Minimal inhibitory concentration of silver nanoparticles was determined. Sublethal concentrations were used in study of nanosilver effect on cells lysis by estimation of the number of cells surv

The biochemical fate of Ag+ ions in Staphylococcus aureus, Escherichia coli, and biological media.

Silver is commonly included in a range of household and medical items to provide bactericidal action. Despite this, the chemical fate of the metal in both mammalian and bacterial systems remains poorly understood. Here, we applied a metallomics approach using X-ray absorption spectroscopy (XAS) and size-exclusion chromatography hyphenated with inductively coupled plasma mass spectrometry (SEC-ICP-MS) to advance our understanding of the biochemical fate of silver ions in bacterial culture and cells, and the chemistry associated with these interactions. When silver ions were added to lysogeny br
