Silybin exerts antioxidant effects and induces mitochondrial biogenesis in liver of rat with secondary biliary cirrhosis.

The accumulation of toxic hydrophobic bile acids in hepatocytes, observed during chronic cholestasis, induces substantial modification in the redox state and in mitochondrial functions. Recent reports have suggested a significant role of impaired lipid metabolism in the progression of chronic cholestasis. In this work we report that changes observed in the expression of the lipogenic enzymes acetyl-CoA carboxylase and fatty acid synthase were associated with a decrease in the activity of citrate carrier (CIC), a protein of the inner mitochondrial membrane closely related to hepatic lipogenesis

Dual-targeting galactose-functionalized hyaluronic acid modified lipid nanoparticles delivering silybin for alleviating alcoholic liver injury.

Alcoholic liver injury stands as a predominant pathogenic contributor to the global burden of liver diseases, with alcohol consumption serving as a significant determinant of worldwide morbidity and mortality. Given that liver-targeted therapy for mitigating alcoholic liver injury remains to be a major clinical challenge due to the poor specificity and instability associated with single targeting modification in actively targeted nanomedicine systems, bifunctional targeting modification may serve as a more promising strategy. Here, galactose-functionalized hyaluronic acid (Gal-HA) coated catio

Baicalein Enhances the Oral Bioavailability and Hepatoprotective Effects of Silybin Through the Inhibition of Efflux Transporters BCRP and MRP2.

Although hepatoprotective properties of silybin are well documented, the clinical therapeutic efficacy is limited by its low bioavailability due to absorption rates, extensive phase II metabolism, and biliary excretion. As our previous study indicated that metabolic enzymes may have limited effects on the pharmacokinetic (PK) behavior of silymarin, here, we intended to increase the oral bioavailability and bio-efficacy of silybin through the inhibition of active efflux. In Caco-2 and transfected MDCKII cell models, flavone baicalein significantly inhibited the efflux of silybin as a BCRP and M

Modulatory effects of curcumin, silybin-phytosome and alpha-R-lipoic acid against thioacetamide-induced liver cirrhosis in rats.

Liver cirrhosis is the final consequence of a progressive fibrotic process characterized by excessive collagen deposition and destruction of the normal liver architecture. This study aimed to investigate the protective effects of curcumin, silybin-phytosome and alpha-R-lipoic acid against thioacetamide-induced cirrhosis. Male rats were allocated into five groups of which one group received saline and served as normal control. Animals from groups 2-5 were treated with thioacetamide administered intraperitoneally at a dose of 200 mg/kg 3 times per week for 7 weeks. Group 2 was left untreated whi

Integrated in-silico and in-vitro evaluation of silybin as a potential quorum-sensing modulator against Acinetobacter baumannii and Staphylococcus aureus.

The emergence of multi-drug resistance in the class of ESKAPE pathogens such as Acinetobacter baumannii and Staphylococcus aureus has led to the search for anti-virulence mechanisms to overcome resistance posed by the existing antibiotics. Quorum sensing (QS), mediated by AbaI/AbaR circuits in A. baumannii and AgrA/AgrC circuits in S. aureus, regulates biofilm formation and virulence factor production in the two species, thereby serving as potential targets for phytocompound action. In the current study silybin, a flavonoid lignan from Silybum marianum, emerged as a potential QS modulator agai

Study on the improvement of bioavailability and hepatoprotective effect of silybin-7-eicosanedioate long-acting injection based on prodrug technology.

Chronic hepatitis (CH) is a prevalent liver disease with poor clinical treatment efficacy due to the low bioavailability and short half-life of oral silybin (SLB) formulations. This study employed a long-chain fatty acid prodrug strategy to synthesize four SLB prodrugs (SLB-7-DDDA, SLB-7-TDDA, SLB-7-HDDA, SLB-7-ODDA), among which SLB-7-ODDA was identified as the optimal candidate owing to its lowest degradation rate (purity > 95%). SLB-7-ODDA long-acting suspension injection (SLB-7-ODDA-LAIS) was prepared via wet ball milling, which showed uniform particle size distribution, excellent redisper

Targeting HSP90ab1: curcumin and silybin inhibit NNV infection in sea perch (Lateolabrax japonicus) via receptor blockade and immune enhancement.

Nervous necrosis virus (NNV), a member of the family Nodaviridae, is a devastating pathogen in global aquaculture, yet effective preventive and therapeutic strategies remain limited. This study aimed to identify natural compounds that inhibit NNV infection by targeting the host receptor Lateolabrax japonicus heat shock protein 90 alpha family class B1 (LjHSP90ab1). Herein, we identified curcumin and silybin as potent inhibitors of LjHSP90ab1. Molecular docking predicted that curcumin and silybin bind to the N-terminal and C-terminal domains of LjHSP90ab1, respectively, and fluorescence spectro

A Potential Approach of Mesenchymal Stem Cells Combined Silybin for Synergistic Treatment in Rheumatoid Arthritis via ICOS/ICOSL.

Achieving clinical remission in rheumatoid arthritis (RA) remains a significant challenge in current therapeutic strategies. While transplantation of human umbilical cord-derived mesenchymal stem cells (UC-MSCs) has shown promising outcomes, therapeutic responses vary considerably among patients. In this study, we characterized the immune profiles of nonresponders and identified elevated expression of inducible costimulator (ICOS) in peripheral immune cells as a critical barrier to effective treatment. This upregulation, combined with the presence of ICOS ligand (ICOSL) on UC-MSCs, activated T

Smart microenvironment-activatable silybin nano-prodrug for precise NIR-II fluorescence monitoring and on-demand therapy of acute liver injury.

Acute liver injury (ALI) is a serious disease characterized by liver function impairment caused by multiple causes in a short period of time. Due to lack of precise diagnosis and timely intervention, many patients with ALI rapidly progress to liver dysfunction and liver failure. Here, a multifunctional silybin nano-prodrug, PTS@IR, was developed that integrated microenvironment-activatable second near-infrared (NIR-II) fluorescence (FL) imaging for precise diagnosis and timely therapy of ALI. The PTS@IR was formed through the self-assembly of a reactive oxygen species (ROS)-cleavable thioketal

Flavonolignans silybin, silychristin and 2,3-dehydrosilybin showed differential cytoprotective, antioxidant and anti-apoptotic effects on splenocytes from Balb/c mice.

Silymarin is an extract obtained from the seeds of milk thistle (Sylibum marianum L., Asteraceae) and contains several structurally related flavonolignans and a small family of flavonoids. Mouse spleen cells represent highly sensitive primary cells suitable for studying the pharmacological potential and biofunctional properties of natural substances. Cultivation of splenocytes for 24 h under standard culture conditions (humidity, 37 °C, 5% CO2, atmospheric oxygen) resulted in decreased viability of splenocytes compared to intact cells. A cytoprotective effect of silybin (SB), silychristin (SCH

Oral Administration of Silybin Protects Against MPTP-Induced Neurotoxicity by Reducing Pro-inflammatory Cytokines and Preserving BDNF Levels in Mice.

Parkinson's disease (PD) is the second most frequent neurodegenerative disease associated with motor dysfunction secondary to the loss of dopaminergic neurons in the nigrostriatal axis. Actual therapy consists mainly of levodopa; however, its long-term use promotes secondary effects. Consequently, finding new therapeutic alternatives, such as neuroprotective molecules, is necessary. Among these alternatives is silybin (Sb), the major bioactive flavonolignan in silymarin. Both exert neuroprotective effects, preserving dopamine levels and dopaminergic neurons when administered in the 1-methyl-4-

Silybin attenuates PERK/IRE1α ER-stress signaling, neuroinflammation, and restores CDNF levels in an MPTP-induced Parkinson's disease model.

Parkinson's disease (PD) is a progressive neurodegenerative disorder in which dopaminergic dysfunction is associated with oxidative stress, chronic neuroinflammation, and endoplasmic reticulum (ER)-stress-related signaling. ER-resident neurotrophic factors, cerebral dopamine neurotrophic factor (CDNF) and mesencephalic astrocyte-derived neurotrophic factor (MANF), have emerged as potential modulators of neuronal stress responses. Silybin, a flavonolignan derived from S. marianum, has antioxidant, anti-inflammatory, and neuroprotective effects in experimental PD models, although the molecular p

Silybin Mitigates Post-Myocardial Infarction Heart Failure in Mice via Modulation of HIF-1α-Driven Glycolysis and Energy Metabolism.

BACKGROUND: Post-myocardial infarction (MI) heart failure (HF) is characterized by myocardial energy metabolism disorder, with excessive glycolysis playing a key role in its progression. Silybin (SIL), a flavonoid derived from Silybum marianum, has demonstrated hepatoprotective and metabolic regulatory effects. However, the role of this flavonoid in ameliorating post-myocardial infarction heart failure (post-MI HF) by modulating energy metabolism remains unclear. METHODS: This study employed an oxygen-glucose deprivation (OGD) model to induce myocardial cell injury in vitro, with YC-1 treatme

Silybin prevented avermectin-induced cardiotoxicity in carp by modulating oxidative stress, inflammation, endoplasmic reticulum stress, mitochondrial apoptosis, and autophagy.

Avermectin is one of the widely used anthelmintics in aquaculture and exhibits substantial toxicity to aquatic organisms. Silybin is extensively used for its anti-inflammatory, antioxidant and anti-apoptotic biological properties. Heart is essential for the survival of fish and plays a vital role in pumping blood oxygen and nutrients. Residual avermectin in water poses harm to carp. However, there is still insufficient research on whether silybin can mitigate the toxicity of avermectin to carp heart tissues. In this research, we established a model involving carp subjected to acute avermectin
