Viruses in Simuliidae: An Updated Systematic Review of Arboviral Diversity and Vector Potential.

Black flies (Diptera: Simuliidae) are important vectors of pathogens, including filarial nematodes, protozoans, and arboviruses, which significantly impact human and animal health. Although their role in arbovirus transmission has not been as thoroughly studied as that of mosquitoes and ticks, advances in molecular tools, particularly metagenomics, have enabled the identification of non-cultivable viruses, significantly enhancing our understanding of black-fly-borne viral diversity and their public and veterinary health implications. However, these methods can also detect insect-specific virus

A method of rearing Simulium damnosum Theobald (Diptera; Simuliidae) under artificial conditions.

The Simuliidae have proved particularly difficult to maintain under artificial conditions, and have resisted all attempts at laboratory colonization. Although a number of species have been successfully reared from egg to adult under laboratory conditions in certain northern countries, the problem of rearing African blackflies has been subjected to less intensive investigation. The development of suitable techniques for rearing and maintaining the vectors of human onchocerciasis in the laboratory would facilitate critical work on many aspects of vector biology. The vectors of human onchocerciasi

Molecular identification of onchocerciasis vector sibling species in black flies (Diptera: Simuliidae).

A number of sibling species in the Simulium damnosum species complex transmit River Blindness (onchocerciasis) in tropical Africa. Black fly sibling species can be reliably recognized only through analysis of the larval polytene chromosomes. The routine, unambiguous assignment of the potentially disease-transmitting adult females to a particular species is virtually impossible. Two molecular assays to identify the sibling species of biting female flies are presented. The first relies on PCR-mediated amplification of internal transcribed spacers of the nuclear rDNA loci while the second involve

Duration of larval development of Simulium yahense (Diptera: Simuliidae) under natural conditions.

The duration of Simulium yahense Vajime & Dunbar larval development on a dam spillway in Harbel, Liberia, was observed to make accurate decisions regarding the frequency of larvicide treatments against this onchocerciasis vector. Larval development required a minimum of 10-12 d from eclosion to first pupation. Initial larvicidal treatment for S. yahense control would require a treatment cycle of 7 d. Once suppression of adult and larval populations is achieved, a 9-12-9-12 day treatment cycle could be adopted.

Sexual mosaicism in the black fly Simulium (Gomphostilbia) hongthaii (Diptera: Simuliidae) in Vietnam.

Sexual mosaicism was found in an adult black fly reared from a pupa in Vietnam. It is considered an antero-posterior type of gynandromorph, showing female phenotype for the head, thorax and wing; male phenotype for the abdomen including the genitalia; and mixed phenotype for the legs. The fly is identified as Simulium (Gomphostilbia) hongthaii in the S. asakoae species-group by having the sensory vesicle medium-long (0.28-0.30 times as long as the third palpal segment), mandible without teeth on its outer margin, and ventral plate transverse, with its lateral margins emarginated basally when v

Taxonomy and polytene chromosomes of the Neotropical black fly Simulium perplexum (Diptera: Simuliidae).

Simulium perplexum Shelley, Maia-Herzog, Luna Dias & Couch is structurally similar in the pupal stage to Simulium guianense Wise, the main vector in the onchocerciasis foci of Amazonian Brazil and Venezuela. We report S. perplexum for the first time beyond its type locality (Guyana, Potaro River), describe its larva, redescribe its pupa, and provide a chromosomal comparison with S. guianense and other morphologically similar species. We collected it in two rivers in Rurópolis municipality, Pará state, Brazil. The larvae can be distinguished from those of related species by having body cuticle

The intervals between the blood-meals of man-biting Simulium damnosum (Diptera: Simuliidae).

Six marking-recapture experiments were carried out with Simulium damnosum in the Cameroon Republic, five in the rain-forest and one in the Sudan-savanna zone. Adult flies were marked by applying a small spot of oil paint to the mesonotum while they were engorging with blood on the legs of volunteers infected with Onchocerca volvulus microfilariae. Catches of wild flies were carried out for 12-13 days after the first day of marking, and all marked flies recaptured were examined. Some of the flies recaptured on days 0, 1 and 2 were nulliparous and had probably been disturbed by the application o

The Biodiversity of Blackflies (Diptera: Simuliidae) in Brazil: New Distribution Records and Updated Checklist.

Currently, according to the most recent Simuliidae World Inventory, there are 97 valid species of blackflies recorded in Brazil, some of which act as vectors for zoonoses such as human onchocerciasis and mansonellosis in the northern and central-western regions of the country. Meanwhile, other species can cause serious socioeconomic problems due to the nuisance of female bites. Therefore, accurate knowledge of their distribution is crucial for the development and implementation of successful preventive strategies. With this aim, this study reviewed and updated the geographical distribution of

Characterization of IgE-binding proteins in the salivary glands of Simulium nigrogilvum (Diptera: Simuliidae).

Simulium dermatitis is an IgE-mediated skin reaction in animals and humans caused by the bites of black flies. Although Simulium nigrogilvum has been incriminated as the main human-biting black fly species in Thailand, information on its salivary allergens is lacking. Salivary gland extract of S. nigrogilvum females was subjected to sodium dodecylsulfate-polyacrylamide gel electrophoresis, and the separated components were applied onto nitrocellulose membranes for immunoblotting, which was performed by probing the protein blots with sera from 17 individuals who were allergic to the bites of S.

Blackflies (Diptera: Simuliidae) of Southern Guyana with keys for the identification of adults and pupae--a review.

A revision is made of the previously poorly studied blackfly fauna from the south-western border of Guyana with Brazil. Notes on the biosystematics of the species found are provided, together with keys and illustrations based on their morphology. Of the 14 species recorded, eight are anthropophilic and two of these (Simulium oyapockense s.l. and S. guianense s.l.) are proven vectors of human onchocerciasis in the nearby Amazonia focus of the disease in neighbouring Brazil.

Cytotaxonomy, morphology and molecular systematics of the Bioko form of Simulium yahense (Diptera: Simuliidae).

Cytotaxonomic analysis of the polytene chromosomes from larvae of the Simulium damnosum Theobald complex from the island of Bioko in Equatorial Guinea is reported, and a new endemic cytoform is described. Chromosomally this cytoform is close to both S. squamosum (Enderlein) and S. yahense Vajime & Dunbar, but is not identical to either. However, it is morphologically and enzymatically identical to S. yahense. The Bioko form was also found to differ from other cytoforms of the S. damnosum complex in West Africa in the copy number or RFLP pattern of several different repetitive DNA sequences. It

Evaluation of larvicides for the control of Simulium damnosum s.l. (Diptera: Simuliidae) in West Africa.

The Onchocerciasis Control Program of the World Health Organization is carrying out an extensive screening program in a search for new larvicides to be used for control of Simulium damnosum s.l. Emphasis has been given to finding a pyrethroid and a carbamate to supplement the organophosphates currently in use. These chemicals with differing modes of action, together with Bacillus thuringiensis H-14, are being used in an attempt to cope with the development and spread of resistance to the organophosphates temephos and chlorphoxim.

Esperanza Window Traps for the collection of anthropophilic blackflies (Diptera: Simuliidae) in Uganda and Tanzania.

There is an increasing need to evaluate the impact of chemotherapeutic and vector-based interventions as onchocerciasis affected countries work towards eliminating the disease. The Esperanza Window Trap (EWT) provides a possible alternative to human landing collections (HLCs) for the collection of anthropophilic blackflies, yet it is not known whether current designs will prove effective for onchocerciasis vectors throughout sub-Saharan Africa. EWTs were deployed for 41 days in northern Uganda and south eastern Tanzania where different Simulium damnosum sibling species are responsible for dise

A patient with severe black fly (Simuliidae) hypersensitivity referred for evaluation of suspected immunodeficiency.

BACKGROUND: Biting flies of the Diptera order and specifically the black fly (Simuliidae family) can be rare causes of severe hypersensitivity reactions. OBJECTIVE: To describe a patient referred for evaluation of immunodeficiency whose clinical course is explained by severe Simuliidae hypersensitivity. METHODS: The patient's immune system was investigated using standard laboratory evaluations. Hypersensitivity to Simuliidae was pursued because of historical features of her presentation and was specifically examined by skin prick and intradermal testing with whole body extract. RESULTS: The

Evidence of multiple mating and hybridization in Simulium damnosum s.l. (Diptera: Simuliidae) in nature.

Beakers with aeration were used to rear individual (single) egg batches of Simulium damnosum (Theobald) s.l. to larvae and adults. Chromosomal analysis of the progeny of individual females indicated that multiple mating occurred within the S. damnosum complex and that hybridization occurred between S. damnosum s.s. and S. sirbanum (Vajime & Dunbar) in nature. These results indicate that possibly more extensive hybridization may be taking place in some areas of the Onchocerciasis Control Program than previously reported. Dramatic changes in the rates of hybridization among members of the S. dam

A cytogenetic analysis of the Simulium ochraceum species complex (Diptera: Simuliidae) in Central America.

A cytobiotaxonomic study of the medically important insect vector Simulium ochraceum s.l. revealed two sibling species and one cytotype from various endemic and nonendemic zones of human onchocerciasis in Guatemala and Mexico. Polytene chromosome maps and idiograms as well as notes on the biology of the three taxa designated S. ochraceum A, S. ochraceum B, and S. ochraceum C within the subgenus Psilopelmia are presented. All three taxa exhibit distinct sex chromosomes and taxon-specific suites of autosomal inversion polymorphisms. Simulium ochraceum C differs from both S. ochraceum A and S. oc

[Evaluation of control measures against Simuliidae in the framework of the OCP (Onchocerciasis Control Program)].

To control the vector S. damnosum s.l., the Onchocerciasis Control Programme in West Africa (OCP) uses larvicides which are sprayed mainly by air. Despite the more or less reversible resistance of some species of the S. damnosum complex to the organophosphorus compounds (temephos and chlorphoxim), the Programme has succeeded in finding effective replacement insecticides. These larvicides, which are used in rotation, taking into account the river discharges, blackfly species present, and their susceptibility to the different insecticides, have made it possible to maintain the good results obtai
