[Effects of CCK-8 of hypothalamus on antral motility in rats].

Cholecystokinin-octapeptide (CCK-8) of 10 ng were injected into lateral hypothalamus (LH) and ventromedial hypothalamus (VMH) in conscious rats, both of which brought about inhibition of antral motility recorded by a strain gauge, the actions were partly antagonised by bilateral vagotomy, but blocked by intravenous infusion of atropine or regitine. It is suggested that both vagus and sympathetic nerves be involved in mediation of function of central CCK-8. In addition, the unit spontaneous discharges of neurons in vagal nuclei were attenuated while CCK-8 injected into LH. On the contrary, the

Immunohistochemical localization of gastrin-cholecystokinin-like material in the central nervous system of the migratory locust.

Brain, corpora cardiaca (CC)-corpora allata (CA) complex, suboesophageal ganglion, thoracic and abdominal ganglia of adults, larvae and embryos of Locusta migratoria have been immunohistochemically screened for gastrin cholecystokinin (CCK-8(s]-like material. In adult, numerous immunoreactive neurons and nerve fibres are located, with a marked symmetry, in various parts of the brain and throughout the ventral nerve cord. In the median part of the brain, cell bodies belonging neither to cellular type A1 nor A2 (following Victoria blue-paraldehyde fuchsin staining) are immunopositive; their proc

Atractylenolide-III attenuates osteoarthritis by repolarizing macrophages through inactivating TLR4/NF-κB signaling.

BACKGROUND: As a common chronic musculoskeletal condition, osteoarthritis (OA) presently lacks particular treatment strategies. The aim of this study was to examine how AT-III therapies affected macrophage repolarity in order to slow down the advancement of OA. METHODS: RAW264.7 macrophages were polarized to M1 subtypes then administered with different concentrations of AT-III. Immunofluorescence, qRT-PCR and flow cytometry were used to assess the polarization of the macrophages. The mechanism of AT-III repolarize macrophages was evaluated by western blot. Furthermore, the effects of macropha

Involvement of cholinergic processes in cholecystokinin (CCK) release [corrected] by luminal oleic acid.

Cholecystokinin (CCK) is an important bioactive peptide that stimulates pancreatic enzyme secretion. Circulating CCK is secreted from endocrine cells in the upper small intestine in response to various luminal stimuli and to vascular administration of gastrin releasing peptides. However, the mechanism of its release has not been fully elucidated. In the present study, the vascularly perfused duodenojejunum was isolated from male Wistar rats. The effects of luminal infusion of sodium oleate (2 or 0.4%) or intra-arterial infusion of neuromedin C(10(-7) M) with or without atropine and with a rece

Two faces of cholecystokinin: anxiety and schizophrenia.

It has been suggested that cholecystokinin (CCK), a gut-brain peptide found in high concentrations in the mammalian brain, might be implicated in the neurobiology of anxiety and panic disorder. The administration of CCK tetrapeptide induced panic attacks analogous to spontaneous ones in patients suffering from panic disorder and to a lesser degree in healthy volunteers. In animal models of anxiety, the pretreatment with CCK agonists and antagonists produced, respectively, anxiogenic- and anxiolytic-like action on the exploratory paradigms. On the other hand, CCK could also play a role in the p

Characteristics of 125I-Bolton-Hunter labelled cholecystokinin binding in human brain.

The high affinity binding of 125I-Bolton-Hunter labelled CCK-8 (125I-BHCCK) was studied in human brain using ligand binding and autoradiographic techniques. High levels of 125I-BHCCK binding were observed in cortex, striatum and cerebellum. In cerebellar membranes 125I-BHCCK binding was inhibited by CCK analogues with an order of potency, caerulein greater than CCK-8 greater than CCK-33 greater than gastrin 1-17 = CCK-8 non-sulphated. CCK-4 was inactive. It is suggested that in human brain 125I-BHCCK labels a population of CCK receptors similar to those observed in guinea pig brain. The proper

Pyloroplasty does not disrupt liquid phase gastric emptying or CCK-induced satiety.

To assess the hypothesis that cholecystokinin (CCK) induces satiety by constricting the pylorus and inhibiting the rate of gastric emptying, we impaired pyloric function in 8 experimental animals using a Heincke-Mikulicz pyloroplasty procedure. Liquid phase gastric emptying was measured with a double-sampling procedure. We found that pyloroplasty did not disrupt the pattern of gastric emptying of saline or nutrient under control conditions. CCK also showed emptying in both pyloroplasty and control animals. Finally, pyloroplasty did not affect the ability of CCK to induce satiety. We provide fu

[Effect of cholecystokinin-octapeptide on the motor function of the digestive tract of the plaice].

The influence of cholecystokinin-octapeptide on the bioelectrical features of gastro-intestinal motility in sea-water fish (plaice) was investigated in chronic experiments. I. v. or intracerebral application of the peptide led to inhibition of the stomach cardia motor activity, to intensification and sparsity of the tonic and peristaltic contractions of the stomach pyloric part, and to an increase in the intestine fore-part activity. Vagotomy prevented the effect of intracerebral application of cholecystokinin. Atropine prevented this effect of cholecystokinin irrespective of the mode of its a

Intracerebroventricular administration of sulphated cholecystokinin octapeptide induces anxiety-like behaviour in goldfish.

Sulphated cholecystokinin octapeptide (CCK-8s) is involved in feeding regulation as an anorexigenic neuropeptide in vertebrates. In rodents, i.c.v. administration of CCK-8s has been shown to affect not only feeding behaviour, but also psychomotor activity. However, there is still no information available concerning the psychophysiological effects of CCK-8s in goldfish. Therefore, in the present study, we examined the effect of synthetic goldfish (gf) CCK-8s on psychomotor activity in this species. Intracerebroventricular administration of gfCCK-8s at 0.1, 0.5 and 2.5 pmol g-1 body weight (BW)

Modulation of apomorphine-induced stereotyped behavior by cholecystokinin.

1. The goal was to verify if central or peripheral sulphated cholecystokinin octapeptide (CCK8) injections can modulate apomorphine (APO)-induced stereotyped behavior. Experiments were designed to determine the involvement of cholecystokinin receptor subtypes as well. 2. Animals which received CCK8 (0.0725, 0.145 and 14.5 nmol, icv) showed a significant (p < 0.05) decrease in APO (0.6 mg/kg, sc)-induced stereotyped behavior. 3. No other statistically significant difference was observed among groups. Since ip CCK8 (1.16 or 2.32 nmol/kg) injections did not interfere with APO-induced stereotypy,

Changes in tone of canine gallbladder measured by an electronic pneumatic barostat.

We developed an electronic pneumatic barostat to measure changes in gallbladder muscle tone in vivo independent of biliary flow dynamics. Dogs were prepared with a gallbladder cannula for insertion of barostat balloon and an inflatable balloon-occluder around the cystic duct. The barostat was validated to maintain a constant pressure of 2 +/- 0.4 (SE) mmHg within a plastic balloon of "infinite" compliance that filled the gallbladder lumen; changes in balloon volume reflected changes in wall tone because intraluminal pressure was kept constant. Gallbladder tone, as measured by the barostat ball

The stomach, cholecystokinin, and satiety.

The stomach of the rhesus monkey empties liquids in a fashion that varies with the character of the solutions. Physiological saline empties exponentially. Glucose solutions empty biphasically--rapidly for the first minutes, then slowly and proportionately to glucose concentration to deliver glucose calories through the pylorus at a regulated rate (0.4 kcal/min). This prolonged and regulated second phase of gastric emptying depends on intestinal inhibition of the stomach. Cholecystokinin (CCK), a hormone released by food in the intestine, is an inhibitor of gastric emptying. In vitro receptor a

Evidence for central neuropeptidergic control of rumination in sheep.

Effects of intracerebroventricular (ICV) vs. intravenous (IV) administration of tetragastrin, pentagastrin, CCK8 and gastrin 17 on rumination were investigated in conscious sheep. Administered at 26 pmoles/kg ICV both tetra and pentagastrin induced a premature short (15-27 min) period of rumination only 24 +/- 7 and 23 +/- 9 min after food distribution in place of 112 +/- 44 min in controls. Similar but less pronounced effects were observed for ICV administration of an equimolar dose of gastrin 17 whereas CCK8 did not promote an early period of rumination despite its anorectic effects. Dual effects of hydrocortisone on exocrine rat pancreas.

The acute and chronic effects of hydrocortisone on exocrine pancreatic function were examined in the isolated perfused rat pancreas. In the first part of this study, rats were given subcutaneous injections of hydrocortisone at doses of 1.25, 2.5, 5, and 10 mg/kg body wt once daily for 7 days. Trypsin and lipase secretion in response to 100 pM cholecystokinin-octapeptide was significantly increased in rats with the two highest doses of hydrocortisone compared with controls, irrespective of whether calculated as the total amount of stimulated output of enzymes or related to the secretion of enzy

Rapid appearance of intraventricularly administered neuropeptides in the peripheral circulation.

125I-Labeled cholecystokinin octapeptide ([125I]CCK-OP) diffuses rapidly by a non-carrier-mediated mechanism into the peripheral blood following injection into the lateral ventricle of a rabbit. In contrast, no [125I]CCK-OP was observed in the CSF following intravenous injection. This unidirectional free transport mechanism from CSF to blood may apply to other neuropeptides as well.

Effect of cholecystokinin on small intestinal motility in suncus murinus.

In a fasting gastrointestinal tract, a characteristic cyclical rhythmic migrating motor complex (MMC) occur that comprises of three phases: I, II, and III. Among these, phase III contractions propagate from the stomach to the lower intestine in mammals, including humans, dogs, and Suncus murinus (suncus). Apart from the phase III of MMC propagating from the stomach, during the gastric phase II, small intestine-originated strong contractions propagate to the lower small intestine; however, the mechanism of contractions originating in the small intestine has not been clarified. In this study, we
