G Protein-Coupled Receptors in Skin Aging.

Skin aging is a complex biological process affected by a plethora of intrinsic and extrinsic factors that alter cutaneous functions through the modulations of signaling pathways and responses. Expressed in various cell types and skin tissue layers, G protein-coupled receptors (GPCRs) play a vital role in regulating skin aging. We have cataloged 156 GPCRs expressed in the skin and reviewed their roles in skin aging, such as pigmentation, loss of elasticity, wrinkles, rough texture, and aging-associated skin disorders. By exploring the GPCRs found in the skin, it may be possible to develop new t

Mitochondria in skin health, aging, and disease.

The skin is a high turnover organ, and its constant renewal depends on the rapid proliferation of its progenitor cells. The energy requirement for these metabolically active cells is met by mitochondrial respiration, an ATP generating process driven by a series of protein complexes collectively known as the electron transport chain (ETC) that is located on the inner membrane of the mitochondria. However, reactive oxygen species (ROS) like superoxide, singlet oxygen, peroxides are inevitably produced during respiration and disrupt macromolecular and cellular structures if not quenched by the an

Werner syndrome: quantitative assessment of skin aging.

BACKGROUND: Werner syndrome (WS) is a rare autosomal recessive disorder characterized by premature aging in adults. Although not sufficient to diagnose WS, persistent short stature and alteration of the dentition are among the few early signs that appear at puberty and can lead to a suspected diagnosis. OBJECTIVE: The study aimed at quantifying the signs of WS skin aging through biophysical parameters to find new parameters to be applied together with clinical observations in order to diagnose the disease early. PATIENTS AND METHODS: The skin disorders induced by the disease were studied usi

Retinoid therapy for aging skin and acne.

Primary care physicians should be familiar with the effects and appropriate uses of retinoids. Topical tretinoin (Retin-A) can reverse photoaging of the skin, although some transient, undesirable side effects usually occur. In patients with acne vulgaris, topical tretinoin and systemic isotretinoin (Accutane) are the only agents that act upon the apparent underlying causes. Recurrence is unlikely after successful results are achieved. Chronic hypervitaminosis A presents insidiously, and physicians must maintain a high index of suspicion. Complete history taking should always include questions

TRPV1: A promising therapeutic target for skin aging and inflammatory skin diseases.

TRPV1 is a non-selective channel receptor widely expressed in skin tissues, including keratinocytes, peripheral sensory nerve fibers and immune cells. It is activated by a variety of exogenous or endogenous inflammatory mediators, triggering neuropeptide release and neurogenic inflammatory response. Previous studies have shown that TRPV1 is closely related to the occurrence and/or development of skin aging and various chronic inflammatory skin diseases, such as psoriasis, atopic dermatitis, rosacea, herpes zoster, allergic contact dermatitis and prurigo nodularis. This review summarizes the st

Artificial Intelligence-Driven Skin Aging Simulation as a Novel Skin Cancer Prevention.

INTRODUCTION: Skin cancer, a prevalent cancer type among fair-skinned patients globally, poses a relevant public health concern due to rising incidence rates. Ultraviolet (UV) radiation poses a major risk factor for skin cancer. However, intentional tanning associated with sunburns remains a common practice, notably among female adults. Appropriate prevention campaigns targeting children and adolescents are needed to improve sun protection behavior particularly in these age groups. The aim of our study was to investigate if an AI-based simulation of facial skin aging can enhance sun protection

Hyaluronic acid: A key molecule in skin aging.

SKIN AGING IS A MULTIFACTORIAL PROCESS CONSISTING OF TWO DISTINCT AND INDEPENDENT MECHANISMS: intrinsic and extrinsic aging. Youthful skin retains its turgor, resilience and pliability, among others, due to its high content of water. Daily external injury, in addition to the normal process of aging, causes loss of moisture. The key molecule involved in skin moisture is hyaluronic acid (HA) that has unique capacity in retaining water. There are multiple sites for the control of HA synthesis, deposition, cell and protein association and degradation, reflecting the complexity of HA metabolism. Th

Genomic-driven insights into changes in aging skin.

Like all tissues, the skin ages due to the passage of time (chronologic aging). However, skin is also exposed to external insults, such as sunlight. Aging due to chronic sun exposure (photoaging) is characterized clinically by wrinkling, dyspigmentation and other changes. Chronologic and photoaging of skin have been distinguished at the structural, cellular and molecular levels. However, many underlying mechanisms remain a mystery. Recent sequencing of the human genome and development of genome-wide microarray platforms now permit analysis of skin aging at the level of gene expression. Analysi

Common skin disorders of aging: diagnosis and treatment.

The incidence of skin disorders increases with age because of changes in skin structures and a lifetime accumulation of environmental insults. Photoaging, resulting from exposure to harmful ultraviolet wavelengths in sunlight, is responsible for many common skin disorders, from wrinkles to melanoma. Older patients also see their physicians with complaints of various types of dermatitis, including contact and seborrheic. Although skin disorders vary in severity, they all adversely affect an older patient's health and/or quality of life. Early recognition, prevention, and treatment of skin disea

Structural and functional changes of normal aging skin.

Solar-induced cutaneous changes are more prevalent and profound in older persons and, thus, are often inappropriately attributed to the aging process, per se. Structural and functional alterations caused by intrinsic aging and independent of environmental insults are now recognized in the skin of elderly individuals. Structurally the aged epidermis likely becomes thinner, the corneocytes become less adherent to one another, and there is flattening of the dermoepidermal interface. The number of melanocytes and Langerhans cells is decreased. The dermis becomes atrophic and it is relatively acell

Understanding exosomes in facial esthetics and skin aging.

BACKGROUND: Skin aging is a multifactorial process mediated by intrinsic (genetic and metabolic) and extrinsic (environmental) factors leading to functional and structural deterioration, including wrinkles, loss of collagen and elastin, as well as various pigmentation disorders. Exosomes, the smallest subset of extracellular vesicles, have emerged as potent cell-free regulators of intercellular communication with significant regenerative potential. This scoping review synthesized available in vitro, in vivo, and clinical evidence investigating the role of exosomes in facial esthetics and skin

Co-regulatory mechanisms of skin and hair aging.

As a cutaneous appendage, the hair follicle is tightly associated with the skin during growth, pigmentation, and aging. These two interconnected tissues share evolutionarily conserved regulatory mechanisms-including stem cell niche signaling, inflammatory cascades, and oxidative stress-and collectively function as hallmarks of systemic organismal aging. Hair abnormalities are well-established concomitant manifestations of aged skin. The crosstalk between skin aging and hair aging is particularly pronounced in several hereditary progeroid disorders, which are typified by concurrent premature ag

Cellular Senescence in Human Skin Aging: Leveraging Senotherapeutics.

BACKGROUND: As the largest organ in the human body, the skin is continuously exposed to intrinsic and extrinsic stimuli that impact its functionality and morphology with aging. Skin aging entails dysregulation of skin cells and loss, fragmentation, or fragility of extracellular matrix fibers that are manifested macroscopically by wrinkling, laxity, and pigmentary abnormalities. Age-related skin changes are the focus of many surgical and nonsurgical treatments aimed at improving overall skin appearance and health. SUMMARY: As a hallmark of aging, cellular senescence, an essentially irreversibl

Two newly developed indices evaluating facial skin aging.

BACKGROUND: Skin assessment methodologies have focused mainly on intuitive aging characteristics, including facial wrinkles and pigmented spots, and usually adopt pattern recognition algorithms. Recently, distinct methods of interpreting skin aging, such as the detection of facial landmarks and age prediction using machine learning techniques, have been conducted. MATERIALS AND METHODS: We defined two indices that represent the severity of facial aging. The first index was the ratio of the bizygomatic distance and bigonial distance. The second index was the ratio of the degrees of the near ma

Immunological Aspects of Skin Aging in Atopic Dermatitis.

The cutaneous immune response is important for the regulation of skin aging well as for the development of immune-mediated skin diseases. Aging of the human skin undergoes immunosenescence with immunological alterations and can be affected by environmental stressors and internal factors, thus leading to various epidermal barrier abnormalities. The dysfunctional epidermal barrier, immune dysregulation, and skin dysbiosis in the advanced age, together with the genetic factors, facilitate the late onset of atopic dermatitis (AD) in the elderly, whose cases have recently been on the rise.
