Effects of Tiagabine on Slow Wave Sleep and Arousal Threshold in Patients With Obstructive Sleep Apnea.

INTRODUCTION: Obstructive sleep apnea (OSA) severity is markedly reduced during slow-wave sleep (SWS) even in patients with a severe disease. The reason for this improvement is uncertain but likely relates to non-anatomical factors (i.e. reduced arousability, chemosensitivity, and increased dilator muscle activity). The anticonvulsant tiagabine produces a dose-dependent increase in SWS in subjects without OSA. This study aimed to test the hypothesis that tiagabine would reduce OSA severity by raising the overall arousal threshold during sleep. AIMS AND METHODS: After a baseline physiology nig

Spatially Extensive LFP Correlations Identify Slow-Wave Sleep in Marmoset Sensorimotor Cortex.

Identifying neural signatures of slow-wave sleep (SWS) is important for a number of reasons including diagnosing potential sleep disorders and examining its role in memory consolidation ( Diekelmann and Born, 2010; Klinzing et al., 2019; Brodt et al., 2023). Studies of sleep in the common marmoset (Callithrix jacchus) have revealed similarities to humans and other nonhuman primates, including distinct sleep stages ( Crofts et al., 2001) and diurnal sleep patterns ( Hoffmann et al., 2012). EEG microstates are correlated with brain functional networks during slow-wave sleep.

Electroencephalography (EEG) microstates have been extensively studied in wakefulness and have been described as the "atoms of thought". Previous studies of EEG have found four microstates, i.e., microstates A, B, C and D, that are consistent among participants across the lifespan during the resting state. Studies using simultaneous EEG and functional magnetic resonance imaging (fMRI) have provided evidence for correlations between EEG microstates and fMRI networks during the resting state. Microstates have also been found during non-rapid eye movement (NREM) sleep. Slow-wave sleep (SWS) is co

Slow-wave sleep deficiency and enhancement: implications for insomnia and its management.

In humans, slow-wave sleep (SWS) consists of stages 3 and 4 of non rapid eye movement (nonREM) sleep. The low-frequency, high-amplitude slow waves that dominate the electroencephalogram (EEG) during SWS can be quantified as slow-wave activity (SWA). SWS and SWA are regulated very accurately in response to variations in the duration and intensity of wakefulness and sleep. SWA declines more or less independently of circadian phase during the course of a sleep episode, indicating that it is primarily under homeostatic rather than circadian control. An age-related decline in SWS and SWA is well es

The GABAergic parafacial zone is a medullary slow wave sleep-promoting center.

Work in animals and humans has suggested the existence of a slow wave sleep (SWS)-promoting/electroencephalogram (EEG)-synchronizing center in the mammalian lower brainstem. Although sleep-active GABAergic neurons in the medullary parafacial zone (PZ) are needed for normal SWS, it remains unclear whether these neurons can initiate and maintain SWS or EEG slow-wave activity (SWA) in behaving mice. We used genetically targeted activation and optogenetically based mapping to examine the downstream circuitry engaged by SWS-promoting PZ neurons, and we found that this circuit uniquely and potently

Fragmentation of slow wave sleep after onset of complete locked-in state.

Locked-in syndrome (LIS) as a result of brainstem lesions or progressive neurodegenerative disorders, such as amyotrophic lateral sclerosis (ALS), is a severe medical condition in which a person is fully conscious but unable to move or talk. LIS can transition into complete locked-in syndrome (CLIS) in which residual abilities to communicate through muscle twitches are entirely lost. It is unknown how CLIS affects circadian rhythm and sleep/wake patterns. Here we report a 39-year-old ALS patient who transitioned from LIS to CLIS while brain activity was continuously recorded using electrocorti

Continuous spike-waves during slow waves sleep. A clinical and electroencephalographic study in fifteen children.

We report on the clinical and EEG features of 15 patients with the syndrome of "continuous spike waves during slow wave sleep" (CSWSS). The differential diagnosis of CSWSS includes benign epilepsy of childhood with centro-temporal spikes, and Landau-Kleffner and Lennox-Gastaut syndromes. We found normal CT and MRI features in 6 cases, periventricular leukomalacia with and without diffuse brain atrophy in 4 cases and hydrocephalus in 1 case. There was no association between specific neurological findings and CSWSS. Nine of our cases had relatively focal discharges, like some cases from the lite

Slow-wave sleep in the TPJ is linked to individual differences in implicit bias.

Implicit biases can profoundly impact social interactions and decision-making processes. Here, we investigate whether the heterogeneity of implicit racial bias can be explained by interindividual differences in the intracortical distribution of slow-wave activity (SWA), a marker of sleep depth. Using high-density EEG, we recorded sleep data from 52 healthy adults (27 females) during a typical night's sleep in their homes. Implicit racial bias was measured using the race Implicit Association Test (IAT). Whole-brain corrected source localization analysis identified a significant association betw

Cortical thinning explains changes in sleep slow waves during adulthood.

Sleep slow waves (SWs) change considerably throughout normal aging. In humans, SWs are generated and propagate on a structural backbone of highly interconnected cortical regions that form most of the default mode network, such as the insula, cingulate cortices, temporal lobe, parietal lobe, and medial frontal lobe. Regions in this network undergo cortical thinning and breakdown in structural and functional connectivity over the course of normal aging. In this study, we investigated how changes in cortical thickness (CT), a measure of gray matter integrity, are involved in modifications of slee

The impact of visual perceptual learning on sleep and local slow-wave initiation.

During non-rapid eye movement (NREM) sleep, a global decrease in synaptic strength associated with slow waves (SWs) would enhance signal-to-noise ratio of neural responses during subsequent wakefulness. To test this prediction, 32 human volunteers were trained to a coarse orientation discrimination task, in either the morning or evening. They were retested after 8 h of wakefulness or sleep, respectively. Performance was enhanced only after a night of sleep, in the absence of any change in the abundance of NREM SWs but in proportion to the number of SWs "initiated" in lateral occipital areas du

Sleep-like slow waves in ADHD: Regional specificity in combined type.

The diagnosis of Attention Deficit/Hyperactivity Disorder (ADHD) remains challenging due to the lack of objective neurophysiological markers, particularly for distinguishing clinical subtypes. Here, we investigated whether sleep-like slow wave activity (SWA) during wakefulness could serve as a subtype-specific neural marker for ADHD. Using EEG recordings during the Attention Network Test (ANT), we analyzed behavioral and neurophysiological data from 120 children, including healthy controls (HC), inattentive-type (ADHD-I), and combined-type (ADHD-C) participants. Our research findings indicated

Slow wave sleep is associated with forgetting in patients with temporal lobe epilepsy.

While time spent in slow wave sleep (SWS) after learning promotes memory consolidation in the healthy brain, it is unclear if the same benefit is obtained in patients with temporal lobe epilepsy (TLE). Interictal epileptiform discharges (IEDs) are potentiated during SWS and thus may disrupt memory consolidation processes thought to depend on hippocampal-neocortical interactions. Here, we explored the relationship between SWS, IEDs, and overnight forgetting in patients with TLE. Nineteen patients with TLE studied object-scene pairs and memory was tested across a day of wakefulness (6 hrs) and a

Dynamics of slow wave sleep in advanced Parkinson's disease: An entropy-based study.

Parkinson's disease (PD) can progress to an advanced stage characterized by motor fluctuations and or levodopa-induced dyskinesia (LID). Although both motor fluctuations and LID reflect altered dopaminergic reactivity, LID has been specifically linked to disrupted synaptic plasticity. Slow wave sleep (N3) reflects homeostatic processes and may offer neurophysiological markers related to the motor phenotype in PD. This study tested whether overnight dynamics of N3 slow-wave activity (SWA) and cortical complexity, i.e., sample entropy (SampEn) differ between PD patients with and without dyskines

Anatomical markers of sleep slow wave activity derived from structural magnetic resonance images.

Sleep studies often observe differences in slow wave activity (SWA) during non-rapid eye movement sleep between subjects. This study investigates to what extent these absolute differences in SWA can be explained with differences in grey matter volume, white matter volume or the thickness of skull and outer liquor rooms. To do this, we selected the 10-min interval showing maximal SWA of 20 young adult subjects and correlated these values lobe-wise with grey matter, skull and liquor thickness and globally with white matter as well as segments of the corpus callosum. Whereas grey matter, skull th

Relationship between locus coeruleus and slow-wave sleep in aging and Alzheimer's disease.

INTRODUCTION: Sleep disruption, particularly loss of slow-wave sleep (SWS), is common in Alzheimer's disease (AD), but its neurobiological underpinnings remain unclear. We investigated whether locus coeruleus (LC) integrity relates to SWS across the AD continuum and whether sex and perivascular spaces (PVSs) modify these associations. METHODS: In a cohort (11 controls, 30 mild cognitive impairment, 17 AD dementia) we combined overnight polysomnography with LC-sensitive magnetic resonance imaging of the LC, basal ganglia and centrum semiovale PVS ratings, and cerebrospinal fluid noradrenaline.

Mirtazapine and Slow-Wave Sleep: A Focused Narrative Review of Human Polysomnographic Evidence.

Sleep disturbance is a common feature of depressive disorders and may be influenced by antidepressant-associated alterations in sleep architecture. Mirtazapine is a noradrenergic and specific serotonergic antidepressant with a distinctive pharmacologic profile involving histamine H1 receptor antagonism, alpha-2 adrenergic antagonism, and serotonergic receptor modulation, which may contribute to its effects on sleep regulation. This focused narrative review examines the neurobiological mechanisms through which mirtazapine may influence sleep architecture and summarizes available human polysomno
