[The use of zolpidem in the treatment of acute and chronic insomnia].

The pharmacological features and practice of using the benzodiazepine receptor agonist zolpidem are analyzed. The issues of definition and clinical significance of insomnia in medical practice and the grounds for choosing a class of hypnotic drugs for its treatment are discussed. The main pharmacokinetic and pharmacodynamic properties of zolpidem are described. The data of studies on its effect on subjective and objective sleep parameters, safety and side effects, features of administration to different groups of patients and long-term use (up to 12 months) are presented.

When Should Pharmacological Interventions for Insomnia Be Recommended?

This commentary on a case describes how social determinants of health also contribute to insomnia and then suggests how to balance risks and benefits of different strategies for managing chronic insomnia. Behaviorally induced insufficient sleep syndrome can exacerbate morning side effects of prescription sleep aids, and there are potentially serious long-term risks (eg, dementia, falls, death) associated with chronic benzodiazepine use. Before trying sleeping pills, chronic insomnia should be treated with cognitive behavioral therapy.

Extracts of Prunella vulgaris Enhanced Pentobarbital-Induced Sleeping Behavior in Mice Potentially via Adenosine A2A Receptor Activity.

The increasing prevalence of sleep dysregulation cases has prompted the search for effective and safe sleep-enhancing agents. Numerous medications used in the treatment of sleep disorders function by enhancing γ-aminobutyric acid neurotransmitter activity. Unfortunately, these substances may induce significant adverse effects in chronic users, such as dependence and motor behavior impairments. Consequently, there is a growing interest in exploring therapeutic sleep-enhancing agents derived from natural sources, with the anticipation of causing less severe side effects. Prunella vulgaris (PV),

Long-term sleep disturbance and prescription sleep aid use among cancer survivors in the United States.

PURPOSE: Insomnia and related sleep disorders are common complaints among cancer patients, and use of prescription sleep aids can be high in the treatment setting. The prevalence of sleep disturbance and prescription sleep aid use in the community-dwelling cancer survivorship population, however, remains relatively unexplored. We aim to ascertain the extent to which a cancer diagnosis is associated with sleep disturbances and prescription sleep aid use as measured in several cross sections of individuals across multiple disease sites and time since cancer diagnosis. METHODS: We used data from

Suvorexant for the treatment of primary insomnia: A systematic review and meta-analysis.

Suvorexant is a dual orexin receptor agonist and is currently approved for the treatment of insomnia in the United States and Japan. We conducted a systematic review and meta-analysis to assess the efficacy and safety of suvorexant for the treatment of primary insomnia. We searched PubMed, EMBASE, and the Cochrane central register of controlled trials, contacted a relevant pharmaceutical company, and accessed websites of the U.S. Food and Drug Administration (FDA) and Pharmaceuticals and Medical Devices Agency (PMDA) for published and unpublished data. A total of four randomized trials involvi

Pharmacological agents for improving sleep quality at high altitude: a systematic review and meta-analysis of randomized controlled trials.

Several hypnotic agents commonly recommended for improving sleep at sea level are discouraged at high altitude. We aimed to evaluate the efficacy and safety of drugs prescribed for improving sleep quality in patients with acute exposure to high altitudes by conducting a systematic review and meta-analysis. An electronic search was executed for randomized controlled trials comparing drug treatments with placebo and no-treatment conditions, which used objective sleep parameters or subjective sleep quality evaluations. Eight studies (152 participants) were included in the meta-analysis and involv

Fermented rice germ extract with high concentration of gamma-aminobutyric acid enhances pentobarbital-induced sleep via GABAergic system in rats.

In modern societies, insomnia stands out as a widespread health concern. Gamma-aminobutyric acid (GABA), an inhibitory neurotransmitter, can exert hypnotic effects. However, despite various clinical trials conducted, scientific evidence regarding the impact of ingested GABA on sleep remains unclear. Therefore, in this study, we investigated the sedative effects of fermented rice germ (RG30) extract with a high GABA concentration through electroencephalography (EEG) analysis in a pentobarbital-induced sleep animal model. The exploration into the neural basis of these positive effects involved e

Copper-cerium layer double hydroxide as a novel tool for melatonin detection in sleep therapy tablets.

Sleep disorders are among the most common health concerns leading to health disorders. To address this problem, a copper-cerium layered double hydroxide (Cu-Ce LDH) was developed as an efficient sensing material for the electrochemical detection of melatonin in sleeping pills. The Cu-Ce LDH enhanced oxidation by adsorbing melatonin through π-π and electrostatic interactions, facilitating electron transfer and improving conductivity from Ce3+ sites. The mixed valency states of Cu and Ce facilitated improved conductivity and high surface area. The enhanced electrochemical behaviour of Cu-Ce LDH

Residual Effects of Acute and Subchronic Zolpidem Treatments on Attentional Processes in Aged Female Rats.

RATIONAL: Zolpidem, a hypnotic Z-drug commonly prescribed to promote sleep, is predominantly used over the age of 50. In this elderly population, adverse behavioral disturbances, impaired driving performance, and an increased risk of falls have been frequently reported. These concerns have raised questions about the residual adverse effects of zolpidem in older adults, particularly on cognitive processes such as executive and attentional functions. OBJECTIVES: This study aimed to investigate the residual effects of zolpidem on attentional performance following either acute or subchronic admin

The effects of cannabidiol on sleep disturbances within a sample of high trait worriers: A double-blind, randomized placebo controlled trial.

Cannabidiol (CBD), a nonintoxicating molecule derived from the cannabis plant, is garnering attention as a novel sleep aid despite a dearth of empirical literature supporting its efficacy for sleep-related indications. The present study aimed to address this gap. Participants were 63 individuals (Mage = 29.27; SDage = 9.58) reporting elevated trait worry who were randomly assigned to take 300 mg CBD, 50 mg CBD, or placebo daily for 2 weeks. Results suggested 300 mg CBD decreased sleep disturbances and sleep quality significantly more than 50 mg CBD (B = -0.39, t = -2.59, p < .05, d = 0.08), bu

Zolpidem Withdrawal Delirium Developed as Severe Psychiatric and Physical Symptoms in a Patient With a History of Ovarian Cancer: A Case Report and Literature Review.

BACKGROUND: Zolpidem is a common medication for sleep disorders prescribed during or after cancer treatment. However, the possible risk of zolpidem withdrawal delirium does not seem to be well recognized by medical staff engaged in cancer treatment. CASE PRESENTATION: A female in her 50s with a diagnosis of ovarian cancer underwent bilateral oophorectomy 6 years earlier. No recurrence was observed after treatment with anticancer drugs. She had been prescribed zolpidem (10 mg/day, oral) by her attending physician for approximately 5 years, although she often took it 2-3 times the dose when her

Orexin 2 Receptor Antagonism is Sufficient to Promote NREM and REM Sleep from Mouse to Man.

Orexin neuropeptides regulate sleep/wake through orexin receptors (OX1R, OX2R); OX2R is the predominant mediator of arousal promotion. The potential for single OX2R antagonism to effectively promote sleep has yet to be demonstrated in humans. MK-1064 is an OX2R-single antagonist. Preclinically, MK-1064 promotes sleep and increases both rapid eye movement (REM) and non-REM (NREM) sleep in rats at OX2R occupancies higher than the range observed for dual orexin receptor antagonists. Similar to dual antagonists, MK-1064 increases NREM and REM sleep in dogs without inducing cataplexy. Two Phase I s

Effect of improving sleep quality the night before surgery with zolpidem on postoperative gastrointestinal function in patients undergoing laparoscopic partial colorectal resection: a randomized, double-blind, controlled trial.

BACKGROUND: Sleep is one of the basic physiological needs of human beings. Preoperative sleep disorders are associated with poor prognosis in surgical patients, and sleep disorders have been shown to be one of the risk factors for gastrointestinal dysfunction. However, there are now few studies to investigate whether improving preoperative sleep disorders can promote the recovery of postoperative gastrointestinal function. This study aimed to investigate the effects and significance of improving preoperative sleep quality with zolpidem on postoperative gastrointestinal function. METHODS: In t
