Sex-Specific Retinal Anomalies Induced by Chronic Social Defeat Stress in Mice.

Major depressive disorder (MDD) is one of the most common consequences of chronic stress. Still, there is currently no reliable biomarker to detect individuals at risk to develop the disease. Recently, the retina emerged as an effective way to investigate psychiatric disorders using the electroretinogram (ERG). In this study, cone and rod ERGs were performed in male and female C57BL/6 mice before and after chronic social defeat stress (CSDS). Mice were then divided as susceptible or resilient to stress. Our results suggest that CSDS reduces the amplitude of both oscillatory potentials and a-wa

Decreased kynurenine pathway potentiate resilience to social defeat effect on cocaine reward.

The kynurenine (KYN) pathway of tryptophan (TRP) degradation is activated by stress and inflammatory factors. It is now well established that social stress induces the activation of the immune system, with central inflammation and KYN metabolism being two of the main factors linking stress with depression. The aim of the present study was to evaluate the long-lasting changes in the KYN pathway induced by social defeat (SD) associated with the resilience or susceptibility to an increase in the conditioned rewarding effects of cocaine. Mice were exposed to repeated SD and 3 weeks later, a condit

Social defeat stress-induced hyperalgesia is mediated by nav 1.8+ nociceptive fibers.

Recently the voltage-gated sodium (Nav) channels began to be studied as possible targets for analgesic drugs. In addition, specific Nav 1.8 blockers are currently being used to treat some types of chronic pain pathologies such as neuropathies and fibromyalgia. Nav 1.8+ fibers convey nociceptive information to brain structures belonging to the limbic system, which is involved in the pathophysiology of major depressive disorders. From this, using a model of chronic social defeat stress (SDS) and intrathecal injections of Nav 1.8 antisense, this study investigated the possible involvement of Nav

A diet enriched with curcumin promotes resilience to chronic social defeat stress.

Chronic exposure to stress is a well-known risk factor for the development of mood and anxiety disorders. Promoting resilience to stress may prevent the development of these disorders, but resilience-enhancing compounds are not yet clinically available. One compound that has shown promise in the clinical setting is curcumin, a polyphenol compound found in the rhizome of the turmeric plant (Curcuma longa) with known anti-inflammatory and antidepressant properties. Here, we tested the efficacy of 1.5% dietary curcumin at promoting resilience to chronic social defeat stress (CSDS) in 129/SvEv mic

Social defeat stress exacerbates the blood abnormalities in Opisthorchis felineus-infected mice.

A model of chronic opisthorchiasis combined with social stress is examined; this situation is more likely for humans and animals than a separate impact of the infectious factor. For this purpose, we evaluated the effects of Opisthorchis felineus ("OP" group) and 30-day social stress (confrontations between males, "SS" group) alone and in combination ("OP + SS" group) in inbred C57BL/6 male mice and compared these effects according to the parameters listed below. The animals exposed to neither factor formed the control group ("CON"). All animals were assayed for blood biochemical parameters, ch

Molecular mechanisms of repeated social defeat-induced glucocorticoid resistance: Role of microRNA.

Glucocorticoid (GC) resistance is a severe problem associated with various inflammatory diseases. Previous studies have shown that repeated social stress induces GC resistance in innate immune cells, but the underlying molecular mechanisms have not been fully elucidated. Therefore, the purpose of this study was to examine potential underlying molecular mechanism(s) of repeated social defeat (RSD) stress on GC resistance in splenic macrophages. It was hypothesized that mRNA expression of receptors for GC and nuclear translocating-associated regulators in splenic macrophages would be affected by

A single social defeat reduces aggression in a highly aggressive strain of Drosophila.

Genes and prior experience both influence the behavior of animals, but the relative contribution of each to fighting behavior in Drosophila remains unclear. To address this issue, we bred hyperaggressive flies by selecting winners of fights over 34-37 generations. Males of this strain initiate fights sooner, retaliate more often, and regularly defeat opponents from the nonselected parent Canton-S strain. After a defeat, however, these highly aggressive flies lose their second fights against socially naïve counterparts. Defeated flies also lunge and retaliate less after experiencing a loss, sug

Social defeat stress and escalation of cocaine and alcohol consumption: Focus on CRF.

Both the ostensibly aversive effects of unpredictable episodes of social stress and the intensely rewarding effects of drugs of abuse activate the mesocorticolimbic dopamine systems. Significant neuroadaptations in interacting stress and reward neurocircuitry may underlie the striking connection between stress and substance use disorders. In rodent models, recurring intermittent exposure to social defeat stress appears to produce a distinct profile of neuroadaptations that translates most readily to the repercussions of social stress in humans. In the present review, preclinical rodent models

Anorexic behavior and elevation of hypothalamic malonyl-CoA in socially defeated rats.

Suppression of body weight and eating disorders, such as anorexia, are one of the major symptoms of psychiatric disorders such as depression. However, the mechanisms of weight loss and reduced appetite in depressive patients and in animal models of depression are largely unknown. In this study, we characterized the mechanism of anorexia resulting from depression using socially defeated rats as an animal model of depression. Socially defeated rats showed suppressed body weight gain, enlarged adrenal glands, decreased home cage activity, decreased food intake, and increased immobility in the for

Modeling vicarious social defeat stress in rodents for investigating emotional stress-induced depression.

Vicarious trauma, a form of emotional stress experienced by observing the suffering of others has gained prominence as a major contributor to depression and other stress-related disorders. Traditional animal models of depression such as Chronic Social Defeat Stress (CSDS) focus on physical stressors but fail to replicate the psychological dimensions of vicarious trauma. The Vicarious Social Defeat Stress (VSDS) model fills this gap by inducing emotional stress without physical injury. This enables us to investigate the neural, behavioral and molecular mechanisms unique to emotional stress-indu

Social defeat stress impairs systemic iron metabolism by activating the hepcidin-ferroportin axis.

Chronic psychological stress has been reported to decrease circulating iron concentrations and impair hematopoiesis. However, the underlying mechanisms remain unclear. This study aimed to investigate the effects of psychological stress on biological iron metabolism by using the social defeat stress (SDS) model, a widely used model of depression. Compared with control mice, mice subjected to SDS (SDS mice) had lower social interaction (SI) behavior. The SDS mice also showed impaired hematopoiesis, as evidenced by reduced circulating red blood cell counts, elevated reticulocyte counts, and decre

T-lymphocyte tyrosine hydroxylase regulates TH17 T-lymphocytes during repeated social defeat stress.

Posttraumatic stress disorder (PTSD) is a debilitating psychiatric disorder which results in deleterious changes to psychological and physical health. Patients with PTSD are especially susceptible to life-threatening co-morbid inflammation-driven pathologies, such as autoimmunity, while also demonstrating increased T-helper 17 (TH17) lymphocyte-driven inflammation. While the exact mechanism of this increased inflammation is unknown, overactivity of the sympathetic nervous system is a hallmark of PTSD. Neurotransmitters of the sympathetic nervous system (i.e., catecholamines) can alter T-lympho

Ethanol intake in male mice exposed to social defeat: Environmental enrichment potentiates resilience.

Large preclinical evidence shows that exposure to social defeat (SD) increases vulnerability to drug abuse, increasing the consumption of ethanol. However, not all subjects are equally affected by the changes induced by stress. Previous reports have evidenced that the resilient phenotype to depressive-like behaviors after SD is associated with the resistant phenotype to cocaine-increased rewarding effects and the smaller neuroinflammatory response. The aim of the present study was to further clarify whether the resilient profile to depressive-like behavior also predicts a protection against th

Oxytocin reverses ethanol consumption and neuroinflammation induced by social defeat in male mice.

Oxytocin (OXT) modulates social interactions, attenuates stressful responses and can decrease drug-seeking and taking behaviors. In previous studies, we observed that social defeat (SD) induced a long-lasting increase in ethanol intake and neuroinflammation in male mice. We also know that OXT blocks the increase in cocaine reward induced by SD. Therefore, in the present study we aimed to evaluate the effect of 1 mg/kg of OXT administered 30 min before each episode of SD on ethanol consumption and the neuroinflammatory response in adult male mice. Three weeks after the last SD, mice underwent o

Maternal Exposure to Witnessed Social Defeat Stress Induces Sex-Specific Disruptions in Juvenile Social Play Behavior in Rats.

Stressing rodent dams during lactation can impair offspring neurodevelopment, altering behavioral and physiological outcomes. Witnessed social defeat stress (WSDS) is a paradigm in which a juvenile or adult rodent observes aggressive encounters, affecting stress physiology and behavior. We developed a novel early-life stress model by exposing lactating rat dams to WSDS and assessed its impact on juvenile social behavior and stress-related physiology. At postnatal day 24, same-sex, age-matched pairs of offspring were tested for social play by scoring pouncing (play initiation), pinning, chasing

Standing strong: Effects of social dominance on neural activity and defeat-induced social avoidance in female Syrian hamsters.

Social dominance modulates stress vulnerability, yet the neural mechanisms by which dominance status alters responses to stress in females remain poorly understood. This study aimed to determine whether activation of neurons in the posterior medial amygdala (MeP) and ventral medial prefrontal cortex (vmPFC) was associated with reduced stress vulnerability in dominant female Syrian hamsters. We created dominance relationships in hamsters, exposed animals to social defeat stress, tested animals for stress-induced social avoidant responses, and collected tissue for c-Fos immunohistochemistry. Cannabinoid type 2 receptors play a crucial role in social defeat-induced depression.

BACKGROUND: The endocannabinoid system plays a crucial role in regulating mood, but the specific involvement of cannabinoid receptor type 2 (CB2R) in depression remains poorly understood. Similarly, the mechanisms by which electroacupuncture (EA) provides therapeutic benefits for depression are not clearly defined. This research aims to explore the function of CB2R in depression and examine if the therapeutic effects of EA are associated with the hippocampal CB2R system. METHODS: Mice experiencing social defeat stress (SDS) were used to model depression and anxiety behaviors. We quantified hi

The effects of propranolol and corticosterone on susceptibility priming in a mouse model of social defeat stress.

Animal models for stress-related mood disorders aim to mirror the diverse spectrum of stress responses observed in humans, which ultimately determine an individual's susceptibility or capacity to cope effectively with the stressor. The acute social defeat stress (ASDS) protocol allows a rapid phenotypic classification of mice subjected to social stress to interrogate the early neural patterns beyond divergent stress outcomes. Although ASDS alone does not cause chronic depression-like behaviors, it "primes" the mouse to be highly susceptible to a subsequent, subthreshold stress (StSDS). Here, w
