Reflex and direct effects of sodium cyanide in anesthetized rats.

Small doses of NaCN (250 micrograms.kg-1) intravenously injected into normal anesthetized Wistar rats, provoked immediate hyperventilation, bradycardia and systemic hypotension. These effects resulted mostly from peripheral chemoreceptor (PCR) stimulation. In totally baro- and chemodenervated rats (CDN), the drug induced hypoventilation, reduction in O2 consumption, prolonged bradycardia and systemic hypotension, characteristics of direct intoxication. In hyperoxic rats, hyperventilation was reduced. Bradycardia was suppressed with reduction in the systemic hypotension. These modifications aro

Failure of cyanide to antagonize sodium nitroprusside in vivo in dogs.

The role of cyanide (CN) in the mechanism of action of sodium nitroprusside (SNP) has not been well defined. Some authors consider accumulation of CN in muscle to be the principal factor responsible for resistance to SNP when given to induce hypotension. To test this hypothesis, SNP was infused in 12 anesthetized dogs. Mean aortic blood pressure was reduced 50% and dogs were maintained at this pressure for 5 minutes. After recovery, eight of the dogs (group 1) received a potassium cyanide (KCN) infusion at a rate of 0.0125 mg/kg/min for 180 minutes; the other four dogs (group 2) were given sal

Effects of acetylcholine and sodium cyanide on cat carotid baroreceptors.

1 The effects of intracarotid (i.a.) injections of acetylcholine (ACh) and sodium cyanide (NaCN) on baroreceptor activity recorded from the sinus nerve have been investigated in cats anaesthetized with pentobarbitone.2 Two types of baroreceptor unit were recorded. The predominant type discharged at least 3 to 4 spikes per pulse wave at normal BP; they are referred to as ;polyspike' units and may have been associated with A fibres. The other type discharged a maximum of 1 to 3 spikes per pulse wave, even at high BP; they are referred to as ;few-spike' units and may have been from C fibres.3 NaC

Cyanide intoxication in sheep: enhancement of efficacy of sodium nitrite, sodium thiosulfate, and cobaltous chloride.

For treatment of cyanide intoxication of ruminants, the present recommended doses of sodium nitrite (5 mg/kg of body weight) and sodium thiosulfate (25 to 50 mg/kg) are smaller than those recommended for other animals; the decrease is partially attributed to the greater susceptibility of ruminants to the toxic effects of sodium nitrite. Based on the high tissue concentration and activity rate of rhodanese in ruminants, sulfur donors such as sodium thiosulfate could be utilized more efficaciously. Doses of sodium nitrite and sodium thiosulfate (up to 22 and 660 mg/kg, respectively) were evaluat

Sodium cyanide antagonism of the vasodilator action of sodium nitroprusside in theisolated rabbit aortic strip.

Resistance to sodium nitroprusside (SNP) is uncommon, but its occurrence has led to massive overdoses of SNP and sometimes death. To examine the mechanism responsible for resistance, aortic smooth muscle strips were prepared and dose-response curves for norepinephrine (NE) obtained. SNP alone caused a shift of the dose-response curve for NE to the right. However, this shift was less when the strips were exposed to both SNP and sodium cyanide (CN-). When CN- alone was added to the aortic strips, the response to NE was unchanged. In a further group of aortic muscle strips first contracted with N

Cyanide release from sodium nitroprusside during coronary bypass in hypothermia.

Erythrocyte cyanide levels were determined by a sensitive fluorimetric method on four occasions during coronary bypass in hypothermia in 18 consecutive patients treated with sodium nitroprusside (SNP) with an infusion rate less than 1 microgram x kg-1 x min-1. Every second patient received the cyanide antidote thiosulphate simultaneously with the SNP-infusion. At normal body temperature, as well as during hypothermia in cases receiving thiosulphate, the cyanide levels rose slowly but significantly with the infusion rate. Higher erythrocyte cyanide levels in relation to the infusion rates, up t

Cyanide and thiocyanate concentrations following sodium nitroprusside infusion in man.

Twenty-six patients, receiving an infusion of sodium nitroprusside (SNP) during surgery, had considerable increases in both red cell and plasma cyanide concentration, but only small changes in plasma thiocyanate concentration. There was a linear relationship between both plasma and RBC cyanide concentrations and the total dose of SNP. The expired cyanide concentration followed the changes in the plasma. We believe that the development of metabolic acidosis, and the recent fatalities involving SNP, are attributable to histotoxic hypoxia as a result of excessive plasma concentrations of cyanide.

Sodium Nitrite and Sodium Thiosulfate Are Effective Against Acute Cyanide Poisoning When Administered by Intramuscular Injection.

STUDY OBJECTIVE: The 2 antidotes for acute cyanide poisoning in the United States must be administered by intravenous injection. In the out-of-hospital setting, intravenous injection is not practical, particularly for mass casualties, and intramuscular injection would be preferred. The purpose of this study is to determine whether sodium nitrite and sodium thiosulfate are effective cyanide antidotes when administered by intramuscular injection. METHODS: We used a randomized, nonblinded, parallel-group study design in 3 mammalian models: cyanide gas inhalation in mice, with treatment postexpos

Intramedullary sodium cyanide injection on respiratory and vasomotor responses in cats.

To examine the effect of hypoxia confined to the ventrolateral medulla we microinjected NaCN into the cat medulla (1.0 mm below the ventral surface) unilaterally and investigated cardio-respiratory changes. We studied anesthetized artificially ventilated animals and measured the electrical activity of phrenic and cervical sympathetic nerves and blood pressure. Histotoxic hypoxia depressed phrenic amplitude and elevated sympathetic tone and blood pressure. These responses were obtained predominantly from the region 5.0-8.0 mm caudal to the foramen caecum and 3.0-5.0 mm lateral to the midline (i

Comparison of effects of cyanide and acetylcholine on renal hemodynamics and sodium excretion.

The present study compared the effects of acetylcholine to cyanide under the same experimental conditions of renal clearance in anesthetized dogs. Since cyanide is one of the few drugs for which the mechanism of action is known (cytotoxic hypoxia), some insight may be gained into the renal effects of acetylcholine since both produce direct natriuresis and diuresis. Infusion of 0.2 microgram/kg/min of acetylcholine and 12.0 microgram/kg/min of sodium cyanide into the left renal artery resulted in similar effects, i.e., increased fractional excretion of sodium, potassium, calcium, and magnesium.

Some physiological and metabolic effects of sodium nitroprusside and cyanide in the dog.

The cardiovascular and acid-base changes following equivalent i.v. bolus doses of sodium nitroprusside (SNP) and potassium cyanide (KCN) have been studied in two groups of anaesthetized dogs. In a third group, the metabolic changes produced by i.v. infusion of SNP 1.5 mg kg-1 at a constant rate over 1 h have been studied. In contrast to a decrease in arterial pressure following SNP, hypertension and tachycardia occurred after the administration of KCN, with hyperventilation and an increase in packed cell volume. During infusion of SNP, increases in plasma cyanide concentrations were associated

[Sodium thiosulfate for acute cyanide poisoning: study in a rat model].

Unlike practices in the United States where it is associated with other antidotes, sodium thiosulfate is not used for emergency therapy for cyanide poisoning in France. The purpose of this study was to develop a rat model using intraperitoneal injections of sodium thiosulfate at a dose of 225 mg/kg to test its therapeutic efficacy for acute cyanide poisoning. Efficacy was assessed directly by quantifying arterial blood cyanide and indirectly using markers of hypoxia: serum lactate and arteriolization of venous blood gases. Cyanide poisoning induced intense biological anomalies which were persi

Effect of cyanide and verapamil on sodium-free contractures in the frog heart.

Sodium-free contractures were studied in myocardial strips from R. pipiens with extracellular sodium (Na0+) replaced by choline chloride and extracellular calcium (Ca20+) varied with EGTA buffer. At calculated Ca02+ below 2.8 X 10(-7) mol/l, no contracture occurred in most of the experiments, even in the presence of cyanide. When Ca02+ was above 2.8 X 10(-7) mol/l, relatively short tension transients of up to 80 sec duration could be avoided if the myocardial strip was previously equilibrated for 20 min in a Na+-Ca2+-free solution. Instead, contractures developed slowly within one to several h

Inhibition of mitochondrial respiration by sodium nitroprusside and the mechanism of cyanide liberation.

The influence of sodium nitroprusside (SNP) on mitochondrial respiration was examined in rat liver mitochondria. The addition of SNP 1 mmol litre-1 during state 3 respiration inhibited the oxygen uptake by 63.4%. A mixture of SNP 1 mmol litre-1 and glutathione (GSH) 1 mmol litre-1 inhibited the oxygen uptake more markedly (by 75.9%). The cyanide concentrations were 0.01 mmol litre-1 with SNP alone and 0.15 mmol litre-1 with the mixture of SNP and GSH. Cyanide production from SNP in the presence of various reducing agents was studied in potassium phosphate 0.1 mol litre-1 buffer solution (pH 7.
