Organization of somatosensory cortex in monotremes: in search of the prototypical plan.

The present investigation was designed to determine the number and internal organization of somatosensory fields in monotremes. Microelectrode mapping methods were used in conjunction with cytochrome oxidase and myelin staining to reveal subdivisions and topography of somatosensory cortex in the platypus and the short-billed echidna. The neocortices of both monotremes were found to contain four representations of the body surface. A large area that contained neurons predominantly responsive to cutaneous stimulation of the contralateral body surface was identified as the primary somatosensory a

[Epileptiform activity in the somatosensory cortex of rats with trigeminal neuralgia].

It was shown in experiments on rats that penicillin 1 microliter microinjection (100 U) into the caudal nucleus of the spinal tract of the trigeminal nerve, accounting for formation of a generator of pathologically enhanced excitation (GREE), brings about in rats the pain syndrome with characteristic for trigeminal neuralgia behavioural manifestations and the emergence of epileptiform activity in the somatosensory cortex, especially pronounced in the contralateral hemisphere. The emergence of this activity reflects, on the one hand, the action of the GREE in the caudal nucleus of the trigemina

Cortical somatosensory evoked potentials. II. Effects of excision of somatosensory or motor cortex in humans and monkeys.

1. To clarify the generators of human short-latency somatosensory evoked potentials (SEPs) thought to arise in sensorimotor cortex, we studied the effects on SEPs of surgical excision of somatosensory or motor cortex in humans and monkeys. 2. Normal median nerve SEPs (P20-N30, N20-P30, and P25-N35) were recorded from the cortical surface of a patient (G13) undergoing a cortical excision for relief of focal seizures. All SEPs were abolished both acutely and chronically after excision of the hand area of somatosensory cortex. Similarly, excision of the hand area of somatosensory cortex abolished

Interconnected cortical networks between primary somatosensory cortex septal columns and posterior parietal cortex in rat.

Visual and somesthetic cues are used for spatial processing in the posterior parietal cortex (PPC) of the mammalian brain. In rats, somatic information collected by the mystacial whiskers is critically involved in constructing a neural representation of the external space. Here, we delineated the topography of the cortical pathway from the primary somatosensory cortex (SI) that may deliver vibrissal cues to PPC for spatial processing. For anterograde tracing, we made small injections of biotinylated dextran amine (BDA) into SI barrel cortex. The injections in the regions directly above the sep

Ocular surface information seen from the somatosensory thalamus and cortex.

Ocular Surface (OS) somatosensory innervation detects external stimuli producing perceptions, such as pain or dryness, the most relevant symptoms in many OS pathologies. Nevertheless, little is known about the central nervous system circuits involved in these perceptions, and how they integrate multimodal inputs in general. Here, we aim to describe the thalamic and cortical activity in response to OS stimulation of different modalities. Electrophysiological extracellular recordings in anaesthetized rats were used to record neural activity, while saline drops at different temperatures were appl

Content Representation of Tactile Mental Imagery in Primary Somatosensory Cortex.

The imagination of tactile stimulation has been shown to activate primary somatosensory cortex (S1) with a somatotopic specificity akin to that seen during the perception of tactile stimuli. Using fMRI and multivariate pattern analysis, we investigate whether this recruitment of sensory regions also reflects content-specific activation (i.e., whether the activation in S1 is specific to the mental content participants imagined). To this end, healthy volunteers (n = 21) either perceived or imagined three types of vibrotactile stimuli (mental content) while fMRI data were acquired. Independent of

Infrared thermal imaging of rat somatosensory cortex with whisker stimulation.

The present study aims to validate the applicability of infrared (IR) thermal imaging for the study of brain function through experiments on the rat barrel cortex. Regional changes in neural activity within the brain produce alterations in local thermal equilibrium via increases in metabolic activity and blood flow. We studied the relationship between temperature change and neural activity in anesthetized rats using IR imaging to visualize stimulus-induced changes in the somatosensory cortex of the brain. Sensory stimulation of the vibrissae (whiskers) was given for 10 s using an oscillating w

mGluR5 regulates glutamate-dependent development of the mouse somatosensory cortex.

We have previously reported that mGluR5 signaling via PLC-beta1 regulates the development of whisker patterns within S1 (barrel) cortex of mice (Hannan et al., 2001). However, whether these defects arise from the loss of postsynaptic mGluR5 signaling, and whether the level of mGluR5 is important for barrel formation, was not examined. Furthermore, whether mGluR5 regulates other developmental processes that occur before or after barrel development is not known. We now show that mGluR5 is present postsynaptically at thalamocortical synapses during barrel formation. In addition, Mglur5(+/-) mice

Seeing the hand being touched modulates the primary somatosensory cortex.

Recent behavioral studies have shown that viewing the stimulated body part can enhance tactile acuity. We examined effects of viewing the stimulated body part by using neuromagnetic source imaging. Study participants were covertly stimulated on their first digit (D1) while watching a video that showed a hand where D1 was stimulated. The video hand appeared in the region where the real hand would be expected. In addition, participants were stimulated on D1 without watching the video. The results demonstrated a shift of D1 to a more inferior position and an increase of the dipole strength of D1

Rapid functional plasticity of the somatosensory cortex after finger amputation.

Recent research indicates that areas of the primary somatosensory (SI) and primary motor cortex show massive cortical reorganization after amputation of the upper arm, forearm or fingers. Most of these studies were carried out months or several years after amputation. In the present study, we describe cortical reorganization of areas in the SI of a patient who underwent amputation of the traumatized middle and ring fingers of his right hand 10 days before cortical magnetic source imaging data were obtained. Somatosensory-evoked magnetic fields (SEF) to mechanical stimuli to the finger tips wer

Layer-specific attentional modulation in the human primary somatosensory cortex.

Selective attention enhances relevant sensory inputs while filtering out distractions, yet its layer-specific effects remain poorly understood. Here, we used microvascular-specific spin-echo BOLD fMRI at 7 T to investigate attentional modulation across cortical layers in the human primary somatosensory cortex (S1) during finger and wrist stimulation. During passive, unattended stimulation, the middle and deep layers exhibited stronger signals than the superficial layer. In the attended finger region, attention significantly increased activity in the superficial layer, consistent with top-down

Intracortical microstimulation of human somatosensory cortex induces natural perceptual biases.

Time-order error, a psychophysical phenomenon in which the duration in between successive stimuli alters perception, has been studied for decades by neuroscientists and psychologists. To date, however, the locus of these effects is unknown. We use intracortical microstimulation of somatosensory cortex in three humans with spinal cord injury as a tool to bypass initial stages of processing and restrict the possible locations that signals could be modified. Using a 2-interval forced choice amplitude discrimination paradigm, we first assessed the extent to which order effects are observed. Compar

Neurodegeneration in the somatosensory cortex after experimental diffuse brain injury.

Disruption and consequent reorganization of central nervous system circuits following traumatic brain injury may manifest as functional deficits and behavioral morbidities. We previously reported axotomy and neuronal atrophy in the ventral basal (VB) complex of the thalamus, without gross degeneration after experimental diffuse brain injury in adult rats. Pathology in VB coincided with the development of late-onset aberrant behavioral responses to whisker stimulation, which lead to the current hypothesis that neurodegeneration after experimental diffuse brain injury includes the primary somato

Somatosensory cells in area PEc of macaque posterior parietal cortex.

PEc is an area defined on cytoarchitectural grounds, located in the posterior part of the superior parietal lobule of macaque brain (Pandya and Seltzer, 1982). The aim of this work was to assess whether passive somatosensory stimulation elicited responses in PEc neurons. Extracellular recordings were performed in three awake Macaca fascicularis. Passive somatosensory stimulation was performed in darkness, and eye movements were monitored continuously. Recording sites were assigned to different areas according to the cytoarchitectonic criteria described by Pandya and Seltzer (1982) and Luppino

Blood flow changes in human somatosensory cortex during anticipated stimulation.

Positron emission tomography (PET) measurements of brain blood flow were used to monitor changes in the human primary and secondary somatosensory cortices during the period when somatosensory stimuli were expected. In anticipation of either focal or innocuous touching, or localized, painful shocks, blood flow decreased in parts of the primary somatosensory cortex map located outside the representation of the skin area that was the target of the expected stimulus. Specifically, attending to an impending stimulus to the fingers produced a significant decrease in blood flow in the somatosensory z

Noninvasive detection of cerebral plasticity in adult human somatosensory cortex.

After upper limb deafferentation, adult macaques exhibit substantial reorganization of cortical somatosensory topography with enlargement of cortical areas responsive to facial stimuli. In the present study non-invasive magnetic source imaging technology has been used to map in detail the bilateral somatosensory homunculi in four neurologically normal controls and two upper arm amputees. Bilateral homuncular maps of normals and of the unaffected hemisphere of both amputees showed a wide hand area. The affected hemisphere of both amputees showed marked intrusion of facial representations into t

Neonatal fluoxetine exposure affects the neuronal structure in the somatosensory cortex and somatosensory-related behaviors in adolescent rats.

Selective serotonin reuptake inhibitor (SSRI)-type antidepressants are often prescribed to depressive pregnant women for their less adverse side effects. However, growing evidences have shown increased congenital malformations and poor neonatal adaptation in the perinatal SSRI-exposed human infants as well as animal pups. In this study, we examined the effects of early exposure of fluoxetine, the most popular SSRI-type antidepressant, on the developing somatosensory system. Physiological saline or fluoxetine (10 mg/kg) was subcutaneously injected into neonatal rats from P0 to P6. Somatosensory

Secondary Somatosensory Cortex Is Required for Learning but Not Execution of a Tactile Discrimination.

The relationship between primary (S1) and secondary (S2) somatosensory cortex is not well understood, and the role of S2 in somatosensory function is not well defined. To test the role of S2 and its interplay with S1 in learning a texture discrimination, we reversibly inhibited primary (S1) and/or secondary somatosensory cortex (S2) bilaterally using DREADDs and measured the effect on the ability of mice to learn a whisker-dependent tactile discrimination. Freely moving mice foraged in an arena that contained two bowls, one of which contained a buried food reward. The bowls could only be disti

Fat perception in the human frontal operculum, insular and somatosensory cortex.

Here, we combined magnetic resonance imaging with lesion-symptom mapping in patients with chronic brain lesions to investigate brain representations of sugar and fat perception. Patients and healthy controls rated chocolate milkshakes that only differed in sugar or fat content. As compared to controls, patients showed an impaired fat, but not sugar perception. Impairments in fat perception overlapped with the anterior insula and frontal operculum, together assumed to underpin gustatory processing. We also identified the mid-dorsal insula as well as the primary and secondary somatosensory corte

Rewiring the primary somatosensory cortex in carpal tunnel syndrome with acupuncture.

Carpal tunnel syndrome is the most common entrapment neuropathy, affecting the median nerve at the wrist. Acupuncture is a minimally-invasive and conservative therapeutic option, and while rooted in a complex practice ritual, acupuncture overlaps significantly with many conventional peripherally-focused neuromodulatory therapies. However, the neurophysiological mechanisms by which acupuncture impacts accepted subjective/psychological and objective/physiological outcomes are not well understood. Eligible patients (n = 80, 65 female, age: 49.3 ± 8.6 years) were enrolled and randomized into three
