Pulsed electric field-assisted biorefinery of soybean hulls for sequential recovery of phenolics, proteins, cellulose, hemicellulose and lignin.

Lignocellulosic biomass recalcitrance remains one of the main barriers to the development of efficient integrated biorefineries. In this study, pulsed electric fields (PEF) were investigated as a structural pretreatment to enhance the sequential fractionation of soybean hulls into phenolics, proteins, pectin-related fractions, cellulose, hemicellulose, and lignin. PEF was applied at electric field strengths of 0.5-2 kV/cm prior to a sequential extraction process involving hydroethanolic extraction, citric acid extraction, and alkaline delignification. PEF significantly increased the extraction

The Major Soybean Allergen Gly m Bd 28K Induces Hypersensitivity Reactions in Mice Sensitized to Cow's Milk Proteins.

Reactions to soy have been reported in a proportion of patients with IgE-mediated cow's milk allergy (CMA). In this work, we analyzed if Gly m Bd 28K/P28, one of the major soybean allergens, is a cross-reactive allergen with cow milk proteins (CMP). We showed that P28 was recognized by IgE sera from CMA patients and activated human peripheral basophils degranulation. Moreover, IgE sera of mice exclusively sensitized to CMP recognized P28. Splenocytes from sensitized animals secreted IL-5 and IL-13 when incubated with CMP or soy proteins, but only IL-13 when treated with P28. In addition, a ski

[Effect of soybean trypsin inhibitor and chicken ovomucoid on the immunogenic and sensitizing properties of various dietary proteins].

Adult male guinea pigs were sensitized by intragastric administration of bovine serum albumin (BSA) and chick ovalbumin (OA) and their mixtures with soybean Kunitz trypsin inhibitor (SBTI) and chick ovomucoid (OM). Sensitization of the animals was evaluated by the anaphylactic shock reaction and also by the levels of serum specific IgG antibodies against BSA and OA as measured in the solid phase radioimmunoassay. The experiment revealed pronounced desensitizing properties of SBTI combined both with OA and BSA. OM produced no effect on the animal sensitization caused by OA and enhanced the BSA-

In vivo evidence of cross-reactivity between cow's milk and soybean proteins in a mouse model of food allergy.

BACKGROUND: Cow's milk allergy (CMA) is an important problem worldwide and the development of an in vivo system to study new immunotherapeutic strategies is of interest. Intolerance to soybean formula has been described in CMA patients, but it is not fully understood. In this work, we used a food allergy model in BALB/c mice to study the cross-reactivity between cow's milk protein (CMP) and soy proteins (SP). METHODS: Mice were orally sensitized with cholera toxin and CMP, and then challenged with CMP or SP to induce allergy. Elicited symptoms, plasma histamine, humoral and cellular immune re

Odorant-Binding Protein 17 Mediates the Recognition of Soybean Volatiles in Adult Riptortus pedestris.

RpedOBP17, an odorant-binding protein highly expressed in adult Riptortus pedestris antennae, was systematically investigated using prokaryotic expression, fluorescence competitive binding, molecular docking, site-directed mutagenesis, and RNA interference. Binding assays revealed affinity to six soybean volatile compounds. Molecular docking and mutagenesis identified Ile46 and Phe129 as critical residues for ligand recognition, since alanine substitutions markedly reduced binding. RNAi-mediated knockdown significantly impaired olfactory chemotaxis, with electroantennogram recordings showing a

Natural variation in GmSOP5 regulates seed oil and protein content during soybean domestication.

Seed oil content, protein content, and yield are agronomically important, correlated traits that determine the economic value of soybean (Glycine max). However, improving seed quality and yield simultaneously is challenging because gains in one breeding target often compromise the other, and the genetic basis of this trade-off is poorly understood. Here, we performed a genome-wide association study of 429 diverse soybean accessions and identified Seed Oil and Protein 5 (SOP5), which encodes a kinesin protein, as a key locus associated with seed oil and protein content. Knockout and overexpress

Effects and action mechanisms of polygonatum sibiricum polysaccharide on gel properties of soybean protein isolate.

In this study, the polygonatum sibiricum polysaccharide (PSP) was extracted using the deep eutectic solvent (DES) and added into soybean protein isolate (SPI) to prepare the SPI - PSP gel. Afterwards, the SPI - PSP gel properties were analyzed to explore how the PSP affected the gel properties and the gelation mechanism. When the PSP addition was from 0% to 2.5% (w/v, g/100 mL), the gel hardness increased from (18.70 ± 2.10) g to (36.10 ± 3.22) g. The gel consistency was enhanced, but the gel flow behavior was inhibited. Besides, the gel water-holding capacity rose from (83.56 ± 1.13) % to (89

Tough sustainable bio-based soybean protein adhesive with salt-bridge hydrogen bonds.

Soy protein adhesives have great potential in replacing traditional petrochemical adhesives to produce eco-friendly and formaldehyde-free plywood. However, weak toughness, poor water resistance and low mold resistance during their usage have severely limited their application in practical production. In this study, we introduced bio-based sodium carboxymethyl cellulose (CMCNa) and lauroyl arginine ethyl ester hydrochloride (LAE) into the soybean meal (SM) system to increase the network interweaving density through the electrostatic interactions and multiple hydrogen bonds between CMCNa, LAE an

Effects of Fermentation with Tetragenococcus halophilus and Zygosaccharomyces rouxii on the Volatile Profiles of Soybean Protein Hydrolysates.

The effects of fermentation with lactic acid bacteria (LAB) and yeast on the aroma of samples were analyzed in this work. The volatile features of different soybean hydrolysates were investigated using both GC-MS and GC-IMS. Only 47 volatile flavor compounds (VFCs) were detected when using GC-IMS, while a combination of GC-MS and GC-IMS resulted in the identification of 150 compounds. LAB-yeast fermentation could significantly increase the diversity and concentrations of VFCs (p < 0.05), including alcohols, acids, esters, and sulfurs, while reduce the contents of aldehydes and ketones. Identification, nomenclature, and evolutionary relationships of mitogen-activated protein kinase (MAPK) genes in soybean.

Mitogen-activated protein kinase (MAPK) genes in eukaryotes regulate various developmental and physiological processes including those associated with biotic and abiotic stresses. Although MAPKs in some plant species including Arabidopsis have been identified, they are yet to be identified in soybean. Major objectives of this study were to identify GmMAPKs, assess their evolutionary relationships, and analyze their functional divergence. We identified a total of 38 MAPKs, eleven MAPKKs, and 150 MAPKKKs in soybean. Within the GmMAPK family, we also identified a new clade of six genes: four gene

[Biological evaluation, in rats and in humans, of a milk product without lactose, and of a soybean protein formula for use in protein-calorie malnutrition].

Protein quality of a lactose-free milk (LFM) was compared with a soy-protein isolate (SPI) by means of the PER and 24-hour nitrogen balance methods performed with 16 infants with protein-energy malnutrition. PER was 1.19 for the SPI, value which is significantly lower than that of 2.28 obtained for LFM, and 2.71 for casein. Nitrogen absorption of LFM and SPI was 80%. The biological value of both formulas did not differ significantly; however, in six month-old babies or younger with severe malnutrition, nitrogen retention was higher with LFM than with SPI. On the other hand, nitrogen retention

Effect of pH on the formation mechanism of riboflavin mediated photocrosslinking hydrogel of soybean protein nanofibrils.

Riboflavin (RF) mediated photocrosslinking provides a green and controllable approach for soy protein nanofibril (SPF) hydrogel, but the pH-dependent reaction efficiency and structural stability remain key challenges to achieve reliable and sustainable applications. In this study, RF was used as a photosensitizer to induce the crosslinking of SPF under 365 nm UV irradiation, and the gelation mechanism of the photocrosslinked hydrogels was systematically investigated under different pH conditions (2, 4, 6, 8, and 10). The results demonstrated that pH not only altered the dispersibility and conf

Effect of extraction pH on the emulsion stability and surface protein structure of soybean oil body.

In this study, the pH was varied (6.0-11.0) during the extraction of soybean oil body (SOB), and the stabilities of the corresponding emulsions as well as the structural properties of SOB surface proteins were investigated. The extraction pH was found to affect both the SOB emulsion stability and surface protein structure. Micromorphological analysis showed that the degree of SOB dispersion was higher under alkaline conditions compared to other treatment conditions (acidic or neutral). Structural changes in the SOB surface proteins, as evaluated using changes in the α-helix and β-sheet content

Effect of Corynebacterium glutamicum Fermentation on the Volatile Flavors of the Enzymatic Hydrolysate of Soybean Protein Isolate.

This study focused on improving the flavor quality of seasonings, and enzymatic hydrolysis of soybean protein isolate (SPI) seasoning via traditional technology may lead to undesirable flavors. Herein, we aimed to develop a new type of SPI seasoning through microbial fermentation to improve its flavor quality. The effect of Corynebacterium glutamicum fermentation on the flavoring compounds of seasonings in SPI enzymatic hydrolysate was examined. Sensory evaluation showed that the SPI seasoning had mainly aromatic and roasted flavor, and the response signals of S18 (aromatic compounds), S24 (al

Role of gellan gum in multiscale assembly and structure-property relationships of potato protein-soybean protein fibril-pea protein fibril composite gels.

Multicomponent gels integrating globular plant proteins with protein fibrils offer a promising soft-matter route for texture design in plant-based meat analogues (PBMAs), yet the polysaccharide-mediated multiscale assembly underlying their structure-property relationships remains poorly understood. In this study, gellan gum (0-4.0 wt%) was used to modulate composite gels comprising potato protein, soybean protein fibrils, and pea protein fibrils (2:1:1, w/w/w). Gellan gum modulated the pre-assembly state, as indicated by changes in apparent soluble-protein fraction, ζ-potential, ANS-derived su

Investigation into the Allergenicity and Functional Properties of Soybean 7S Protein upon Ultrasound-Assisted Punicalagin Binding.

Soybean ranks among the commonly reported allergenic foods with soy 7S protein (7S) acting as its key allergenic component. In this study, in vitro and in vivo experiments collectively confirmed that ultrasound-assisted punicalagin (PUN) binding effectively reduced 7S protein allergenicity. This effect was achieved through multiple pathways: reducing allergen-specific antibody binding capacity, inhibiting mast cell degranulation, adjusting Th1/Th2 immune homeostasis, improving intestinal barrier integrity, and reshaping gut microbiota composition. Additionally, this treatment modulated the fun

Sources of Microbial and Organic Contaminants in the Production of Soybean Whey Protein for Feed and Potential Food Applications.

Soybean whey wastewater (SWW), a rich source of soybean whey protein (SWP), is prone to microbial rancidity, posing environmental and resource challenges. This study explores the causes of rancidity-characterized by a pungent, sour, and putrid odor-in the effluents of sealed buffer tank during SWP recovery via pneumatic flotation. Metagenome, bacterial diversity, and HPLC analyses showed the obligate anaerobe Megasphaera spp. dominated rancid effluents (up to 44% abundance), consumed lactate (decreasing from 10.2 g/L in influent to 2.7 g/L in effluent), and produced malodorous propionate and b
