A Parallelized, Automated System for Higher-Throughput Spatial Navigation Testing of Rats.

Spatial navigation tasks for rodents have been an extremely useful tool for studying cognitive processes for decades but are often laborious to administer and sensitive to experimenter-induced variability. To address these limitations, we have developed a parallelized maze system which runs four highly flexible maze environments simultaneously with automated reward delivery, enabling reduced experimenter involvement, increased throughput, and streamlined data collection. Further, our software tracks subject performance and manages the progression through a preset curriculum as subjects learn,

Spatial navigation questionnaires as a supportive diagnostic tool in early Alzheimer's disease.

Impaired spatial navigation is early marker of Alzheimer's disease (AD). We examined ability of self- and informant-reported navigation questionnaires to discriminate between clinically and biomarker-defined participants, and associations of questionnaires with navigation performance, regional brain atrophy, AD biomarkers, and biomarker status. 262 participants (cognitively normal, with subjective cognitive decline, amnestic mild cognitive impairment [aMCI], and mild dementia) and their informants completed three navigation questionnaires. High-Throughput Task to Study Memory Recall During Spatial Navigation in Rodents.

Spatial navigation is one of the most frequently used behavioral paradigms to study memory formation in rodents. Commonly used tasks to study memory are labor-intensive, preventing the simultaneous testing of multiple animals with the tendency to yield a low number of trials, curtailing the statistical power. Moreover, they are not tailored to be combined with neurophysiology recordings because they are not based on overt stereotyped behavioral responses that can be precisely timed. Here we present a novel task to study long-term memory formation and recall during spatial navigation. The task

The Spatial Structure of Neural Encoding in Mouse Posterior Cortex during Navigation.

Navigation engages many cortical areas, including visual, parietal, and retrosplenial cortices. These regions have been mapped anatomically and with sensory stimuli and studied individually during behavior. Here, we investigated how behaviorally driven neural activity is distributed and combined across these regions. We performed dense sampling of single-neuron activity across the mouse posterior cortex and developed unbiased methods to relate neural activity to behavior and anatomical space. Most parts of the posterior cortex encoded most behavior-related features. However, the relative stren

The anterior thalamus provides a subcortical circuit supporting memory and spatial navigation.

The anterior thalamic nuclei (ATN), a central component of Papez' circuit, are generally assumed to be key constituents of the neural circuits responsible for certain categories of learning and memory. Supporting evidence for this contention is that damage to either of two brain regions, the medial temporal lobe and the medial diencephalon, is most consistently associated with anterograde amnesia. Within these respective regions, the hippocampal formation and the ATN (anteromedial, anteroventral, and anterodorsal) are the particular structures of interest. The extensive direct and indirect hip

Spatial performance of unilateral vestibular defective patients in nonvisual versus visual navigation.

The purpose of this study was to investigate the effects of unilateral vestibular neurotomy on humans ability to perform navigation tasks. These tasks provided self-motion feedback by way of either locomotor activity only (nonvisual navigation or "locomotor task") or visual motion cues only (visually simulated navigation or "visual task"). After exploration of an environment in which 4 locations were marked by different objects, subjects attempted to navigate to those locations either by reproducing the same paths as those followed during exploration, by reversing routes, or by making spatial

Spatially periodic computation in the entorhinal-hippocampal circuit during navigation.

To enable navigation in both physical and mental spaces, the human brain employs a cognitive map constructed from the global metrics of the entorhinal cortex and the local representations of the hippocampus. However, how these two regions coordinate to enable navigation remains poorly understood. Here, we designed an object-matching task where human participants unknowingly manipulated object variants arranged in a ring-like structure around a central prototype. Functional MRI revealed a threefold spatial periodicity in the hippocampal activity that tracked navigation directions from object va

Collaborative Navigation Improves Spatial Learning Across Symmetric and Asymmetric Locomotion in Virtual Reality.

Multi-user virtual reality (VR) systems support multiple locomotion methods, but coordination may be challenging when partners use different methods. We studied dyads navigating a shared virtual maze using symmetric (same) or asymmetric (different) locomotion methods-specifically, steering (continuous movement with physical turning) and teleportation (point-and-teleport within 30 meters with physical turning)-and assessed survey knowledge using direction and straightline distance estimates between objects. We found that (1) dyads acquired better survey knowledge than individuals across locomot

Embodied Spatial Navigation Training in Mild Cognitive Impairment: A Proof-of-Concept Trial.

BACKGROUND: Egocentric and allocentric spatial memory impairments affect the navigation abilities of older adults with mild cognitive impairment (MCI). Embodied cognition research hints that specific aids can be implemented into virtual reality (VR) training to enhance spatial memory. OBJECTIVE: In this study, we preliminarily tested 'ANTaging', an embodied-based immersive VR training for egocentric and allocentric memory, compared to treatment as usual (TAU) spatial training in MCI. METHODS: MCI patients were recruited for this controlled trial. A cognitive battery was administered at pre-t

Assessing Human Spatial Navigation in a Virtual Space and its Sensitivity to Exercise.

Spatial navigation (SN) is the ability to locomote through the environment, which requires an understanding of where one is located in time and space. This capacity is known to rely on the sequential firing of place cells within the hippocampus. SN is an important behavior to investigate as this process deteriorates with age, especially in neurodegenerative disorders. However, the investigation of SN is limited by the lack of sophisticated behavioral techniques to assess this hippocampal-dependent task. Therefore, the goal of this protocol was to develop a novel, real-world approach to studyin

Spatial navigation and the basal forebrain in Alzheimer's disease and other neurodegenerative diseases.

Neurodegenerative diseases such as Alzheimer's disease (AD) and Lewy body disease (LBD) are frequently associated with spatial navigation impairments. However, these impairments are less pronounced in frontotemporal dementia and limbic-predominant age-related TDP-43 encephalopathy. While basal forebrain degeneration occurs in all of these diseases, disruption of cholinergic pathways and related cholinergic deficits has been primarily observed in AD and LBD, and in both of these conditions, basal forebrain degeneration appears to be associated with spatial navigation deficits. In this review, w

Direction-selective laterodorsal thalamic nucleus encodes optic flow and turning in spatial navigation.

Vision is crucial for efficient navigation, but how the brain transforms visual signals into navigational cues remains unclear. The laterodorsal thalamic nucleus (LD), with strong connections to visual and navigation circuits, is well positioned for this function, yet its role has been largely unexplored. In mice, we found that LD neurons exhibit strong direction selectivity, with a dominant directional preference for contralateral-to-ipsilateral visual motion, mirroring horizontal optic flow during contraversive turning. In freely moving animals, LD ensembles were preferentially activated dur

Sleep-dependent structural neuroplasticity after a spatial navigation task: A diffusion imaging study.

Evidence for sleep-dependent changes in microstructural neuroplasticity remains scarce, despite the fact that it is a mandatory correlate of the reorganization of learning-related functional networks. We investigated the effects of post-training sleep on structural neuroplasticity markers measuring standard diffusion tensor imaging (DTI), mean diffusivity (MD), and the revised biophysical neurite orientation dispersion and density imaging (NODDI), free water fraction (FWF), and neurite density (NDI) parameters that enable disentangling whether MD changes result from modifications in neurites o

Hippocampal acidity and volume are differentially associated with spatial navigation in older adults.

The hippocampus is negatively affected by aging and is critical for spatial navigation. While there is evidence that wayfinding navigation tasks are especially sensitive to preclinical hippocampal deterioration, these studies have primarily used volumetric hippocampal imaging without considering microstructural properties or anatomical variation within the hippocampus. T1ρ is an MRI measure sensitive to regional pH, with longer relaxation rates reflecting acidosis as a marker of metabolic dysfunction and neuropathological burden. For the first time, we investigate how measures of wayfinding in

Visual cortical prosthesis with a geomagnetic compass restores spatial navigation in blind rats.

Allocentric sense is one of the major components that underlie spatial navigation. In blind patients, the difficulty in spatial exploration is attributed, at least partly, to the deficit of absolute direction perception. In support of this notion, we announce that blind adult rats can perform spatial tasks normally when externally provided with real-time feedback of their head directions. Head-mountable microstimulators coupled with a digital geomagnetic compass were bilaterally implanted in the primary visual cortex of adult rats whose eyelids had been sutured. These "blind" rats were trained

Retrospective Grid Cells in the Medial Entorhinal Cortex Encode Past Paths During Spatial Navigation.

Neurons dynamically switch between prospective and retrospective coding depending on task demands. Prospective coding optimizes future actions, while retrospective coding helps learn from past experiences. The grid cells in the medial entorhinal cortex (MEC) play a key role in spatial navigation, with periodic firing fields to encode current position. While their role in prospective coding has been reported recently, whether and how grid cells exhibit retrospective coding (encoding past paths) remains unclear. Here, we combined large-scale two-photon imaging in freely moving mice with well-est

Spatial navigation is associated with subcortical alterations and progression risk in subjective cognitive decline.

BACKGROUND: Subjective cognitive decline (SCD) may serve as a symptomatic indicator for preclinical Alzheimer's disease; however, SCD is a heterogeneous entity regarding clinical progression. We aimed to investigate whether spatial navigation could reveal subcortical structural alterations and the risk of progression to objective cognitive impairment in SCD individuals. METHODS: One hundred and eighty participants were enrolled: those with SCD (n = 80), normal controls (NCs, n = 77), and mild cognitive impairment (MCI, n = 23). SCD participants were further divided into the SCD-good (G-SCD, n

Hippocampal spatial view cells for memory and navigation, and their underlying connectivity in humans.

Hippocampal and parahippocampal gyrus spatial view neurons in primates respond to the spatial location being looked at. The representation is allocentric, in that the responses are to locations "out there" in the world, and are relatively invariant with respect to retinal position, eye position, head direction, and the place where the individual is located. The underlying connectivity in humans is from ventromedial visual cortical regions to the parahippocampal scene area, leading to the theory that spatial view cells are formed by combinations of overlapping feature inputs self-organized base

The Influence of the Encoding Modality on Spatial Navigation for Sighted and Late-Blind People.

People usually rely on sight to encode spatial information, becoming aware of other sensory cues when deprived of vision. In the absence of vision, it has been demonstrated that physical movements and spatial descriptions can effectively provide the spatial information that is necessary for the construction of an adequate spatial mental model. However, no study has previously compared the influence of these encoding modalities on complex movements such as human spatial navigation within real room-size environments. Thus, we investigated whether the encoding of a spatial layout through verbal c

Evidence for spatial navigational impairments in hydrocephalus patients without spina bifida.

The cognitive sequelae of hydrocephalus have mostly been explored with standardised clinical tasks. The aim of the present research was determine whether impairments on these abstract tasks extend to everyday spatial and navigational behaviour. Patients with hydrocephalus, but without spina bifida, were compared to a control group on tests of searching behaviour, landmark memory, route learning, and path integration. Participants with hydrocephalus displayed reduced sensitivity to spatial cueing, less accurate route-learning, and significantly less accurate spatial updating. These data represe
