Possibility of averting cytokine storm in SARS-COV 2 patients using specialized pro-resolving lipid mediators.

Fatal "cytokine storms (CS)" observed in critically ill COVID-19 patients are consequences of dysregulated host immune system and over-exuberant inflammatory response. Acute respiratory distress syndrome (ARDS), multi-system organ failure, and eventual death are distinctive symptoms, attributed to higher morbidity and mortality rates among these patients. Consequent efforts to save critical COVID-19 patients via the usage of several novel therapeutic options are put in force. Strategically, drugs being used in such patients are dexamethasone, remdesivir, hydroxychloroquine, etc. along with the

Current experimental methods to investigate the impact of specialized pro-resolving lipid mediators on Sjögren's syndrome.

Sjögren's syndrome is a chronic inflammatory autoimmune disease characterized by diminished secretory function of the exocrine glands. Although extensive investigation has been done to understand Sjögren's syndrome, the causes of the disease are as yet unknown and treatments remain largely ineffective, with established therapeutic interventions being limited to use of saliva substitutes with modest effectiveness. A primary feature of Sjögren's syndrome is uncontrolled inflammation of exocrine tissues and previous studies have demonstrated that lipid-based specialized pro-resolving mediators re

Specialized Pro-Resolving Mediators Do Not Inhibit the Synthesis of Inflammatory Mediators Induced by Tumor Necrosis Factor-α in Synovial Fibroblasts.

BACKGROUND: Tumor necrosis factor (TNF)-α, a proinflammatory cytokine, is involved in the pathogenesis of rheumatoid arthritis (RA). The omega-3 unsaturated fatty acid-derived metabolites resolvin (Rv) D1, RvE1, and maresin-1 (MaR1) have been reported as anti-inflammatory lipid mediators and are known as specialized pro-resolving mediators (SPMs). In this study, we aimed to investigate the anti-inflammatory effects of SPMs on TNF-α-induced responses in synovial fibroblasts. METHODS: We investigated the effects of SPMs on gene expression and/or production of cyclooxygenase-2 (COX-2), microsoma

Profiles of specialized pro-resolving mediators in dairy cows with a healthy postpartum period or with endometritis.

In humans and laboratory species, specialized pro-resolving mediators (SPM), a class of cell-signaling molecules originating from the metabolism of PUFA, emerged as mediators of inflammation resolution, but their role in other species remains almost unknown. In dairy cows, postpartum uterine involution involves a physiological transient inflammation, which may evolve into a pathologic inflammation (endometritis). This observational retrospective cohort study characterized the SPM profiles of plasma (2, 10, and 21 d postpartum; DPP) and uterine fluid (21 DPP) of healthy cows (n = 19), and their

Cannabinoid receptors orchestrate distinct anti-tumour pathways in gastric cancer via and beyond specialized pro-resolving mediators.

Endocannabinoids (ECS) and specialized pro-resolving mediators (SPMs) are both lipid-based compounds, but differ significantly in origin, mechanisms, and functions. Their mechanistic interaction in cancer remains undefined, particularly in gastric cancer (GC). Several interconnections have been described between these two "bioactive lipids" involved in inflammation resolution, homeostatic and anti-tumour functions. Cannabinoid signalling can modulate SPM biosynthesis in immune cells, thus we investigated whether this crosstalk operates in GC cells, and whether SPMs mediate part of the anti-tum

Anti-inflammatory celastrol promotes a switch from leukotriene biosynthesis to formation of specialized pro-resolving lipid mediators.

The pentacyclic triterpenoid quinone methide celastrol (CS) from Tripterygium wilfordii Hook. F. effectively ameliorates inflammation with potential as therapeutics for inflammatory diseases. However, the molecular mechanisms underlying the anti-inflammatory and inflammation-resolving features of CS are incompletely understood. Here we demonstrate that CS potently inhibits the activity of human 5-lipoxygenase (5-LOX), the key enzyme in pro-inflammatory leukotriene (LT) formation, in cell-free assays with IC50 = 0.19-0.49 µM. Specialized Pro-Resolving Mediators Derived from N-3 Polyunsaturated Fatty Acids: Role in Metabolic Syndrome and Related Complications.

Significance: Metabolic syndrome (MetS) prevalence continues to grow and represents a serious public health issue worldwide. This multifactorial condition carries the risk of hastening the development of type 2 diabetes (T2D), non-alcoholic fatty liver disease (NAFLD), and cardiovascular diseases (CVD). Another troubling aspect of MetS is the requirement of poly-pharmacological therapy not devoid of side effects. Therefore, there is an urgent need for prospecting alternative nutraceuticals as effective therapeutic agents for MetS. Recent Advances: Currently, there is an increased interest in u

The effect of perinatal supplementation of DHA on specialized pro-resolving lipid mediators in the brain of offspring.

The perinatal period is crucial for fetal neurological development, relying on omega-3 polyunsaturated fatty acids (PUFAs) for essential processes. Omega-3 PUFA, including docosahexaenoic acid (DHA), are precursors to a novel class of bioactive metabolites called specialized pro-resolving mediators (SPMs), which have been suggested to have a dual purpose in mitigating neuroinflammation while simultaneously supporting cognitive outcomes, implicating a role in offspring neurodevelopment. DHA is evidenced for its role in early brain development, but the underlying mechanism it exerts its cognitiv

Development of an Optimized LC-MS Method for the Detection of Specialized Pro-Resolving Mediators in Biological Samples.

The cardioprotective and anti-inflammatory effects of long chain omega-3 polyunsaturated fatty acids (n3 PUFA) are believed to be partly mediated by their oxygenated metabolites (oxylipins). In the last two decades interest in a novel group of autacoids termed specialized pro-resolving mediators (SPMs) increased. These are actively involved in the resolution of inflammation. SPMs are multiple hydroxylated fatty acids including resolvins, maresins, and protectins derived from the n3 PUFA eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) as well as lipoxins derived from arachidonic acid

Specialized Pro-Resolving Mediators MaR1 and LXA4 Resolve Inflammation During Acute Chemical Lung Injury in the Absence of Neutrophils.

Gastric aspiration pneumonia involves chemical injury to the alveoli of the lungs with inflammation, tissue damage, and recruitment of polymorphonuclear leukocytes (PMNs). PMNs are also known to be involved in the production of specialized pro-resolving mediators (SPMs), small lipid molecules that contribute to the resolution of inflammation. This study aimed to identify target PMN-produced SPMs and interrogate their actions and potential use for therapeutic treatment after chemical injury. The data revealed that maresin 1 (MaR1), lipoxin A4, and 18-HEPE are produced after chemical injury in t

MSC administration resolves experimental acute gout increasing specialized pro-resolving mediators synthesis through a super-induction of prostaglandin E2.

BACKGROUND: The resolution of inflammation is an active process triggered in the acute phase of inflammation and mainly directed by specialized pro-resolving mediators (SPMs). Modulating the inflammatory response in favor of resolution is a therapeutic strategy of enormous interest and value. Previous studies have shown that human adipose mesenchymal stem cells (MSCs) possess the ability to shorten the acute inflammatory response by hinting an early cyclooxygenase (COX)-2 induction. We studied the potential of MSC to accelerate the resolution of inflammation through the direct promotion of the

The Natural Combination Medicine Traumeel (Tr14) Improves Resolution of Inflammation by Promoting the Biosynthesis of Specialized Pro-Resolving Mediators.

The resolution of inflammation is an integral part of the acute inflammatory response and eventually leads to the return to homeostasis. It is supported by specialized pro-resolving mediators (SPMs) that act as immunoresolvents via specific G-protein-coupled receptors. In contrast to classical non-steroidal anti-inflammatory drugs (NSAIDs) that suppress the formation of pro-inflammatory lipid mediators such as prostaglandins, novel pharmacotherapeutic concepts propose to foster the biosynthesis of beneficial SPMs. Here, we demonstrate that the natural combination medicine Traumeel (Tr14) impro

From anti-inflammation to pro-resolution: a new paradigm for specialized pro-resolving mediators in regulating neuroinflammation and repair after cerebral ischemia-reperfusion.

Uncontrolled neuroinflammation following cerebral ischemia-reperfusion is a core pathophysiological process driving secondary brain injury and leading to long-term neurological dysfunction. For decades, traditional "anti-inflammatory" strategies targeting the inhibition of key pro-inflammatory pathways have repeatedly failed in clinical translation, compelling a fundamental re-evaluation of the biological nature of inflammation. Inflammation is not a process that passively subsides upon stimulus removal but a dynamic one that requires active "resolution" through endogenous programs to restore

Omega-3 fatty acids, specialized pro-resolving mediators, and depression: A mechanistic meta-analysis of precursors and clinical implications.

BACKGROUND: Omega-3 polyunsaturated fatty acids (n-3 PUFAs), especially eicosapentaenoic acid (EPA), exhibit adjunctive antidepressant efficacy despite negligible detectable levels in the brain. Emerging evidence implicates specialized pro‑resolving mediators (SPMs); bioactive n-3 PUFA metabolites offer a coherent pathway by actively resolving inflammation, enhancing tissue repair, and supporting neuronal function. To test whether n‑3 PUFA supplementation activates pro‑resolving biology in humans, this meta‑analysis quantified effects on circulating SPM precursors. METHODS: We systematically

Differential contribution of lipoxygenase isoforms to the sequential biosynthesis of specialized pro-resolving mediators in monocellular and cell co-incubations.

Oxylipins, produced from polyunsaturated fatty acids (PUFA) by lipoxygenases (LOX), play pivotal roles in inflammation. While 5-LOX-derived leukotrienes promote inflammation, the specialized pro-resolving mediators (SPMs) contribute to inflammation resolution. The biosynthesis of the SPM members lipoxins and resolvins (RVs) supposedly relies on two sequential PUFA oxygenation steps, involving metabolism by 5-LOX and 12-/15-lipoxygenating paralogues (e.g., 12-LOX, 15-LOX-1/-2). These oxygenations may occur within one cell where both LOX isoforms are expressed or via transcellular metabolism bet

Resolution of Skeletal Muscle Inflammation: Role of Specialized Pro-resolving Lipid Mediators in the Recovery from Exercise, Injury, and Disease.

This chapter examines the resolution biology and pharmacology of skeletal muscle inflammation across the physiological contexts of exercise, acute injury, and chronic musculoskeletal disease. Central to these processes are specialized pro-resolving lipid mediators (SPMs), bioactive metabolites of omega-6 (n-6) arachidonic acid (ARA), omega-3 (n-3) eicosapentaenoic acid (EPA), and n-3 docosahexaenoic acid (DHA), formed by the coordinated action of mammalian lipoxygenase (LOX) enzymes. Unlike traditional anti-inflammatory mechanisms that passively dissipate, SPMs actively coordinate resolution b

A high seizure burden increases brain concentrations of specialized pro-resolving mediators in the Scn1a+/- mouse model of Dravet syndrome.

OBJECTIVE: Dravet syndrome is a severe, intractable epilepsy in which 80 % of patients have a de novo mutation in the gene SCN1A. We recently reported that a high seizure burden increased hippocampal concentrations of an array of pro-inflammatory prostaglandins in the Scn1a+/- mouse model of Dravet syndrome. This raised the possibility that a high seizure burden might also trigger the accumulation of specialized pro-resolving mediators that facilitate the resolution of neuroinflammation and brain repair. The present study therefore aimed to examine whether a high seizure burden increased hippo
