Copper (Cu) is an essential trace element required for the normal development of living organisms. Due to its redox potential, copper is a cofactor in many enzymes responsible for important processes in cells. Copper deficiency has a significant influence on the reduction or the total eradication of copper-dependent enzymes in the body, thereby inhibiting cell life processes. On the other hand, copper is a very reactive element and in its free state, it can trigger the production of large amounts of free radicals, which will consequently lead to the damage of proteins and DNA. Because of those

Indiscriminate use of synthetic chemical compounds and the unregulated presence of heavy metals threatens the integral reproducibility of mankind and other living organisms. The toxicity of these compounds far outweighs the usefulness of these compounds. Male reproductive health is linked to the process of spermatogenesis and there is a general consensus that males are more sensitive to these environmental contaminants and so significantly affected when compared to their female counterparts. The review discusses the various toxic contaminants polluting the environment and the effect of these c

Appropriate level of thyroid hormone is essential for normal development and metabolism in most vertebrate tissues and altered thyroid status impacts adversely on them. For many years the testis was regarded as a thyroid hormone unresponsive organ, but consistent evidence accumulated in the past two decades has definitively changed this classical view. Currently, the concept that thyroid hormone plays a critical role in testis development, in rats and other vertebrate species, is clearly established. Although the effects of thyroid hormone on Sertoli and Leydig cells in the immature testis are

Spermatogenesis is a multi-step process, regulated by endocrine, paracrine and autocrine signals, through which immature germ cells differentiate into spermatozoa. The cAMP-dependent transcription factor CREM directs in the testis the expression of genes related to the structuring of mature spermatozoa. CREM gene expression originates different isoforms, which in turn can be divided into activators or repressors of gene expression. Whereas only CREM repressors are expressed in pre-meiotic germ cells in mice, a switch to the expression of the CREM activator tau occurs in post-meiotic germ cells

The most common cause of male infertility is idiopathic. Fresh insights based on genetic and molecular analysis of the human genome permit classification of formerly unexplained disorders in spermatogenesis. In this article, we review new procedures that expand diagnostic and therapeutic approaches to male infertility. Recombinant DNA technology makes it possible to detect specific chromosomal and/or genetic defects among infertile patients. The identification of genes linked to disorders in spermatogenesis and male sexual differentiation has increased exponentially in the past decade. Genetic

Many drugs and chemicals have been found which interfere with the process of spermatogenesis. Among these substances are, for example, sex-hormones (androgens, antiandrogens, estrogens, progestogens, anabolics), chemotherapeutics, antibiotics, antifungal drugs, anticancer drugs, nonsteroidal antiinflammatory drugs, antihypertensives, neuroleptics, dopaminantagonists, indols, tranquilizers, tricyclic antidepressives, heavy metals (Co, Cd), MAO-inhibitors, antimetabolites, barbiturates, immunosuppressives (glucocorticoids), aldosteronantagonists, anticonvulsives, and perhaps alcohol and nicotine

The male gamete, the spermatozoon, is the carrier of paternal inheritance. Besides paternal genome, the spermatozoon contributes essential factors for successful fertilization, embryonic development, and continuation of species. A dynamic and extraordinarily complex process called spermatogenesis produces spermatozoa capable of maturing and acquiring full fertilizing capacity. Spermatogenesis proceeds through a series of events that involve cell division (proliferative phase assuring continuous restocking of spermatogenic stem cells), meiosis (recombination and haploidization of paternal genom

Spermatid production is a complex regulatory process in which coordination between hormonal control and apoptosis plays a pivotal role in maintaining a balanced number of sperm cells. Apoptosis in spermatogenesis is controlled by pro-apoptotic and anti-apoptotic molecules. Hormones involved in the apoptotic process during spermatogenesis include gonadotrophins, sex hormones, and glucocorticoid (GC). GC acts broadly as an apoptosis inducer by binding to its receptor (glucocorticoid receptor: GR) during organ development processes, such as spermatogenesis. However, the downstream pathway induced

The male reproductive system consists of testes, a series of ducts connecting the testes to the external urethral orifice, accessory sex glands, and the penis. Spermatogonial stem cells differentiate and mature in testes and epididymides, and spermatozoa are ejaculated with exocrine fluids secreted by accessory sex glands. Many studies have clarified the detailed structure and function of the male reproductive system, and have shown that various biologic controls, including genomics, epigenetics, and the neuroendocrine-immune system regulate proliferation, differentiation, and maturation of ge

Sperm production starts from puberty in the seminiferous tubules providing testosterone production by the Leydig cells that takes place in the interstice of the testicles. Normal spermatogenesis depends on specific signalling from the hypothalamic-pituitary-gonadal axis. GnRH, FSH and LH are the main hormones involved in the production and maturation of spermatozoa. Exogenous administration of androgens influences the hypothalamic- pituitary-gonadal axis with negative feedback that may lead to a partial or complete cessation of spermatogenesis by decreasing FSH and LH. Despite the fact that ma

Abnormal spermatogenesis is an important pathophysiological process underlying male infertility. Apoptosis of spermatogenic cells and disruption of ectoplasmic specialization (ES) have been characterized as the key biological events of this disorder. Under physiological and pathophysiological conditions (such as exposure to starvation, environmental chemicals, radiation), autophagy is activated in spermatogenic or Sertoli cells in order to maintain survival of the spermatogenic cells by inhibiting spermatogenic cell apoptosis and stabilizing the integrity of ES via degradation of PDZ and LIM d

CIB1 is a 22-kDa calcium binding, regulatory protein with approximately 50% homology to calmodulin and calcineurin B. CIB1 is widely expressed and binds to a number of effectors, such as integrin alphaIIb, PAK1, and polo-like kinases, in different tissues. However, the in vivo functions of CIB1 are not well understood. To elucidate the function of CIB1 in whole animals, we used homologous recombination in embryonic stem cells to generate Cib1(-/-) mice. Although Cib1(-/-) mice grow normally, the males are sterile due to disruption of the haploid phase of spermatogenesis. This is associated wit

A reduction in recombination in the pseudoautosomal region is associated with an increased frequency of aneuploid 24,XY human sperm. Similarly, individuals with paternally derived Klinefelter syndrome (47,XXY) also have a paucity of recombination in the chromosomes that have undergone nondisjunction. Meiotic studies using newly developed immunocytogenetic techniques have demonstrated errors of chromosome synapsis and significantly reduced recombination in infertile men with nonobstructive azoospermia. These men have an increased risk of aneuploidy in sperm that have been surgically removed fro

One hundred and twenty cases of big unilateral hydrocele of the tunica vaginalis testis have been studied to ascertain the effect on the structure and function of the testis, taking the normal side as control. There was no pressure effect from the hydrocele on the structure of the testis in 70 per cent, a flattening of testis in 22 per cent, and atrophy of testis in 8 per cent of cases. There was partial arrest of spermatogenesis in 10 per cent and total arrest of spermatogenesis in 8 per cent of cases. The remaining 82 per cent showed normal spermatogenesis.

Cultivation of neonatal mouse testis tissue can induce spermatogenesis and produce fertile sperms. However, in vitro spermatogenesis mediated by the current organ culture method comes short in fully mimicking the in vivo counterpart, partly due to a lack of knowledge underlying molecular phenotypes of in vitro spermatogenesis. In this study, we investigated transcriptome of cultured testis tissues using microarray method. Principle component analysis of the transcriptome data revealed delay and/or arrest of spermatogenesis and immediate radical immune reactions in the cultured testis tissues.

The role of motor proteins in supporting intracellular transports of vesicles and organelles in mammalian cells has been known for decades. On the other hand, the function of motor proteins that support spermatogenesis is also well established since the deletion of motor protein genes leads to subfertility and/or infertility. Furthermore, mutations and genetic variations of motor protein genes affect fertility in men, but also a wide range of developmental defects in humans including multiple organs besides the testis. In this review, we seek to provide a summary of microtubule and actin-depen
