Spinal cord regeneration induced by a voltage-gated calcium channel agonist.

Regeneration in the central nervous system (CNS) is prohibitive. This is likely due to an interplay of cellular (gene expression, growth factors) and environmental (inhibition by CNS myelin) factors. Calcium supports various intracellular functions, and multiple in vitro studies have shown a role of calcium in axonal growth. In this study, we examine the role of a calcium agonist, S(-)-Bay K 8644, in promoting or impeding CNS growth in vivo, in an effort to understand further the relationship between the voltage-gated L type calcium channel and regeneration. Using a well-established rat spinal

Hapln1-HA signaling promotes progenitor cell proliferation and spinal cord regeneration.

Adult zebrafish exhibit scarless repair and functional recovery following spinal cord injury. Their regenerative capacity is attributed to potent stem-like progenitors that mediate neuronal and glial repair. Zebrafish are thought to lack anti-regenerative extracellular matrix (ECM) components abundant in mammalian SCI, but the positive contributions of ECM to spontaneous spinal cord repair are less understood. By employing cross-species single-cell transcriptomics, we found the hyaluran modifying enzyme Hapln1 is upregulated in zebrafish progenitors but not in mouse progenitors following injur

Microtubules in Spinal Cord Injury: From Cytoskeletal Dysregulation to Therapeutic Regeneration.

Spinal cord injury (SCI) triggers an immediate and sustained disruption of the composition and organization of the neuronal cytoskeleton. Radical alterations in axonal and dendritic microtubules characterize both the acute injury phase and the protracted recovery period. For decades, researchers have sought to correct these microtubule defects as a therapeutic strategy to encourage axonal regeneration, collateral sprouting, and the functional rewiring of neuronal circuits. Recent studies have demonstrated that taxol and related microtubule-active drugs improve outcomes in rodent models. These

Biphasic inflammation control by fibroblasts enables spinal cord regeneration in zebrafish.

Fibrosis and persistent inflammation are interconnected processes that inhibit axon regeneration in the mammalian central nervous system (CNS). Here, we show that in zebrafish, the interplay between fibroblasts and immune cells is required for regeneration. Using temporally resolved single-cell transcriptomics, we identify a transient, injury-induced cthrc1a+ fibroblast state with an inflammation-associated, less differentiated, non-fibrotic profile. Ca2+-dependent induction of this fibroblast state precedes and is critical for the initiation of the inflammatory response. Subsequently, cthrc1a

Operation spinal cord regeneration: Patterning information residing in extracellular matrix glycosaminoglycans.

INTRODUCTION: Spinal cord injuries are devastating, with many complications beyond paralysis and loss of sensory function. Although spinal cord regeneration can revolutionize treatment for spinal cord injuries, the goal has not yet been achieved. The regenerative mechanism of axolotls demonstrates that the regeneration is a repeat of developmental process that all animals have all the genes, but axolotls have both the genes and the patterning information to do it at the adult stage. METHODS: A narrative review was conducted. Relevant studies were collected via an English-language PubMed datab

Chondroitin sulfates do not impede axonal regeneration in goldfish spinal cord.

Chondroitin sulfate proteoglycans produced in glial scar tissue are a major inhibitory factor for axonal regeneration after central nervous system injury in mammals. The inhibition is largely due to chondroitin sulfates, whose effects differ according to the sulfation pattern. In contrast to mammals, fish nerves spontaneously regenerate beyond the scar tissue after spinal cord injury, although the mechanisms that allow for axons to pass through the scar are unclear. Here, we used immunohistochemistry to examine the expression of two chondroitin sulfates with different sulfation variants at the

Spinal interneuron axons spontaneously regenerate after spinal cord injury in the adult feline.

It is well established that long, descending axons of the adult mammalian spinal cord do not regenerate after a spinal cord injury (SCI). These axons do not regenerate because they do not mount an adequate regenerative response and growth is inhibited at the injury site by growth cone collapsing molecules, such as chondroitin sulfate proteoglycans (CSPGs). However, whether axons of axotomized spinal interneurons regenerate through the inhibitory environment of an SCI site remains unknown. Here, we show that cut axons from adult mammalian spinal interneurons can regenerate through an SCI site a

Axial Identity of Spinal Cord Neural Progenitor Cell Grafts Is Dispensable for Regeneration and Functional Recovery After Spinal Cord Injury.

Neural progenitor cell (NPC) transplantation is a promising strategy for spinal cord injury repair, as graft-derived neurons can integrate into host circuitry and promote functional recovery. While the brain-regional and dorsoventral identities of NPCs are known to influence graft composition and performance, the importance of axial (rostrocaudal) identity, specifically whether NPCs must be matched to the spinal level of injury, remains poorly understood. To address this, we compared outcomes following transplantation of NPCs isolated from the anterior embryonic spinal cord (A-NPCs) versus the

GelMA Hydrogel Encapsulating iPSC-Derived Human Spinal Cord Organoids Enhances Neural Regeneration and Restores Motor Function in Rat Spinal Cord Injury.

Spinal cord injury (SCI) severely compromises neural regeneration due to limited intrinsic repair capacity. Combining induced pluripotent stem cell (iPSC)-derived organoids with biomaterial scaffolds offers a promising regenerative strategy. This study investigated the therapeutic potential of human spinal cord organoids (hSCOs) encapsulated within gelatin methacryloyl (GelMA) hydrogel for SCI repair. hSCOs were generated from iPSCs via stage-specific patterning (dorsoventral inhibition followed by retinoic acid/SAG-induced motor neuron specification) and encapsulated in GelMA hydrogel. The th

Transient activation of potent progenitor cells is required for spinal cord regeneration.

Adult zebrafish exhibit full recovery following spinal cord injury. Transient expansion of stem cell-like progenitors is thought to underlie their regenerative capacity. Yet, our understanding of the identities and contributions of the crucial stem cell populations that direct spontaneous neural repair remains limited. Moreover, while most neural regeneration research is centered on promoting proliferative repair, the regulatory mechanisms that reinstate quiescence post-repair are unknown. Here, we determined the molecular identities and cellular contributions of sox2 + progenitors during spin

Interplay of axon regeneration genes and immune infiltration in spinal cord injury.

BACKGROUND: Spinal Cord Injury (SCI) impacts neural function and regeneration. This study aimed to identify key axon regeneration genes in SCI and their correlations with immune infiltration and SCI subtyping. METHODS: Gene expression profiles of 30 sham-operated mice and 29 SCI mice were obtained from GSE5296, GSE47681, and GSE93561 datasets. A PPI network of axon regeneration genes was constructed. Consensus clustering classified SCI subtypes. Differential expression analysis identified genes associated with SCI and its subtypes. Immune infiltration was assessed. WGCNA identified key genes.

Multifunctional Hyaluronic Acid/Graphite Nanoplatelet Hydrogels as Tools for Spinal Cord Regeneration.

Spinal cord injury (SCI) is a condition that interrupts neural electrical conduction, resulting in significant motor and sensory dysfunction. Current treatments remain inadequate for fully restoring neuronal activity, highlighting the urgent need for advanced neurodegenerative materials to reconnect disrupted nerve pathways and recover neural connectivity and function. Herein, conductive and adhesive hydrogel composites based on hyaluronic acid (HA) filled with exfoliated graphite nanoplatelets (EG) or pyrrolidine-functionalized EG (f-EG) are developed. Adhesive catechol-conjugated HA (HA-Cat)

Relation of selenium status to neuro-regeneration after traumatic spinal cord injury.

INTRODUCTION: The trace element selenium (Se) is crucial for the biosynthesis of selenoproteins. Both neurodevelopment and the survival of neurons that are subject to stress depend on a regular selenoprotein biosynthesis and sufficient Se supply by selenoprotein P (SELENOP). HYPOTHESIS: Neuro-regeneration after traumatic spinal cord injury (TSCI) is related to the Se status. STUDY DESIGN: Single-centre prospective observational study. PATIENTS AND METHODS: Three groups of patients with comparable injuries were studied; vertebral fractures without neurological impairment (n = 10, group C), p

Chemotropic guidance facilitates axonal regeneration and synapse formation after spinal cord injury.

A principal objective of spinal cord injury (SCI) research is the restoration of axonal connectivity to denervated targets. We tested the hypothesis that chemotropic mechanisms would guide regenerating spinal cord axons to appropriate brainstem targets. We subjected rats to cervical level 1 (C1) lesions and combinatorial treatments to elicit axonal bridging into and beyond lesion sites. Lentiviral vectors expressing neurotrophin-3 (NT-3) were then injected into an appropriate brainstem target, the nucleus gracilis, and an inappropriate target, the reticular formation. NT-3 expression in the co

Real-Time Feedback Strategically Regulates Optoelectronics for Customized Optogenetic Spinal Cord Regeneration.

Previous optogenetic bioelectronic systems have enabled a highly selective way of modulating neural populations by delivering a certain wavelength of light to engage with exogenously expressed light-sensitive proteins, which lay the foundation of therapeutic interventions of neural circuits. However, real-time biofeedback and strategic modulation are crucial for adjusting customized clinical treatment adjustment. To achieve this purpose, we integrated illumination, temperature, and electromyographic (EMG) sensing elements into the optogenetic bioelectronic system to avoid overexposure caused b

Recent Advances in Hydrogels as Therapeutic Tools for Spinal Cord Injury Regeneration.

INTRODUCTION: It is estimated that more than 27 million people worldwide suffer from spinal cord injury (SCI) due to road traffic accidents, falls, and other causes, with 1 million new cases occurring each year. The primary objectives of SCI repair are to prevent further damage to nerves, inhibit glial scar and inflammatory responses, and facilitate nerve regeneration. However, at present, there is no curative therapy available. Despite the long-standing utilization of conventional pharmaceutical interventions and surgical approaches in clinical settings, these approaches have not yielded opti

SOX Genes in Spinal Cord Injury: Redefining Neural Stem Cell Regeneration Strategies.

The study design is literature review. The sex-determining region Y gene (SRY)-related high mobility group box (HMG)-box (SOX) gene family has primarily been associated with neural development and sex determination and is a key component of human embryonic development. Recent studies on zebrafish models have demonstrated that the unique ability of the latter for central nervous tissue (CNS) repair following injury is largely mediated by SOX genes. Given that efforts aimed at the structural regeneration and functional restoration of neural tissue still represent a major therapeutic challenge in
