Antipodal central effects of apomorphine and dopamine in chickens.

Dopamine, infused into the hypothalamus or injected into the 3rd cerebral ventricle of chickens pretreated with mebanazine, induced behavioural and electrocortical sleep, suppressed motor activity and lowered body temperature and CO2 elimination, effects prevented by phenoxybenzamine but not spiperone. The hypothermic effects of dopamine were intensified by testing chicks below thermoneutrality. Ampomorphine, infused into the hypothalamus, injected into the 3rd cerebral ventricle or given intravenously, evoked behavioural and electrocortical arousal in the intact chicken and in the encéphale i

Elevation of brain neuroleptic/dopamine receptors in schizophrenia.

To test the dopamine hypothesis of schizophrenia the authors measured specific 3H-neuroleptic/dopamine binding sites in three dopamin-rich regions of 59 postmortem normal human brains and 50 postmortem brains from schizophrenic patients using 3H-haloperidol and 3H-spiperone. The binding of 2 nM 3H-haloperidol and of 1 nM 3H-spiperone was significantly elevated in the brains from schizophrenic patients. The brain regions from patients who had no history of being treated with neuroleptic drugs also exhibited significantly higher binding of the 3H-neuroleptics. These results are compatible with t

Implication of acidic lipids in 5-hydroxytryptamine receptor mechanisms.

To establish the possible involvement of acidic lipids in 5-HT receptor mechanisms, we subjected whole rat brain synaptic plasma membranes to treatment with several kinds of lipid-modifying reagents and examined the [3H]5-HT and [3H]spiperone binding properties of the membranes. [3H]5-HT binding was decreased by treatment with Azure A, while [3H]spiperone binding was not altered. Similarly, prior treatment with arylsulphatase reduced the former binding, but had no effect on the latter binding. On the other hand, neither [3H]ligand binding was sensitive to phospholipases C and D. In contrast, p

When administered to rats in a cold environment, 3,4-methylenedioxymethamphetamine reduces brown adipose tissue thermogenesis and increases tail blood flow: effects of pretreatment with 5-HT1A and dopamine D2 antagonists.

When given in a warm environment MDMA (3,4-methylenedioxymethamphetamine, ecstasy) causes hyperthermia by increasing interscapular brown adipose tissue (iBAT) heat production and decreasing heat loss via cutaneous vasoconstriction. When given in a cold environment, however, MDMA causes hypothermia by an unknown mechanism. This paper addresses these mechanisms and in addition examines whether antagonists at 5-HT(1A) and D(2) receptors reduce the hypothermic action of MDMA. Male Sprague-Dawley rats instrumented with a Doppler probe for measuring tail blood flow, and probes for measuring core and

Vascular and cardiac effects of alpha-methyl-5-HT and DOI are mediated by different 5-HT receptors in the pithed rat.

In pithed rats, alpha-methyl-5-hydroxytryptamine (alpha-methyl-5-HT) increased blood pressure and heart rate, 1-(2,5-dimethoxy-4-iodophenyl)-2-aminopropane (DOI) almost exclusively increased blood pressure and 1-(3-chlorophenyl)piperazine (mCPP), heart rate. The maximal responses relative to 5-HT indicate that alpha-methyl-5-HT may be a full agonist at vascular and cardiac 5-HT receptors, DOI a partial agonist at both receptors and mCPP a full agonist at cardiac 5-HT receptors but a partial agonist at vascular 5-HT receptors. Kainate lesion dissociates striatal dopamine receptor radioligand binding sites.

Kainic acid lesion of rat striata reduces the specific dopamine receptor binding of the butyrophenone antagonist [3H]spiperone and the butyrophenone-like antagonist [3H]domperidone by 56% and 59% respectively. Significantly greater decreases in binding were observed with the agonist [3H]N-propylnorapomorphine (NPA) and the antagonist [3H]flupentixol which showed 79% and 73% losses of high affinity binding respectively. These data indicate that, in part, [3H]spiperone and [3H]domperidone label distinct dopamine receptors with different neuronal localizations from those labeled by [3H]flupentixo

Distinct Dopamine D₂ Receptor Antagonists Differentially Impact D₂ Receptor Oligomerization.

Dopamine D₂ receptors (D₂R) are known to form transient homodimer complexes, of which the increased formation has already been associated with development of schizophrenia. Pharmacological targeting and modulation of the equilibrium of these receptor homodimers might lead to a better understanding of the critical role played by these complexes in physiological and pathological conditions. Whereas agonist addition has shown to prolong the D₂R dimer lifetime and increase the level of dimer formation, the possible influence of D₂R antagonists on dimerization has remained rather unexplored. Here,

Dopamine receptor binding of a novel dibenzodioxazocine derivative, EGYT-2509.

The binding of novel dibenzodioxazocine derivatives to rat striatal dopamine receptors was studied in vitro, using 3H-spiperone as radioligand. The biochemical-pharmacological characteristics of the effect of a selected representative, EGYT-2509 are discussed in details. The parameters of specific spiperone binding to rat striatal membrane preparation (KD = 0.550 nM, Bmax = 465 fmole/mg protein) as well as the displacing potencies of known dopamine receptor ligands matched closely the corresponding values in the literature. Using 0.4 nM radioligand, a Ki value of 404 nM was obtained for EGYT-2

Alterations in striatal and extrastriatal D-1 and D-2 dopamine receptors in the MPTP-treated common marmoset: an autoradiographic study.

In adult common marmosets (Callithrix jacchus), MPTP (1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) treatment induced almost total depletion of cells in the substantia nigra pars compacts (SNc) but partial cell loss in the ventral tegmental area (VTA). There was severe depletion of [3H]-mazindol binding to dopamine (DA) uptake sites in the caudate, putamen, and SNc. The loss of [3H]-mazindol binding in the nucleus accumbens (NAc) and olfactory tubercle (OT) was less marked. [3H]-mazindol binding in the body of caudate nucleus showed a small but significant recovery with increasing post-lesion

Decreased striatal dopamine receptor binding in primary focal dystonia: a D2 or D3 defect?

Dystonia is an involuntary movement disorder characterized by repetitive patterned or sustained muscle contractions causing twisting or abnormal postures. Several lines of evidence suggest that abnormalities of dopaminergic pathways contribute to the pathophysiology of dystonia. In particular, dysfunction of D2-like receptors that mediate function of the indirect pathway in the basal ganglia may play a key role. We have demonstrated with positron emission tomography that patients with primary focal cranial or hand dystonia have reduced putamenal specific binding of [(18)F]spiperone, a nonselec

Identification of serotonin 5HT2 receptors in bovine pineal gland.

The concentration of serotonin in the pineal gland is extremely high, which prompted speculation that in addition to serving as a precursor of melatonin, serotonin may have an independent function of its own. By using [3H]-spiperone as a ligand, and ketanserine as a selective serotonin 5HT2 receptor antagonist, we have identified 5HT2 receptor in the bovine pineal gland, revealing a single population of binding sites with a dissociation equilibrium constant (Kd) value of 1.26 +/- 0.41 nM and a receptor density (Bmax) value of 193 +/- 38.85 fmol/mg protein. Specific [3H]piflutixol binding to CHAPS-solubilised rat striatal preparations involves dopamine D-2 but not D-1 binding sites.

[3H]Piflutixol binding to rat striatal membrane preparations identifies both D-1 and D-2 sites. We used [3H]piflutixol to characterise those binding sites present in 3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate (CHAPS)-solubilised rat striatal preparations. The specific binding of [3H]piflutixol, as defined using cis-flupenthixol, to CHAPS-solubilised rat striatal tissue was saturable and of high affinity. Specific [3H]piflutixol binding to the solubilised preparations was displaced stereoselectively by the isomers of butaclamol and to an equal extent by both cis-flupenthixol and

Potency of antipsychotics in reversing the effects of a hallucinogenic drug on locus coeruleus neurons correlates with 5-HT2 binding affinity.

Systemic administration of the phenethylamine hallucinogen 2,5-dimethoxy-4-methylamphetamine (DOM) has previously been shown to decrease spontaneous activity but increase the response to peripheral nerve stimulation of locus coeruleus (LC) neurons in anesthetized rats. Five antipsychotic drugs (spiperone, chlorpromazine, clozapine, haloperidol, and sulpiride) were tested for their ability to antagonize these effects of DOM in the LC. Spiperone, chlorpromazine, clozapine, and haloperidol were able to completely reverse the effects of a standard dose of DOM, while sulpiride was ineffective. The

Studies on the interactions of retinal dopamine with choroidal thickness in the chicken.

PURPOSE: Recently, an increasing number of studies relied on the assumption that visually induced changes in choroidal thickness can serve as a proxy to predict future axial eye growth. The retinal signals controlling choroidal thickness are, however, not well defined. We have studied the potential roles of dopamine, released from the retina, in the choroidal response in the chicken. METHODS: Changes in retinal dopamine release and choroidal thickness changes were induced by intravitreal injections of either atropine (250 µg or 360 nMol), atropine combined with a dopamine antagonist, spiperon

[The inhibition of dopamine D1 and D2 receptors in human putamen by antipsychotic drugs].

The inhibition of 3H-SCH 23390 (D1), 3H-spiperone (D2), and 3H-YM-09151 (D2) binding to human putamen membranes by four antipsychotic drugs was studied. Two substituted benzamides, (-)-sulpiride and YM-09151, weakly inhibited specific 3H-SCH 23390 binding to D1 receptor sites. The inhibition of 3H-SCH 23390 binding by haloperidol and chlorpromazine was also weak in the 10(-8)-10(-6) M range. All four drugs potently inhibited 3H-spiperone binding, with the following rank order of potency: haloperidol greater than YM-09151 greater than (-)-sulpiride greater than chlorpromazine. 3H-YM-09151 bindi

3-Methoxytyramine accumulation: effects of typical neuroleptics and various atypical compounds.

The accumulation of 3-methoxytyramine (3-MT), the O-methylated metabolite of dopamine (DA), in rat striatum was used to assess the effects of drugs on dopaminergic activity. This was accomplished by pretreating rats with pargyline to completely inhibit 3-MT catabolism. Under the conditions used, 3-MT accumulation was linear over time for at least 90 minutes. Apomorphine and gamma-butyrolactone, drugs which depress the activity of DA-containing neurons, decreased striatal 3-MT accumulation; whereas typical neuroleptics (haloperidol, fluphenazine, chlorpromazine), which increase the activity of

Indian red scorpion (Buthus tamulus) venom-induced augmentation of cardiac reflexes is mediated through the involvement of peripheral 5-HT3 and central 5-HT1A receptor subtypes.

The present study was undertaken to identify 5-HT receptor subtypes involved in Buthus tamulus (BT) venom-induced augmentation of cardiac reflexes elicited by phenyldiguanide (PDG). Intravenous injection of PDG (10 microg/kg) produced parallel decrease in mean arterial pressure (MAP) and heart rate (HR) in urethane anaesthetized rats (r=0.82; p < 0.001). Injection of PDG (1-40 microg/kg, i.v.) produced concentration-dependent decrease in time-response area of the HR. After BT venom (20 microg/kg) the concentration-response curve was shifted to the left. Further, fall of MAP and HR in response

Hallucinogenic and neuroleptic drug interactions with potential neurotransmitter receptors in parasitic nematodes.

Receptors potentially involved in neurotransmitting have been characterised in the muscle tissue and in whole worms of the nematodes Ascaris suum and Onchocerca volvulus, respectively. Binding studies revealed a high affinity for LSD with apparent KD values of 94 nM for A. suum and 120 nM for O. volvulus, whereas those of the neuroleptics haloperidol, spiperone and mianserin were found to be in the micromolar range. A variety of neurotransmitter antagonists, known to bind with high affinities either to mammalian D1/2 or to 5-HT1/2 receptors, were tested for their ability to bind to the nematod

Evidence that the serotonin agonist, DOI, increases renin secretion and blood pressure through both central and peripheral 5-HT2 receptors.

DOI [(+/-)-1-(2,5-dimethoxy-4-iodophenyl)-2-aminopropane HCI] is a serotonin (5-HT1C/5-HT2) agonist, with potent cardiovascular effects. The purpose of the present studies was to determine the identity and location of the 5-HT receptor subtype(s) mediating the renin and blood pressure responses to DOI. Injection (i.p.) of DOI to conscious male rats elevated plasma renin activity in a dose-dependent manner. The 5-HT1C/5-HT2 antagonist ritanserin completely blocked the DOI-induced increase in plasma renin activity. In order to distinguish the 5-HT2- from the 5-HT1C- mediated effect of DOI, spipe
