BACKGROUND: The F116V mutation in the substrate recognition site 1 (SRS1) of Spodoptera exigua CYP9A186 has been demonstrated to confer ~200-fold resistance to emamectin benzoate (EB). In this study, a novel mutation (F116I) in CYP9A25, orthologous to CYP9A186, was detected in a field population of Spodoptera litura (YJ22) collected from Yuanjiang, Yunnan province, China in 2022. The association of this mutation with EB resistance was investigated.
RESULTS: Two homozygous strains, YJ22-116F (wild-type at 116 position of CYP9A25) and YJ22-116I (mutant) were isolated from YJ22 through two round

BACKGROUND: Spodoptera frugiperda, a global agricultural pest, can be effectively controlled through the sterile insect technique. However, exposure to low-dose radiation below the sterilization threshold may induce hormetic effects. Here, the biphasic aspects of the fertile progeny population of S. frugiperda were analyzed using an age-stage, two-sex life table after dosing male and female pupae with 10-350 Gy gamma radiation.
RESULTS: The parental sterilizing dose for 6-day-old female and male pupae was 200 and 350 Gy, respectively. The total longevity, pre-adult survival rate, net reproduc

The sources of fungal symbionts of insects are not well understood, yet the acquisition and assembly of fungal communities in mobile insect hosts have important implications for the ecology of migratory insects and their plant hosts. To determine potential sources of fungi associated with the fall armyworm (Spodoptera frugiperda), we characterized the fungal communities associated with four different ecological compartments (insects, infested leaves, uninfested leaves, and soil) and estimated the contributions of each of these potential sources to the insect's fungal microbiome. Results show t

Spinosad (Spi) is a promising insecticide with a novel mode of action and favorable environmental safety, but its field efficacy is severely limited by environmental instability, which restricts its preventive application. In this study, spinosad-loaded chitosan-sodium alginate particles (CS-ALG@Spi) were successfully fabricated and systematically characterized. The prepared microspheres exhibited regular morphology, suitable particle size, good dispersion, and satisfactory dilution stability. Compared with free Spi, CS-ALG@Spi showed significantly enhanced UV stability and an obvious acid-res

Spodoptera frugiperda (fall armyworm) has emerged as a major global pest of maize and other crops. Spinetoram, a widely used spinosyn insecticide, has played a central role in its management; however, increasing reports of resistance threaten the long-term sustainability of this compound. This review synthesizes current knowledge on spinetoram resistance in S. frugiperda, encompassing field reports, underlying molecular mechanisms, and emerging diagnostic approaches. Clear evidence of resistance has been reported from Brazil, whereas studies from China and Pakistan indicate emerging resistance

Spodoptera litura is a polyphagous lepidopteran pest with rapidly evolving insecticide resistance. Peptide toxins from venomous organisms, many stabilized by the inhibitor cystine knot motif, offer bioinsecticidal potential but show limited oral efficacy. Seven previously reported candidates were recombinantly produced in Pichia pastoris and screened against S. litura by injection and leaf-disc feeding bioassays, followed by whole-plant spray assays. The marine nemertean toxin nemertide α-1, designated A1, showed the strongest oral activity, with a median lethal concentration of 3.97 μg μL-1.

Spodoptera frugiperda (J.E. Smith, 1797) (Lepidoptera: Noctuidae) is a pest of great economic importance which can feed on more than 300 plant species. As it is polyphagous, its host plants may have variable physical and chemical constitutions. This may influence larval development, as protein and carbohydrate levels are important factors for adequate biological development. The aim of this study was to evaluate insect developmental parameters as well as to compare the food consumption of S. frugiperda larvae reared using diets with different protein levels under laboratory conditions. The objective of this study was to develop an alternative strategy for the control of Spodoptera litura larvae by using botanical insecticides. Rhizomes of Alpinia galanga were extracted with hexane, dichloromethane, ethyl acetate and methanol, respectively. Each crude extract was examined for toxicity against the second instars of S. litura according to a topical application method. The dichloromethane extract was found to show the highest toxicity of all the extracts, with LD50 = 3177 and 2099 ppm at 24 and 48 h post-treatment, respectively.

Clinical features and pathological and mycological findings in a field outbreak of intoxication in dairy cattle grazing kikuyu grass are reported. The outbreak followed invasion of the grass by the army worm (Spodoptera exempta). Clinical signs included drooling of saliva, depression, apparent inco-ordination, sunken eyes, ruminal distension and atony, recumbency, moderate diarrhoea and "sham drinking". Seventy-seven cows (64%) were clinically affected over a period of 12 days. Of these, 37 died. Necropsies performed on 4 affected cattle revealed necrosis of the epithelium of the forestomach,

Spodoptera frugiperda (J.E. Smith) is a notoriously destructive and invasive agricultural pest, causing substantial economic losses worldwide. Research has shown that cytochrome P450s play an important role in detoxifying insecticides. However, the precise molecular mechanism of P450-mediated detoxification of pyrethroids in S. frugiperda remains largely unknown. Enzyme activity assays demonstrated the involvement of P450 monooxygenases in pyrethroid detoxification in S. frugiperda. Subsequently, the results of induction experiments revealed that CYP9A58 and CYP9A59 were significantly upregula

Lepidopterans produce two distinct types of sperm: nucleated eupyrene sperm for fertilization and anucleate apyrene sperm for auxiliary functions. However, the mechanisms underlying sperm dimorphism in fall armyworm Spodoptera frugiperda remain poorly understood. Serine-Arginine Protein Kinases (SRPKs) are a class of kinases that catalyze the phosphorylation of SR proteins, but recent studies have shown that SRPK is critical for chromatin remodeling of sperm in mammals. Whether SRPK is involved in lepidopteran spermatogenesis is completely unknown. Here, we describe the entire process of elong

The seeds of Phaseolus lathyroides were extracted with hexane, dichloromethane, ethyl acetate and methanol, respectively by Soxhlet apparatus. Each crude extract was examined for toxicity against the second instars of Spodoptera litura using a topical application method under laboratory conditions. The ethyl acetate extract showed the most effective mortality (LD₅₀ = 11,964 and 9,169 ppm after treated at 24 and 48 hours, respectively). Furthermore, in vivo enzyme based experiments revealed that acetylcholinesterase activity of survived S. litura (24 hours post-treatment) was increased by 12% c

0The fall armyworm (Spodoptera frugiperda) is an important pest insect due to high degree of polyphagia. In order to better understand its adaptation mechanism against plant protease inhibitors, bioassays were carried supplementing diet with the Kunitz trypsin inhibitor from Entada acaciifolia seeds (EATI). In vitro assays showed an increase of proteolytic activity in EATI-fed larvae midgut. Moreover, the trypsin enzymes showed insensitivity to inhibition with EATI. In order to understand what genes were overexpressed after chronic exposition to EATI, quantitative RT-PCR analyses were performe

Curcumin, a plant-polyphenolic compound, has been reported to induce autophagy and autophagic cell death in insect cells but its impact on cell cycle regulation is not well understood. In this study, we investigated the short-term effects of curcumin on the cell proliferation, cell cycle progression and autophagy-associated cellular responses in the Sf9 insect cells of Spodoptera frugiperda. Short-term treatment of Sf9 cells with low-dose curcumin (5-15 µg/mL) resulted in a continuous decrease in cell proliferation in a dose and time dependent manner. Flow cytometric analysis showed a signific

Itol A, an isoryanodane diterpene derived from Itoa orientalis Hemsl. (Flacourtiaceae), is a potential plant-based insecticide. However, the effect of itol A on the tobacco cutworm [Spodoptera litura (Fab.) (Lepidoptera: Noctuidae)], an important and widely distributed insect pest, remains unclear. In this study, the toxicity and inhibitory potency of itol A on S. litura were evaluated. The results indicated that itol A exhibited larvicidal activity against the third instar larvae in a concentration-dependent manner (LC50 875.48 mg/L at 96 h). Antifeedant activity also was observed, and the 24

The fall armyworm (Spodoptera frugiperda, Noctuidae, Lepidoptera) is one of the most important crop pests in the Americas, causing significant damage to maize, rice and sorghum. The mechanisms that determine its defences against pathogens are particularly relevant for the development of management and control strategies. We used an in silico approach to identify and characterize pathogen response genes (repat) present in different tissue libraries of S. fugiperda. The analyses revealed complete cDNA for nine repat genes; of these, repat15 and repat39 were found in libraries from a specific tis

Exposure to Bacillus thuringiensis (Bt) toxins in low- and moderate-dose transgenic crops may induce sublethal effects and increase the rate of Bt resistance evolution, potentially compromising control efficacy against target pests. We tested this hypothesis using the fall armyworm Spodoptera frugiperda, a major polyphagous lepidopteran pest relatively tolerant to Bt notorious for evolving field-relevant resistance to single-gene Bt maize. Late-instar larvae were collected from Bt Cry1Ab and non-Bt maize fields in five locations in Brazil, and their offspring was compared for survival, develop

The role of salicylic acid (SA) in plant responses to pathogens has been well documented, but its direct and indirect effects on plant responses to insects are not so well understood. We examined the effects of SA, alone and in combination with jasmonic acid (JA), on the performance of the generalist herbivore, Spodoptera exigua, in wild-type and mutant Arabidopsis thaliana genotypes that varied genetically in their ability to mount SA- and JA-mediated defence responses. In one experiment, growth of S. exigua larvae was highest on the Wassilewskija wild-type, intermediate on the Columbia wild-
