Dietary squalene modifies plasma lipoproteins and hepatic cholesterol metabolism in rabbits.

To evaluate the effects of squalene, the main unsaponifiable component of virgin olive oil, on lipid metabolism, two groups of male New Zealand rabbits were fed a 1% sunflower oil-enriched regular diet or the same diet containing 0.5% squalene for 4 weeks. Plasma triglycerides, total- and HDL-cholesterol and their lipoproteins were assayed. Analyses of hepatic lipid droplets, triglycerides, total- and non-esterified cholesterol, squalene, protein and gene expression, and cholesterol precursors were carried out. In the jejunum, the squalene content and mRNA and protein APOB expressions were mea

Interdependence of Anti-Inflammatory and Antioxidant Properties of Squalene-Implication for Cardiovascular Health.

Cardiovascular diseases (CVD) have been recognized as the leading cause of mortality worldwide, accounting for 31% of global mortality. Among the risk factors of CVD, hyperlipidemia has been established as the most potent risk factor. Statins, a class of drug that reduces lower-density lipoprotein cholesterol (LDL-C), are the preferred medical treatment. However, due to the development of statin-associated muscle symptoms, statins are associated with patients' discontinuation and nonadherence. Other statin-induced side effects, such as hepatotoxicity and gastrointestinal upset, all contribute

Alterations on the growth and ultrastructure of Leishmania chagasi induced by squalene synthase inhibitors.

Leishmaniasis is an important disease in widely dispersed regions of the world. In South America, visceral leishmaniasis (VL) is mainly caused by Leishmania chagasi. The morbidity associated with the infection is high, and death may occur in some untreated patients. Treatment has been based upon pentavalent antimonial drugs for more than half a century and problems, including development of resistance to antimonials and lack of efficacy against VL/HIV co-infections, have emphasized the need for new drugs. Squalene synthase (SQS) is an essential enzyme for the biosynthesis of protozoal sterol m

Squalene-adenosine nanoparticles enhance cardioprotection in myocardial infarction: Efficacy and mechanistic insights.

Reperfusion injury following myocardial infarction (MI) poses a significant challenge, as it can exacerbate heart failure despite being the primary treatment. Adenosine is a promising cardioprotective drug, but its rapid degradation in circulation limits its therapeutic potential. Here, we report the design of self-assembled squalene adenosine nanoparticles (SQAdNPs) as a novel strategy to enhance adenosine's therapeutic potential in MI. In a rat ischemia/reperfusion MI model, it was observed that fluorescent SQAd NPs accumulated in the ischemic myocardium for at least 24 h post-intravenous ad

Hypoxia truncates and constitutively activates the key cholesterol synthesis enzyme squalene monooxygenase.

Cholesterol synthesis is both energy- and oxygen-intensive, yet relatively little is known of the regulatory effects of hypoxia on pathway enzymes. We previously showed that the rate-limiting and first oxygen-dependent enzyme of the committed cholesterol synthesis pathway, squalene monooxygenase (SM), can undergo partial proteasomal degradation that renders it constitutively active. Here, we show hypoxia is a physiological trigger for this truncation, which occurs through a two-part mechanism: (1) increased targeting of SM to the proteasome via stabilization of the E3 ubiquitin ligase MARCHF6

Development and Efficacy of Enzyme-Responsive Squalene-Chidamide Nanoparticles for Pancreatic Cancer.

Here, we present a protocol to address the limited intratumoral penetration of small-molecule drugs caused by the dense extracellular matrix (ECM) of pancreatic ductal adenocarcinoma (PDAC). Prodrugs offer great potential to overcome this challenge by enhancing drug penetration and tumor-killing efficacy. Squalene (SQ), a natural precursor for cholesterol biosynthesis with excellent biosafety and biocompatibility, can improve the membrane compatibility of hydrophilic drugs and enhance their cellular uptake when conjugated to chemotherapeutic agents or bioactive small molecules. Kinetics of Squalene Quenching Singlet Oxygen and the Thermal Degradation Products Identification.

Squalene has been proven to possess various bioactive functions that are widely present in vegetable oils. A more comprehensive understanding of the reaction behavior of squalene under oxidative conditions was achieved by studying its antioxidant capacity and thermal degradation products. The total singlet oxygen quenching rate constant (kr + kq) of squalene was 3.8 × 107 M-1 s-1, and both physical and chemical quenching mechanisms equally contribute to the overall singlet oxygen quenching. Fourteen degradation products of squalene were identified at 180 °C by using gas chromatography-mass spe

Aspergillus flavus squalene synthase as an antifungal target: Expression, activity, and inhibition.

Invasive aspergillosis (IA) is a life-threatening disease impacting immunocompromised individuals. Standard treatments of IA, including polyenes and azoles, suffer from high toxicity and emerging resistance, leading to the need to develop new antifungal agents with novel mechanisms of action. Ergosterol biosynthesis is a classic target for antifungals, and squalene synthase (SQS) catalyzes the first committed step in ergosterol biosynthesis in Aspergillus spp. making SQS of interest in the context of antifungal development. Here, we cloned, expressed, purified and characterized SQS from the pa

Alleviating the Crabtree effect for high-level production of squalene in Saccharomyces cerevisiae.

Saccharomyces cerevisiae is a widely used biomanufacturing chassis; however, its inherent Crabtree effect severely limits energy yield and diverts carbon flux away from the desired non-ethanol products. To address this challenge, we developed a multidimensional metabolic engineering strategy to alleviate the Crabtree effect, using squalene high-level production as a model. Central metabolism was first modulated through three coordinated interventions: modulation of the SNF1/AMPK energy-sensing system, introduction of an A81D variant of the global transcriptional regulator MTH1, and enhancement

Survey on the Prevalence of Terbinafine-Resistant Trichophyton spp. with Squalene Epoxidase Mutations.

Terbinafine (TBF) and azoles are commonly used to treat fungal infections such as tinea pedis and tinea unguium. TBF-resistant Trichophyton species have been increasingly reported globally; however, the research has primarily focused on Trichophyton rubrum. In other words, there are limited studies that exist on other causative Trichophyton species, such as Trichophyton interdigitale, Trichophyton mentagrophytes, and Trichophyton indotineae. This study aimed to determine the prevalence and resistance mechanisms of TBF-resistant Trichophyton isolates in Japan. Screening of 701 clinical isolates

Squalene synthase inhibitors: An update on the search for new antihyperlipidemic and antiatherosclerotic agents.

Atherosclerosis and related heart disease is strongly associated with elevated blood levels of total (and LDL) cholesterol. Due to the widespread incidence as well as severity of this pathological condition, major efforts have been made for the discovery and development of hypocholesteroleamic agents. In the past few decades, HMG-CoA reductase inhibitors (statins) are being extensively used as lipid lowering drugs. These agents act predominantly by inhibiting the enzyme 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR) that is the rate limiting step of cholesterol biosynthesis. Both the s

Enhancement of an analytical method for the determination of squalene in anthrax vaccine adsorbed formulations.

Specific lots of anthrax vaccine adsorbed administered to members of the U.S. Armed Forces have been alleged to contain squalene, a chemical purported to be associated with illnesses of Gulf War veterans. A method of enhanced sensitivity for determining squalene in anthrax vaccine adsorbed using high-performance liquid chromatography with photodiode array detection has been developed, validated, and applied to 44 bottles of 38 lots of anthrax vaccine. In 43 bottles of 37 lots, no squalene was detected within a detection limit of 1ng/0.5ml dose (2 parts-per-billion). One lot, FAV008, was found

Computational exploration of squalene analog 4,4'diapophytofluene as a potential anti-aging phytotherapeutic.

Cellular senescence is a complex biological process characterized by several unique features including cell-cycle arrest, macromolecular damage, secretory phenotypes (SASPs), and deregulated metabolism. These factors are essential for understanding their impact on aging and diseases. Extensive studies on various biochemical pathways associated with mammalian aging have identified SIRT-1, Bcl-xL, Hsp-90, MDM-2, AMPK and mTOR as some key regulatory proteins. So, preserving the functions of these proteins could potentially decelerate the aging process. A previous study had demonstrated that 4,4'-

A novel, synthesized, amphiphilic ethylene glycol squalene derivative suppresses BBN-induced bladder carcinogenesis.

Squalene, a natural triterpene with antioxidant, anti-inflammatory, and immunostimulatory properties, holds promise for cancer therapy. Here, we examined a previously developed, diethylene glycol derivative of squalene (SQ-diEG) and investigated its in vivo anti-carcinogenic effects in bladder cancer. C57BL/6 mice were treated with 0.025% N-butyl-N-(4-hydroxybutyl) nitrosamine (BBN) to induce bladder cancer, with SQ-diEG or PBS (control) administered orally from Week 0. SQ-diEG significantly reduced bladder cancer incidence to 3.7% after 8 weeks, compared to 21.4% in controls (p = 0.025). Tran

Ruthenium polypyridyl squalene derivative: a novel self-assembling lipophilic probe for cellular imaging.

Transition metal complexes provide a promising avenue for designing new therapeutic and diagnostic agents. In particular, ruthenium(II) polypyridyl complexes are useful for studying cellular uptake, due to their easy synthesis and unique photophysical properties. Dyes are frequently combined with material substrates to modulate their properties, enhance stability, reduce toxicity, and improve delivery. A novel Ru polypyridyl complex linked to a derivative of the natural lipid squalene (Ru-BIPPBI-hx-SQ) is described. Using the solvent displacement method, Ru-BIPPBI-hx-SQ easily self-assembles i
