Characterization of Newly Isolated Rosenblumvirus Phage Infecting Staphylococcus aureus from Different Sources.

Staphylococcus aureus is a globally significant pathogen associated with severe infections, foodborne illnesses, and animal diseases. Its control has become increasingly challenging due to the spread of antibiotic-resistant strains, highlighting the urgent need for effective alternatives. In this context, bacteriophages have emerged as promising biocontrol agents. This study aimed to characterize the newly isolated Staphylococcus phage CapO46 and evaluate its efficacy in reducing S. aureus in milk. Identified as a new species within the Rosenblumvirus genus, CapO46 exhibited a podovirus-like s

Antibiotic Exposure Leads to Reduced Phage Susceptibility in Vancomycin Intermediate Staphylococcus aureus (VISA).

In the time of antimicrobial resistance, phage therapy is frequently suggested as a possible solution for such difficult-to-treat infections. Vancomycin-intermediate Staphylococcus aureus (VISA) remains a relatively rare yet increasing occurrence in the clinic for which phage therapy may be an option. However, the data presented herein suggest a potential cross-resistance mechanism to phage following vancomycin exposure in VISA strains. When comparing genetically similar strains differing in their susceptibility to vancomycin, those with intermediate levels of vancomycin resistance displayed d

Design and Preclinical Development of a Phage Product for the Treatment of Antibiotic-Resistant Staphylococcus aureus Infections.

Bacteriophages, viruses that only kill specific bacteria, are receiving substantial attention as nontraditional antibacterial agents that may help alleviate the growing antibiotic resistance problem in medicine. We describe the design and preclinical development of AB-SA01, a fixed-composition bacteriophage product intended to treat Staphylococcus aureus infections. AB-SA01 contains three naturally occurring, obligately lytic myoviruses related to Staphylococcus phage K. AB-SA01 component phages have been sequenced and contain no identifiable bacterial virulence or antibiotic resistance genes.

Changing pattern of phage group II Staphylococcus aureus infections: from community to hospital.

Changes in Staphylococcus aureus strains of phage group II were evaluated over a 30-year period. Strains of this group were previously known to be mainly community-acquired, to be rather sensitive to antibiotics and to occur in infections with better clinical outcome. During the period 1961 to 1990 a total of 551, 563 S. aureus strains was characterized according to antibiotic resistance, phage type and site of isolation. Clinical information was obtained on 16,781 bacteraemia cases. Strains of group II occurred until 1983 with a rather constant frequency around 16% of the S. aureus population

Correlation of penicillinase production with phage type and susceptibility to antibiotics and heavy metals in Staphylococcus aureus.

One hundred and thirty-nine bacteraemia strains of Staphylococcus aureus, representing different combinations of phage type and susceptibility to antibiotics and to cadmium (Cd), arsenate (As) and mercury (Hg), were investigated for penicillinase production. The determination of enzyme activity in induced and uninduced conditions was performed by iodometric titration. The amount of penicillinase produced could be correlated with phage pattern. Epidemically occurring strains of the 94,96 and the 83A complexes produced the largest amount of penicillinase, whereas strains of the 52,52A,80,81 comp

Staphylococcus aureus associated with toxic shock syndrome: phage typing and toxin capability testing.

Phage type 29 Staphylococcus aureus was identified singly or with type 52 in 64.1% of 248 coded isolates from patients with toxic shock syndrome. These phage types also have a high capability of producing pyrogenic exotoxin C and enterotoxin F. The origin and development of these toxigenic strains were explored by studying 25,220 isolates of S. aureus stored in a staphylococcal bank between 1960 and 1979. A small percentage of phage types 29, 52 were found in 1960, but their prevalence increased between 1961 and 1970, and continued at elevated levels through 1979. The toxigenic capabilities of

Precision of a phage susceptibility spot assay assessed with 154 clinical Staphylococcus aureus isolates.

Lytic phages are a promising consideration to combat recalcitrant bacterial infections. To advance the possibility of using phage therapy in the clinical routine, standardized phage susceptibility testing methods are needed. Here, staphylococcal phage K was tested against 154 periprosthetic joint infection-associated Staphylococcus aureus isolates using a double overlay spot assay in technical and biological triplicates. Individual plaques were assessed using a morphology scoring system ranging from 0 (no plaques) to 4 (large, clear plaques), with scores ≥2 considered indicative of infectivity

Phage protein Gp11 blocks Staphylococcus aureus cell division by inhibiting peptidoglycan biosynthesis.

UNLABELLED: Phages and bacteria have a long history of co-evolution. However, these dynamics of phage-host interactions are still largely unknown; identification of phage inhibitors that remodel host metabolism will provide valuable information for target development for antimicrobials. Here, we perform a comprehensive screen for early-gene products of ΦNM1 that inhibit cell growth in Staphylococcus aureus. A small membrane protein, Gp11, with inhibitory effects on S. aureus cell division was identified. A bacterial two-hybrid library containing 345 essential S. aureus genes was constructed to

Antigenic properties and susceptibility to antibiotics of strains of Staphylococcus aureus in the 94, 96 phage complex.

Out of the 2,527 strains typed in 1976, 1977, 1978 and 1979, 226 strains (8.94%) were lysed by phages 94, 96 or 94 + 96. The two phages lyse together 83.17% of strains, whilst phage 94 alone lyses only 3.96% and phage 96 alone 12.87%. 94/96 strains have the S. aureus species antigens, particularly that of biotype A (human) at levels similar to those previously described: Protein A 96%, beta Ribitol teichoic acid (93%). Very characteristic is the presence in these strains of type antigens c1 (89%) and sigma (92%), which are significantly unusual or absent in the strains of other phages types. 9

Genomic diversity of two lineages of exfoliative toxin A-converting phages predominating in Staphylococcus aureus strains in the Czech Republic.

We have isolated and characterized two distinct types of exfoliative toxin A (ETA)-converting bacteriophages originating from Staphylococcus aureus strains responsible for massive outbreaks of pemphigus neonatorum in the Czech Republic. Three induced phages designated as ph iB531, phi B557 and phi B122 were found to be capable of transferring the eta gene into the prophageless non-toxigenic S. aureus strain and converting it into an ETA producer. Comparisons of the phage sequences derived from 12 selected genes and 2 genomic segments (polymorphic P2 and conserved C4) revealed that phi B531 and

Metagenome analysis of Russian and Georgian Pyophage cocktails and a placebo-controlled safety trial of single phage versus phage cocktail in healthy Staphylococcus aureus carriers.

Bacteriophage therapy is a commonly used treatment for Staphylococcus aureus infections in countries of the former Soviet Union, using both single phages and phage cocktails. The scarce data available on Eastern phage cocktails prompted an investigation into commercially-available Pyophage cocktails from two different manufacturers used to treat skin and wound infections. Comparison of the metagenomic composition of two Pyophage products from Georgia and Russia revealed substantial differences in phage-types targeting Escherichia, Enterococcus, Salmonella, Pseudomonas aeruginosa and Proteus, t

Antibacterial efficacy of recombinant phage lysin JD007-LY01 against Staphylococcus aureus in food matrices.

Bacteriophage endolysins can specifically bind to and efficiently lyse host bacteria, and therefore considered promising alternatives to antibiotics. Natural endolysins, however, often have limitations such as a narrow lytic spectrum or low lytic activity, which restrict their broader application in food antibacterial control. In this study, a recombinant endolysin JD007-LY01 with a stable protein structure was constructed by fusing the binding domain of the native endolysin LY01 with the lytic domain of the exogenous endolysin JD007. Experimental results showed that JD007-LY01 lysed all Staph

First characterization of Staphylococcus felis in diabetic foot osteomyelitis: from intracellular persistence to phage treatment.

Staphylococcus felis is a coagulase-negative Staphylococcus (CoNS) primarily associated with the feline microbiota and only rarely reported in human disease. Here, we report its implication in diabetic foot osteomyelitis, and provide the first comprehensive characterization of its pathogenic potential. Two isolates (NSF001 and NSF002), recovered 5 months apart from bone biopsies of the same patient, were analyzed for growth kinetics, biofilm formation, and intracellular persistence in macrophages and osteoblasts. Both isolates proliferated efficiently, produced robust biofilm, and persisted wi
