Necrotizing 'malignant' external otitis caused by Staphylococcus epidermidis.

Necrotizing "malignant" external otitis is a life-threatening skull base infection that originates in the external auditory canal and is characterized by otalgia and purulent aural discharge with external auditory canal cellulitis and granulation. Necrotizing external otitis, seen almost exclusively in elderly diabetics, is almost always caused by Pseudomonas aeruginosa. To our knowledge, there have been only six nonpseudomonal cases reported to date. We describe a 70-year-old diabetic man with necrotizing external otitis caused by Staphylococcus epidermidis, confirmed by serial cultures. This

Methicillin-resistant Staphylococcus epidermidis keratitis treated with vancomycin.

Staphylococcus epidermidis accounts for nearly one third of all cases of bacterial keratitis in certain geographic areas. Recently, the sensitivity of this organism has changed dramatically so that nearly half of nosocomially acquired systemic S epidermidis infections are resistant to methicillin sodium, cephalosporins, and aminoglycosides. Methicillin-resistant and gentamicin sulfate-resistant S epidermidis causing infectious blepharoconjunctivitis and endophthalmitis has previously been reported. Two cases of methicillin- and gentamicin-resistant S epidermidis keratitis occurred that were tr

Common R-plasmids in Staphylococcus aureus and Staphylococcus epidermidis during a nosocomial Staphylococcus aureus outbreak.

During a 7-month period in 1978 to 1979, 31 patients and personnel at a Kentucky hospital were colonized or infected with a Staphylococcus aureus strain resistant to clindamycin, erythromycin, gentamicin, methicillin, penicillin, and tetracycline. S. epidermidis with similar antibiotic resistance patterns had been isolated in this hospital in the year before the S. aureus outbreak. A 32-megadalton R-plasmid, pUW3626, mediating resistance to penicillin and gentamicin, was present in these isolates and in coisolated S. epidermidis from the same outbreak. By colony hybridization, pUW3626 was homo

Recognition of (Sesc) for Easy Identification of Staphylococcus Epidermidis and Molecular and Phenotypic Study of Β-Lactam Resistance in Staphylococcus Epidermidis Isolates in Isfahan.

BACKGROUND: Not only is it crucial to rapidly detect Staphylococcus epidermidis (S. epidermidis) isolates from a broad range of bacteria, but recognizing resistance agents can greatly improve current diagnostic and therapeutic strategies. METHODS: The current cross-sectional study investigated 120 clinical isolates from a nosocomial S. epidermidis infection. The isolates were identified using common biochemical tests, and specific S. epidermidis surface protein C (SesC) primers were used to confirm the presence of S. epidermidis. PCR and special primers were used to detect the β-lactamase gen

Staphylococcus epidermidis DnaK alters biofilm formation and proteome in Staphylococcus aureus CIP 107093.

Staphylococcus aureus and Staphylococcus epidermidis, two Gram-positive bacteria of the human skin microbiota, form biofilms that contribute to dysbiosis and inflammatory skin diseases such as psoriasis and atopic dermatitis. The human calcitonin gene-related peptide (CGRP), involved in skin inflammation, was previously shown to enhance the virulence of S. epidermidis MFP04. We previously observed a significant increase in the level of the molecular chaperone DnaK/Hsp70 in the secretome of CGRP-activated S. epidermidis. Here, we investigated the role of recombinant S. epidermidis DnaK in biofi

Genetic Diversity of Composite Enterotoxigenic Staphylococcus epidermidis Pathogenicity Islands.

The only known elements encoding enterotoxins in coagulase-negative staphylococci are composite Staphylococcus epidermidis pathogenicity islands (SePIs), including SePI and S. epidermidis composite insertion (SeCI) regions. We investigated 1545 Staphylococcus spp. genomes using whole-genome MLST, and queried them for genes of staphylococcal enterotoxin family and for 29 ORFs identified in prototype SePI from S. epidermidis FRI909. Enterotoxin-encoding genes were identified in 97% of Staphylococcus aureus genomes, in one Staphylococcus argenteus genome and in nine S. epidermidis genomes. All en

Staphylococcus epidermidis biofilms control by N-acetylcysteine and rifampicin.

Medical device-associated infections caused by Staphylococcus epidermidis usually involve biofilm formation and its eradication is particularly challenging. Although rifampicin has been proving to be one of the most effective antibiotics against S. epidermidis biofilms, its use as a single agent can lead to the acquisition of resistance. Therefore, we assessed the combined effect of rifampicin with N-acetylcysteine (NAC) known by its mucolytic effect, in the control of S. epidermidis biofilms. Biofilms of 2 S. epidermidis strains (9142 and 1457) were treated with 1x minimum inhibitory concentr

Clinical behavior of implant infections due to Staphylococcus epidermidis.

UNLABELLED: Surgical implants and other foreign material are increasingly used in modern medicine to restore or to improve the function of the human body. Infection of an implant is associated with considerable morbidity due to frequent hospitalizations, surgery and antimicrobial treatment. The underlying mechanism is the formation of a bacterial biofilm on the surface of the implanted body. The recognition and diagnosis of implant infections is essential for further therapy and, above all, the decision to remove and exchange the implant. METHODS: We compared the data of 60 patients with impl

Quorum-sensing control of biofilm factors in Staphylococcus epidermidis.

Staphylococcus epidermidis is the most frequent cause of nosocomial sepsis and catheter-related infections, in which biofilm formation is considered to be the main virulence mechanism. Quorum-sensing systems have been recognized as important regulators of virulence and biofilm formation in many bacteria. There is a single quorum-sensing system in S. epidermidis encoded by the agr operon. To investigate quorum-sensing control of biofilm formation, we constructed an agr deletion mutant, assayed for the different stages of biofilm formation, and determined agr-dependent regulation of biofilm fact

Staphylococcus epidermidis: emerging resistance and need for alternative agents.

Previously, Staphylococcus epidermidis and other coagulase-negative staphylococci isolated from the blood of hospitalized patients were often considered contaminants. Now, coagulase-negative staphylococci are among the leading causes of nosocomial blood infections. Multidrug resistance could predict a true nosocomial infection rather than a blood culture contaminant. Recent studies indicated the emergence of resistance to the quinolones, particularly to ciprofloxacin. Tolerance and occasional resistance to vancomycin have been reported recently. In addition, several reports indicated that vanc

Staphylococcus epidermidis adhesion to films deposited from hydroxyethylmethacrylate plasma.

The adhesion of S. epidermidis ATCC 35984 strain on polystyrene (PS) disks coated by films deposited from hydroxyethylmethacrylate (HEMA) plasma was evaluated and compared to adhesion on untreated PS and oxygen-plasma-treated PS. Films were deposited keeping constant the monomer flow rate while the discharge power ranged from 40-100 W in order to obtain coating with different surface properties. Surface chemistry, energetics, and morphology were evaluated by Electron Spectroscopy for Chemical Analysis (ESCA), contact angle measurement, and Atomic Force Microscopy (AFM), respectively. Topical vancomycin formulation for methicillin-resistant Staphylococcus epidermidis blepharoconjunctivitis.

We successfully treated two patients with severe Staphylococcus epidermidis blepharoconjunctivitis by means of a topical vancomycin hydrochloride solution (50 mg/ml) prepared with sterile water. Aqueous vancomycin preparations, however, cause significant ocular irritation probably because of low pH and osmolality values. Solutions prepared with normal saline (5 mg/ml) and phosphate-buffered artificial tears (5 mg/ml and 50 mg/ml) were significantly less irritating and possessed equivalent in vitro antimicrobial activity. Topical vancomycin should be used only when commercially available antibi

Topical vancomycin for the treatment of Staphylococcus epidermidis and methicillin-resistant Staphylococcus aureus conjunctivitis.

Staphylococcus aureus and Staphylococcus epidermidis are organisms that frequently cause conjunctivitis or blepharoconjunctivitis. We describe a patient with methicillin-resistant S. aureus and S. epidermidis conjunctivitis who was treated successfully using an extemporaneously prepared topical ophthalmic solution of vancomycin hydrochloride 31 mg/mL. Studies describing the preparation, stability, and comfort of this solution, as well as reports pertaining to efficacy, are reviewed. Controlled clinical trials evaluating the safety and efficacy of vancomycin ophthalmic solution have not yet bee

Characteristics of Staphylococcus epidermidis Isolated from Humans and Animals.

Staphylococcus epidermidis is an important opportunistic pathogen and a potential reservoir of antimicrobial resistance determinants, yet comparatively little is known about isolates originating from both humans and companion animals. This study characterized 320 S. epidermidis isolates obtained from humans, cats, and dogs within a One Health framework using phenotypic, molecular, proteomic, and genomic approaches. Antimicrobial susceptibility testing revealed high frequencies of resistance to oxacillin (67.19%), erythromycin (66.56%), tetracycline (55.63%), and clindamycin (53.75%) by disk di

Antibacterial and Antibiofilm Activity of Closantel Against Staphylococcus epidermidis.

Staphylococcus epidermidis is recognized as the major cause of implanted indwelling medical device-related infections. The ability of S. epidermidis to form biofilms largely increases its resistance to conventional antibiotics, which is the major cause of treatment failure. Therefore, there is a pressing need to discover novel antimicrobials against S. epidermidis biofilms. In this study, Closantel (Clos), an antiparasitic drug, was repurposed to be effective against S. epidermidis planktonic cells with the minimal inhibitory concentration values of 0.25-0.5 μg/mL. Clos exhibited potent biofil
