Bifidobacterium longum S3 alleviates loperamide-induced constipation by modulating intestinal acetic acid and stearic acid levels in mice.

Constipation is a major gastrointestinal (GI) symptom worldwide, with diverse causes of formation, and requires effective and safe therapeutic measures. In the present study, we used loperamide hydrochloride to establish a constipation model and assessed the effect of Bifidobacterium on constipation and its possible mechanism of relief. The results showed that B. longum S3 exerted a constipation-relieving effect primarily by improving the gut microbiota, enriching genera including Lactobacillus, Alistipes, and Ruminococcaceae UCG-007, and decreasing the bacteria Lachnospiraceae NK4B4 group. Mixed Monolayer Characteristics of Tangeretin with Stearic Acid and Dipalmitoyl Phosphatidyl Choline at the Air/Water Interface.

As a natural compound with various biological activities, tangeretin (TAN) has numerous potential advantages in food and medical fields. However, its poor aqueous solubility and bioavailability limit its effective application. In this paper, the mixed monolayer characteristics of different mass fractions of TAN with stearic acid (SA) and dipalmitoyl phosphatidyl choline (DPPC) were investigated. The optimal film-forming composition was obtained by analyzing the molecular area, monolayer compressibility, shrinkage and expansion, and other thermodynamic parameters. The results indicated that the

The signature 10-hydroxy stearic acid thought to correlate with infectivity in oocysts of Cryptosporidium species is an artifact.

Heating or freezing leads to loss in infectivity of oocysts of Cryptosporidium parvum toward neonatal BALB/c mice and is reflected in the profile of the polar lipid fatty acids. Upon loss of infectivity, the ratio of polar lipid to neutral lipid fatty acid decreased and the relative proportions of 18:1n-9 also decreased; proportions of 18:2n-6 and 20:5n-6 increased, whereas the proportions of 16:0 remained constant with freezing. During these investigations, a novel fatty acid, 10-OH 18:0, was discovered in the glycolipid fraction. The identification of a fatty acid unique to species of Crypto

Stearic acid methyl ester affords neuroprotection and improves functional outcomes after cardiac arrest.

Cardiac arrest causes neuronal damage and functional impairments that can result in learning/memory dysfunction after ischemia. We previously identified a saturated fatty acid (stearic acid methyl ester, SAME) that was released from the superior cervical ganglion (sympathetic ganglion). The function of stearic acid methyl ester is currently unknown. Here, we show that SAME can inhibit the detrimental effects of global cerebral ischemia (i.e. cardiac arrest). Treatment with SAME in the presence of asphyxial cardiac arrest (ACA) revived learning and working memory deficits. Similarly, SAME-treat

Tumor microenvironment-responsive nanoparticles composed of hyaluronic acid and stearic acid for the treatment of lung metastasis in triple-negative breast cancer.

Triple-negative breast cancer lung metastasis is a key challenge in its treatment. We herein present an intelligent nanoparticle delivery platform, HCS, which was fabricated by synthesizing a polymer long-chain SA-CY-HA (HCS) through a two-step amidation reaction utilizing stearic acid (SA), cystamine dihydrochloride (CY), and hyaluronic acid (HA). This platform facilitates the co-delivery of paclitaxel (PTX) and honokiol (Hol), thereby concurrently suppressing tumor growth and lung metastasis. HCS can encapsulate the hydrophobic soluble drugs PTX and Hol internally, enhancing their solubility

Design and In Vitro Evaluation of Gambogic Acid-Conjugated Stearic Acid Solid Lipid Nanoparticles for Transferrin Receptor-Mediated Drug Delivery.

Solid lipid nanoparticles (SLNs) have garnered significant interest for their safety and efficacy, especially following the success of COVID-19 mRNA vaccines. This study presents the synthesis and characterization of a novel stearic acid (SA)-gambogic acid (GA) conjugate, where GA, a xanthonoid, exhibits high affinity for the transferrin receptor (TfR) without competing with endogenous transferrin. The SA-GA conjugate was employed to formulate SLNs using a hot homogenization-ultrasonication-solvent evaporation technique for the peroral delivery of cyclosporine (CsA), paclitaxel (PTX), and urol

Stearic acid-functionalized high amylose starch nanoparticles: Tailoring interfacial wettability and thermal stability via fatty acid localization.

High amylose starch nanoparticles (HSNPs) were functionalized with stearic acid (SA) to enhance their efficacy as Pickering emulsion stabilizers. Through ethanol pre-washing, removable and residual SA fractions were differentiated, revealing that removable SA primarily dictates surface hydrophobicity. Specifically, 8% SA loading increased the contact angle from 68° to 80°, indicating enhanced surface hydrophobicity and favoring particle adsorption at the oil-water interface. Emulsions stabilized by HSNP-SA composites exhibited superior stability, with unwashed 8% SA samples achieving a 100% em

Bio-degradable superhydrophobic cellulose-based paper straws fabricated through coating of citric acid-crosslinking CNF & stearic acid.

To address the poor water resistance and mechanical degradation of conventional paper straws, this study develops an environmentally benign, fully biodegradable superhydrophobic coating using cellulose nanofibers (CNF), stearic acid (SA) and citric acid (CA). CNF was first dispersed in ethanol, followed by chemical grafting of hydrophobic alkyl chains onto its surface; a synergistic micro-nano hierarchical structure was then constructed by optimizing thermal soaking and drying processes. The hybrid material's morphology was comprehensively characterized via FTIR, SEM, and AFM. The resulting co

Stearic acid-modified PSMA-targeting peptide-drug conjugate for long-acting prostate cancer therapy.

Prostate-specific membrane antigen (PSMA)-targeting peptide-drug conjugates (PDCs) offer promise for the treatment of PSMA-positive prostate cancer, but their applications are limited by rapid clearance, poor pharmacokinetics, and low efficacy. Herein, we design a series of PDCs incorporating rational functional moieties, including a Glu-Ureido-Lys PSMA-targeting ligand, the cytotoxic payload monomethyl auristatin E (MMAE), and a cathepsin B-cleavable Val-Cit linker, combined with structural modifications such as free amine exposure, acetylation, stearic acid acylation, or p-iodophenylbutyric

Atmosphere-Dependent Radiation Stabilization of Stearic Acid on Vaterite CaCO3: A Comparison of Gamma and Electron-Beam Irradiation.

Calcium carbonate is a widely used filler in polymer composites due to its low cost and ability to improve stiffness, dimensional stability, and impact resistance. However, its hydrophilic surface limits compatibility with nonpolar polymer matrices, making surface modification essential to improve filler dispersion and interfacial adhesion. Stearic acid is commonly applied as a surface modifier for calcium carbonate because it readily chemisorbs onto the mineral surface and forms densely packed self-assembled monolayers that improve hydrophobic character. Despite its widespread use, stearic ac

Thiolated Chitosan-Stearic Acid Thiol-Linked Biomimetic Hydrogels for Axonal Regrowth and Neuronal Tissue Regeneration.

Neuronal disorders like spinal cord injury, stroke, peripheral neuropathy, and neurodegenerative diseases like Alzheimer's disease, Parkinson's disease, etc., need regenerative approaches and therapeutics like tissue engineering and cell therapy for neuronal systems. Significant factors such as neuronal damage, impaired immune response, and glial scar formation create a hostile environment for neural repair and functional recovery. To overcome these challenges, the development of biocompatible and bioactive biomaterials is essential for therapeutic applications. In this work, a thiolated chito

Lipophilic 5-fluorouracil prodrug encapsulated xylan-stearic acid conjugates nanoparticles for colon cancer therapy.

In this study, self-assembled nanoparticles based on amphiphilic xylan-stearic acid (Xyl-SA) conjugates have been developed for the efficient delivery of 5-fluorouracil (5-FU) in cancer therapy. The self-assembled behavior of Xyl-SA conjugates in aqueous medium was investigated using pyrene as fluorescent probe. To enhance the loading efficacy of 5-FU, the lipophilic 5-fluorouracil-stearic acid (5-FUSA) prodrug was synthesized and subsequently encapsulated into the hydrophobic core of Xyl-SA NPs. The obtained Xyl-SA/5-FUSA NPs had an appropriate size (~278 nm), high drug loading of 5-FUSA (~14

Preparation and evaluation of novel directly-compressed fast-disintegrating furosemide tablets with sucrose stearic acid ester.

Fast-disintegrating tablets of furosemide (FS) were prepared by the novel direct compression method. FS, microcrystalline cellulose (MC), croscarmellose sodium (CC), xylitol (XL) and sucrose stearic acid esters (SSEs) with an hydrophilic-lipophilic balance (HLB) of 16, 15 and 11, named S1670, S1570 and S1170, were used. An FS/SSE/MC mixed powder was obtained by solvent evaporation of a suspension of MC in ethanol solution containing FS and SSE, and the resultant mixed powder was mixed with CC and XL, and directly compressed. The tablets with hardness of more than 40 N and disintegration time o

Topical application of docosanol- or stearic acid-containing creams reduces severity of phenol burn wounds in mice.

Because of their reported antiviral and anti-inflammatory activities, cream formulations containing n-docosanol (docosanol) or stearic acid were tested for effects on chemically-induced burns in mice. In this model, injury was induced by painting the abdomens of mice with a chloroform solution of phenol. This was followed by the topical application of test substances 0.5, 3, and 6 h later. Progression of the wounds was assessed by a single evaluator after 8 h, using a numerical score of gross morphology. Docosanol- and stearic acid-containing creams substantially and reproducibly lessened the

Elovl6 inhibits colorectal cancer progression through stearic acid-mediated mitochondrial fusion and metabolic reprogramming.

Lipid metabolic reprogramming is a hallmark of colorectal cancer (CRC), yet the precise molecular mechanisms underlying lipid-mediated oncogenesis and the specific lipid metabolic enzymes involved remain largely elusive. Here, we identify elongation of very-long-chain fatty acid protein 6 (Elovl6) as a critical regulator in CRC progression. Clinical data reveal significant down-regulation of Elovl6 in colon cancer tissues, with low expression levels correlating with unfavorable patient prognosis. We demonstrate that Elovl6 exerts potent tumor-suppressive effects, significantly inhibiting cellu

Mechanisms of testicular toxicity uncovered by multi-omics: 9,10-epoxy stearic acid as a foodborne contaminant in mice.

High-temperature cooking processes generate oxidized lipids such as 9,10-epoxy stearic acid (ESA), yet its reproductive toxicity remains poorly characterized. Here, we demonstrated that ESA exposure in mice induces hepatotoxicity, reduced serum testosterone, and male reproductive dysfunction, manifested by testicular injury, disrupted spermatogenesis, and impaired sperm quality. Integrated metabolomics and proteomics analysis revealed that ESA disrupts amino acid and carbohydrate metabolic homeostasis, and analysis of these two datasets suggests that mitochondrial dysfunction is likely a key f

Effect of a blend of palmitic and stearic acid on transfer of preformed fatty acids into milk fat using an 18 C n-3 fatty acid perturbation model.

The transfer of dietary fatty acids (FA) into milk fat is variable and has been reported to be higher on lower fat diets, which may be a homeorhetic adaptation to support milk fat production. The objective of this study was to determine the effect of increasing dietary fat on the kinetics of transfer of preformed FA into milk fat using a perturbation model based on abomasal infusion of a bolus of n-3 FA. Eight ruminally cannulated multiparous Holstein cows (115 ± 32 DIM and producing 41.5 ± 4.43 kg of milk; mean ± SD) were used in a switchback design with three 21-d periods. Treatments were a

Hydrophobically modified PEG-driven morphology control in thermo-responsive biosourced 12-hydroxy-stearic acid assemblies.

HYPOTHESIS: We have recently shown that mixing green fatty acid 12-hydroxystearic acid (12-HSA) molecules with a low amount of various end-capped polyethylene glycol (PEG) chains (mono- or di-functionalized by 12-HSA or stearic acid (SA) moieties) provides a straightforward one-pot route to design PEGylated self-assemblies, either vesicles or multilamellar tubes. Tuning the concentration of end-capped PEG chains offers a simple strategy to precisely control the morphology of these PEGylated architectures. EXPERIMENTAL: Four types of 4 kDa end-capped PEG chains, functionalized at one or both e
