Modified β-glucans promote saponin and fucosylated chondroitin sulfate biosynthesis in sea cucumber Apostichopus japonicus in association with activation of the TLR-MAPK signaling pathway.

Sea cucumber is a high-value marine food that contains characteristic bioactive components, notably saponins and polysaccharides. Strategies promoting their biosynthesis to improve sea cucumber quality are vital for the industry. The impacts of three dietary yeast β-glucans (native β-glucan, ultrasonic-degraded low-molecular-weight β-glucan, and carboxymethylated β-glucan) on the biosynthesis of saponin and fucosylated chondroitin sulfate (FCS) in A. japonicus and the underlying mechanism were investigated. A 28-day feeding trial showed that modified β-glucans improved growth performance and i

First record of Japanese sea cucumber Apostichopus japonicus (Selenka, 1867) (Aspidochirotida: Stichopodidae) from the Mediterranean Sea, Turkey.

The Japanese sea cucumber Apostichopus japonicus (Selenka, 1867), native to the East Asian coasts, was recorded for the first time in the Mediterranean Sea along the east Aegean coast (Izmir, Turkey). Fifty-eight specimens were collected by scuba diving at depths up to 10 m between July and August 2024. Species identity was confirmed by morphological examination and molecular analyses using partial regions of the cytochrome oxidase subunit I (COI) and 16S ribosomal RNA (16S rRNA) genes. Phylogenetic analyses based on COI and 16S rRNA sequences confirmed species identity, with haplotypes closel

Anti-inflammatory Effects of the Fucoidan from Sea Cucumber Apostichopus japonicus.

Fucoidan from Apostichopus japonicus (Aj-FUC) has shown anti-inflammatory activity, whereas its mechanism was not explicated. This study investigated the anti-inflammatory potential and mechanism of the fucoidan from green and purple A. japonicus (G-FUC and P-FUC) in lipopolysaccharide (LPS)-treated RAW264.7 cells. Results showed that Aj-FUCs at 25-400 µg/mL had no toxicity to cells after 24 h stimulation and promoted cell phagocytic activity. ELISA results indicated that Aj-FUC reduced the nitric oxide (NO), tumor necrosis factor α (TNF-α), interleukin-1β (IL-1β), and IL-6 levels and increase

High-value components and bioactives from sea cucumbers for functional foods--a review.

Sea cucumbers, belonging to the class Holothuroidea, are marine invertebrates, habitually found in the benthic areas and deep seas across the world. They have high commercial value coupled with increasing global production and trade. Sea cucumbers, informally named as bêche-de-mer, or gamat, have long been used for food and folk medicine in the communities of Asia and Middle East. Nutritionally, sea cucumbers have an impressive profile of valuable nutrients such as Vitamin A, Vitamin B1 (thiamine), Vitamin B2 (riboflavin), Vitamin B3 (niacin), and minerals, especially calcium, magnesium, iron

Efficacy of 'Gamadent' toothpaste on the healing of gingival tissues: a preliminary report.

The aim of this randomised, parallel, double-blind study, in which 28 adult patients diagnosed with chronic gingivitis or early stages of chronic periodontitis were recruited, was to evaluate the efficacy of 'Gamadent' toothpaste compared to a placebo toothpaste. 'Gamadent' toothpaste has all the basic constituents of a toothpaste with the addition of a sea cucumber extract (SCE) of the species Stichopus sp. 1 to improve the healing potential of tissues. The placebo has the same basic constituents minus the extract. Out of the 28 patients, 14 were placed in the test group who used the 'Gamaden

The nuclear receptor coactivator 4 regulates ferritinophagy induced by vibrio splendidus in coelomocytes of Apostichopus japonicus.

Iron homeostasis is vital for the host's defense against pathogenic invasion and the ferritinophagy is a crucial mechanism in maintaining intracellular iron homeostasis by facilitating the degradation and recycling of stored iron. The nuclear receptor coactivator 4 (NCOA4) serves as a ferritinophagy receptor, facilitating the binding and delivery of ferritin to the autophagosome and lysosome. However, NCOA4 of the sea cucumber Apostichopus japonicus (AjNCOA4) has not been reported until now. In this study, we identified and characterized AjNCOA4 in A. japonicus. This gene encodes a polypeptide

A Peptide HEPFYGNEGALR from Apostichopus japonicus Alleviates Acute Alcoholic Liver Injury by Enhancing Antioxidant Response in Male C57BL/6J Mice.

Liver-related disease caused by alcohol is a frequent disorder of the hepatic tract. Heavy consumption of alcohol in a short period causes oxidative damage to the liver. Sea cucumber is abundant in nutrients and its various extracts have been studied for antioxidant properties. One peptide was isolated and identified from Apostichopus japonicus in our recent study. We investigated the benefits of the peptide in a model of acute ethanol-induced male C57BL/6J mice. Dietary intake of the peptide could attenuate hepatomegaly, hepatitis and the accumulation of lipid droplets, and increase antioxida

Antidepressant effect of the peptide EIILEAGK from Apostichopus japonicus via inflammatory and mitochondrial regulations in a lipopolysaccharide mouse model.

An increasing number of studies support the hypothesis of inflammation-induced depression, and several anti-inflammatory drugs are known to target tumour necrosis factor ɑ (TNF-ɑ) and interleukin 6 (IL-6) to show antidepressant potential. Here, a novel peptide derived from the sea cucumber Apostichopus japonicus was found to have an anti-inflammatory effect, as demonstrated by inhibiting the activation of NF-κB and reducing the levels of pro-inflammatory cytokines (TNF-α, IL-6, and IL-1β) in the brain tissue and blood of lipopolysaccharide (LPS)-treated mice. Interestingly, LPS-induced depress

Fucoidan from Apostichopus japonicus ameliorates alcoholic liver disease by regulating gut-liver axis homeostasis.

Long-term and excessive alcohol consumption can lead to the development of alcoholic liver disease (ALD), characterized by oxidative damage, intestinal barrier injury, and disruption of intestinal microbiota. In this study, we extracted fucoidan (Aj-FUC) from Apostichopus japonicus using enzymatic methods and characterized its structure. The ALD model was established in male Balb/c mice using 56° Baijiu, with silymarin as a positive control. Mice were orally administered 100 mg/kg·bw and 300 mg/kg·bw of Aj-FUC for 28 days to evaluate its effects on liver injury in ALD mice and explore its pote

A novel C-type lectin from the sea cucumber Apostichopus japonicus (AjCTL-2) with preferential binding of d-galactose.

C-type lectins (CTLs) are Ca2+ dependent carbohydrate-binding proteins that share structural homology in their carbohydrate-recognition domains (CRDs). In the present study, a novel CTL was identified from sea cucumber Apostichopus japonicus (named as AjCTL-2). The deduced amino acid sequence of AjCTL-2 was homologous to CTLs from other animals with the identities ranging from 33% to 40%. It contained a canonical signal peptide at the N-terminus, a low density lipoprotein receptor class A (LDLa), a C1r/C1s/Uegf/bone morphogenetic protein 1 (CUB), and a CRD with two motifs Glu-Pro-Asn (EPN) and

Metabolic reprogramming in cancer and senescence.

The rising trend in global cancer incidence has caused widespread concern, one of the main reasons being the aging of the global population. Statistical data show that cancer incidence and mortality rates show a clear upward trend with age. Although there is a commonality between dysregulated nutrient sensing, which is one of the main features of aging, and metabolic reprogramming of tumor cells, the specific regulatory relationship is not clear. This manuscript intends to comprehensively analyze the relationship between senescence and tumor metabolic reprogramming; as well as reveal the impac
