Exploring congenital sucrase-isomaltase deficiency in autism spectrum disorder patients with irritable bowel syndrome symptoms: A prospective SI gene sequencing study.

Congenital sucrase-isomaltase deficiency (CSID) is an inherited metabolic disorder causing chronic gastrointestinal symptoms and malnutrition when untreated. Most CSID patients are likely to remain under- or misdiagnosed. This study aimed to investigate prevalence of CSID among patients with autism spectrum disorder (ASD) presenting with irritable bowel syndrome (IBS) symptoms via prospective SI gene sequencing. A prospective cross-sectional study was conducted on 98 ASD patients exhibiting gastrointestinal symptoms consistent with IBS. Participants were assessed according to Rome IV criteria

Structural and physicochemical characterisation of branched dextrans produced by an active α-(1→2) branching sucrase from Apilactobacillus kunkeei PDER37.

Recently, branching sucrases encoded in the genomes of certain Lactic Acid Bacteria (LAB) strains have become novel enzymes to obtain branched α-glucans. In this study an active α-(1 → 2) branching sucrase from Apilactobacillus kunkeei PDER37 was expressed, characterised and distinct branched dextrans was obtained with reactions under different sucrose: dextran ratio. Structural characterisation by 1H and 13C NMR analysis demonstrated the branching of the dextran with (1 → 2)-linked α-d-glucose units with no alteration in the final structure depending on sucrose: dextran ratio (D0) but this ra

Severe pathogenic variants of intestinal sucrase-isomaltase interact avidly with the wild type enzyme and negatively impact its function and trafficking.

Sucrase-isomaltase (SI) is the major disaccharidase of the small intestine, exhibiting a broad α-glucosidase activity profile. The importance of SI in gut health is typified by the development of sucrose and starch maldigestion in individuals carrying mutations in the SI gene, like in congenital sucrase-isomaltase deficiency (CSID). Common and rare defective SI gene variants (SIGVs) have also been shown to increase the risk of irritable bowel syndrome (IBS) with symptoms and clinical features similar to CSID and also in symptomatic heterozygote carriers. Here, we investigate the impact of the

Interaction between the α-glucosidases, sucrase-isomaltase and maltase-glucoamylase, in human intestinal brush border membranes and its potential impact on disaccharide digestion.

The two major intestinal α-glycosidases, sucrase-isomaltase (SI) and maltase-glucoamylase (MGAM), are active towards α-1,4 glycosidic linkages that prevail in starch. These enzymes share striking structural similarities and follow similar biosynthetic pathways. It has been hypothesized that starch digestion can be modulated via "toggling" of activities of these mucosal α-glycosidases, suggesting a possible interaction between these two enzyme complexes in the intestinal brush border membrane (BBM). Here, the potential interaction between SI and MGAM was investigated in solubilized BBMs utilizi

Potent sucrase inhibition by purple sweet potato anthocyanins for glycemic control: unveiling peonidin-3-caffeoyl-feruloylsophoroside-5-glucoside as the key functional monomer and its interaction mechanism.

Purple sweet potato anthocyanins (APSP) are promising natural sucrase-inhibitory agents for postprandial glycemic control; however, their key bioactive constituents and underlying mechanisms remain unclear. In this study, APSP was found to interact with sucrase (493.33 ± 29.02 nm), leading conformational changes through spontaneous binding. Peonidin-3-caffeoyl-feruloylsophoroside-5-glucoside (P-3-C-FS-5-G) was identified and isolated (purity ≥90.5%) as the major sucrase-inhibitory monomer from APSP. P-3-C-FS-5-G produced dose-dependent competitive inhibition (inhibition rate 24.11 ± 3.05%). It

The Associations of Sucrase-Isomaltase Hypomorphic Variants With Long-Term Outcomes and Dietary Intake in an Australian Irritable Bowel Syndrome Population Educated on the FODMAP Diet: A Cross-Sectional and Retrospective Study.

BACKGROUND: Hypomorphic variants of sucrase-isomaltase (SI) have been associated with irritable bowel syndrome (IBS) in adults, but how their presence influences therapeutic outcomes is uncertain. AIMS: To investigate the frequency of sucrase-isomaltase hypomorphic variants in patients with IBS and their association with short- and long-term outcomes after initiation of a FODMAP diet. METHODS: Clinical outcomes in patients with IBS were retrospectively examined at mean 7.1 (range 2.5-13.4) years after being educated on a FODMAP diet by a gastrointestinal dietitian and their current food inta

[Genetics of sucrose metabolism disorders in different population groups].

The study of the genetic determinants of the disaccharidase activity opens up new prospects for improving diagnostics and choosing medical tactics in gastroenterology. The aim of the study was to systematize the data on the role of the sucrase-isomaltase gene (SI) in regulating sucrose metabolism and the contribution of SI mutations to the prevalence of sucrose malabsorption disorders (sucrase-isomaltase deficiency, SID) and certain forms of enterological pathology in different population groups. Material and methods. A review of the peer-reviewed scientific literature, mainly in the PubMed da

Precancerous changes in the stomach.

Intestinal metaplasia is often associated with human gastric carcinoma. Intestinalization seems to be a typical example of abnormal differentiation and is possibly a precancerous state. For investigation of intestinal metaplasia, a method for visualizing disaccharidases using Tes-Tape was developed; this method was applied to many specimens of stomach surgically removed for the treatment of gastric carcinoma. More than 130 specimens of human stomach were investigated. Intestinalization was classified into types I and II intestinal metaplasia. In type I intestinal metaplasia, sucrase, maltase,

Use of the Biphasic (13)C-Sucrose/Glucose Breath Test to Assess Sucrose Maldigestion in Adults with Functional Bowel Disorders.

Sucrase insufficiency has been observed in children with of functional bowel disorders (FBD) and symptoms of dietary carbohydrate intolerance may be indistinguishable from those of FBD. A two-phase (13)C-sucrose/(13)C-glucose breath test ((13)C-S/GBT) was used to assess sucrase activity because disaccharidase assays are seldom performed in adults. When (13)C-sucrose is hydrolyzed to liberate monosaccharides, oxidation to (13)CO2 is a proportional indicator of sucrase activity. Subsequently, (13)C-glucose oxidation rate was determined after a secondary substrate ingestion (superdose) to adjust

Differentiation status of rat enterocytes after intestinal adaptation to jejunoileal bypass.

The differentiation status of epithelial cells in intestinal adaptation remains unclear. To determine whether enterocytes reach optimum maturity following adaptation after 85% shortening of the rat gut by jejunoileal bypass surgery, activities of two brush border enzymatic markers of differentiation, alkaline phosphatase and sucrase, were examined in subpopulations of epithelial cells isolated sequentially from the villus/crypt axis of normal (sham operated) and hyperplastic mucosa. In jejunal villi, adaptational hyperplasia was associated with an increase in total epithelial alkaline phosphat

Colonic and small-intestinal phenotypes in gastric cancers: relationships with clinicopathological findings.

The clinicopathological significance of colonic and small-intestinal phenotypes has hitherto remained unclear in gastric cancers. The purpose of the present study was therefore to examine 86 gastric carcinomas histologically and phenotypically using several phenotypic markers, including colon-specific carbonic anhydrase 1 (CA1) and sucrase as small-intestine specific marker. Of 86 gastric cancers, sucrase and CA1 expression was observed in 12 (14.0%) and only in two cases (2.3%), respectively, associated with other intestinal markers such as villin and mucin core protein (MUC)2. In the sucrase

Effects of Sepiolite and Biochar on Enzyme Activity of Soil Contaminated by Cd and Atrazine.

The effects of sepiolite and biochar on the enzymatic activities of the soil in Cd- and atrazine-contaminated soils were studied. During the growth of pakchoi, the activities of acid phosphatase, sucrase, acid protease, and cellulase decreased, catalase activity increased, and urease activity decreased first and then increased. At the first harvest, compared with that for the control group, the soil pH after treatment with remediation materials increased from 5.41 to 7.43; the activities of urease, acid protease, and catalase increased by 62.8%, 38.6%, and 86.1%, respectively. Detection of primary and secondary sucrose malabsorption in children by means of the breath hydrogen technique.

Eighty-one breath hydrogen tests were performed in 72 children who were investigated either because of chronic diarrhoea or recurrent abdominal pain, or because they were relatives of patients with congenital sucrase-isomaltase deficiency. The results of tests on 16 patients were positive (sucrase-isomaltase deficiency, seven patients; secondary sucrose malabsorption, nine patients). Hydrogen production was higher in patients with sucrase-isomaltase deficiency (mean, 96 ppm; range, 20 ppm to 432ppm) than in those with secondary sucrose malabsorption (mean, 21 ppm; range, 12ppm to 51ppm). Effect of chronic hypo and hypervitaminosis C on the brush border enzymes and the intestinal uptake of glucose and alanine.

Brush border sucrase and alkaline phosphatase activities are considerably enhanced in the intestine of ascorbic acid deficient guinea-pigs. Similar increase in the uptake of D-glucose and L-alanine also occurs in chronic vitamin C deficiency. However the permeability of D-glucose and L-alanine in the intestine of animals fed with large doses of vitamin C is severely depressed, with a reduction in the levels of sucrase and alkaline phosphatase activities.

Differentiation of human umbilical cord mesenchymal stem cells into functional intestinal epithelial cells via conditioned medium co-culture.

BACKGROUND: The induction of stem cell differentiation to generate intestinal epithelial cells (IECs) with absorptive functions offers significant therapeutic potential for treating conditions such as Crohn's disease, ulcerative colitis, radiation enteritis, and other refractory intestinal epithelial injuries. Human umbilical cord mesenchymal stem cells (hUC-MSCs) are capable of differentiating into functional IEC-like cells. OBJECTIVE: This study aimed to induce the differentiation of hUC-MSCs into IECs using a conditioned medium co-culture method. METHOD: A culture medium derived from huma

Effect of epidermal growth factor on disaccharidase activity in intestinal epithelial cells of mice.

The effects of epidermal growth factor (EGF) on disaccharidase activity in intestinal epithelial cells were studied in mice. In salivectomised mice, the sucrase and maltase activities decreased significantly compared to the control group. When EGF was given to salivectomised mice, the sucrase and maltase activities rose, but still remained below the normal values. These results show that the submaxillary glands produced another factor, beside EGF, which induced glycoprotein enzyme formation in the Golgi complex.

Associations of 13C-Sucrose Breath Test Dynamics with Anthropometry and Demographics: A Comparison of Studies in the United Kingdom and Zambia.

BACKGROUND: The 13C-sucrose breath test (13C-SBT) is a noninvasive diagnostic test that has been used to assess intestinal sucrase-isomaltase activity, which may be altered in gut function disorders such as environmental enteric dysfunction (EED), a syndrome characterized by chronic inflammation and histologic changes in the small intestine and thought to be ubiquitous among people living without access to improved water and sanitation. However, characterizing associations between 13C-SBT breath curves and gut function status requires disaggregating associations with sucrase-isomaltase activit

[Enzymatic activity of the microsomal fraction of the mucosa of the small intestine in guinea pigs with vitamin D and C deficiencies].

The influence of vitamin D and C deficiency on the kinetic parameters of sucrase and alkali phosphatase activities was studied in the microsomal fraction of the small intestinal mucosa of guinea pigs. It was found that Km values for these enzymes did not depend on the animal providing with these vitamins. Deficiency of one of these vitamins did not influence sucrase activity, however, simultaneous elimination of vitamins D and C resulted in the activity rise by 92%. Alkali phosphatase and Ca-ATPase activities proved to be similarly dependent on providing with vitamin D in the presence of vitam
