Cytochrome P450-Catalyzed Hydroxylation of Δ8-Tetrahydrocannabinol and Implications for Pharmacogenetics.

Introduction: The use of Δ8-tetrahydrocannabinol (Δ8-THC) has become increasingly prevalent in the United States, but its metabolic pathways remain poorly characterized. This study aimed to identify the key enzymes responsible for the 11'-hydroxylation of Δ8-THC, its primary metabolic pathway, and assess the effect of metabolizing enzyme genotype on this activity. Results: The intrinsic clearance of recombinant (r) CYP2C9 was >6200-fold higher than that observed for rCYP2C19, the only other tested CYP450 with detectable activity against Δ8-THC. The Km,u of rCYP2C9 (0.013 ± 0.0032 µM) was compa

Potential Role of CYP3A4 in Determining In Vivo Exposure to Cannabidiol (CBD) and its Active Metabolite 7-OH-CBD: Evidence from an In Vitro Study.

BACKGROUND: Plasma concentration of cannabidiol (CBD) is a determining factor for its antiseizure efficacy. Since CBD bioavailability decreases with increasing dose, pharmacokinetic drug-drug interactions that reduce its metabolic clearance may represent an alternative strategy to increase systemic exposure without further dose escalation. OBJECTIVE: To investigate in vitro how combinations of antiseizure medications (ASMs) with varying cytochrome P450 (CYP450) inhibitory properties influence the metabolism of CBD and 7-OH-CBD. METHODS: CBD was incubated with human liver microsomes (HLMs) ei

Dermatomyositis: immunological landscape, biomarkers, and potential candidate drugs.

INTRODUCTION: Dermatomyositis (DM) is a rare inflammatory disease characterized by the invasion of the skin and muscles. Environmental, genetic, and immunological factors contribute to disease pathology. To date, no bioinformatics studies have been conducted on the potential pathogenic genes and immune cell infiltration in DM. Therefore, we aimed to identify differentially expressed genes (DEGs) and immune cells, as well as potential pathogenic genes and immune characteristics, which may be useful for the diagnosis and treatment of DM. METHOD: GSE1551, GSE5370, GSE39454, and GSE48280 from Gen

Fraction metabolized by cytochrome P450 enzymes: A comprehensive evaluation of the translatability of an in vitro HepatoPac assay.

In our previous study, long-term cocultured hepatocytes were used to estimate the fraction of a drug metabolized by CYP3A4 (fm,CYP3A4). Metabolic turnover was measured with and without a CYP3A4 selective inhibitor, and the results were verified against in vivo reference data. The current study followed a similar approach using direct or time-dependent inhibitors to evaluate fm,CYP1A2, fm,CYP2C8, fm,CYP2C9, fm,CYP2C19, and fm,CYP2D6 for a set of marketed drugs. The used inhibitors were for CYP1A2 (20 μM furafylline), CYP2C8 (40 μM montelukast), CYP2C9 (40 μM sulfaphenazole), CYP2C19 (3 μM (-)N-

Two-dimensional high-performance liquid chromatographic method to assay p-hydroxyphenylphenylhydantoin enantiomers in biological fluids and stereoselectivity of enzyme induction in phenytoin metabolism.

A two-dimensional high-performance liquid chromatographic method was developed to assay the enantiomers of a major phenytoin metabolite, p-hydroxyphenylphenylhydantoin (p-HPPH). Racemic p-HPPH was first separated from phenytoin and other interfering peaks by a reversed-phase column and monitored by an ultraviolet detector. At the retention time of p-HPPH, the racemic p-HPPH peak was automatically transferred to a chiral ligand-exchange column to separate R-p-HPPH and S-p-HPPH by a time-programmed column-switching valve. The ratio of enantiomers was measured by a second ultraviolet detector. Th

Overlapping substrate specificities of cytochrome P450 3A and P-glycoprotein for a novel cysteine protease inhibitor.

K02 (morpholine-urea-Phe-Hphe-vinylsulfone), a newly developed peptidomimetic, acts as a potent cysteine protease inhibitor, especially of cathepsins B and L (which are associated with cancer progression) and cruzain (a cysteine protease of Trypanosoma cruzi, which is responsible for Chagas' disease). Here we investigated features of the disposition of K02 using in vitro systems, characterizing the interaction of the drug with human cytochrome P450 (CYP) 3A and P-glycoprotein (P-gp), a mediator of multidrug resistance (MDR) to cancer chemotherapy and a countertransporter in the intestine that

Repurposing sulfonamide drugs as anticancer ligands and understanding its properties through density functional theory.

Drug repurposing represents a promising approach towards drug discovery that has the potential to improve patient outcomes and address unmet medical needs. This study attempted to repurpose existing sulfonamide drugs in search of novel anticancer drugs because of their effectiveness in treating bacterial infections. A search was made in DrugBank for Sulfonamide, and 25 drugs with functional groups like SH, OSO, CS, and -S- were chosen for our study. The drug properties, such as dipole moment, volume, polarisability, highest occupied molecular orbital (HOMO), lowest unoccupied molecular orbital

Human reductive halothane metabolism in vitro is catalyzed by cytochrome P450 2A6 and 3A4.

The anesthetic halothane undergoes extensive oxidative and reductive biotransformation, resulting in metabolites that cause hepatotoxicity. Halothane is reduced anaerobically by cytochrome P450 (P450) to the volatile metabolites 2-chloro-1,1-difluoroethene (CDE) and 2-chloro-1,1,1-trifluoroethane (CTE). The purpose of this investigation was to identify the human P450 isoform(s) responsible for reductive halothane metabolism. CDE and CTE formation from halothane metabolism by human liver microsomes was determined by GC/MS analysis. Halothane metabolism to CDE and CTE under reductive conditions

Cytotoxicity of ginkgolic acid in HepG2 cells and primary rat hepatocytes.

Ginkgolic acids and related alkylphenols (e.g. cardanols and cardols) have been recognized as hazardous compounds with suspected cytotoxic, allergenic, mutagenic and carcinogenic properties. To determine whether the phase I metabolism could contribute to their cytotoxicity, we investigated the cytotoxicity of one model compound, ginkgolic acid (15:1), using in vitro bioassay systems. In the first step, cytochrome P450 enzymes involved in ginkgolic acid metabolism were investigated in rat liver microsomes; then, two in vitro cell-based assay systems, primary rat hepatocytes and HepG2 cells, wer

The aliphatic oxidation of salmeterol to alpha-hydroxysalmeterol in human liver microsomes is catalyzed by CYP3A.

Salmeterol xinafoate (Serevent) is a long-acting beta2-adrenoceptor agonist, used in the treatment of asthma, that has bronchodilator and anti-inflammatory action. Salmeterol is extensively metabolized by aliphatic oxidation in humans, with the major metabolite being alpha-hydroxysalmeterol. The aim of this investigation was to identify the specific cytochrome P450 (P450) isoform or isoforms involved in the formation of alpha-hydroxysalmeterol in human liver microsomes. Critical role of free cytosolic calcium, but not uncoupling, in mitochondrial permeability transition and cell death induced by diclofenac oxidative metabolites in immortalized human hepatocytes.

Diclofenac is a widely used nonsteroidal anti-inflammatory drug that has been associated with rare but serious hepatotoxicity. Experimental evidence indicates that diclofenac targets mitochondria and induces the permeability transition (mPT) which leads to apoptotic cell death in hepatocytes. While the downstream effector mechanisms have been well characterized, the more proximal pathways leading to the mPT are not known. The purpose of this study was to explore the role of free cytosolic calcium (Ca(2+)(c)) in diclofenac-induced cell injury in immortalized human hepatocytes. We show that expo

Metabolism of ezlopitant, a nonpeptidic substance P receptor antagonist, in liver microsomes: enzyme kinetics, cytochrome P450 isoform identity, and in vitro-in vivo correlation.

The enzyme kinetics of the metabolism of ezlopitant in liver microsomes from various species have been determined. The rank order of the species with regard to the in vitro intrinsic clearance of ezlopitant was monkey >> guinea pig > rat >> dog > human. CJ-12,764, a benzyl alcohol analog, was observed as a major metabolite, and a dehydrogenated metabolite (CJ-12,458) was equally important in human liver microsomes. Scale-up of the liver microsomal intrinsic clearance data and correcting for both serum protein binding and nonspecific microsomal binding yielded predicted hepatic clearance values
