[Anatomy of the central auditory pathway. Demonstration with horseradish peroxidase in the guinea pig].

The neurones of the afferent and efferent auditory pathways were labelled by axonally transported horseradish peroxidase. The first neurone of the ascending system is the acoustic nerve with the spiral ganglion. These fibres terminate mainly in the ventral cochlear nucleus (VCN). The neurones of the latter chiefly run to the contralateral superior olivary complex (SOC), whereas the neurones of the dorsal cochlear nucleus (DCN) terminate mainly in the central nucleus of the contralateral inferior colliculus (IC). The nerve cells of the superior olivary complex and nuclei of the lateral lemniscu

Projections of the lateral terminal accessory optic nucleus of the common marmoset (Callithrix jacchus).

The connections of the lateral terminal nucleus (LTN) of the accessory optic system (AOS) of the marmoset monkey were studied with anterograde 3H-amino acid light autoradiography and horseradish peroxidase retrograde labeling techniques. Results show a first and largest LTN projection to the pretectal and AOS nuclei including the ipsilateral nucleus of the optic tract, dorsal terminal nucleus, and interstitial nucleus of the superior fasciculus (posterior fibers); smaller contralateral projections are to the olivary pretectal nucleus, dorsal terminal nucleus, and LTN. Breathing under Anesthesia: A Key Role for the Retrotrapezoid Nucleus Revealed by Conditional Phox2b Mutant Mice.

BACKGROUND: Optimal management of anesthesia-induced respiratory depression requires identification of the neural pathways that are most effective in maintaining breathing during anesthesia. Lesion studies point to the brainstem retrotrapezoid nucleus. We therefore examined the respiratory effects of common anesthetic/analgesic agents in mice with selective genetic loss of retrotrapezoid nucleus neurons (Phox2b mice, hereafter designated "mutants"). METHODS: All mice received intraperitoneal ketamine doses ranging from 100 mg/kg at postnatal day (P) 8 to 250 mg/kg at P60 to P62. Anesthesia ef

Ketamine-xylazine anesthesia depth affects auditory neuronal responses in the lateral superior olive complex of the gerbil.

Studies of in vivo neuronal responses to auditory inputs in the superior olive complex (SOC) are usually done under anesthesia. However, little attention has been paid to the effect of anesthesia itself on response properties. Here, we assessed the effect of anesthesia depth under ketamine-xylazine anesthetics on auditory evoked response properties of lateral SOC neurons. Anesthesia depth was tracked by monitoring EEG spectral peak frequencies. An increase in anesthesia depth led to a decrease of spontaneous discharge activities and an elevated response threshold. The temporal responses to sup

The ion channels and synapses responsible for the physiological diversity of mammalian lower brainstem auditory neurons.

The auditory part of the brainstem is composed of several nuclei specialized in the computation of the different spectral and temporal features of the sound before it reaches the higher auditory regions. There are a high diversity of neuronal types in these nuclei, many with remarkable electrophysiological and synaptic properties unique to these structures. This diversity reflects specializations necessary to process the different auditory signals in order to extract precisely the acoustic information necessary for the auditory perception by the animal. Low threshold Kv1 channels and HCN chann

Spontaneous otoacoustic emission recordings during contralateral pure-tone activation of medial olivocochlear reflex.

We hypothesized that cochlear frequency discrimination occurs through medial olivocochlear efferent (MOCE)-induced alterations in outer hair cell (OHC) electromotility, which is independent from basilar membrane traveling waves. After obtaining informed consent, volunteers with normal hearing (n = 10; mean age: 20.6 ± 1.2 years) and patients with unilateral deafness (n = 10; mean age: 30.2 ± 17.9 years) or bilateral deafness (n = 8; mean age: 30.7 ± 13.8 years) underwent a complete physical and audiological examination, and audiological tests including transient evoked otoacoustic emission and

Increased contralateral suppression of otoacoustic emissions indicates a hyperresponsive medial olivocochlear system in humans with tinnitus and hyperacusis.

Atypical medial olivocochlear (MOC) feedback from brain stem to cochlea has been proposed to play a role in tinnitus, but even well-constructed tests of this idea have yielded inconsistent results. In the present study, it was hypothesized that low sound tolerance (mild to moderate hyperacusis), which can accompany tinnitus or occur on its own, might contribute to the inconsistency. Sound-level tolerance (SLT) was assessed in subjects (all men) with clinically normal or near-normal thresholds to form threshold-, age-, and sex-matched groups: 1) no tinnitus/high SLT, 2) no tinnitus/low SLT, 3)

Changes in calcium-binding protein expression in the auditory brainstem nuclei of the jaundiced Gunn rat.

Sensorineural hearing loss and auditory dysfunction are major sequelae of neonatal hyperbilirubinemia. The sites and cellular effects of bilirubin toxicity in the auditory brainstem pathway are not easily detected. Since altered intracellular calcium homeostasis may play a role in neuronal cell death, we hypothesized that the expression of calcium-binding proteins may be altered in the classic animal model of bilirubin neurotoxicity. The expression of the calcium-binding proteins, calbindin-D28k and parvalbumin, in the brainstem auditory pathway of homozygous recessive jaundiced (jj) Gunn rats

Functional organization of auditory cortical fields in the Mongolian gerbil (Meriones unguiculatus): binaural 2-deoxyglucose patterns.

Unilaterally deafened (cochlear destruction) gerbils were exposed to white noise after injection of 14-C-2-deoxyglucose. The labelling patterns were compared to those of unstimulated operated animals, noise stimulated control animals and bilaterally ear plugged animals. Serial transverse, horizontal and tangential autoradiographs through the cortex were analysed. In lesioned animals, labelling was strongly reduced on the side contralateral to the lesion in the high frequency regions of A1 and the anterior auditory field (AAF). We assume that these regions correspond to the high frequency EI ce

Distinct cell classes in the superior paraolivary nucleus (SPN) region in the gerbil auditory brainstem revealed by in vivo physiological and anatomical characterization.

The superior para-olivary nucleus (SPN or SPON) is a prominent nucleus in the superior olivary complex of the auditory brainstem. The cellular composition of the nucleus reportedly differs between species, but a prominent recurring feature is the unusual characteristic to not respond during a sound but at its offset. Blocking glycine has shown that sound-induced inhibition is the mechanism, but the time course of the responsible synaptic events has not been directly measured in vivo. We obtained intracellular recordings in the Mongolian gerbil (meriones unguiculatus) with patch electrodes cont

MR imaging of the excised human brainstem: a correlative neuroanatomic study.

The purpose of this study was to examine an excised human brainstem with high-resolution MR imaging. The MR images were correlated with myelin-stained histologic specimens and standard anatomic descriptions. Structures visualized included nuclei and/or fibers of all the cranial nerves, with the exception of the trochlear nerve. The topography of the brainstem is discussed. Major tracts such as the corticospinal spinothalamic, spinocerebellar, medial and lateral lemniscus, and other ascending and descending tracts and intrinsic brainstem nuclei such as the olivary complex, pontine nuclei, red n

The medial olivocochlear reflex in children during active listening.

OBJECTIVE: To determine if active listening modulates the strength of the medial olivocochlear (MOC) reflex in children. DESIGN: Click-evoked otoacoustic emissions (CEOAEs) were recorded from the right ear in quiet and in four test conditions: one with contralateral broadband noise (BBN) only, and three with active listening tasks wherein attention was directed to speech embedded in contralateral BBN. STUDY SAMPLE: Fifteen typically-developing children (ranging in age from 8 to14 years) with normal hearing. RESULTS: CEOAE levels were reduced in every condition with contralateral acoustic st

Efferent modulation of pre-neural and neural distortion products.

Distortion product otoacoustic emissions (DPOAEs) and distortion product frequency following responses (DPFFRs) are respectively pre-neural and neural measurements associated with cochlear nonlinearity. Because cochlear nonlinearity is putatively linked to outer hair cell electromotility, DPOAEs and DPFFRs may provide complementary measurements of the human medial olivocochlear (MOC) reflex, which directly modulates outer hair cell function. In this study, we first quantified MOC reflex-induced DPOAE inhibition at spectral fine structure peaks in 22 young human adults with normal hearing. The

Central acoustic tract in an echolocating bat: an extralemniscal auditory pathway to the thalamus.

To determine the sources and targets of auditory pathways that bypass the inferior colliculus in the mustache bat, we injected WGA-HRP in the medial geniculate body and related auditory nuclei of the thalamus as well as in the lower brainstem. We used electrophysiological methods to verify that the injection electrode was in an area responsive to sound. The only thalamic injections that produced retrograde transport to cells in auditory nuclei caudal to the inferior colliculus were those that included the suprageniculate nucleus. These injections labeled a group of large multipolar cells lying

Robustness of neuronal tuning to binaural sound localization cues against age-related loss of inhibitory synaptic inputs.

Sound localization relies on minute differences in the timing and intensity of sound arriving at both ears. Neurons of the lateral superior olive (LSO) in the brainstem process these interaural disparities by precisely detecting excitatory and inhibitory synaptic inputs. Aging generally induces selective loss of inhibitory synaptic transmission along the entire auditory pathways, including the reduction of inhibitory afferents to LSO. Electrophysiological recordings in animals, however, reported only minor functional changes in aged LSO. The perplexing discrepancy between anatomical and physio

A physiological study of acoustic blinking reflex in the cat.

Blinking reflex was elicited by acoustic stimulus to investigate the reflex pathway in the encéphalo isolé cat. Reflex responses were recorded using electromyography (EMG) of the orbicularis oculi. The investigation was as follows: After stimulation by a loud click sound, the EMGs of the orbicularis oculi were elicited bilaterally. Latencies were 17-20 msec on the ipsilateral side, and 19-21 msec on the contralateral side. By electrical stimulation of the peripheral nerve in the cochlea and the acoustic nerve EMGs were also obtained. The latencies of the electrical stimulation responses correl

Functional asymmetries of crossed and uncrossed medial olivocochlear efferent pathways in humans.

In a previous study, a peripheral auditory asymmetry in right-handers was found. The activity of the uncrossed medial olivocochlear efferent pathway has been shown to be more effective in the right than in the left ear. Our aim was to compare right and left medial olivocochlear inhibition during contralateral, ipsilateral and binaural ear stimulation with a broad band noise, in order to better localize this asymmetry. Uncrossed pathway asymmetry was confirmed and the crossed pathway showed a reverse asymmetry: a left ear advantage. These results are in accordance with the assumption that the m

Far-field acoustic response: origins in the cat.

Short-latency evoked potentials recorded from the vertex of adult cats in response to click stimulation (the far-field acoustic response) were analyzed in a series of lesion experiments to determine the origins of each component. The resultant data indicate that the primary generator of potential is the acoustic nerve; of potential 2, the cochlear nucleus; of potential 3, neurons of the superior olivary complex activated by projections crossing the midline; of potential 4, neurons of the ventral nucleus of the lateral lemniscus and preolivary region activated equally by crossed and uncrossed p

GABA corelease guides the functional maturation of glycinergic synapses in an auditory sound localization circuit.

In the mammalian brainstem and spinal cord, glycine is the primary inhibitory neurotransmitter. However, during development, many glycinergic neurons also corelease the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). Although the acute effects of GABA corelease on immature synaptic transmission have been increasingly characterized, its role in synapse maturation and circuit formation remains poorly understood. Here, we investigated the developmental roles of GABA corelease at glycinergic synapses from the medial nucleus of the trapezoid body (MNTB) to the lateral superior olive (LS

Preventing presbycusis in mice with enhanced medial olivocochlear feedback.

"Growing old" is the most common cause of hearing loss. Age-related hearing loss (ARHL) (presbycusis) first affects the ability to understand speech in background noise, even when auditory thresholds in quiet are normal. It has been suggested that cochlear denervation ("synaptopathy") is an early contributor to age-related auditory decline. In the present work, we characterized age-related cochlear synaptic degeneration and hair cell loss in mice with enhanced α9α10 cholinergic nicotinic receptors gating kinetics ("gain of function" nAChRs). These mediate inhibitory olivocochlear feedback thro
