Allogeneic stem cells from deciduous teeth in treatment for periodontitis in miniature swine.

BACKGROUND: Regeneration of lost periodontium in periodontitis is a challenge in that alveolar bone, cementum, and periodontal ligament need to be restored to their original architecture. Stem cells from exfoliated deciduous teeth (SHEDs) appear to be an attractive candidate for periodontium tissue regeneration. Previously, the authors successfully regenerated periodontal defects using autologous and allogeneic periodontal ligament stem cells (PDLSCs). The purpose of the present study is to investigate the ability of allogeneic SHEDs to regenerate lost periodontium in a swine periodontitis mod

Development and resuscitation of a sedated, mature male miniature swine severe hemorrhage model.

BACKGROUND: A sedated, mature male miniature swine hemorrhage model has been specifically developed to evaluate resuscitation products for the Defense Advanced Research Projects Agency Surviving Blood Loss program. METHODS: Animals were placed in a sling, sedated with midazolam, and hemorrhaged 60% of estimated blood volume (∼39 mL/kg) exponentially for 1 hour with no resuscitation (control; n = 16). An additional 26 swine were treated similarly, then resuscitated with 1 mL/kg/min of Hextend to a systolic blood pressure of either 65 mm Hg ± 2 mm Hg (n = 7) or 80 mm Hg ± 5 mm Hg (n = 7) and wi

Tolerance to composite tissue allografts across a major histocompatibility barrier in miniature swine.

BACKGROUND: Tolerance to composite tissue allografts might allow the widespread clinical use of reconstructive allotransplantation if protocols to achieve this could be rendered sufficiently nontoxic. The authors investigated whether tolerance could be generated in miniature swine to composite tissue allografts across a major histocompatibility (MHC) barrier. A clinically relevant tolerance protocol involving hematopoietic cell transplantation without the need for irradiation or myelosuppressive drugs was tested. METHODS: Seven recipient animals were transiently T-cell depleted and a short co

Miniature swine as an animal model in photodermatology: factors influencing sunburn cell formation.

Swine have played an important role as animal models in dermatological research. In order to evaluate the usefulness of pig skin in photodermatology, sunburn cell (SBC) formation in the epidermis of miniature swine and erythema production were studied. SBCs were readily formed in the epidermis of the swine by ultraviolet radiation. Suberythemal doses of broad-band UVB radiation were effective in producing SBCs, and there was a dose-response relationship between SBC formation and ultraviolet radiation. When SBC formation was related to ultraviolet radiation in terms of biological dose (i.e. min

Development of histological sectioning techniques for the cochlear implanted inner ear in miniature swine.

BACKGROUND: Cochlear pathological sectioning is essential for studying inner ear structural changes. Traditional methods like paraffin embedding, frozen sectioning, and collodion embedding are limited to decalcified tissues. For cochlear specimens with rigid implants (e.g., cochlear implants), the implant must be separated before decalcification and sectioning. This process often damages the delicate cochlear architecture, compromising pathological integrity. Thus, there is a need for a precise and efficient method for examining cochlear tissues with implants. NEW METHOD: We introduce a novel

Expression of porcine endogenous retroviruses (PERVs) in melanomas of Munich miniature swine (MMS) Troll.

Porcine endogenous retroviruses (PERVs) are integrated in the genome of all pig breeds. Since some of them are able to infect human cells, they might represent a risk for xenotransplantation using pig cells or organs. However, the expression and biological role of PERVs in healthy pigs as well as in porcine tumours is largely unknown. Since we and others have recently shown overexpression of a human endogenous retrovirus, HERV-K, in human melanomas, we studied the expression of PERVs in melanomas of selectively bred Munich miniature swine (MMS) Troll. This breeding herd of MMS Troll is charact

Induction of tolerance to renal allografts across single-haplotype MHC disparities in miniature swine.

We have previously demonstrated that a 12-day course of cyclosporine A (CsA) leads to the induction of tolerance to renal allografts in 100% of recipients selectively mismatched at class I for both haplotypes, and in 71% of recipients selectively mismatched at class II for both haplotypes, but in 0% of recipients mismatched for two haplotypes at both class I and class II. We have postulated that the mechanism by which tolerance is induced may therefore require matching for either class I or class II antigens. One might predict from this hypothesis that tolerance would also be induced in donor-

Heterotopic intestine and abdominal wall transplantation in a miniature swine: A model for graft immunology.

In order to accurately monitor graft immunology, we have developed a method for performing intestine and abdominal wall transplantation heterotopically in miniature swine. The procedure consisted of simultaneous segmental terminal ileum and full-thickness abdominal wall transplantation in Lanyu miniature swine, with the intestinal and the abdominal wall grafts being placed on the recipient's bilateral rear flank. Five transplantations were technically successful. One animal died on the first post-transplant day due to anesthesia-related issues, three abdominal wall and four intestinal grafts s

Mandible-derived extracellular vesicles regulate early tooth development in miniature swine via targeting KDM2B.

Tissue interactions play a crucial role in tooth development. Notably, extracellular vesicle-mediated interactions between the mandible and tooth germ are considered essential. Here, we revealed that mandible extracellular vesicles could modulate the proliferation and differentiation of dental mesenchymal cells by regulating the histone demethylase KDM2B. Further investigation showed that mandible derived extracellular vesicles could deliver miR-206 to KDM2B, thereby regulating tooth development. An animal study demonstrated that the miR-206/KDM2B pathway affected tooth morphogenesis and miner

Miniature-swine iPSC-derived GABA progenitor cells function in a rat Parkinson's disease model.

Induced pluripotent stem cells (iPS cells) are considered a promising source of cell-based therapy for the treatment of Parkinson's disease (PD). Recent studies have shown forebrain GABA interneurons have crucial roles in many psychiatric disorders, and secondary changes in the GABA system play a directly effect on the pathogenesis of PD. Here, we first describe an efficient differentiation procedure of GABA progenitors (MiPSC-iGABAPs) from miniature-swine iPSCs through two major developmental stages. Then, the MiPSC-iGABAPs were stereotactically transplanted into the right medial forebrain bu

Obesity induced during sexual maturation is linked to LDL-triacylglycerols in Yucatan miniature swine.

The incidence of childhood obesity is rising dramatically throughout industrialised countries. To evaluate and study the impact of childhood obesity on lipoprotein metabolism, we developed a new animal model of premature obesity. Yucatan mini-pigs aged 4 months were studied over a 12-month period from childhood to adulthood. Animals were divided into two groups: the first group were overfed a Western misbalanced diet; the second group were normally fed a recommended human-type diet. Cholesterol and triacylglycerol concentrations in VLDL-, LDL- and HDL-lipoproteins were followed from baseline t

Effects of mobilization regimens in donors on outcomes of hematopoietic cell transplantation in miniature Swine.

Toxicities and complications associated with hematopoietic cell transplantation currently limit this potentially curative therapy for malignant and nonmalignant blood disorders. Miniature swine provide a clinically relevant model for studies to improve posttransplantation outcomes. Miniature swine recipients of high-dose haploidentical hepatopoietic cell transplantation after reduced-intensity conditioning consisting of low-dose (100 cGy) total-body irradiation, partial T-cell depletion by using a CD3 immunotoxin, and a 45-d course of cyclosporine A typically successfully engraft without graft

Role of the thymus and kidney graft in the maintenance of tolerance to heart grafts in miniature swine.

BACKGROUND: The authors have examined the mechanism whereby co-transplantation of a kidney and heart from the same donor induces and maintains tolerance to both organs in miniature swine. METHODS: Transplants were performed across a major histocompatibility complex class I mismatch, and recipients received cyclosporine for 12 days. Group 1 animals received heart transplants alone (n=5), and all other groups received both heart and kidney allografts. Group 2 animals received no further intervention (n=2). Group 3 animals underwent transplant nephrectomy 8 days after heart and kidney co-transpl

Efficacy of Octacalcium Phosphate/Gelatin (OCP/Gel) Composite Implantation for Miniature Swine Lumbar Interbody Fusion.

Synthetic octacalcium phosphate (OCP) has emerged as a potential precursor for bone apatite crystals, promoting faster bone formation and better biodegradability compared to hydroxyapatite and β-tricalcium phosphate materials. Combining OCP with various polymeric biomaterials enhances its ductility, making it suitable for clinical applications, including dentistry. Preclinical studies on OCP/gelatin (OCP/Gel) composites have shown excellent osteoconductive and osteoinductive properties, indicating potential for bone defect repairs. This study investigates the efficacy of OCP/Gel as a filler fo

Isolation and characterization of vascular endothelial cells derived from fetal tooth buds of miniature swine.

The aim of the present study was to isolate endothelial cells from tooth buds (unerupted deciduous teeth) of miniature swine. Mandibular molar tooth buds harvested from swine fetuses at fetal days 90-110 were cultured in growth medium supplemented with 15% fetal bovine serum in 100-mm culture dishes until the primary cells outgrown from the tooth buds reached confluence. A morphologically defined set of pavement-shaped primary cells were picked up manually with filter paper containing trypsin/ethylenediamine tetraacetic acid solution and transferred to a separate dish.
