Cyanobacteria engineered for production of biofuels and biochemicals from carbon dioxide represent a promising area of research in relation to a sustainable economy. Previously, we have succeeded in producing isopropanol from cellular acetyl-CoA by means of Synechococcus elongatus PCC 7942 into which a synthetic metabolic pathway was introduced. The isopropanol production by this synthetic metabolic pathway requires acetate; therefore, the cells grown under photosynthetic conditions have to be transferred to a dark and anaerobic conditions to produce acetate. In this study, we achieved acetate

The marine cyanobacterium Synechococcus sp. PCC 7002 (Syn7002) is a model organism that lacks the gene cluster required for vitamin B12 biosynthesis, necessitating cooperative interactions with other microbes. In this study, we established a synthetic microbial consortium by co-culturing Syn7002 with a bloom-forming Microcystis community, followed by purification, and subsequently investigated the interactions between Syn7002 and the associated microbial community. Electron microscopy revealed numerous rod-shaped bacteria clustered around Syn7002 cells, indicating close spatial associations be

Coastal California experiences large-scale blooms of Synechococcus cyanobacteria, which are predicted to become more prevalent by the end of the 21st century as a result of global climate change. This study investigated whether exposure to bloom-like concentrations of two Synechococcus strains, CC9311 and CC9902, alters fish behaviour. Black perch (Embiotoca jacksoni) were exposed to Synechococcus strain CC9311 or CC9902 (1.5 × 10(6) cells ml(-1)) or to control seawater in experimental aquaria for 3 days. Fish movement inside a testing arena was then recorded and analysed using video camera-ba

The recombinant two-on-two hemoglobin from the cyanobacterium Synechoccocus sp. PCC 7002 (S7002 rHb) is a bishistidine hexacoordinate globin capable of forming a covalent cross-link between a heme vinyl and a histidine in the C-terminal helix (H helix). Of the two heme axial histidines, His46 (in the E helix, distal side) and His70 (in the F helix, proximal histidine), His46 is displaced by exogenous ligands. S7002 rHb can be readily prepared as an apoglobin (apo-rHb), a non-cross-linked hemichrome (ferric iron and histidine axial ligands, rHb-R), and a cross-linked hemichrome (rHb-A). To dete

This study aimed to investigate whether that microwave pretreatment of glycogen-containing Synechococcus elongatus UTEX 2973 could enhance subsequent enzymatic saccharification and ethanol fermentation. The first experiment found that pretreatment with microwave was more effective than either pretreatment with ultrasound or lysozyme under the specific operational conditions examined, when a glycogen-containing cyanobacterial suspension (100 g/L) was pretreated and subjected to the saccharification assay at low biomass loading (10 g/L). Next, to investigate the appropriate microwave pretreatmen

The adaptive evolution of cyanobacteria over a prolonged period has allowed them to utilize carbon dioxide (CO2) at the low concentrations found in the atmosphere (0.04% CO2) for growth. However, whether the exposure of cyanobacteria to high concentrations of CO2 results in oxidative stress and the activation of antioxidant defense response remains unknown, albeit fluctuations in other culture conditions have been reported to exert these effects. The current study reveals the physiological regulation of the model cyanobacterium Synechococcus elongatus PCC 7942 upon exposure to 1% CO2 and the u

Phosphate availability fluctuates widely in aquatic environments, yet how engineering the regulatory context of polyphosphate kinase affects cyanobacterial responses to such fluctuations remains unclear. Here, the wild-type strain of Synechococcus sp. PCC 7002 and Dppk, a modified ppk-expression strain derived from the WT background, were subjected to phosphate starvation followed by phosphate resupply. Dense RT-qPCR sampling was used to resolve rapid ppk-expression dynamics, and time-resolved RNA sequencing was performed at seven stages with four biological replicates per condition. WT displa

Phytoplankton-bacteria interactions underpin primary production and nutrient cycling in both oligotrophic and eutrophic aquatic environments, profoundly influencing marine biogeochemical cycles. Despite their importance, how these interactions vary under simultaneous environmental stressors such as warming and iron (Fe) limitation remains largely unexplored, especially across differing ecotypes. Here, we compared the responses of oligotrophic (strain YX04-1) and eutrophic (strain XM-24) ecotype Synechococcus-heterotrophic bacteria interactions to concurrent warming and Fe limitation, using the

Iron is an essential component in many protein complexes involved in photosynthesis, but environmental iron availability is often low as oxidized forms of iron are insoluble in water. To adjust to low environmental iron levels, cyanobacteria undergo numerous changes to balance their iron budget and mitigate the physiological effects of iron depletion. We investigated changes in key protein abundances and photophysiological parameters in the model cyanobacteria Synechococcus PCC 7942 and Synechocystis PCC 6803 over a 120 hour time course of iron deprivation. The iron stress induced protein (Isi

A mathematical model of the Mossbauer spectrum (80K) of native membranes of Synechococcus elongatus was constructed on the basis of values of the quadruple splitting (Delta) and the isomeric shift (delta) of the iron-containing components of the photosynthetic apparatus obtained from the literature. Thermally induced changes in the intensity of the spectral components of membranes and isolated preparations of photosystem (PS) I were studied using this model. It was shown that exposure of membranes to 70-80 degrees C causes a decrease in the intensity of the components related to the FX, FA, an

Predictive control of gene expression is an essential tool for developing synthetic biological systems. The current toolbox for controlling gene expression in cyanobacteria is a barrier to more in-depth genetic analysis and manipulation. Towards relieving this bottleneck, this work describes the use of synthetic biology to construct an anhydrotetracycline-based induction system and adapt a trans-acting small RNA (sRNA) system for use in the cyanobacterium Synechococcus sp. strain PCC 7002. An anhydrotetracycline-inducible promoter was developed to maximize intrinsic strength and dynamic range.

Genetic manipulation in cyanobacteria enables the direct production of valuable chemicals from carbon dioxide. However, there are still very few reports of the production of highly effective photosynthetic chemicals. Several synthetic metabolic pathways (e.g., isopropanol, acetone, isoprene, and fatty acids) have been constructed by branching from acetyl-CoA and malonyl-CoA, which are key intermediates for photosynthetic chemical production downstream of pyruvate decarboxylation. Recent reports of the absolute determination of cellular metabolites in Synechococcus elongatus PCC 7942 have shown

One of the most abundant and ubiquitous members of the phytoplankton in the marine ecosystem is the unicellular cyanobacteria of the genus Synechococcus, which contributes to a substantial fraction of the ocean's primary productivity. Nevertheless, like any other organism, Synechococcus is infected by a specific kind of virus: the cyanophages. These viruses, highly abundant and widely distributed throughout the world's oceans, shape the bacterial communities by regulating their abundance, diversity, and physiology. However, the information regarding the occurrence of Synechococcus-infecting ph

The marine cyanobacterium Synecococcus sp. (CCMP 1334) was grown in a continuous culture system on a 12:12 h light:dark cycle at all combinations of low and high pCO2 (400 and 1000 ppmv, respectively), nutrient availability (nitrate-limited and nutrient-replete conditions), and temperatures of 21, 24, 28, 32, and 35°C. The maximum nutrient-replete growth rate was ~1.15 day-1 at 32-35°C. Median nutrient-replete growth rates were higher at 1000 ppmv than at 400 ppmv pCO2 at all temperatures. Carbon:nitrogen ratios were independent of pCO2 at a fixed relative growth rate (i.e., growth rate ÷ nutr

The production of alcohols directly from carbon dioxide by engineered cyanobacteria is an attractive technology for a sustainable future. Enhanced tolerance to the produced alcohols would be a desirable feature of the engineered cyanobacterial strains with higher alcohol productivity. We have recently obtained the mutant strains of Synechococcus elongatus PCC 7942 with higher tolerance to isopropanol using a single-cell screening system (Arai et al., Biotechnol. Bioeng., 114, 1771-1778, 2017). Among the mutant strains, SY1043 showed the highest isopropanol tolerance. Interestingly, SY1043 also

Eutrophication and deoxygenation possibly occur in coastal waters due to excessive nutrients from agricultural and aquacultural activities, leading to sulfide accumulation. Cyanobacteria, as photosynthetic prokaryotes, play significant roles in carbon fixation in the ocean. Although some cyanobacteria can use sulfide as the electron donor for photosynthesis under anaerobic conditions, little is known on how they interact with sulfide under aerobic conditions. In this study, we report that Synechococcus sp. strain PCC7002 (PCC7002), harboring an sqr gene encoding sulfide:quinone oxidoreductase

A variety of contemporary techniques were used to investigate the vertical distribution of thermophilic unicellular cyanobacteria, Synechococcus spp., and their activity within the upper 1-mm-thick photic zone of the mat community found in an alkaline siliceous hot spring in Yellowstone National Park in Wyoming. Detailed measurements were made over a diel cycle at a 61 degrees C site. Net oxygenic photosynthesis measured with oxygen microelectrodes was highest within the uppermost 100- to 200-microm-thick layer until midmorning, but as the day progressed, the peak of net activity shifted to de

Synechococcus HS-9 is being recognized as one of the potential strains for biodiesel production due to its high levels of fatty acid methyl ester (FAME), which are around 70-78%. The first stage in producing microalgae biodiesel involves the biomass production process through a photobioreactor cultivation process. In addition to microalgae strains, the optimization of the photobioreactor's performance is essential for producing high biomass. In this case, biotic factors (microalgae inoculum) and abiotic factors (nutrients, temperature, light, pH, carbon dioxide (CO2), fluid flow, hydrodynamic

Marine phytoplankton and heterotrophic bacteria share a very close but usually changeable relationship. However, the ultimate fate of their unstable relationship on a long-term scale is unclear. Here, the relationship between Synechococcus and heterotrophic bacterial communities underwent a dramatic shift from antagonism to commensalism and eventually to mutualism during long-term cocultivation. The relationship change is attributed to the different (even opposite) effects of diverse bacterial members on Synechococcus and the ratio of beneficial to harmful bacteria. Different bacterial members
