Characterization of the expression pattern of adrenergic receptors in rat taste buds.

Taste buds signal the presence of chemical stimuli in the oral cavity to the central nervous system using both early transduction mechanisms, which allow single cells to be depolarized via receptor-mediated signaling pathways, and late transduction mechanisms, which involve extensive cell-to-cell communication among the cells in the bud. The latter mechanisms, which involve a large number of neurotransmitters and neuropeptides, are less well understood. Among neurotransmitters, multiple lines of evidence suggest that norepinephrine plays a yet unknown role in the taste bud. This study investig

Palatal taste buds in man: topographical arrangement in islands of keratinized epithelium.

In an attempt to find and reconstruct the exact anatomical correlate of taste sensitivity in the human soft palate, a novel approach was used to examine the mucosal surface in conjunction with serial tissue sectioning. Six human subjects, 15 to 29 years of age, 3 females and 3 males, served to take precision impressions of the hard and the anterior soft palate, using non-commercial trays and a self-curing resin. From these impressions, replicas were cast in Epon and subsequently sputter-coated with gold and examined in a scanning electron microscope. In addition, one biopsy was taken from one

New details of the ultrastructure (TEM, SEM) of taste buds in fishes.

Quantitative distribution of the taste buds (TB) in different parts of the body and the fine structure of the TB components are described in Cobitis taenia L. No evidence of synaptic contacts between any cellular components in the TB has been found. The afferent synapses have been recognized only at the gustatory cells and at the basal cell. The microvillar processes on the upper side of the basal cell are demonstrated for the first time in fishes. These processes resemble those at the basal cells in the TB of tetrapods and at the Merkel cells, scattered in the epidermis of all vertebrates. Si

Give-and-take of gustation: the interplay between gustatory neurons and taste buds.

Mammalian taste buds are highly regenerative and can restore themselves after normal wear and tear of the lingual epithelium or following physical and chemical insults, including burns, chemotherapy, and nerve injury. This is due to the continual proliferation, differentiation, and maturation of taste progenitor cells, which then must reconnect with peripheral gustatory neurons to relay taste signals to the brain. The turnover and re-establishment of peripheral taste synapses are vital to maintain this complex sensory system. Over the past several decades, the signal transduction and neurotran

The Role of 5-HT3 Receptors in Signaling from Taste Buds to Nerves.

Activation of taste buds triggers the release of several neurotransmitters, including ATP and serotonin (5-hydroxytryptamine; 5-HT). Type III taste cells release 5-HT directly in response to acidic (sour) stimuli and indirectly in response to bitter and sweet tasting stimuli. Although ATP is necessary for activation of nerve fibers for all taste stimuli, the role of 5-HT is unclear. We investigated whether gustatory afferents express functional 5-HT3 receptors and, if so, whether these receptors play a role in transmission of taste information from taste buds to nerves. In mice expressing GFP

Expression of ATP-gated P2X3 receptors in rat gustatory papillae and taste buds.

It has recently become evident that ATP and other extracellular nucleotides could play an important role in signal transductions. ATP mediates excitatory signaling by means of P2X receptors. P2X3, one of its subtypes, a membrane ligand-gated ion channel, is strongly expressed in peripheral sensory neurons. The aim of the present study was to examine the distribution of nerve fibers expressing P2X3 receptors in taste buds in the gustatory papillae and soft palate of rats by immunohistochemistry. We found that the fluorescence ATP marker quinacrine stained subsets of taste bud cells. Numerous ne

Neurocalcin-immunoreactive neurons in the petrosal ganglion innervate the taste bud.

The distribution and origin of neurocalcin-immunoreactive (NC-ir) nerve fibers in the taste bud and carotid body were examined by an immunofluorescence method. In the circumvallate papilla of the tongue, NC-ir nerve fibers made subepithelial nerve plexuses and occasionally penetrated the taste bud. However, the carotid body was devoid of ir nerve fibers. In the petrosal ganglion, 32% of neurons were immunoreactive for NC. Such neurons were mostly medium-sized to large, and scattered throughout the ganglion. In the superior cervical and intralingual ganglia, numerous ir varicose fibers surround

Age-related alteration of taste bud distribution in the common marmoset.

Alteration in the number of taste buds on the soft palate (SP), fungiform (FF), foliate (FL) and circumvallate (CV) papillae in the common marmoset at different postnatal ages was examined histologically. After paraffin embedding, complete serial sections at 10 microm thickness were made and stained by HE. Digitized images for each section were examined carefully. The number of FF taste buds at day 1 was 334. While only 20% of all the taste buds at birth possessed a taste pore, 39% of 174 SP taste buds at day 1 possessed a taste pore. The number of taste buds with pores at day 1 was small for

The effects of sialoadenectomy and exogenous EGF on taste bud morphology and maintenance.

Taste buds on the dorsal tongue surface are continually bathed in saliva rich in epidermal growth factor (EGF). In the following experiment, taste bud number and morphology were monitored following submandibular and sublingual salivary gland removal (sialoadenectomy), to determine if EGF plays a role in the maintenance and formation of taste buds. Adult male rats were divided into four groups: sialoadenectomized (SX, n = 4); sialoadenectomized with EGF replacement (SX + EGF, n = 5); sham-operated (SH, n = 4); and sham-operated with exogenous EGF (SH + EGF, n = 5). After a 3 week recovery, SX +

The fine structural effect of sialectomy on the taste bud cells in the rat.

Taste buds in the rat and other mammals share a secretory activity with their transduction function as taste receptor. The present work shows the effect of bilateral removal of the main salivary glands on taste bud cells' components related to secretion in the vallate papilla of the rat. In the sialectomized rats remarkable changes were evidence in the dark and intermediate types of taste bud cells, which are known to be the secretory components. Such changes involve hypertrophy of either the protein synthetizing machinery, the smooth endoplasmic reticulum or the Golgi complex. Lucent and coat

Voltage-gated channels involved in taste responses and characterizing taste bud cells in mouse soft palates.

Taste bud cells (TBCs) on soft palates differ from those on tongues in innervation and chemosensitivity. We investigated voltage-gated channels involved in the taste responses of TBCs on mouse soft palates under in-situ tight-seal voltage/current-clamp conditions. Under the cell-attached mode, TBCs spontaneously fired action currents, which were blocked by application of 1 microM TTX to TBC basolateral membranes. Firing frequencies increased in response to taste substances applied to TBC receptor membranes. Under the whole-cell clamp mode, as expected, TBCs produced various voltage-gated curre

Expression of GAD67 and Dlx5 in the taste buds of mice genetically lacking Mash1.

It has been reported that a subset of type III taste cells express glutamate decarboxylase (GAD)67, which is a molecule that synthesizes gamma-aminobutyric acid (GABA), and that Mash1 could be a potential regulator of the development of GABAnergic neurons via Dlx transcription factors in the central nervous system. In this study, we investigated the expression of GAD67 and Dlx in the embryonic taste buds of the soft palate and circumvallate papilla using Mash1 knockout (KO)/GAD67-GFP knock-in mice. In the wild-type animal, a subset of type III taste cells contained GAD67 in the taste buds of t

A new gene (rmSTG) specific for taste buds is found by laser capture microdissection.

Getting pure populations of taste buds suitable for molecular analysis has hampered the characterization of genes specifically expressed in taste cells. To solve this problem, we prepared specific cDNA libraries from small numbers of taste cells and surrounding epithelium isolated by laser capture microdissection (LCM) and report the discovery of a rhesus monkey novel gene (rmSTG) expressed specifically in taste cells, as found by differential screening of the cDNA libraries and RNA in situ hybridization. RNA in situ hybridization shows the preferential expression of this gene in taste buds fr

Epithelial differentiation and taste buds in the soft palate of the monkey, Macaca irus.

A combination of light, transmission and scanning electron microscopy was employed to demonstrate the occurrence, arrangement and structure of taste buds in the oral mucosa of the soft palate of monkeys (Macaca irus). Taste buds are found in aggregates confined to 0.15 to 0.3 mm wide, round islands of keratinizing epithelium embedded in the normally non-keratinizing integument. Topography, configuration and structure of these epithelial islands and their taste buds are described, and the question of a developmental and functional interrelationship between epithelial differentiation and propert

Taste bud development and patterning in sighted and blind morphs of Astyanax mexicanus.

In the blind cave-dwelling morph of A. mexicanus, the eye degenerates while other sensory systems, such as gustation, are expanded compared to their sighted (surface-dwelling) ancestor. This study compares the development of taste buds along the jaws of each morph. To determine whether cavefish have an altered onset or rate of taste bud development, we fluorescently labeled basal and receptor cells within taste buds over a developmental series. Our results show that taste bud number increases during development in both morphs. The rate of development is, however, accelerated in cavefish; a sma

Nerve Injury-Induced Immune Responses in the Taste Bud Target Field.

Damage to the chorda tympani (CT) nerve through trauma or experimental nerve axotomy results in the degeneration of anterior taste buds and taste loss. Our previous work demonstrated that interleukin-1 receptor 1 (Il1r) signaling is required for taste bud regeneration and the recovery of taste function. However, the effects of experimental axotomy on immune responses in the absence of Il1r signaling remain unclear. Here we performed unilateral CT sectioning in Il1r KO or wild-type mice to observe changes in innate immune cell populations in the anterior taste field. We found that CD45+ immune
