Dual binding mode of "bitter sugars" to their human bitter taste receptor target.

The 25 human bitter taste receptors (hTAS2Rs) are responsible for detecting bitter molecules present in food, and they also play several physiological and pathological roles in extraoral compartments. Therefore, understanding their ligand specificity is important both for food research and for pharmacological applications. Here we provide a molecular insight into the exquisite molecular recognition of bitter β-glycopyranosides by one of the members of this receptor subclass, hTAS2R16. Most of its agonists have in common the presence of a β-glycopyranose unit along with an extremely structurall

Postprandial dynamics of sensory receptor and neural mechanisms underlying feeding regulation in mule ducks.

The gustatory system plays a central role in nutrient detection and feeding regulation in birds, yet the underlying molecular mechanisms in ducks remain poorly characterized. Here, we provide the first integrated analysis of nutrient detection pathways, from peripheral oral sensing to central brain integration, in mule ducks at 4 weeks of age. Nutritional status was confirmed via plasma free fatty acids and triglycerides. We quantified taste receptors mediating sweet (Taste Receptor type 1 member 1 and 3), bitter (Taste Receptor type 2 member 9 and 40), sour (Acid-Sensing Ion Channels), umami

The Taste Receptor TAS1R3 Regulates Small Intestinal Tuft Cell Homeostasis.

Tuft cells are an epithelial cell type critical for initiating type 2 immune responses to parasites and protozoa in the small intestine. To respond to these stimuli, intestinal tuft cells use taste chemosensory signaling pathways, but the role of taste receptors in type 2 immunity is poorly understood. In this study, we show that the taste receptor TAS1R3, which detects sweet and umami in the tongue, also regulates tuft cell responses in the distal small intestine. BALB/c mice, which have an inactive form of TAS1R3, as well as Tas1r3-deficient C57BL6/J mice both have severely impaired response

Evaluation of Bronchial Receptors with Concentration-Response Curves, Bitterness Assessment, and Computer Simulations.

This research aims to explore the human-like taste response of three bitter molecules (chloroquine, denatonium, and saccharin) on the human bronchi receptor T2R agonist using a statistical physics approach. By fitting the concentration-taste response curves, we estimate three model parameters (RM, n, C1/2) to assess the intermolecular interactions with T2R receptor sites. In addition, the transduction coefficient (α) of the three tasting molecules has been determined. This analysis provides valuable insights into the binding energy spectrum (AES) and establishes a common gustatory band for the

BitterTranslate: A Natural Language Processing and Machine Learning-Based Framework for Mapping Bitter Taste Receptor Agonism.

Bitter taste receptors (TAS2Rs) are G protein-coupled receptors (GPCRs) expressed most notably on the tongue, but also in many extraoral tissues. TAS2R ligands are used for improving bitter-drug compliance, treating various illnesses, and studying the receptors' extraoral functions. Although machine learning, a promising drug discovery tool, can be used to predict TAS2R activators, obtaining high-quality features for training is time-intensive and reliant on specialized software. This work explores the potential of transformers (a neural network architecture for extracting features from sequen

Bitter tastants and artificial sweeteners activate a subset of epithelial cells in acute tissue slices of the rat trachea.

Bitter and sweet receptors (T2Rs and T1Rs) are expressed in many extra-oral tissues including upper and lower airways. To investigate if bitter tastants and artificial sweeteners could activate physiological responses in tracheal epithelial cells we performed confocal Ca2+ imaging recordings on acute tracheal slices. We stimulated the cells with denatonium benzoate, a T2R agonist, and with the artificial sweeteners sucralose, saccharin and acesulfame-K. To test cell viability we measured responses to ATP. We found that 39% of the epithelial cells responding to ATP also responded to bitter stim

Beef-Derived Peptides Mediated Desensitization of Bitter Taste Receptor T2R14 Through GPCR Kinase 2.

Background/Objectives: Humans have at least 26 bitter taste receptors (T2Rs), and among these, bitter taste receptor 14 (T2R14) is highly expressed in both oral and extraoral tissues. Over 100 bitter ligands can activate T2R14, including hormones, vitamins, plant compounds, and peptides. Previous studies suggest that bitter tastants such as quinine and caffeine can inhibit G protein-coupled receptor kinases (GRKs) and delay T2R signal termination. Our earlier research showed that peptides from alcalase and chymotrypsin hydrolysates of beef proteins inhibited quinine-dependent calcium release t

The pharmacology of bitter taste receptors and their role in human airways.

The receptors involved in bitter taste perception (bitter taste receptors--T2Rs) constitute a family of G-protein-coupled receptors, of which around 29 subtypes have been identified in humans. T2R expression was initially thought to be confined to the oral cavity but has recently been described in a range of other tissues (such as the heart, gut, nasal cavity and lungs) and cell types (chemosensory, smooth muscle, endothelial, epithelial and inflammatory cells). Although it is still not clear whether endogenous T2R agonists exist, the T2R receptors recognize many natural and synthetic compound

Defects in the peripheral taste structure and function in the MRL/lpr mouse model of autoimmune disease.

While our understanding of the molecular and cellular aspects of taste reception and signaling continues to improve, the aberrations in these processes that lead to taste dysfunction remain largely unexplored. Abnormalities in taste can develop in a variety of diseases, including infections and autoimmune disorders. In this study, we used a mouse model of autoimmune disease to investigate the underlying mechanisms of taste disorders. MRL/MpJ-Fas(lpr)/J (MRL/lpr) mice develop a systemic autoimmunity with phenotypic similarities to human systemic lupus erythematosus and Sjögren's syndrome. Our r

Infection by the parasitic helminth Trichinella spiralis activates a Tas2r-mediated signaling pathway in intestinal tuft cells.

The parasitic helminth Trichinella spiralis, which poses a serious health risk to animals and humans, can be found worldwide. Recent findings indicate that a rare type of gut epithelial cell, tuft cells, can detect the helminth, triggering type 2 immune responses. However, the underlying molecular mechanisms remain to be fully understood. Here we show that both excretory-secretory products (E-S) and extract of T. spiralis can stimulate the release of the cytokine interleukin 25 (IL-25) from the mouse small intestinal villi and evoke calcium responses from tuft cells in the intestinal organoids

Improving the palatability of Platycodonic Radix for pediatric use: Integrating LC-MS-based bitter component profiling with an HP-β-Cyclodextrin/xylitol taste-masking strategy.

Platycodonic Radix (PR) is a key herb in traditional Chinese prescriptions for pediatric respiratory disorders. However, its intense bitterness and persistent throat astringency reduce treatment compliance. This study established a mechanism-informed taste-masking strategy integrating chemical profiling, sensory evaluation, and formulation optimization. PR sensory characteristics were first quantified using human panels and an electronic tongue. Subsequently, PR water extracts were characterized by non-targeted profiling based on liquid chromatography-mass spectrometry (LC-MS). Putative bitter

Effects of insulin signaling on mouse taste cell proliferation.

Expression of insulin and its receptor (IR) in rodent taste cells has been proposed, but exactly which types of taste cells express IR and the function of insulin signaling in taste organ have yet to be determined. In this study, we analyzed expression of IR mRNA and protein in mouse taste bud cells in vivo and explored its function ex vivo in organoids, using RT-PCR, immunohistochemistry, and quantitative PCR. In mouse taste tissue, IR was expressed broadly in taste buds, including in type II and III taste cells. With using 3-D taste bud organoids, we found insulin in the culture medium signi

The Pathogenesis of COVID-19-Related Taste Disorder and Treatments.

COVID-19, mainly manifested as acute respiratory distress syndrome, has afflicted millions of people worldwide since 2019. Taste dysfunction is a common early-stage symptom of COVID-19 infection that burdens patients for weeks or even permanently in some cases. Owing to its subjectivity and complexity, the mechanism of taste disorder is poorly studied. Previous studies have reported that the COVID-19 entry receptors are highly expressed in taste buds, thereby intensifying the cytocidal effect. Taste receptor cells are vulnerable to inflammation, and the COVID-19-induced cytokine storm causes s

Expression profile of the zinc transporter ZnT3 in taste cells of rat circumvallate papillae and its role in zinc release, a potential mechanism for taste stimulation.

Zinc is an essential trace element, and its deficiency causes taste dysfunction. Zinc accumulates in zinc transporter (ZnT)3-expressing presynaptic vesicles in hippocampal neurons and acts as a neurotransmitter in the central nervous system. However, the distribution of zinc and its role as a signal transmitter in taste buds remain unknown. Therefore, we examined the distribution of zinc and expression profiles of ZnT3 in taste cells and evaluated zinc release from isolated taste cells upon taste stimuli. Taste cells with a spindle or pyriform morphology were revealed by staining with the fluo

Structural similarity analysis and AI-predicted binding mode of bitter flavonoids in pomelo (Citrus maxima) using HRMS-based metabolomics and molecular fingerprinting.

The bitterness of pomelo is primarily attributed to flavonoids, which significantly impacts consumer acceptance. However, the molecular basis by which specific structural features of flavonoids interact with bitter taste receptors remains unclear. This study combined UHPLC-Orbitrap-MS, electronic tongue, and in silico prediction to analyze flavonoids in eight pomelo cultivars. A total of 35 flavonoids were identified, among which 25 were classified as potentially bitter. The key findings revealed that the neohesperidosyl group and the 4'/5'-hydroxyls groups on the B-ring are crucial for bitter

Converging Signaling Networks Drive Taste Bud Morphogenesis, Turnover, and Regeneration.

Buds are continuously renewed sensory organs in which development, adult maintenance, and repair share overlapping molecular circuitry. During embryogenesis, WNT/β-catenin signaling promotes taste placode formation and placodal Shh expression, while SHH refines papilla spacing and restricts neighboring papilla formation. SOX2 functions as a taste-competence and progenitor maintenance factor. In adults, LGR5/LGR6-RSPO-WNT signaling sustains progenitor activity, and gustatory neurons are an important source of RSPO2; available genetic evidence is consistent with a neuron-derived contribution to

Activation of TAS2R4 signaling attenuates podocyte injury induced by high glucose.

Bitter taste receptors (TAS2Rs) Tas2r108 gene possesses a high abundance in mouse kidney; however, the biological functions of Tas2r108 encoded receptor TAS2Rs member 4 (TAS2R4) are still unknown. In the present study, we found that mouse TAS2R4 (mTAS2R4) signaling was inactivated in chronic high glucose-stimulated mouse podocyte cell line MPC, evidenced by the decreased protein expressions of mTAS2R4 and phospholipase C β2 (PLCβ2), a key downstream molecule of mTAS2R4 signaling. Nonetheless, agonism of mTAS2R4 by quinine recovered mTAS2R4 and PLCβ2 levels, and increased podocyte cell viabilit
