A placental phenotype for intrahepatic cholestasis of pregnancy.

Intrahepatic cholestasis of pregnancy (ICP) is a pregnancy specific liver disease associated with significant risk of fetal complications. It is hypothesised that the risk of adverse fetal outcomes relates to the toxic effects of bile acids, the levels of which are increased in both maternal and fetal serum. Human and rodent studies have shown that transplacental transfer of bile acids is impaired in ICP. Furthermore, the morphology of placentas from the rodent model of ICP is markedly abnormal, and is associated with increased expression of apoptotic markers and oxidative stress. Using placen

Berberine Protects against High-Energy and Low-Protein Diet-Induced Hepatic Steatosis: Modulation of Gut Microbiota and Bile Acid Metabolism in Laying Hens.

Berberine (BBR) is a natural alkaloid with multiple biotical effects that has potential as a treatment for fatty liver hemorrhagic syndrome (FLHS). However, the mechanism underlying the protective effect of BBR against FLHS remains unclear. The present study aimed to investigate the effect of BBR on FLHS induced by a high-energy, low-protein (HELP) diet and explore the involvement of the gut microbiota and bile acid metabolism in the protective effects. A total of 90 healthy 140-day-old Hy-line laying hens were randomly divided into three groups, including a control group (fed a basic diet), a

Parabacteroides distasonis ameliorates hepatic fibrosis potentially via modulating intestinal bile acid metabolism and hepatocyte pyroptosis in male mice.

Parabacteroides distasonis (P. distasonis) plays an important role in human health, including diabetes, colorectal cancer and inflammatory bowel disease. Here, we show that P. distasonis is decreased in patients with hepatic fibrosis, and that administration of P. distasonis to male mice improves thioacetamide (TAA)- and methionine and choline-deficient (MCD) diet-induced hepatic fibrosis. Administration of P. distasonis also leads to increased bile salt hydrolase (BSH) activity, inhibition of intestinal farnesoid X receptor (FXR) signaling and decreased taurochenodeoxycholic acid (TCDCA) leve

Co-immobilised 7α- and 7β-HSDH as recyclable biocatalyst: high-performance production of TUDCA from waste chicken bile.

Chicken gallbladder has long been considered to be worthless and discarded as waste. The main composition of chicken bile is taurochenodeoxycholic acid (TCDCA), which is the isomeride of tauroursodeoxycholic acid (TUDCA). TUDCA has been effectively used for treatment of many diseases. In this paper, 7α- and 7β-hydroxysteroid dehydrogenases (HSDH) were co-immobilised on modified chitosan microspheres, and used as recyclable biocatalyst for the catalysis of chicken bile. The catalytic reaction reached equilibrium within 4 h compared with 1 h using TCDCA as substrate. After four continuous batch

Exposure to Grilled Lamb-Borne Carbon Quantum Dots Induces Intrahepatic Cholestasis by Activating the Intestinal Microbial-Derived Lipopolysaccharide-TLR4 Pathway.

The by-products of thermally processed foods, carbon quantum dots (CQDs), pose unpredictable risks to human health due to their nanoscale size and abundant surface functional groups that can readily accumulate in organs. Herein, mice were oral-exposed to grilled lamb-derived CQDs (25 mg kg-1) for 9 weeks. The results indicated that CQDs exposure resulted in liver and intestinal barrier injury, as well as an increase in intestinal microbiota-derived lipopolysaccharide (LPS). CQDs exposure directly and indirectly upregulated the expression of bile acid (BA) synthesis genes (Cyp7a1, Cyp8b1, and C

TUDCA receptors and their role on pancreatic beta cells.

Bile acids have received increasing attention over the past years as their multiple alternative roles became clearer. Tauroursodeoxycholic Acid (TUDCA) in specific has generated special interest due to its ability to promote pancreatic survival and function, as well as reduce endoplasmic reticulum stress. However, there are few studies explaining the molecular mechanisms behind TUDCA's beneficial actions on pancreatic beta cells. In this review, we decided to review the literature in order to craft a primer for researchers on what is known about TUDCA's receptors and the molecular pathways inv

Proteomic response of Turicibacter bilis MMM721 to chicken bile and its bile acids.

OBJECTIVE: Bile and its individual components, mainly bile acids, are important for digestion and drive bacterial community dynamics in the upper gastrointestinal tract of chickens. However, specific responses to bile acids have been characterized in only a few commensal bacteria, and it is unclear how other members of the microbiota respond to biliary stress. Here, we used label-free LC-MS/MS to assess the proteomic response of a common inhabitant of the chicken small intestine, Turicibacter bilis MMM721, to 24 h of growth in anaerobic growth media supplemented with 0.1% whole chicken bile, 0

Acute effects of melatonin on spontaneous and picrotoxin-evoked sleep-wake behaviour in the rat.

BACKGROUND: Infections classified as group 1 biological carcinogens include the helminthiases caused by Schistosoma haematobium and Opisthorchis viverrini. The molecular mediators underlying the infection with these parasites and cancer remain unclear. Although carcinogenesis is a multistep process, we have postulated that these parasites release metabolites including oxysterols and estrogen-like metabolites that interact with host cell DNA. How and why the parasite produce/excrete these metabolites remain unclear. A gene encoding a CYP enzyme was identified in schistosomes and opisthorchiids.

Lithocholic acid feeding results in direct hepato-toxicity independent of neutrophil function in mice.

Lithocholic acid (LCA) supplementation in the diet results in intrahepatic cholestasis and bile infarcts. Previously we showed that an innate immune response is critical for cholestatic liver injury in the bile duct ligated mice. Thus, the purpose of this study was to investigate the role of neutrophils in the mechanism of liver injury caused by feeding mice a diet containing LCA. C57BL/6 mice were given control or 1% LCA containing diet for 24-96 h and then examined for parameters of hepatotoxicity. Plasma ALT levels were significantly increased by 48 h after LCA feeding, which correlated wit
