Cd34+ Stromal Cells/Telocytes in Normal and Pathological Skin.

We studied CD34+ stromal cells/telocytes (CD34+SCs/TCs) in pathologic skin, after briefly examining them in normal conditions. We confirm previous studies by other authors in the normal dermis regarding CD34+SC/TC characteristics and distribution around vessels, nerves and cutaneous annexes, highlighting their practical absence in the papillary dermis and presence in the bulge region of perifollicular groups of very small CD34+ stromal cells. In non-tumoral skin pathology, we studied examples of the principal histologic patterns in which CD34+SCs/TCs have (1) a fundamental pathophysiological r

Telocytes: ultrastructural, immunohistochemical and electrophysiological characteristics in human myometrium.

Telocytes (TCs) have been described in various organs and species (www.telocytes.com) as cells with telopodes (Tps) - very long cellular extensions with an alternation of thin segments (podomers) and dilated portions (podoms). We examined TCs using electron microscopy (EM), immunohistochemistry (IHC), immunofluorescence (IF), time-lapse videomicroscopy and whole-cell patch voltage clamp. EM showed a three-dimensional network of dichotomous-branching Tps, a labyrinthine system with homocellular and heterocellular junctions. Tps release extracellular vesicles (mean diameter of 160.6±6.9 nm in no

Modulation of Telocytes in Women with Preeclampsia: A Prospective Comparative Study.

BACKGROUND: Telocytes (TCs) are networking cells with enigmatic functions. Placenta is a noninnervated organ with the TCs could have function of signal transmission to placental myofibroblasts, being likely a regulator for maternal blood flow. Preeclampsia (PE) is a disease complicating the second half of pregnancy associated with hypoxia probably due to failure of vascular remodeling of spiral arteries resulting in poor placental perfusion. We hypothesized that disturbance in the morphology of TCs may have a role in the pathogenesis of PE. MATERIALS AND METHODS: Women with normal or physiolo

The Autonomic Innervation and Uterine Telocyte Interplay in Leiomyoma Formation.

The autonomic innervation of the uterus is involved in multiple pathophysiological processes in both humans and animals. Pathological conditions such as adenomyosis or inflammatory pelvic disease are usually accompanied by significant alterations in uterine innervation. In the current study, we focused on autonomic innervation of uterine fibroids, the identification of recently described interstitial cells, telocytes, and the possible interplay between these structures. In this work, uterine telocytes were identified by immunopositivity for c-kit, CD34, and PDGFRα. Nerves were revealed by immu

A comprehensive guide to telocytes and their great potential in cardiovascular system.

Telocytes, a recently discovered type of interstitial cells, have a very distinctive morphology - the small cell body with long extensions, named telopodes. In our review, apart from introducing general aspects of telocytes, we focus on properties, functions and future potential of those cells in cardiovascular system. However, physiological functions of telocytes in cardiovascular system are still regarded as quite enigmatic. Previous studies claim that they play a role in organogenesis and regeneration, bioelectrical signalling, mechanoelectrical coupling, anti-oxidative protection, angiogen

The Third Dimension of Telocytes Revealed by FIB-SEM Tomography.

Lately, spatial three-dimensional (3D) identity of cells and their interrelations with the environment that surrounds it represent a challenging trend with the purpose to achieve a holistic view over the functions. Combining data from different imaging of cells in the third dimension can offer insight into behavior modalities making a world of difference. This chapter outlines a breakthrough in telocyte research by volume electron microscopy with the aid of focused ion beam scanning electron microscopy (FIB-SEM). Reconstructing 3D (three-dimensional) appearance of telocytes from a set of two-d

Telopodes of telocytes are influenced in vitro by redox conditions and ageing.

Telocytes (TCs) are a novel cell type identified among interstitial cells in various organs. TCs are characterized by very long cell processes (tens to hundreds micrometres) named telopodes (Tps) with uneven calibre: dilations (podoms) and very thin segments (podomers). However, little is known about the factors which influence Tps conformation. Recently, extracellular matrix proteins were found to influence Tps extension, adherence and spreading. Here, we show that oxidative stress and ageing influence formation of new Tps of TCs cultivated from human non-pregnant myometrium. Using real-time

The primary cilium of telocytes in the vasculature: electron microscope imaging.

Blood vessels are highly organized and complex structure, which are far more than simple tubes conducting the blood to almost any tissue of the body. The fine structure of the wall of blood vessels has been studied previously using the electron microscope, but the presence the telocytes associated with vasculature, a specific new cellular entity, has not been studied in depth. Interestingly, telocytes have been recently found in the epicardium, myocardium, endocardium, human term placenta, duodenal lamina propria and pleura. We show the presence of telocytes located on the extracellular matrix

Distribution of telocytes in the choroid (eye) of a teleost: An ultrastructural observation.

This paper reports the presence of telocytes in the choroid, a pigmented vascular tissue layer of the eye, as examined by scanning and transmission electron microscopy. These cells are reported to occur in the stroma of heart, reproductive tissues, digestive tract, skin, lungs and excretory organs. In the choroid of the teleost Clarius gariepinus, telocytes are widespread over the entire area of the choroidal stroma. Their processes, called telopodes, are rather thin (<0.2 µm in diameter) and exceptionally longer (>50 µm), which branch into 3-5 short processes. These processes are intimately a

Morphological and histochemical identification of telocytes in adult yak epididymis.

Telocytes (TCs) are a newly discovered type of mesenchymal cell that are closely related to the tissue's internal environment. The study aimed to investigate the morphological identification of TCs in the epididymis of adult yak and their role in the local microenvironment. In this study, transmission electron microscopy (TEM), scanning electron microscopy, immunofluorescence, qRT-PCR, and western blotting were used to analyze the cell morphology of TCs. The results showed that there are two types of TCs in the epididymal stroma of yak by TEM; one type is distributed around the capillaries wit

Relation between ureteral telocytes and the hydronephrosis development in children.

INTRODUCTION: Hydronephrosis is an actual pediatric problem, affecting children in the ante- and neonatal periods. Intrinsic stenosis is due to external obstruction and creates a pathophysiological basis of this urological pathology. Co-localization of ureter with a renal vasculature also could not be omitted from this point of view. Mesenchymal cells, partially telocytes, are important for local fibrosis development and hydronephrosis formation as well. In the current study, we focused on identification of telocytes in the human ureters to hypothesize their role in hydronephrosis pathophysiol

Telocytes in the reproductive organs: Current understanding and future challenges.

Over the past decades, we were witnessing spectacular molecular medicine advances. However, many of the reproductive medicine problems, such as fertility issues and premature birth still represent major challenges for obstetrics and gynecology worldwide. A new cell population - the telocytes (TCs) - were described in the interstitial space of many organs, and their possible implications in many important physiological and pathological processes should not be overlooked. In this article, we present a historical perspective outlining the landmarks in the discovery, evolution and achievements in

Telocytes/CD34+ Stromal Cells in Pathologically Affected White Adipose Tissue.

We studied telocytes/CD34+ stromal cells (TCs/CD34+SCs) in pathologically affected white adipose tissue after briefly examining them in normal fat. To this aim, we reviewed pathological processes, including original contributions, in which TCs/CD34+SCs are conserved, increased, and lost, or acquire a specific arrangement. The pathologic processes in which TCs/CD34+SCs are studied in adipose tissue include inflammation and repair through granulation tissue, iatrogenic insulin-amyloid type amyloidosis, non-adipose tissue components (nerve fascicles and fibres in neuromas and hyperplastic neuroge

An Enriched Primary Culture Reveals Morphological Heterogeneity of Rat Testicular Telocytes.

Telocytes are stromal cells characterized by small cell bodies and long cytoplasmic extensions termed telopodes. Although identified in the rat testicular interstitium, no standardized protocol for their isolation and in vitro enrichment has been established. This study aimed to develop an enriched primary culture model for rat testicular telocytes and characterize their morphology in vitro. Testicular cells from adult male Wistar rats were isolated by enzymatic digestion, subjected to short-term culture with differential adhesion, and enriched by fluorescence-activated cell sorting based on a

Morphological evidence of telocytes in DHEA-induced polycystic ovary syndrome model.

Polycystic Ovary Syndrome (PCOS) is a condition causing histopathological alterations in the ovarian stroma. Telocytes (TCs) are specialized interstitial/stromal cells present in the connective tissue of various organs. In this study, we investigated the presence and spatial organization of TCs in the ovaries of a rat model of PCOS induced by dehydroepiandrosterone (DHEA). The ovarian tissues from both PCOS and control groups were stained using hematoxylin-eosin (H&E), Bielschowsky's silver stain, methylene blue, and toluidine blue for light microscopy analysis, and scanned digitally. Ovaries

Telocytes Mediating the Effects of GABA-Herbal Medicine Composite on Insomnia.

Insomnia is one of the main manifestations of subhealth, a condition that has become increasingly prevalent and seriously affects the physical and mental well-being of many individuals. This study aims to investigate the effects of γ-aminobutyric acid (GABA)-herbal medicine composite (GABA-HMC) on sleep in both healthy and insomnia-plagued rats. According to traditional Chinese medicine theory, the Du Meridian, situated along the dorsal midline fascia of the body, plays a crucial role in regulating neuropsychiatric disorders, such as insomnia. Recent reports morphologically confirmed that telo

Cardiac telocytes and fibroblasts in primary culture: different morphologies and immunophenotypes.

Telocytes (TCs) are a peculiar type of interstitial cells with very long prolongations termed telopodes. TCs have previously been identified in different anatomic structures of the heart, and have also been isolated and cultured from heart tissues in vitro. TCs and fibroblasts, both located in the interstitial spaces of the heart, have different morphologies and functionality. However, other than microscopic observation, a reliable means to make differential diagnosis of cardiac TCs from fibroblasts remains unclear. In the present study, we isolated and cultured cardiac TCs and fibroblasts fro

Transcriptomic and Histological Characterization of Telocytes in the Human Dorsal Root Ganglion.

Telocytes are interstitial cells characterized by long processes that span considerable distances within tissues, likely facilitating coordination and interaction with various cell types. Although present in central and peripheral neuronal tissues, their role remains elusive. Dorsal root ganglia (DRG) house pseudounipolar afferent neurons responsible for transmitting signals related to temperature, proprioception, and nociception. This study aimed to investigate the presence and function of telocytes in human DRG by examining their transcriptional profile, anatomical location, and ultrastructu

Are telocytes related to maintenance of vascular homeostasis in normal and pathological aorta?

The telocyte (TC) is a new interstitial cell type described in a wide variety of organs and loose connective tissues around small vessels, but its presence in large arteries remains unexplored. TCs have small cell bodies and remarkably thin, long, moniliform processes called telopods (Tps). Using transmission electron microscopy and immunofluorescence, we identified TCs in normal human thoracic aortas and in those with aneurysm or acute dissection (TAAD). In normal aortas the TCs were distributed throughout the connective tissue of the adventitial layer, in its innermost portion and at the zon
