Insecticide susceptibility status of Culex quinquefasciatus Say, the vector of bancroftian filariasis against temephos in Delhi and National Capital Region.

Lymphatic filariasis is the second leading cause of long-term disability in the world. In India, filariasis is endemic in 250 districts in 20 states and union territories with about 553 million people at risk of infection. Temephos is being used as a larvicide to control the population of Culex quinquefasciatus mosquito, the vector of bancroftian filariasis. The susceptibility status of Cx. quinquefasciatus to temephos in various urban zones of Delhi and the national capital region (NCR) was evaluated using the WHO standard susceptibility test kit. Results of the test revealed that the larval

Temephos resistance in two forms of Aedes aegypti and its significance for the resistance mechanism.

Aedes aegypti, at the larval stage, has been subjected to the temephos selection in laboratory. The level of temephos resistance was detected in a microplate by biochemical assay using WHO bioassay technique. The major enzyme-based resistance mechanisms involved in temephos resistance include elevated nonspecific esterase, oxidase and insensitive acetylcholinesterase. After 19 generations of temephos selection, the selected group showed resistance ratios of 4.64 and 16.92, when compared with a non-selected group and the WHO susceptible strain, respectively. The two seperated forms, type form a

[Effect of temephos on acetylcholinesterase activity in the brain of Tilapia guineensis. 3: Comparative effect of temephos and 3 substitute insecticides].

The Onchocerciasis Control Programme (OCP) is realized in the major part of the treated area by weekly applications of temephos in biotopes of simulles larvae. This insecticide is very effective and its impact on the aquatic fauna is evaluated by means of periodic samplings of the fauna. Therefore, the most sensitive way of tracking down fish poisoning is by studying the brain acetylcholinesterase depression. Evaluated on Tilapia guineensis this lowering is moderate when operational doses are used by OCP. The discovery of resistence to temephos incited researchers of OCP to try remplacement in

Melatonin Abates TMJOA Chronic Pain by MT2R in Trigeminal Ganglion Neurons.

Temporomandibular joint osteoarthritis (TMJOA) is one of the most common diseases causing chronic pain in the oral and maxillofacial region. So far, there are few ways to relieve the pain of TMJOA. Melatonin (MT) has a good analgesic effect in many diseases, including fibromyalgia, neuropathic pain, chronic headache, and burn pain, with very low acute toxicity and side effects. This study was to investigate the role and mechanism of MT in TMJOA chronic pain. In rats TMJOA chronic pain occurred at day 14 after an intra-temporomandibular joint injection of monosodium iodoacetate, which we previo

Successful control of onchocerciasis vectors in San Vicente Pacaya, Guatemala, 1984-1989.

Between 1984 and 1989, the onchocerciasis-vector control zone on the pilot area of San Vicente Pacaya, Guatemala, which had been subject to experimental control since 1979, was extended from 91.3 to 148.6 km2. Temephos was used as a larvicide against Simulium ochraceum s.l. the target species. As a new strategy, only breeding sites with water discharges of 0.1-10 litres/s were treated, every 2 weeks. This approach provided a substantial reduction in effort, number of treated sites, time and cost. To assess the effect of the temephos, nine sites were selected in which standardized collections o

Simulium larvae susceptibility to temephos and the effect of 10 weeks of treatment of the Mbende tributary in the Nkam-Wouri River drainage of Cameroon on larval density and adult fly biting rates.

BACKGROUND: Despite over 18 years of annual ivermectin mass drug administration (MDA) in Cameroon's Nkam-Wouri River drainage, onchocerciasis transmission persists. Several reasons, including multiple breeding sites and abundant vector populations, contribute to ongoing transmission. High vector abundance also causes a biting nuisance to local populations. The change in paradigm from onchocerciasis control to elimination may not be achieved if alternative control measures are not used. There is a need to complement ivermectin MDA with other strategies. This study tested the susceptibility of S

Effect of lambda cyhalothrin and temephos on detoxification enzyme systems in Culex quinquefasciatus (Diptera: Culicidae).

Mosquitoes serve as vector for transmitting diseases. Among mosquitoes, Culex quinquefasciatus transmits lymphatic filariasis, yellow fever Japanese encephalitis etc. Application of chemical insecticides is still the best option for vector control programmes. Continuous use of these chemicals on mosquito reduces its effects. The present study determined the baseline susceptibility of Cx. quinquefasciatus in response to λ-cyhalothrin and temephos treatments. In addition, the biochemical mechanisms and zymogram analysis involved in insecticide detoxification among larval mosquitoes were studied.

Resistance to organophosphate and pyrethroid and their underlying mechanism in Aedes albopictus (Diptera: Culicidae) from dengue endemic sub-Himalayan West Bengal, India.

Dengue cases are increasing every year in the sub-Himalayan part of West Bengal, where Aedes albopictus (Skuse 1894) has emerged as a predominant dengue vector. Dengue management heavily relies on the recurrent application of chemical insecticides such as temephos (larvicide) and deltamethrin (adulticide). However Ae. albopictus can quickly develop resistance which complicates their management. Thus, detailed population-specific studies on resistance status and mechanism of resistance are useful for designing an effective dengue management strategy. In this study, wild mosquitoes were collecte

Temephos Decreases Sperm Quality and Fertilization Rate and Is Metabolized in Rat Reproductive Tissues at Low-Dose Exposure.

Temephos is an organophosphorus pesticide used in control campaigns against vectors that transmit diseases, including dengue, a public health concern. The WHO classifies temephos in category III and its safe concentration (low-observable-adverse-effect level) in male rats is 100 mg/kg/day for up to 44 days. Temephos inhibits acetylcholinesterase (AChE) and is metabolized in different tissues, probably by mixed-function oxidases; one of its metabolites is bisphenol S (BPS), which is considered an endocrine disruptor. The aim of this study was to evaluate the effects of temephos on sperm functio

How heterogeneous is the involvement of ABC transporters against insecticides?

Understanding the molecular mechanisms underlying cellular defense against xenobiotic compounds is a main research issue in medical and veterinary entomology, as insecticide/acaricide resistance is a major threat in the control of arthropods. ABC transporters are recognized as a component of the detoxifying mechanism in arthropods. We investigated the possible involvement of ABC transporters in defense to the organophosphate insecticide temephos in the malarial vector Anopheles stephensi. We performed bioassays on larvae of An. stephensi, using insecticide alone and in combination with ABC-tra

Spatial and temporal country-wide survey of temephos resistance in Brazilian populations of Aedes aegypti.

The organophosphate temephos has been the main insecticide used against larvae of the dengue and yellow fever mosquito (Aedes aegypti) in Brazil since the mid-1980s. Reports of resistance date back to 1995; however, no systematic reports of widespread temephos resistance have occurred to date. As resistance investigation is paramount for strategic decision-making by health officials, our objective here was to investigate the spatial and temporal spread of temephos resistance in Ae. aegypti in Brazil for the last 12 years using discriminating temephos concentrations and the bioassay protocols o

Synthesis and crystal structure of new temephos analogues as cholinesterase inhibitor: molecular docking, QSAR study, and hydrogen bonding analysis of solid state.

A series of temephos (Tem) derivatives were synthesized and characterized by 31P, 13C, and 1H NMR and FT-IR spectral techniques. Also, the crystal structure of compound 9 was investigated. The hydrogen bonding energies (E2) were calculated by NBO analysis of the crystal cluster. The activities and the mixed-type mechanism of Tem derivatives were evaluated using the modified Ellman's and Lineweaver-Burk's methods on cholinesterase (ChE) enzymes. The inhibitory activities of Tem derivatives with a P═S moiety were higher than those with a P═O moiety. Docking analysis disclosed that the hydrogen b

Insecticide susceptibility and knockdown resistance (Kdr) mutations (F1534C and V1016I) in Aedes aegypti populations from Dar es Salaam, Tanzania.

BACKGROUND: Dengue, Zika and chikungunya are Aedes-borne arboviruses that pose a major global public health threat. In the absence of effective treatments or universally available vaccines, disease control in sub-Saharan Africa relies primarily on insecticidal vector control. However, sustained insecticide application imposes selection pressure for insecticide resistance that compromises intervention effectiveness. This study assessed the insecticide resistance profile and knockdown resistance of Aedes aegypti populations in Dar es Salaam, Tanzania. METHODS: Phenotypic resistance of Ae. aegyp
