Genomic Structure and Molecular Characterization of Toll-like Receptors in Black Scraper Thamnaconus Modestus and Their Expression Response to Two Types of Pathogens.

The black scraper (Thamnaconus modestus) is an important commercial species in China. However, with the rapid expansion of aquaculture, the culture of this species faces substantial economic losses due to infectious diseases. Toll-like receptors (TLRs) recognize a wide range of pathogen-associated molecular patterns (PAMPs) and play a crucial role in disease resistance by initiating innate immune responses in the host. The genome of the black scraper comprises eight TLR members, which can be classified into five subfamilies based on evolutionary analysis. Moreover, the TmTLRs were identified o

New collections of blood flukes (Aporocotylidae) from fishes of the tropical Indo-west Pacific, including a new genus, two new species and molecular evidence that Elaphrobates chaetodontis (Yamaguti, 1970) is widespread in the region.

We report new collections of the Aporocotylidae from Australia, French Polynesia, and Japan. A new species of Cardicola Short, 1953 is described from Scomberomorus commerson (Lacépède) (Scombridae), off Lizard Island. Cardicola nolani n. sp. can be distinguished from its congeners based on the position of the oötype, the position of the male genital pore, and the absence of an oral sucker. A new species is described from Abalistes stellatus (Anonymous) (Balistidae), also from off Lizard Island. Phylogenetically the new species forms a strongly-supported clade with Cardicola yuelao Yong, Cutmor

Anterior migration of lateral plate mesodermal cells during embryogenesis of the pufferfish Takifugu niphobles: insight into the rostral positioning of pelvic fins.

In vertebrates, paired appendages (limbs and fins) are derived from the somatic mesoderm subsequent to the separation of the lateral plate mesoderm into somatic and splanchnic layers. This is less clear for teleosts, however, because the developmental processes of separation into two layers and of extension over the yolk have rarely been studied. During teleost evolution, the position of pelvic fins has generally shifted rostrally (Rosen; Nelson, 1982, 1994), although at the early embryonic stage the presumptive pelvic fin cells are initially located near the future anus region - the anterior

LPXRFamide peptide stimulates growth hormone and prolactin gene expression during the spawning period in the grass puffer, a semi-lunar synchronized spawner.

Gonadotropin-inhibitory hormone (GnIH) plays as a multifunctional neurohormone that controls reproduction in birds and mammals. LPXRFamide (LPXRFa) peptide, the fish ortholog of GnIH, has been shown to regulate the secretion of not only gonadotropin (GTH) but also growth hormone (GH) and prolactin (PRL), which are potentially important for gonadal function. To investigate the role of LPXRFa peptide on reproduction of the grass puffer, which spawns in semilunar cycles, we examined changes in the levels of gh and prl expression over the several months during the reproductive cycle, and the effec

Necessity of redefinition and revision of mouse unit for tetrodotoxin: Relationship between lethal time and mouse unit of tetrodotoxin in mouse bioassay.

BACKGROUND: An assay that utilizes the relationship between the amount of toxin administered and the lethal time of mice is called a mouse bioassay (MBA) and has been used as an assay for marine neurotoxins such as paralytic shellfish poison toxins and pufferfish toxins. The MBA for tetrodotoxin (TTX), which is possessed by pufferfish, has been used for many years in Japan. In the Japanese reference method, equivalent to the official method, the toxicity of TTX is expressed in mouse units (M.U.), and 1 M.U. is defined as the amount of the toxins required to kill a 20 g mouse in 30 min and is s

A new species of the fossil pufferfish genus †Archaeotetraodon (Tetraodontidae) from the Middle Miocene of the North Caucasus, Russia.

The fossil puffer fish genus †Archaeotetraodon Tyler & Bannikov, 1994 is unique among the Tetraodontidae in having the upright central spinule of the dermal scale plates bifid rather than singular and undivided. Six species of this genus have previously been described variously from the Oligocene and Miocene of Russia, Italy, Algeria, and Ukraine. Described herein is a seventh species of this genus, †Archaeotetraodon bemisae sp. nov., based upon two specimens from the Pshekha River, Krasnodar Region, North Caucasus, Russia. These Maikopian specimens are no less than 15 mya, in the basal-most M

Isometric growth in the world's largest bony fishes (genus Mola)? Morphological insights from fisheries bycatch data.

For teleost fishes, the relationship between morphometric traits can provide significant insight into species life history, however gathering such data for noncommercial species can prove challenging. Here, we use data collected opportunistically from fisheries bycatch and stranding events to assess growth scaling over orders of magnitude in the ocean sunfish (genus Mola). Intriguingly, the confidence intervals for the relationship between length and mass suggests that isometric scaling is likely, a growth pattern rarely observed in fishes owing to the scaling of supportive structures. These d

Consequences of experimental cortisol manipulations on the thermal biology of the checkered puffer (Sphoeroides testudineus) in laboratory and field environments.

Anthropogenic climate change is altering temperature regimes for coastal marine fishes. However, given that temperature changes will not occur in isolation of other stressors, it is necessary to explore the potential consequences of stress on the thermal tolerances and preferences of tropical marine fish in order to understand the thresholds for survival, and predict the associated coastal ecological consequences. In this study, we used exogenous cortisol injections to investigate the effects of a thermal challenge on checkered puffers (Sphoeroides testudineus) as a secondary stressor. There w

Spatial and quantitative gene expression analysis of SREB receptors in the gonads of green-spotted pufferfish (Dichotomyctere nigroviridis).

Super-conserved Receptors Expressed in Brain (SREB) are a highly conserved family of orphan G protein-coupled receptors that consist of three members in most vertebrates: SREB1 (GPR27), SREB2 (GPR85), and SREB3 (GPR173). Each receptor is associated with diverse physiological processes and expressed in both ovaries and testes, but reproductive functions are only beginning to be understood. In addition, some fishes gained a novel fourth gene, SREB3B, which may have unique functions. The purpose of this study was to conduct a spatial and quantitative analysis of SREBs in the gonads of pufferfish

Is neostigmine effective in severe pufferfish-associated tetrodotoxin poisoning?

CONTEXT: Pufferfish-associated tetrodotoxin poisoning occurs occasionally in coastal regions of Asian countries and sporadically elsewhere. While anecdotal reports have suggested neostigmine use, evidence for its clinical effectiveness remains unclear. OBJECTIVE: We sought to determine whether neostigmine was likely to be effective in tetrodotoxin-poisoned patients with acute respiratory failure caused by pufferfish consumption. METHODS: We searched in PubMed, EMBASE, CINAHL, Google Scholar, and other databases to collect cases of acute pufferfish-associated tetrodotoxin poisoning ever publi

Molecular phylogenetic analysis of the problematic genus Cardicola (Digenea: Aporocotylidae) indicates massive polyphyly, dramatic morphological radiation and host-switching.

Novel multi-locus sequence data were used to assess the molecular phylogenetic relationships of fish blood flukes showing similarity to the genus Cardicola Short, 1953 (Trematoda: Aporocotylidae). Analyses of three ribosomal (ITS2, 28S & 18S) subregions and one mitochondrial (cox1) DNA subregion shows that the hitherto-monophyletic clade formed by species of Cardicola Short, 1953 also includes species of three other genera - Braya Nolan & Cribb, 2006, Elaphrobates Bullard & Overstreet, 2003 and Rhaphidotrema Yong & Cribb, 2011 - as well as a new, morphologically distinct species discovered fro

Aptamer-antibody sandwich lateral flow test for rapid visual detection of tetrodotoxin in pufferfish.

Tetrodotoxin (TTX) is a highly potent marine toxin which can cause severe poisoning following consumption of contaminated fish and seafood. Thus, a sensitive, reliable and simple test is required for rapid screening of samples and prevention of intoxication. Herein, we translated a previously reported microtiter plate hybrid aptamer-antibody assay into a rapid lateral flow assay (LFA) test. The test relies on an aptamer immobilized on the membrane and an antibody conjugated with gold nanoparticles to provide a visual result when TTX is present in the sample. The optimized test is simple (one-s

Potential Threats Posed by Tetrodotoxins in UK Waters: Examination of Detection Methodology Used in Their Control.

Tetrodotoxin is a neurotoxin responsible for many human fatalities, most commonly following the consumption of pufferfish. Whilst the source of the toxin has not been conclusively proven, it is thought to be associated with various species of marine bacteria. Whilst the toxins are well studied in fish and gastropods, in recent years, there have been a number of reports of tetrodotoxin occurring in bivalve shellfish, including those harvested from the UK and other parts of Europe. This paper reviews evidence concerning the prevalence of tetrodotoxins in the UK together with methodologies curren

Adaptative responses of myenteric neurons of Sphoeroides testudineus to environmental pollution.

Contamination in estuarine regions affects the local biota damaging the ecosystems and reaching humans. The gastrointestinal tract is a dynamic environment capable of obtaining nutrients and energy from food while it protects the host against harmful toxins and pathogens from the external environment. These functions are modulated by the enteric nervous system and changes in its structure can result in gastrointestinal disorders. The objective of this study was to evaluate if the environmental contaminants have effects on the myenteric neuronal plasticity of pufferfish Sphoeroides testudineus.
