Noninactivating, tetrodotoxin-sensitive Na+ conductance in rat optic nerve axons.

The ionic current underlying the upstroke of axonal action potentials is carried by rapidly activating, voltage-dependent Na+ channels. Termination of the action potential is mediated in part by the rapid inactivation of these Na+ channels. We previously demonstrated that an influx of Na+ plays a critical role in the cascade leading to irreversible anoxic injury in central nervous system white matter. We speculated that a noninactivating Na+ conductance mediates this pathological Na+ influx and persists at depolarized membrane potentials as seen in anoxic axons. In the present study we measure

Partial characterization of a tetrodotoxin-binding component from nerve membrane.

Tetrodotoxin from Japanese puffer fish has been labeled with tritium and purified from the crude mixture obtained. The interaction between the purified [(3)H]tetrodotoxin and membrane suspensions from the olfactory nerve of long-nosed garfish has been investigated by equilibrium dialysis. Tetrodotoxin binds to membrane suspensions with a dissociation constant K(D) = 8.3 nM. The nerve preparation binds 42 pmol of [(3)H]tetrodotoxin/g of wet tissue at saturating toxin concentrations. With various hydrolytic enzymes, the binding component is shown to be a protein embedded in a phospholipid enviro

Cooperative Chemical Biosynthesis of Tetrodotoxin: Evidence from Marine Microbial Contributors.

Tetrodotoxin (TTX) is a potent neurotoxin widely distributed in pufferfish and other marine organisms, yet its microbial biosynthetic basis remains unresolved because no definitive bacterial TTX gene cluster has been identified. This study develops a hypothesis-generating comparative-genomic framework to prioritize bacterial genomes for experimental investigation of TTX-related metabolism; it does not demonstrate bacterial TTX biosynthesis. Thirteen genomes representing Pseudoalteromonas, Cytobacillus, Shewanella, and Vibrio were analyzed using whole-genome comparison, average amino acid ident

The skin microbiome facilitates adaptive tetrodotoxin production in poisonous newts.

This study of the outcome and prognostic factors in prenatally diagnosed agenesis of the corpus callosum (ACC) was undertaken to see if there are any differences between subgroups, what relationship they have to neurodevelopmental outcome and whether this information aids the counselling of parents of fetuses with the condition. The outcome of 14 prenatally diagnosed fetuses with ACC and 61 postnatally diagnosed patients was assessed in terms of clinical problems, developmental milestones and neurological signs; each patient was then given a score out of 10, 0 being a normal outcome and 10 bei

Tetrodotoxin blocks L-type Ca2+ channels in canine ventricular cardiomyocytes.

Tetrodotoxin (TTX) is believed to be the most selective inhibitor of voltage-gated fast Na(+) channels in excitable tissues, including nerve, skeletal muscle, and heart, although TTX sensitivity of the latter is lower than the former by at least three orders of magnitude. In the present study, the TTX sensitivity of L-type Ca(2+) current (I (Ca)) was studied in isolated canine ventricular cells using conventional voltage clamp and action potential voltage clamp techniques. TTX was found to block I (Ca) in a reversible manner without altering inactivation kinetics of I (Ca). Fitting results to

Prostaglandins and tetrodotoxin-insensitive relaxation of opossum lower esophageal sphincter.

Field stimulation with pulses of 0.5 or 5 ms relaxed isolated strips of lower esophageal sphincter (LES) of the opossum; only responses to 0.5-ms pulses were inhibited by tetrodotoxin. Black widow spider venom prevented relaxation to both stimuli; thus both stimuli may release nonadrenergic inhibitory mediator. Isoproterenol, but not PGEs or ATP, was a consistent relaxant of LES. PGF2alpha (approximately 1 microgram/ml) and stable endoperoxides (approximately 10 ng/ml) stimulated LES muscle. Electrocardiogram indicators for risk of respiratory failure in patients with tetrodotoxin poisoning.

Tetrodotoxin blocks voltage-gated sodium channels, which are responsible for the initiation and propagation of action potentials. Impairment of action potential conduction in myocardial cells affects the cardiac conduction system and can influence electrocardiogram output. In this retrospective study, we investigated whether tetrodotoxin poisoning prolong the PR, QRS and QTc intervals, which might be used as prognostic indices of respiratory failure after tetrodotoxin poisoning. Additionally, we evaluated whether these changes correlate with tetrodotoxin concentration in the body. Eighty-four

Detection of the Potential Inactivation of Tetrodotoxin by Lactic Acid Bacterial Exopolysaccharide.

Screening for compounds that can neutralize the toxicity of tetrodotoxin (TTX) or reduce its negative effects is necessary. Our study tested the TTX detoxification capacity of exopolysaccharide (EPS) extracted from lactic acid bacteria. EPS of Leuconostoc mesenteroides N3 isolated from the Vung Tau sea (Vietnam), Lactobacillus plantarum PN05, and Lactobacillus rhamnosus PN04 were used in the study. To more completely evaluate the importance of EPS in detoxification, EPS samples of Leuconostoc mesenteroides N3, Lactobacillus plantarum PN05 and Lactobacillus rhamnosus PN04 were also tested. The

Binding and Pharmacokinetics of the Sodium Channel Blocking Toxins (Saxitoxin and the Tetrodotoxins).

Tetrodotoxin (TTX) found in diverse variety of animals including puffer fishes, some newts, frogs and limited number of non-vertebrate species (6 different phyla). The saxitoxin (STX) and the TTX are small molecules composed of 7,8,9 guanidinium and 1,2,3 guanidinium groups, respectively in their structures. These groups provide positive charge to the molecules and are believed to interact with negatively charged Glu755 and Asp400 residues in domain II and I of the sodium channel strongly. The pharmacokinetic studies (absorption, distribution and accumulation) reported on Takifugu rubripes, Ta

Effect of purified phospholipases on the binding of tetrodotoxin to axon plasma membrane.

The role of phospholipids in the binding of [3H]tetrodotoxin to garfish olfactory nerve axon plasma membrane was studied by the use of purified phospholipases. Treatment of the membranes with low concentrations of either phospholipase A2 (Crotalus adamanteus and Naja naja) or phospholipase C (Bacillus cereus and Clostridium perfringens) resulted in a marked reduction in tetrodotoxin binding activity. A 90% reduction in the activity occurred with about 45% hydrolysis of membrane phospholipids by phospholipase A2, and with phospholipase C the lipid hydrolysis was about 60--70% for a 70--80% redu

Effects of tetrodotoxin and anaesthetics on brain metabolism and transport during anoxia.

1. Tetrodotoxin, at concentrations at which it abolishes generation of action potentials in the nervous system, enhances by about 300% the rate of anaerobic glycolysis of brain-cortex slices from adult rats, or from adult and infant guinea pigs. This occurs to a greater extent in Ca(2+)-deficient incubation media than in Ca(2+)-rich media. Tetrodotoxin has no accelerative effect on cerebral aerobic glycolysis. 2. Tetrodotoxin does not affect the rate of anaerobic glycolysis of 2-day-old rat brain-cortex slices, nor that of adult rat kidney medulla, nor that of an extract of an acetone-dried po

Multi-Omics Reveals Tetrodotoxin Transport and Accumulation Mechanisms in Takifugu bimaculatus.

The potent toxicity of tetrodotoxin (TTX) has long constrained sustainable growth in pufferfish aquaculture. Takifugu bimaculatus, an economically important species farmed along the coast of Fujian, China, remains poorly understood regarding how it transports and accumulates this potent neurotoxin. To address this gap, we combined transcriptomic and proteomic analyses to characterize the molecular responses of T. bimaculatus to TTX exposure. After oral administration, TTX primarily accumulated in the liver, ovaries, and skin. Multi-omics profiling revealed 163 differentially expressed genes (D

Reversible cerebral vasoconstriction syndrome associated with tetrodotoxin poisoning: A case report.

An 81-year-old woman with a history of hypertension and Alzheimer's disease presented to the emergency department because of impaired consciousness. Physical examination revealed acute progressive generalized flaccid paralysis, hypertension, respiratory failure, and pupillary dilation. Although the patient did not complain of headache, head magnetic resonance angiography and magnetic resonance imaging showed multifocal segmental cerebral vasospasm and cerebral infarction in the left occipital lobe. Her family reported that although she did not have a license to cook pufferfish, she was in the

Prefrontal cortex pyramidal neurons express functional Nav1.8 tetrodotoxin-resistant sodium currents.

It has been repeatedly proved that Nav1.8 tetrodotoxin (TTX)-resistant sodium currents are expressed in peripheral sensory neurons where they play important role in nociception. There are very few publications that show the presence of TTX-resistant sodium currents in central neurons. The aim of this study was to assess if functional Nav1.8 TTX-resistant sodium currents are expressed in prefrontal cortex pyramidal neurons. All recordings were performed in the presence of TTX in the extracellular solution to block TTX-sensitive sodium currents. The TTX-resistant sodium current recorded in this

Hemolymph-Mediated Dynamics of Paralytic Shellfish Toxins and Tetrodotoxin in Scallops.

To investigate the dynamics of paralytic shellfish toxins (PSTs) and tetrodotoxins (TTX) in scallops, PSTs and TTX were analyzed in the hemolymph supernatant (hemolymph-S) and digestive gland in Yesso and Akazara scallops cultured in eastern Japan. In Yesso scallops sampled between 22 April 2025 and 21 October 2025, the PST concentrations in the hemolymph-S were 1.6-17% of those in the digestive gland, showing a positive correlation (r = 0.753). From Poison to Promise: The Evolution of Tetrodotoxin and Its Potential as a Therapeutic.

Tetrodotoxin (TTX) is a potent neurotoxin that was first identified in pufferfish but has since been isolated from an array of taxa that host TTX-producing bacteria. However, determining its origin, ecosystem roles, and biomedical applications has challenged researchers for decades. Recognized as a poison and for its lethal effects on humans when ingested, TTX is primarily a powerful sodium channel inhibitor that targets voltage-gated sodium channels, including six of the nine mammalian isoforms. Although lethal doses for humans range from 1.5-2.0 mg TTX (blood level 9 ng/mL), when it is admin

Antihyperalgesic effect of tetrodotoxin in rat models of persistent muscle pain.

Persistent muscle pain is a common and disabling symptom for which available treatments have limited efficacy. Since tetrodotoxin (TTX) displays a marked antinociceptive effect in models of persistent cutaneous pain, we tested its local antinociceptive effect in rat models of muscle pain induced by inflammation, ergonomic injury and chemotherapy-induced neuropathy. While local injection of TTX (0.03-1 μg) into the gastrocnemius muscle did not affect the mechanical nociceptive threshold in naïve rats, exposure to the inflammogen carrageenan produced a marked muscle mechanical hyperalgesia, whic
