The thalamus is a subcortical structure formed by different nuclei that relay information to the neocortex. Several reports have already described alterations of this structure in patients of schizophrenia that experience auditory hallucinations. However, to date no study has addressed whether the volumes of specific thalamic nuclei are altered in chronic patients experiencing persistent auditory hallucinations. We have processed structural MRI images using Freesurfer, and have segmented them into 25 nuclei using the probabilistic atlas developed by Iglesias and collaborators (Iglesias et al.,

AIM: Although the thalamus is a key structure in the pathophysiology of obsessive-compulsive disorder (OCD), reports regarding thalamic volume alterations in OCD patients have been inconsistent. Because the thalamus has a complex structure with distinct functions, we investigated subregional volume changes in the thalamus and their relationship with clinical attributes in a large sample of medication-free OCD patients.
METHODS: We collected T1-weighted magnetic resonance imaging data from 177 OCD patients and 152 healthy controls (HCs). Using FreeSurfer, we segmented the thalamus into 12 nucl

OBJECTIVES: Down syndrome (DS) is a genetic condition characterized by cognitive disability starting from infancy. Children with DS exhibit deficits in several cognitive domains, including executive function, i.e., a set of cognitive processes that heavily depend on higher-order thalamic nuclei. The goal of this study was to establish whether executive function-related thalamic nuclei of fetuses with DS exhibit neuroanatomical alterations that may contribute to the defects in higher-order control processes seen in children with DS.
PATIENTS AND METHODS: In brain sections from fetuses with DS

OBJECTIVE: Thalamus models of psychosis implicate association nuclei in the pathogenesis of psychosis and mechanisms of cognitive impairment. Studies to date have provided conflicting findings for structural deficits specific to these nuclei. The authors sought to characterize thalamic structural abnormalities in psychosis and a neurodevelopmental cohort, and to determine whether nuclear volumes were associated with cognitive function.
METHODS: Thalamic nuclei volumes were tested in a cross-sectional sample of 472 adults (293 with psychosis) and the Philadelphia Neurodevelopmental Cohort (PNC

The rostral intralaminar thalamic nuclei (ILn) are organized to activate pathways originating in medial prefrontal cortex (mPF) that mediate memory-guided responding during alert, wakeful states. Previous studies have shown that rostral ILn or mPF lesions produce deficits in delayed matching to position (DMTP). Here, we manipulated rostral ILn activity in rats by microinjecting drugs or applying electrical current and examined effects on DMTP. Inhibiting activity with the GABA(A) agonist muscimol impaired DMTP. Decreasing GABA(A) tone with FG-7142 (N-methyl-beta-carboline-3-carboxamide) improv

Deep brain stimulation (DBS) of the thalamic ventral intermediate nucleus (Vim) has been a standard therapy for essential tremor. It has been shown that high-frequency (≥100 Hz) DBS suppresses Vim firing and tremor activity; however, the underlying mechanisms are not fully understood. Here, we investigate whether neuronal suppression during high-frequency DBS occurs at cellular levels or is influenced by network-level effects. Using in vivo recordings of Vim neurons, from people of unknown sex, during different DBS frequencies, we detected a positive-going evoked-field potential, quasi-evoked

INTRODUCTION: Childhood growth hormone deficiency (GHD) is an endocrine disorder characterized by reduced secretion of growth hormone (GH), leading to impaired linear growth and delayed developmental milestones. Recent studies suggest that GHD is associated with cognitive, socio-emotional, and behavioral impairments, potentially via altered neurodevelopment, with neuroimaging studies revealing changes in brain morphology and broader GH-related effects on the central nervous system. The thalamus, a major subcortical relay integrating sensory, motor and cognitive information, has received limite

INTRODUCTION: Essential tremor (ET) is a neurological disorder primarily characterized by upper limb action tremor. It is widely recognized that the thalamus is implicated in ET pathophysiology, playing a central role in treatment approaches. This study aimed to explore thalamic morphology, assessing macrostructural changes and intrinsic thalamic networks in ET patients.
METHODS: A total of 109 ET (41 with and 68 without resting tremor) and 81 healthy controls (HC) were enrolled in the study. An automatic probabilistic segmentation of thalamic nuclei was employed on T1-weighted MRI images usi

Injury to the anterior thalamic nuclei (ATN) and their neural connections is the most consistent neuropathology associated with diencephalic amnesia. ATN lesions in rats produce memory impairments that support a key role for this region within an extended hippocampal system of complex overlapping neural connections. Environmental enrichment is a therapeutic tool that produces substantial, although incomplete, recovery of memory function after ATN lesions, even after the lesion-induced deficit has become established. Similarly, the neurotrophic agent cerebrolysin, also counters the negative eff

The projections from the midline and intralaminar thalamic nuclei to the cerebral cortex were studied in the rat by means of anterograde tracing with Phaseolus vulgaris-leucoagglutinin. The midline and intralaminar nuclear complex taken as a whole projects to widespread, predominantly frontal, cortical areas. Each of the constituent thalamic nuclei has a restricted cortical projection field that overlaps only slightly with the projection fields of adjacent midline and intralaminar nuclei. The projections of the intralaminar nuclei cover a larger cortical area than those of the midline nuclei.

BACKGROUND: The thalamus serves as a critical relay hub for transmitting peripheral nociceptive information to the cerebral cortex and plays a central role in the sensory, motor, affective, and cognitive dimensions of pain processing. However, the dynamic response patterns of distinct thalamic nuclei during stimulus-pain processing remain poorly understood.
METHODS: In this study, fiber photometry was employed to record calcium (Ca2+) signals from multiple thalamic nuclei under two intensities of nociceptive somatic stimulation.
RESULTS: The results showed that all recorded nuclei exhibited

Background: The thalamus, as a key relay of neuronal information flow between subcortical structures and cortical networks, has been implicated in focal limbic seizures propagation, awareness maintenance, and seizure-related cognitive deficits. However, the specific functional alterations between different thalamic nuclei and subcortical-cortical systems in temporal lobe epilepsy (TLE) remain largely unknown. Methods: We examined thalamic functional connectivity (FC) in 26 TLE patients and 30 healthy controls matched for sex, age, and education. The anterior (ANT), ventral posterior medial, an

Thalamic nuclei play critical roles in regulation of neurological functions like sleep and wakefulness. They are increasingly implicated in neurodegenerative and neurological diseases such as multiple sclerosis and essential tremor. However, segmentation of thalamic nuclei is difficult due to their poor visibility in conventional MRI scans. Sophisticated methods have been proposed which require specialized MRI acquisitions and complex post processing. There are few high spatial resolution (1 mm3 or higher) in vivo MRI thalamic atlases available currently. The goal of this work is the developme

Objective. Postinfarction transneuronal degeneration refers to secondary neuronal death that occurs within a few days to weeks following the disruption of input or output to synapsed neurons sustaining ischemic insults. The thalamus receives its blood supply from the posterior circulation; however, infarctions of the middle cerebral arterial may cause secondary transneuronal degeneration in the thalamus. In this study, we presented the areas of ischemia and associated transneuronal degeneration following MCAo in a rat model. Materials and Methods. Eighteen 12-week-old male Sprague-Dawley rats

Background/Objectives: Epilepsy is a long-term condition marked by recurring seizures that greatly affect daily life. The thalamus, with its extensive connections to the cortex, plays a vital role in epileptic processes. Changes in the volume of the thalamic nuclei are believed to occur in epilepsy. Our study aimed to examine the volumetric characteristics of thalamic nuclei in individuals diagnosed with generalized epilepsy and to explore the potential role of changes in these nuclei in the process of epileptogenesis. Methods: In this retrospective study, a total of 232 participants were incl

The head direction (HD) circuit is a complex interconnected network of brain regions ranging from the brain stem to the cortex. Recent work found that HD cells corecorded ipsilaterally in the anterodorsal nucleus (ADN) of the thalamus displayed coordinated firing patterns. A high-frequency oscillation pattern (130-160 Hz) was visible in the cross-correlograms of these HD cell pairs. Spectral analysis further found that the power of this oscillation was greatest at 0 ms and decreased at greater lags, and demonstrated that there was greater synchrony between HD cells with similar preferred firin

OBJECTIVE: To evaluate the respective roles of the anterior thalamic nucleus (ANT) and the medial pulvinar (PuM) during mesial temporal lobe seizures recorded by stereoelectroencephalography (SEEG).
METHODS: We assessed functional connectivity (FC) in 15 SEEG recorded seizures from 6 patients using a non-linear correlation method. Functional interactions were explored between the mesial temporal region, the temporal neocortex, ANT and PuM. The node total-strength (the summed connectivity of the node with all other nodes) as well as the directionality of the links (IN and OUT strengths) were c

The thalamus is primarily known as a relay for sensory information; however, it also critically contributes to higher-order cortical processing and coordination. Thalamocortical hyperconnectivity is associated with hallucinatory phenomena that occur in various psychopathologies (e.g., psychosis, migraine aura) and altered states of consciousness (ASC; e.g., induced by psychedelic drugs). However, the exact functional contribution of thalamocortical hyperconnectivity in forming hallucinatory experiences is unclear. Flicker light stimulation (FLS) can be used as an experimental tool to induce tr

The thalamus is a central integration structure in the brain, receiving and distributing information among the cerebral cortex, subcortical structures, and the peripheral nervous system. Prior studies clearly show that the thalamus atrophies in cognitively unimpaired aging. However, the thalamus is comprised of multiple nuclei involved in a wide range of functions, and the age-related atrophy of individual thalamic nuclei remains unknown. Using a recently developed automated method of identifying thalamic nuclei (3T or 7T MRI with white-matter-nulled MPRAGE contrast and THOMAS segmentation) an
