Molecular detection of Theileria and Babesia infections in cattle.

This study was carried out to determine the presence and distribution of tick-borne haemoprotozoan parasites (Theileria and Babesia) in apparently healthy cattle in the East Black Sea Region of Turkey. A total of 389 blood samples were collected from the animals of various ages in six provinces in the region. Prevalence of infection was determined by reverse line blot (RLB) assay. The hypervariable V4 region of the 18S ribosomal RNA (rRNA) gene was amplified with a set of primers for members of the genera Theileria and Babesia. Amplified PCR products were hybridized onto a membrane to which ge

Recent development in control of Theileria annulata in Iran.

During 1973-1990 cattle immunization, in Iran, was induced by two strains within an interval of one month: first the milder and then the mild strain. Although this method of vaccination rendered satisfactory results in the field, yet production, maintenance in deep-freezers and transportation in liquid nitrogen particularly to remote areas of the country proved to be uneconomical and time consuming. Therefore, in order to reduce cost and save time, a new method involving only one local and live attenuated vaccine strain was sought. Reports received from different ecological areas of the countr

Theileria infectivity of Hyalomma ticks in Haryana, India.

Theileria infection of Hyalomma ticks collected from three districts of Haryana was assessed in whole salivary glands by the methyl-green pyronin staining method. Of 1,662 ticks screened, 546 (32.8%) were found Theileria positive. Infection rate in 935 female ticks (36.9%) was more than that of 727 male ticks (27.6%). Density of Theileria infection (number of infected acini per infected tick) was also higher in female ticks. Theileria infection rate of ticks varied greatly in the three districts viz. only 12.0% in Rohtak, 25.8% in Hisar and 48.3% in Karnal. Per cent infection rate was high (63

Epidemiology and transmission of Theileria sp of cattle in Australia.

Theileria sp occurs in cattle in Australia where Boophilus microplus is not present. Two species of ixodid ticks namely Haemaphysalis longicornis and H. bancrofti have been implicated as natural vectors. Experimental transmission was obtained with these two species but attempts to transmit the infection with B. microplus, Ixodes holocyclus and Amblyomma triguttatum were not successful. Although calves born in the enzootic area may become infected before one-month-old most show evidence of infection by 5 months of age. Calves born in the winter also become infected earlier than those born at ot

Theileria orientalis Ikeda Genotype in Cattle, Virginia, USA.

Theileria orientalis Ikeda genotype is a parasite that causes a disease in cattle that results in major economic issues in Asia, New Zealand, and Australia. The parasite is transmitted by Haemaphysalis longicornis ticks, which have recently been reported in numerous states throughout the eastern United States. Concurrently, cattle in Virginia showed clinical signs consistent with a hemoprotozoan infection. We used amplicons specific for the major piroplasm surface protein and small subunit rDNA of piroplasms to test blood samples from the cattle by PCR. Bidirectional Sanger sequencing showed s

Quantitative detection of Theileria haneyi in South African horses.

Theileria haneyi is an apicomplexan parasite closely related to Theileria equi, a known causative agent of equine piroplasmosis. The molecular distinction between these parasites relies on a nested polymerase chain reaction (PCR) assay, which has been reported to be unreliable. A recently reported indirect ELISA based on equi merozoite antigen 11 (Thema-11) of T. haneyi can detect geographically diverse T. haneyi strains. Since the ema-11 gene is exclusive to T. haneyi, it was chosen as the target for developing a TaqMan minor groove binder (MGB™) quantitative real-time PCR (qPCR). Published T

Epidrugs: alternative chemotherapy targeting Theileria annulata schizont stage parasites.

The growing emergence of resistance to current anti-theilerial agents necessitates the exploration of alternative approaches to drug discovery. This study evaluated the antiparasitic efficacy of 148 compounds derived from an epigenetic inhibitor library against the schizont stage of a Theileria annulata-infected cell line. Initial screening at a concentration of 10 µM identified 27 compounds exhibiting promising anti-theilerial activity. Further investigation, including determination of the 50% inhibitory concentration (IC50) and host cell cytotoxicity assay, highlighted seven highly effective

Theileria orientalis outbreak in an organized cattle breeding farm.

Theileriosis is an important tick borne disease of cattle caused by a haemoprotozoan of genus Theileria. Clinical bovine theileriosis is mainly caused by T. annulata or T. parva but the clinical disease due to T. orientalis is rare. T. orientalis mainly infect RBCs and causes "Oriental theileriosis" or Theileria-associated bovine anaemia in cattle and other livestock species. Two genotypes of T. orientalis (Chitose and Ikeda) are reported to cause severe disease in some countries. In this report, a spontaneous outbreak of Oriental theileriosis was studied in an organized Holstein-Friesian catt

The pulmonary involvement in Theileria lestoquardi naturally infected sheep.

Malignant Ovine Theileriosis (MOT) caused by Theileria lestoquardi is considered a major constraint for sheep production in many areas of the world including Sudan. Pulmonary oedema is thought to be the main cause of animal death, but the mechanism, the cell types involved and/or the probable cause of this pneumonia has yet to be defined. The present study was carried out to investigate the pulmonary involvement post T. lestoquardi infection and to identify the cell types involved in pneumonia. Apparently healthy sheep were exposed to ticks challenge in T. lestoquardi endemic area. Lungs impre

Characterization of Theileria parva which infects waterbuck (Kobus defassa).

Theileria-free waterbuck (Kobus defassa) born in captivity were successfully infected with Theileria parva sporozoites derived from ticks infected by feeding on African buffalo (Syncerus caffer). All waterbuck underwent mild infections with the development of sporadic schizont and piroplasm parasitosis when inoculated with sporozoite doses lethal to cattle. A carrier state of T. parva was demonstrated by feeding clean R. appendiculatus nymphs on two of these infected waterbuck. Tick batches from these waterbuck on 2 of 5 occasions transmitted lethal Theileria infections to cattle. In a separat

Microscopic and molecular detection of Anaplasma and Theileria species in goats from Egypt, with the first report of the emerging zoonotic Anaplasma capra and Theileria orientalis.

INTRODUCTION: Goats constitute a fundamental component of the livestock sector in Egypt, contributing to rural livelihoods, food security, and national income. However, their productivity is increasingly affected by tick-borne diseases. Despite their importance, data on tick-borne pathogens affecting goats in Egypt are still lacking. Anaplasmosis and theileriosis are among the most important tick-borne diseases, posing serious threats to animal health and economic stability. METHODS: This study investigated the microscopic and molecular detection of Anaplasma and Theileria species in goats fr

Stage-specific proteomic and phosphoproteomic remodeling in Theileria annulata.

Theileria annulata, an apicomplexan parasite, causes lymphoproliferative disease in cattle and threatens the bovine industry in tropical and subtropical regions. To clarify life-cycle mechanisms and parasite-host interactions, we conducted high-resolution Orbitrap proteomic and phosphoproteomic analyses of the schizont and piroplasm stages. In total, we confidently identified and quantified 2,093 proteins; among these, 459 proteins were observed in a phosphorylated state, comprising 5,069 distinct phosphorylation sites. Functional enrichment analysis of differentially abundant proteins indicat

Investigation of carbohydrate-based molecules of Theileria parva parasites.

Theileria parva (Tp), the causative agent of East Coast Fever, poses a major threat to Bos taurus cattle in sub-Saharan Africa. Progress towards a stable, affordable, cross-protective subunit vaccine covering more than one major histocompatibility complex (MHC)-presented Tp antigen has been hindered by the limited knowledge of antigens. Given the importance of glycoconjugates in protozoan pathogen biology, especially during the invasion process, we investigated the glycosylation potential of Tp. Screening of Tp Muguga (TpM) schizonts with C-type lectin (-like) receptors showed that ruminant, b

First Molecular Characterization of Theileria equi from Northeastern Algeria.

PURPOSE: Equine piroplasmosis (EP) is a tick-borne disease that affects equids and is caused by the parasites Theileria equi, Theileria haneyi and Babesia caballi. This disease leads to significant economic losses for the global equine industry. The current study aimed to evaluate the prevalence of Piroplasmida infections in equines from El Tarf, located in northeastern Algeria, and investigate the phylogenetic diversity within isolates obtained from infected equines in this study and between those from previous studies. METHODS: A total of 120 blood samples were collected from equines in thr

Molecular detection of Theileria cervi in equids from México.

Equine piroplasmosis is a parasitic illness caused by various protozoa of the Babesia and Theileria genera, which parasitize within red blood cells. The transmission of these pathogens occurs through certain genus of ticks, including Amblyomma, Haemaphysalis, Hyalomma, and Rhipicephalus. In recent times, an increase in the identification of new Theileria species and genotypes has been observed. This is further complicated by the presence of mixed Theileria infections in both mammals and tick vectors, particularly in regions where wildlife and livestock share habitats and vectors. Therefore, th

Babesia (Theileria) equi genotype A among Indian equine population.

Equine piroplasmosis, caused by Babesia (Theileria) equi, is well reported from many parts of India. However, literature regarding its prevalence from semi arid India is limited. Alongside, there is complete absence of information about genetic characterization of B.(T.) equi and the associated genotypes from India. In the present study, the prevalence of B.(T.) equi was studied from semi arid India using 18S ribosomal gene based PCR assay. An overall prevalence rate of 10.46% was recorded. PCR was more sensitive and specific in comparison with blood smears. The found isolates were sequenced.

Phylogenetic analysis of Theileria species transmitted by Haemaphysalis qinghaiensis.

The phylogenetic relationships between six isolates of Theileria spp. infective to small ruminants, and two isolates of Theileria spp. infective to yak, all transmitted by Haemaphysalis qinghaiensis, together with the Theileria orientalis/sergenti/buffeli group and T. sinensis, were analyzed using the 18S ssrRNA gene sequence. The target DNA segment was amplified by polymerase chain reaction (PCR). The PCR product was used either for direct sequencing or was ligated to the PCR II vector for sequencing. The length of the 18S ssrRNA gene of all Theileria spp. involved in this study was around 1,

Immunization against diseases caused by Theileria parva: a review.

Theileria parva is the causative agent of three epidemiologically different diseases, East Coast fever (ECF), Corridor disease and January disease, caused by 3 types of T. parva, T. p. parva, T. p. lawrencei and T. p. bovis, respectively. The history of immunization against these diseases has been marked by salient discoveries such as the immune status in recovered animals, the activity of tetracyclines during the incubation period, the possibility for cryopreserving supernatant of prefed ticks and the development of useful serological tests. The possibility of simultaneous administration of s

Theileria parva: expression of a sporozoite surface coat antigen.

A monoclonal antibody specific for the Theileria parva sporozoite, which recognizes a determinant on the surface coat and blocks sporozoite infectivity, was used to investigate the presence of the determinant on other stages of the parasite lifecycle. Immunofluorescence techniques did not demonstrate this determinant on the kinete, schizont, merozoite, or piroplasm stages of the parasite. Immunoautoradiography, using a tritiated form of the monoclonal antibody, on sections of infected salivary glands collected from ticks that had fed for 0, 1, 2, 3, or 4 days revealed that the determinant reco
